Cloning of Pectate Lyase Gene in Erwinia rhapontici (Erwinia rhapontici의 Pectate Lyase 유전자 Cloniong)

  • Erwinia rhapontici causes soft-rot disease in a number of plants such as onion, garlic and hyacinth. There has been no report that E. rhapontici produces pectate lyase. Pel gene was cloned from genomic DNA of the parasitic soft-rot E. rhapontici polymerase chain reaction by using synthetic oligonulceotide primers designed from the pel 1 to E. carotovora. The recombinant plasmid pJE101 containing pectate lyase gene, when introduced into E. coli DH5$\alpha$, produced pectate lyase an macerated hyacinth tissue.

Studies on the Cotyledon Culture of Panax ginseng (고려인삼의 자엽배양에 관한 연구)

  • Cotyledon of Panax ginseng was cultured in the growth regulator-free Knudson C medium comprising only several kinds of mineral salts and sucrose. Shoot primordium or callus developed profusely from the cotyledonary tissue and finally plantlets were produced directly from the shoot primordium or indirectly through callus. Microscopic examination revealed that the epidermal cell as well as the mesophyll cell of the cotyledon became meristematic and divided, changing into multinucleate cell or multicellular body, eventually developing into either a shoot primordium or callus.

Effect of Polyamine on Auxin Polar Transport in Corn Coleoptile Segments (Polyamine이 옥수수 자엽초 절편에서 오옥신 이동에 미치는 영향)

  • The effects of polyamines on auxin polar transport were studied in corn coleoptile segments. Among putresine, spermidine and spermine tested in labelled auxin transport, spermidine inhibited auxin polar transport most strongly. Its inhibitory effect appeared after 1 h of transport period. Spermidine inhibited labelled auxin and 14C-benzoic acid accumulation into the tissue in the various pH range tested (pH 4.0-8.0). These results suggest that the inhibition of auxin transport may not be due to decrease in pH by spermine the effect of decreased pH in the extracellular space.

Scab Disease of Aralia elata Caused by Elsinoe araliae (Elsinoe araliae에 의한 두릅나무 더뎅이병)

  • Scab disease of Aralia elata (Miq.) Seemann occurred severely in Kangwon province, Korea. First, brown spots were formed on leaves and enlarged along vein. The infected leaves including petiole and midrib were twisted forming scab and died eventually. Also the disease infected twigs and trunks of the tree forming scab. Conidia, ascospores and asci developed in locule were observed in the infected tissue. The causal organism of the scab disease of Araliae elata was isolated and identified as Elsinoe araliae Yamamoto (imperfect stage: Sphaceloma araliae Jenkins) based on the morphological and cultural characteristics, and pathogenic test.

  • Explants from stem, leaf and root tissue of Cissus juttae were plased on Ms medium supplemented ifferent combination of 2.4-D, NAA, Kinetin and BA in a actorial experiment. some callus formed in every treatment, but the best callus growth was on media containing $1.0{\;}mg/{\ell}$ NAA and $1.0{\;}mg/{\ell}$ BA. Formation of advantitious emboyos was evident in callus to MS medium supplemented with $0.1{\;}mg/{\ell}$.
