Callus Induction and Embryogenesis Through Pollen Culture in Paeonia albiflora PALL (작약의 화분배양에 의한 캘러스 및 배발생)

  • In order to induce haploid plant through pollen culture, pollens of Paeonia albiflora were cultured on MS liquid medium The development of micospore through pollen culture was examined The effect of low temperature (5$^{\circ}C$, 10 days) pretreatment on callus induction and embryogenesis in pollen culture was not evident Calli derived from pollen gave rise to globular embryos when transferred onto solid medium containing 0.5 mg/, 2,4-L. The effect of low temperature pretreatment and medium. combination to pollen viability was unrecognized. Pollen viability was reduced as the culture proceeded.

Virulence Attenuation of Pectobacterium carotovorum Using N-Acyl-homoserine Lactone Degrading Bacteria Isolated from Potato Rhizosphere

  • Several soil bacteria were found to degrade N-Acylhomoserine lactones (NAHLs), thereby interfering with the bacterial quorum sensing system. In this research, fifteen strains of NAHL degrading rhizobacteria were isolated from potato rhizosphere. Based on phenotypic characteristics and 16S rDNA sequence analyses, the strains were identified as members of genera Bacillus, Streptomyces, Arthrobacter, Pseudomonas and Mesorhizobium. All tested isolates were capable to degrade both synthetic and natural NAHL produced by Pectobacterium carotovorum subsp. carotovorum (Pcc) strain EMPCC. In quorum quenching experiments selected isolates, especially Mesorhizobium sp., were markedly reduced the pathogenicity of Pcc strain EMPCC in potato tubers and totally suppressed tissue maceration on potato tubers. These led to consider the latter as a useful biocontrol agent against Pectobacterium spp.

  • Mass production of forest medicinal plants is related to quality control of raw medicinal materials. Plant tissue culture is an important technology to produce high-quality plant materials. Numerous factors are reported to influence the success of in vitro regeneration of medicinal plants. Embryogenesis is known to be the most effective techniques and it has developed in some medicinal plant species. Various in vitro cultural condition for direct and/or indirect somatic embryogenesis systems have developed in Epimedium koreaum, Bupleurum falcatum, Paeonia lactiflora, Chrysanthemum zawadskii, Houttuynia cordata etc. In this study, we provide the present statue and information of in vitro propagation techniques that is able to apply as an efficient system for rootstock propagation system of forest medicinal plants.

  • Twenty six per cent of Canada's core biotech companies are Ag-biotechnology, compared with five per cent in the United States. Activities include the use of microorganisms, plant cells to create commercially viable products, and transformation of plants to improve specific qualities. Goals ate to increase the world's food supply, enable crops and animals to resist pests and diseases, increase the nutritional content of food, and improve production efficiency. I believe that there are business opportunities in the agriculture sector for Canada and other countries such as Korea to collaborate. These are research to research : institute to institute: business to business, and combinations of these areas. However, for these opportunities to become a reality we need to increase our awareness of each other's activities and needs. My presentation today was not designed with specifics, but to encourage some strategic thinking by both countries on how we can do business together in agriculture and biotechnology.

On the Possible Function of Telfairia occidentalis Agglutinin in the Plant

  • The fate of Telfairia occidentalis seed Agglutinin, TOA, has been monitored during germination. While the level of the Agglutinin in the cotyledons decreased sharply in the first three days to about half of the initial level, it stabilises at this level for the following twelve days. In this interval, Agglutinin activity becomes manifest in the radicle on the fourth day, peaking on the fifth and decreasing rapidly thereafter. In the plumule, the lectin activity becomes manifest on the sixth day, peaks on the seventh and decreases rapidly thereafter. No lectin activity is detectable in any plant tissue including the rump of the cotyledons twenty-seven days after germination. The implications of these observations on the possible role of lectins in plants are discussed.

Researches Using Radio-labelled Insecticides in Korea

  • The scientific articles using radio labelled insecticides performed by Korean researchers were reviewed. The research works were divided into 4 categories such as soil, plant, animal and insect. All researches used $^{14}C$-labelled chemicals, and the $^{14}C$-carbofuran was widely used among them. Fate of insecticides, bound-residues and metabolic process were staple concerning area in soil study. And the uptake and translocation, metabolism and metabolites also a major interests in plant study. As well as the degradation, metabolic pathway and metabolites, and distribution of chemicals in animal tissue were another point of consideration in animal study. And finally, the penetration ratio into body and resistant mechanism were the major concerning views of study with insects.

Studies on the Culture of Haploid Tobacco Leaf (담배 반수성의 유엽배양에 관한 연구)

  • Young haploid leaf derived from the anthers of tobacco plant was cultuerd and plantlets of various ploidies were obtained. When the leaf was put on the medium supplemented with kinetin as growth regulator, plantlets developed directly from the leaf, and the plants coming out in early stage of culture were all haploid. Plants developing in later stage were mostly haploids with some exception of diploid and aneuploid. Leaves were also cultured on the callus-inducing media supplemented with 2,4-D and kinetiion, and the calluses were sub-cultured for six months. Plants developed from these calluses were mostly aneuploids of various chromosome numbers. In view of the fact that the plants directly developed from the leaf were all haploid, the tissue of the original leaf explant was assumed to be uniform as far as chromosome number was concerned. On the other hand, it seemed that the occurrence of various ploidies in the plants derived from the calluses of same origin was the result of the influence of in vitro culture. Apical meristem tissues and various multicellular bodies were formed in the epidermal and inner mesophyll tissues as well as in the sub-epidermal cells.

Regeneration from Storage Root Disk Culture of Purple Sweet Potato

  • Sweet potato has low regeneration capacity, which is a serious obstacle for the fruitful production of transgenic plants. Simple and rapid regeneration method from storage root explants of purple sweet potato (Ipomoea batatas L.) was investigated. The embryogenic callus was observed from 4 cultivars and its highest rate was induced at 1 μM 2,4-D after 5 weeks of culture. Result revealed that a low concentration of 2,4-D and low light intensity was important factors for embryogenic callus formation. After subculture on medium with 5 μM ABA for 4 days, subsequently, occurred the regeneration of shoots within 4 weeks when these embryogenic callus was transferred onto the MS hormone free medium. Regenerated shoots were developed into platelets, and grown normal plants in the greenhouse. We developed a simple and quickly protocol to regenerate plantlets in storage root explants of purple sweet potato. This regeneration system will facilitate tissue culture and gene transfer research of purple sweet potato.

Construction of Binary Vectors for the Rice Transformation Using a Rice Actin Promoter and Replication Origin of pTi12 Isolated from Agrobacterium tumefaciens KU12 (Agrobactrium tumefaciens KU12로부터 분리한 pli12의 Replication Origin과 벼의 Actin 유전자 프로모터를 이용한 벼의 Binary Vector 제조)

  • Binary vectors, pBI-ActR1, pBI-ActF1 and pBSH-ActR1, were constructed using pGA642, the replication origin of pTi12 and the rice actin promoter. The sizes of pBI-ActR1, pBI-ActF1 and pBSH-ActR1 were 12.9 kb, 13.2 kb and 11.95 kb, respectively. These vectors containing a rice actin promoter followed by a GUS structural gene could induce stronly the expression of GUS gene in transformed rice cells. Rice explants from 3-4 day old seedlings after germinatin were cocultured with A. tumefaceins harboring pBI-ActR1, pBI-ActF1 or pBSH-ActR1, and then GUS expression in the explants was assayed. Transformation of rice explants by these binary vectors was tissue-specific, such that the meristematic regions of shoot apex, root and hypocotyl were transformed by these binary vectors.

Discoloration Pattern of Lettuce Leaf Disks as Influenced by Sulfur Dioxide (아황산에 의한 상치 잎구조의 변색패턴)

  • Development of a model system for mode of action studies of $SO_2$ was attempted with a plant tissue. Leaf disks, 1.0cm diameter, cut from the lamina of lettuce leaves, were floated on the testing medium and placed in light or dark condition to investigate the discoloration pattern with various sources of $SO_2$. Discoloration of leaf disks tended to be more serious with higher concentrations of $SO_2$ and on exposure to the light. Leaf disks were more severely discolored at lower pH with constant SO2 concentration. These discoloration patterns were highly reproducible and similar in all sources of $SO_2$. Spectrophotometric evidence suggested that light-mediated discoloration of leaf disks in the presence of $SO_2$ might occur mainly through chlorophyll ${\alpha}$ degradation.
