Plant Cell Contact-Dependent Virulence Regulation of hrp Genes in Pseudomonas syringae pv. tabaci 11528 (Pseudomonas syringae pv. tabaci 에서 식물세포접촉에 의한 병원성 유전자의 조절)

  • The hrp gene cluster in the plant pathogen Pseudomonas syringae is a key determinant of pathogenicity. Recent studies have demonstrated that specific host cell induction of the Ralstonia solanacearum hrp gene cluster is controlled by the PrhA (plant regulator of hrp) receptor. To characterize the role that P. syringae PrhA plays in the virulence of plant cells, a prhA homolog was isolated from P. syringae pv. tabaci and a $\Delta$prhA mutant was constructed by allelic exchange. The $\Delta$prhA mutant had reduced virulence in the host plant, and co-culture of P. syringae pv. tabaci and plant cell suspensions induced a much higher level of hrpA gene transcription than culture in hrp-inducing minimal medium. These results indicate that PrhA of P. syringae is a putative pathogen-plant cell contact sensor, therefore, we used a hrpA-gfp reporter fusion to monitor the in situ expression of PrhA. The results of this study demonstrated that PrhA induces hrp gene expression in P. syringae pv. tabaci in the presence of plant cells.

Proteomic Analysis of the GacA Response Regulator in Pseudomonas chlororaphis O6

  • The GacS/GacA system in the root colonizer Pseudomonas chlororaphis O6 is a key regulatory system of many traits relevant to the plant probiotic nature of this bacterium. The work in this paper elucidates proteins using proteomics approach in P. chlororaphis O6 under the control of the cytoplasmic regulatory protein, GacA. A gacA mutant of P. chlororaphis O6 showed loss in production of phenazines, acyl homoserine lactones, hydrogen cyanide, and protease, changes that were associated with reduced in vitro antifungal activity against plant fungal pathogens. Production of iron-chelating siderophore was significantly enhanced in the gacA mutant, also paralleling changes in a gacS mutant. However, proteomic analysis revealed proteins (13 downregulated and 7 upregulated proteins in the mutant compared to parental strain) under GacA control that were not apparent by a proteomic study of a gacS mutant. The putative identity of the downregulated proteins suggested that a gacA mutant would have altered transport potentials. Notable would be a predicted loss of type-VI secretion and PEP-dependent transport. Study of mutants of these GacA-regulated proteins will indicate further the features required for probiotic potential in this rhizobacterium.

  • This paper develops a pole-zero assignment self-tuning regulator utilizing the method of a neural network in the plant parameter estimation. An approach to parameter estimation of the plant with a Hopfield neural network model is proposed, and the control characteristics of the plant are evaluated by means of a simulation for a second-order linear time invariant plant. The results obtained with those of Exponentially Weighted Recursive Least Squares(EWRLS) method are also shown.

  • This experiments were conducted to investigate growth regulator concentrations and a period of chilling treatment for increasing the size of bulblets of Narcissus pseudo-narcissus cv King Alfred. Combination of 2.5 mg/L BA and 2.5 mg/L NAA remarkedly increased the diameter and growth of regenerated bulblets. Chilling period favorable for the growth of regenerated bulblets was found to be 8 weeks, and addition of 1.0 mg/L NAA alone to MS medium was the most effective for increasing the size of bulblet for the culture after the chilling treatment.

  • When the fuzzy controller apply to a real plant, We have not excepted result of a satisfactory control by modeling error and lacking information about an plant. In this case, we have to adjust the control factors for improvement of the control performance and this method need a lot of time and cost for perform a trial and error. In this paper, we proposed the fuzzy regulator with the automatic adjustment of scaling factors. It was improve upon the control performance using a adequate scale factor by fuzzy inference. We implemented the controller using the DSP processor and applied in a hydraulic servo system. And then we observed an experimental results.

  • Flowering time (or heading date) is controlled by intrinsic genetic programs in response to environmental cues, such as photoperiod and temperature. Rice, a facultative short-day (SD) plant, flowers early in SD and late in long-day (LD) conditions. Casein kinases (CKs) generally act as positive regulators in many signaling pathways in plants. In rice, Heading date 6 (Hd6) and Hd16 encode $CK2{\alpha}$ and CKI, respectively, and mainly function to delay flowering time. Additionally, the major LD-dependent floral repressors Hd2/Oryza sativa Pseudo-Response Regulator 37 (OsPRR37;hereafter PRR37) and Ghd7 also confer strong photoperiod sensitivity. In floral induction, Hd16 acts upstream of Ghd7 and CKI interacts with and phosphorylates Ghd7. In addition, Hd6 and Hd16 also act upstream of Hd2. However, whether CKI and $CK2{\alpha}$ directly regulate the function of PRR37 remains unclear. Here, we use in vitro pull-down and in vivo bimolecular fluorescence complementation assays to show that CKI and $CK2{\alpha}$ interact with PRR37. We further use in vitro kinase assays to show that CKI and $CK2{\alpha}$ phosphorylate different regions of PRR37. Our results indicate that direct posttranslational modification of PRR37 mediates the genetic interactions between these two protein kinases and PRR37. The significance of CK-mediated phosphorylation for PRR37 and Ghd7 function is discussed.

An Analytical Design of Feedback Regulator and Signal State Estimator in Discrete Linear Systems (이산 선형시스템에서의 피이드백 조정기 및 신호상태 추정기의 해석적 설계)

  • This paper deals with an analytic design of feedback regulator and signal state estimator in discrete linear systems. On the way of developing the deadbeat regulator, some necessary conditions for control policy have been derived, it is proved that the q periods delay in the control causes q periods delay in the point at which deadbeat response occurs. We have derived some relations such that the eigenvalue of system plant can be arbitrarily changed by the characteristics of minor loop compensator which is introduced in feedback path. And also we show that the signal state estimator which estimates the state of given signal sequence must satisfy some conditions. Theorems and conclusions are described with some simplel nontrivial numerical examples and signal state tracking application problems.

Robust regulator design for an interval plant (구간 플랜트에 대한 견실한 레귤레이타 설계)

  • In this paper, we present an algorithmic technique for determining a feedback compensator which will stabilize the interval dynamic system, specifically, the robust regulator design for interval plants. The approach taken here is to allow the system parameters to live within prescribed intervals then design a dynamic feedback compensator which guarantees closed-loop system stable. The main contribution of this paper is the idea of introducing a "simplified Kharitonov's result" for low order polynomials to search for suitable compensator parameters in the compensator parameter space to make the uncertain syste robust. We also design the robust regulator which will D-stabilize (have the closed-loop poles in the left sector only) the dynamic interval system while having good performance. The nuerical examples are given to show the substantially improved robustness which results from our approach. approach.

  • A multi-heat pump provides the benefits of comfort, energy conservation and easy maintenance. Recently, the multi-heat pump has been widely employed in small and medium-sized buildings. However, the control algorithm of the multi-heat pump are limited in the open literature due to complicated operating conditions. In this study, the MIMO control algorithm using integral optimum regulator was designed and the control performance of it was analyzed. In addition, system model of the control plant was developed by PRBS system identification scheme. The MIMO controller adopting the integral optimum regulator yielded satisfactory control performance results.
