본 연구에서는 식물세포배양으로부터 항암제 paclitaxel 정제를 위한 전처리 공정으로 계면활성제 (surfactant)를 이용한 마이셀(micelle) 공정을 개발하였으며, 특히 마이셀 공정에서의 계면활성제 영향에 대해 조사하였다. 마이셀은 친수성 그룹과 소수성 그룹으로 구성되어 있어 마이셀 내부로는 물에 녹지 않는 paclitaxel을 소수성 그룹이 감싸게 되고 외부는 친수성 그룹이 되어 수용액에 용해될 수 있게 된다. 따라서 식물세포인 biomass로부터 유기용매 추출액을 건조하여 상 분리에 적합한 유기용매에 녹인 후 마이셀 형성에 적합한 계면활성제를 첨가하고 교반하여 정체시키면 물에 녹지 않는 paclitaxel이 마이셀 내부의 소수성 그룹에 감싸여 수용액 상로 이동하고 paclitaxel을 제외한 여러 가지 불순물들 (특히 lipid 성분들)은 유기용매 상에 그대로 존재하게 되어 paclitaxel과 불순물들을 효율적으로 분리하게 된다. 본 실험에서는 분리 및 정제에 많이 쓰이는 대표적인 양이온(CPC, CTMAC, LTMAC), 음이온 (SDS, AOT), 비이온 (Tween, PEG, Triton) 계면활성제의 영향에 대한 실험을 수행하였다. 양이온 계면활성제인 CTMAC (5%, w/v)을 이용한 마이셀 공정에서 가장 높은 수율 (${\sim}99%$)과 순도 (${\sim}21%$)를 얻었으며 또한 마이셀 형성 후 상 분리에 30 min 정도 소요되어 가장 단시간에 상 분리가 이루어짐을 알 수 있었다. 전처리 공정에서의 마이셀 형성 및 상 분리 방법은 불순물로부터 효과적으로 높은 순도의 paclitaxel을 고수율로 회수할 수 있어 최종 정제를 위한 전처리 공정으로 매우 유용하게 활용될 수 있을 것으로 판단된다.
잔대의 70% 에탄올 추출물은 강한 항산화 효과 및 산화적 스트레스 조건에서 뇌신경세포 사멸 억제 효과가 있었다. 6개의 분획물 중에 잔대 추출물의 클로로포름 분획물(AT-CH)은 높은 DPPH 라디칼 소거작용 및 세포 내 활성산소종(ROS) 소거작용을 가지고 있었고 $H_2O_2$에 대항하여 뇌신경 세포(SK-N-SH) 보호 효과가 가장 뛰어났다. 활성이 가장 우수한 클로로포름 분획물을 대상으로 주요 물질을 순수분리한 후 $^1H-NMR$, $^{13}C-NMR$ 그리고 EI mass spectra를 이용하여 화학구조를 분석한 결과 phytosterols(${\beta}$-sitosterol과 daucosterol)이라는 것을 알 수 있었다. ${\beta}$-Sitosterol과 daucosterol 역시 산화적 스트레스에 대항하여 SK-N-SH 세포를 보호하는 작용이 있었다. AT-CH 그리고 이것으로부터 분리된 화합물은 p38 경로를 저해하였다. 이들 결과는 AT-CH는 p38 경로를 저해하고 세포 내 ROS 소거작용에 의해 산화적 스트레스로부터 뇌신경세포 보호 효과를 가질 것이라 제안하였다.
전기방사는 높은 비표면적을 가지는 마이크로~나노 단위 직경의 나노섬유를 생산하는 간단하고 효율적인 공정이다. 따라서 식물 추출물과 폴리머를 혼합한 방사용액으로 손쉽게 의료용 나노섬유의 제조가 가능하다. 향나무는 라디칼 생성, 화상, 세포손상과 같은 자외선과 SLS에 의한 피부손상을 방지하는데 효과적이라고 알려져 있다. 또한 방미효과와 함께 집먼지 진드기 방지 효과가 보고된 바 있다. 전기방사로 향나무 추출물을 함유하여 제조한 PVA 나노섬유를 연구하였다. 향나무 추출물의 서로 다른 농도(0.25, 0.5, 1.5 wt. %)를 함유하는 PVA/향나무 추출물 나노 복합섬유를 제조하였으며 방사용액의 농도, 인가전압, TCD 등의 전기방사 조건을 최적화 하였다. 연구결과 균일한 PVA/향나무 추출물 나노 복합섬유을 얻을 수 있는 최적 조건으로 PVA 농도는 12wt%, 인가전압은 10 Kv, TCD는 10~20 cm로 나타나났다. 제조된 전기방사 나노 복합섬유의 형태 및 미세구조를 SEM을 통해 관찰하였다. 향나무 추출물의 첨가에 의해 나노섬유의 직경이 증가하는 것으로 나타났다. 결과적으로 310~360 nm의 직경범위를 가지는 PVA/향나무 추출물 복합 나노섬유가 전기방사를 통해 성공적으로 얻어졌다.
Wnt/${\beta}$-catenin 신호전달체계는 세포의 분화와 증식, 기관의 발생과 조절을 담당하는 중요한 세포내 신호전달체계이다. 발생과정에서 Wnt/${\beta}$-catenin 신호전달체계의 작용이 지방세포로의 분화를 억제하고 조골세포와 신경세포로의 분화는 촉진한다는 많은 연구들이 보고되어 있으며, 현재 Wnt/${\beta}$-catenin 신호전달체계의 조절을 통한 여러 질병의 치료와 예방에 대한 관심이 대두되고 있다. 본 연구에서는 세포를 기반으로 한 초고속 저분자 스크리닝 시스템을 이용하여 Wnt의 상승제인 silybin을 발굴하였다. silybin은 Wnt가 존재 않을 경우에는 ${\beta}$-catenin 단백질의 수준에 영향을 미치지 않지만 Wnt가 존재할 경우, mRNA 발현양의 변화 없이 세포질내의 ${\beta}$-catenin 단백질의 수준을 증가시킨다. 또한 silybin에 의해 증가된 ${\beta}$-catenin으로 인해 지방세포분화에 중요한 전사인자라고 알려진 PPAR-${\gamma}$와 C/EBP-${\alpha}$의 발현을 억제한다. 따라서 이 연구에서는 silybin이 세포질내 ${\beta}$-catenin 단백질의 수준을 증가시킴으로써 Wnt/${\beta}$-catenin 신호전달체계를 활성한다는 사실을 제시하였다.
인버타제는 설탕을 포도당과 과당으로 가수분해하며, 광합성 조직과 다양한 수용체 조직 사이의 탄수화물 재배치에서 중요한 역할을 수행한다. 인버타제는 식물에서 다양한 기능을 수행하기 위하여 여러가지 isoforms으로 존재한다. 불용성 인버타제는 고농도의 염을 포함하는 버퍼용액에서만 추출되며, 이 종류의 인버타제 중 산성 인버타제는 산성 pH(pH 4-5)에서 최적의 활성을 갖는다. 물리적인 상처에 반응하여 잎, 줄기 및 뿌리에서 불용성 산성 인버타제(INAC-INV)가 유도되는 것이 연구되어 왔다. 식물에서 효소의 localization을 검출하기 위한 면역조직화 연구를 수행하였다. 이 연구에서 인버타제의 축적은 기계적 손상 후 72시간에 최고수준에 도달하였다. INAC-INV의 활성은 대조구 잎보다 상처받은 조직에서 3배까지 증가하였다. 면역조직화 결과는 INAC-INV가 세포벽과 세포간극에 축적되어 있음을 보여주었다. INAC-INV는 또한 상처와 가까운 사부조직의 체관세포벽에 위치하였다. 종합해 볼 때, 이 연구는 상처에 의한 INAC-INV 유도는 상처치료 과정에서 필요한 높은 에너지 요구에 대한 반응에 역할을 할 수 있음을 추측케 한다.
Saprolegniasis is one of the most devastating oomycete diseases in freshwater fish which is caused by species in the genus Saprolegnia including Saprolegnia parasitica. In this study, we isolated the strain of S. parasitica from diseased rainbow trout in Korea. Morphological and molecular based identification confirmed that isolated oomycete belongs to the member of S. parasitica, supported by its typical features including cotton-like mycelium, zoospores and phylogenetic analysis with internal transcribed spacer region. Pathogenicity of isolated S. parasitica was developed in embryo, juvenile, and adult zebrafish as a disease model. Host-pathogen interaction in adult zebrafish was investigated at transcriptional level. Upon infection with S. parasitica, pathogen/antigen recognition and signaling (TLR2, TLR4b, TLR5b, NOD1, and major histocompatibility complex class I), pro/anti-inflammatory cytokines (interleukin $[IL]-1{\beta}$, tumor necrosis factor ${\alpha}$, IL-6, IL-8, interferon ${\gamma}$, IL-12, and IL-10), matrix metalloproteinase (MMP9 and MMP13), cell surface molecules ($CD8^+$ and $CD4^+$) and antioxidant enzymes (superoxide dismutase, catalase) related genes were differentially modulated at 3- and 12-hr post infection. As an anti-Saprolegnia agent, plant based lawsone was applied to investigate on the susceptibility of S. parasitica showing the minimum inhibitory concentration and percentage inhibition of radial growth as $200{\mu}g/mL$ and 31.8%, respectively. Moreover, natural lawsone changed the membrane permeability of S. parasitica mycelium and caused irreversible damage and disintegration to the cellular membranes of S. parasitica. Transcriptional responses of the genes of S. parasitica mycelium exposed to lawsone were altered, indicating that lawsone could be a potential anti-S. parasitica agent for controlling S. parasitica infection.
The Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and probably the most common plant among the various herbal folk remedies being used in the treatment of abdominal pain hepatitis chronic liver disease, jaundice and coughing in Korea. Recently the biological and pharmacological actions of herb have been studied well such as antibacterial, antidiabetic and antitumor activities. This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillarix extracts against Hepa-1c1c7 and Sarcoma 180 cancer cells on in vivo experimental tests. On in vivo experimental tests using 280 ICR mice the gain of body weight in the control-group mice bearing Sarcoma 180 ascites tumor was 1.5 times more than that of the normal-group mice after 33 days. However, the gain of body weight in all experimental groups administered with Artemisia capillaris extracts was significantly lower than that of the control-group mice (P<0.05). The mean survival times of mice administered with Artemisia capillaris extracts of 25 and 100 mg/kg for 28 days were shown to be 25.39% and 15.39% longer than that of the control-group mice injected with saline (P<0.05). Artemisia capillaris extracts showed the highest tumor inhibition effects, which were 42.4% and 27.2% when intraperitoneally injected with doses of 25 and 100 mg/kg once a day for 28 days in inoculated ICR mice with Sarcoma 180 solid tumor cells (P<0.05). The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.
Insomuch as it is important to manage water quality, from the perspective of water management, it is essential to understand the effect of the weirs on water quality and phytoplankton dynamics in various regions. The purpose of this study is to investigate the characteristics of nitro-nutrients, as well as occurrences and succession patterns of phytoplankton, in the river sections of the two weirs in the Yeongsan River for the five years (from 2012 to 2016) after the weir construction. In respect to this data, the average water temperature measured at the representative point in the section of the Seungchon Weir ($17.1^{\circ}C$) was higher than that of the Juksan Weir ($16.6^{\circ}C$) by comparison. By way of an analysis of this data, it was found that the water quality variables such as, organic matter, nitrogen nutrients and phosphorus nutrients were improved gradually during the period, but the degree of the improvement differs as noted and measured between the weirs. Under the circumstances, it is especially noted that the $NH_3-N$ concentration was higher for the point of the Seungchon Weir (2.204 mg/L) than that of the Juksan Weir (1.157 mg/L). This indicates that effluent as seen from sewage treatment plants and hydrological feature near the densely population area, could be the main cause for the incidence of water pollution in the upstream section of the Seungchon Weir. Additionally, the phytoplankton analysis showed that a relative abundance of diatoms and green algae were 56.9 % and 25.8 % respectively. However, it is noted that the cyanobacteria was measured lower as 10.7 %. Also, in the study sites cell density and occurrence frequency of cyanobacteria were relatively lower than compared to the same measurements noted in other rivers.
덩굴마름병균인 Didymella bryoniae KACC 40669에 대하여 높은 항진균 활성을 나타내는 Brevibacillus sp. KMU-391은 Russia의 Sanktpeterburg 지역의 토양 시료로부터 분리하였다. Brevibacillus sp. KMU-391이 생산하는 항진균 물질은 D. bryoniae의 생육을 강하게 억제할 수 있었으며 TSB를 기본배지로 하여 탄소원으로 1.0% sucrose와 1.0%의 polypeptone을 첨가한 후 pH를 7.0으로 조정하여 $30^{\circ}C$에서 180 rpm으로 2일간 배양하였을 때 가장 높은 항진균 활성을 나타내었다. 배양액에 $30{\sim}60%$의 ammonium sulfate를 처리하여 Brevibacillus sp. KMU-391이 생산하는 항진균 물질을 회수한 후 D. bryoniae를 액체배양을 통한 항진균 활성을 조사한 결과 대조구와 비교하여 41%의 생육을 저해하는 것으로 확인되었다. 또한 butanol을 이용하여 항잔균 물질을 회수한 후, 다양한 작물병원성 곰팡이를 대상으로 항진균 활성 spectrum을 조사한 결과 조사, 장미 잿빛곰팡이병, 잠두 붉은점무늬병, 고추 탄저병, 참외 탄저병, 수박 등의 박과작물의 덩굴마름병, 보리 질병, 토마토 시들음병, 글라디올러스 마른썩음병, 토마토 질병, 수박 덩굴쪼김병, 수박 질병, 사과나무 역병, 벼 잎집무늬마름병, 참외 줄기썩음병, 그리고 고추균핵병에 대하여 높은 항진균 활성을 나타내었다. 이러한 결과를 바탕으로 Brevibacillus sp. KMU-391을 이용하여 작물병원성 곰팡이에 대한 생물농약으로서의 응용 가능성을 확인 할 수 있었다.
