Isolation, Cultivation, and Antifungal Activity of a Lichen-Forming Fungus

  • A lichen-forming fungus was successfully isolated by discharged spore method from Korean lichen (Hetero-dermia sp.) and cultivated in pure culture. The isolate JR0012 inhibited mycelial growth of several plant-pathogenic fungi. Mycelial growth of the four Pythium spp. tested was completely inhibited. Potato dextrose broth was found to be the medium favorable for large-scale production of antibiotics from the isolate. Anti-fungal substances produced in axenic culture were partially purified. This is the first report in Korea of lichen-forming fungus successfully isolated and which exhibited strong antifungal activity against plant-pathogenic fungi, especially the four Pythium spp..

속성배추를 이용한 식물 탐구 수업이 초등학생의 식물 개념 변화에 미치는 효과 (Effects of the Plant Inquiry Instruction using Rapid-cycling Brassica rapa on the Change of Plant Concept of the Elementary School Students)

  • This study is intended to develop plant inquiry instruction using Rapid-cycling Brassica rapa (RcBr) whose life cycle is relatively short, and to apply it to the elementary science instruction of the 4th grader and examine their plant concept, science inquiry skills and science-related attitudes. The materials were consisted of laboratory manuals for the students as well as teachers' guide. By observing the life cycle of RcBr, students can experience the conceptual learning of the plant's life cycle. In addition, this study investigated the cause of change in science inquiry skills and science-related attitudes by interviewing 12 students. It has shown that plant inquiry instruction using RcBr has meaningful effects on students' understanding of the plant concept, improving students' science inquiry skills, and changing students' science-related attitudes. Students who showed improvement in science inquiry skills were able to answer questions regarding science knowledge correctly. And students whose science related attitudes were improved had a positive attitude on cultivating RcBr. Students told that RcBr was an interesting and good material to inquire plant. Because of its small size and its relatively short life cycle of RcBR, it should be a desirable plant material for the inquiry instruction which can give rise to useful and meaningful results for the elementary school students.

Inhibitory Effects of Super Reductive Water on Plant Pathogenic Fungi

  • The antifungal activity of super reductive water (SRW) against plant pathogenic fungi was examined to extend its application to integrated pest management (IPM) for plant diseases. Diluted solutions ($\times$1/10, $\times$1/25, and $\times$1/50) of SRW inhibited fungal growth of kiwifruit soft rot pathogen, Diaporthe actinidiae, in a concentration dependent manner, When kiwifruits were inoculated on wounds with mycelium blocks, stock and diluted solutions successfully inhibited the disease development. In addition to the high pH of the SRW, fungistatic activity was also considered as the cause of the antifungal effect against the pathogen. Whereas conidial germination of Magnaporthe grisea was not affected by the diluted SRW solutions, appressorium formation was significantly inhibited in a concentration dependent manner, With little harmfulness to human health and environment SRW could be used to control plant pathogenic fungi, particularly appressorium-forming fungal pathogens.

식물형태학에서 사용하는 피자식물의 꽃 구조에 관한 한글 용어의 분석 (Analysis of Some Korean Terminologies on the Flower Structures of Angiosperms in Plant Morphology)

  • Korean terminologies on flower structures of angiosperms in plant morphology, written incorrectly in many books, were analysed to suggest accurately expressed terminologies. 24 books in areas such as general biology, plant biology, plant morphology, and biological dictionaries and glossaries were selected to analyse the accuracy of the terminologies for seed structures, e.g., crassinucellate (ovule), tenuinucellate (ovule), placentation, free cental placentation, herkogamy or hercogamy, heterostyly or heterostylous, distyly or distylous, tristyly or tristylous, and hypanthium. The definition and etymology of the terminologies were traced in 4 textbooks of plant anatomy and 2 dictionaries of biology and botany written in English. On the basis of the definition, etymology, and principles for terminology formation according to the International Organization for Standardization (ISO 704:2000), accurately and reasonably expressed Korean terminologies were proposed. The 9 terminologies examined in this study were found in the glossary of biological terminologies, published by the Korean Association of Biological Sciences in 2005, and designated as an editorial source for science and biology textbooks for middle and high schools by Ministry of Education in 2007. However, none of the 9 terminologies described in the glossary were consistent with those proposed in the present study. The inconsistencies indicated the need for a reassessment of this glossary of biological terminologies. The new, proposed terminologies would facilitate mutual understanding between teachers and students of plant biology.

초등학교 학급 운영의 일환으로서 식물 관찰 활동의 효과 (Effects of Plant Observation Activities as a Strategy for Classroom Management in the Elementary School)

  • 본 연구는 초등학교 3학년 학급에서 학급 운영의 일환으로 식물 관찰 활동을 1년 동안 진행하며 학생들에게 미친 효과가 무엇인지 탐색한 연구이다. 식물 관찰 활동의 효과를 살펴보기 위해 식물 관찰 활동을 시간적인 순서에 따라 기술하며 관찰 유형을 분석하였고, 학생들이 느낀 인식적 정서를 분석하였다. 본 연구의 결론은 다음과 같다. 첫째, 꾸준한 식물 관찰 활동은 학생들의 관찰 유형에 의미 있는 변화를 일으켰다. 둘째, 식물 관찰 활동은 학생들이 긍정적인 정서를 가지고 참여하도록 유도하였다. 즉, 꾸준한 식물 관찰 활동은 학생들이 다양한 감각을 활용하여 관찰하도록 하였고, 흥미를 높이고 호기심을 키우며 재밌게 참여한 후에는 만족을 주는 배움을 이끌어냈다.

초등학교에 설치된 식물이름표 현황 조사연구 - 광주·전남권을 중심으로 - (A Survey on the Installation Status of Plant-label in Elementary Schools: Focusing on Gwangju·Jeollanam-do)

  • 광주광역시 및 전라남도 소재 초등학교 35곳을 대상으로 하여 식물이름표 설치 현황을 조사하였다. 식물이름표의 설치 여부 및 상태 등을 종합하여 평가한 결과, 35개 초등학교 중 12곳이 '우수'한 것으로, 13곳은 '양호'한 것으로, 그리고 8곳이 '미흡'한 것으로 나타났다. 학교 소재지나 역사, 부지 면적, 구성원 규모 등은 식물이름표의 현황이나 상태에 직접적인 영향을 미치진 않은 것으로 확인되었다. 다양한 종류의 식물들에 여러 유형의 식물이름표를 설치하고 계속적으로 갱신함으로써 해당 학교가 식물이름표를 중요한 교구로서 인식하고 있다는 인상을 주는 경우가 있던 반면, 단 하나의 식물이름표도 설치돼 있지 않은 학교들도 있었다. 또한, 식물이름표가 설치돼 있긴 하였으나 지속적인 유지·관리가 이뤄지고 있지 못한 채 방치되고 있는 사례들도 있었다. 전문업체에 의뢰하여 식물이름표를 제작·설치한 사례들도 있었으나 그 내용에 오류가 있거나 설치 이후 제대로 관리되질 못해 되려 학교의 경관을 훼손하고 있는 사례들도 있었다. 이에, 학교별 개성을 살릴 수 있을 뿐만 아니라 지속가능한 형태의 식물이름표를 제작하고 설치하는 과정 자체를 교육적으로 활용할 수 있도록 식물이름표를 자체적으로 제작하고 설치하는 것을 제안하는 바이다.

Restriction Analyses of PCR Amplified Partial SSU Ribosomal DNA to Distinguish Arbuscular Mycorrhizal Fungi from Other Fungi Colonizing Plant Roots

  • Roots of Glycine max and Miscanthus sinensis and soil samples were collected from various field sites at Goesan, Chungbuk in Korea. Microscopic observations of the roots indicated high colonization rates of both arbuscular mycorrhizal fungi(AMF) and other fungi. The partial small subunit of ribosomal DNA genes were amplified with the genomic DNA extracted from their roots by nested polymerase chain reaction(PCR) with universal primer NS1 and fungal specific primers AML Restriction fragment length polymorphism(RFLP) was analyzed using the combinations of three restriction enzymes, HinfI, AluI and AsuC21. Nucleotides sequence analysis revealed that ten sequences from Miscanthus sinensis and one sequence from Glycine max were close to those of arbuscular mycorrhizal fungi. Also, 33% of total clones amplified with NS31-AM1 primers from M. sinensis and 97% from G. max were close to Fusarium oxysporum or other pathogenic fungi, and they were successfully distinguished from AME Results suggested that these techniques could help to distinguish arbuscular mycorrhizal fungi from root pathogenic fungi in the plant roots. Especially, DNA amplified by these primers showed distinct polymorphisms between AMF and plant pathogenic species of Fusarium when digested with AsuC21.

교재원 식물을 활용한 환경놀이 프로그램 개발 및 적용 (Effectiveness of Environmental Play Teaching Program using the Plant in Neighborhood Learning Gardens)

  • The purpose of this research is placed on the development and application of environmental play programs for elementary school students their utilizing the neighborhood learning gardens in their districts to find its effectiveness. The research questions drawn to attain the purpose of the research are as follows. Initially, analyze the contents related to the plant appearing in the textbooks of wise life, science and practical arts of the 7th elementary school curriculum. Secondly, develop a plant utilizing environmental play program targeted towards the 4th grade elementary school students with the results of the textbook analysis as the foundation. Third, apply the plant utilizing environmental play program into the classroom to verify its effectiveness. Based on the conclusion of this research, the following is to be proposed. First, the revitalization of regional environmental education utilizing the various surroundings of the region is essential. Also, the current method of education, which is focused on theoretical knowledge, and being operated in most of the schools need to be changed over to diverse environmental education programs that are linked with the region. For this, an internet based database, information sharing and exchange program centralized around the regional environmental education center needs to be prepared. Moreover, since this research had developed an environmental play program utilizing plants that focused on the 4th grade elementary school students to verify its effectiveness, the development for environmental play programs dealing with various themes for each grades is required. Lastly, although this research has verified the effectiveness in the variables of ecological knowledge, environmental susceptibility, environmental attitude, environmental concern, environmental function and responsible environmental behavior amongst the variables of environmental literacy but there requires a succeeding research considering the variables that haven't been included in this research.

국내 건설현장 외국인 근로자 안전교육 모델 개발 (A Development of Safety Education Model for a Foreign Worker in Domestic Construction Site)

  • 국내 건설현장에서는 외국인 근로자들의 수가 증가함에 따라 그들에게 적합한 안전교육체계의 개발이 요구되고 있다. 본 연구에서는 외국인 근로자들을 위한 안전교육 모델을 제안하고 실제로 프로토타입의 안전교육시스템을 구현하였다. 그 결과로 이러닝 시스템 기반의 안전교육 모델을 개발하였다. 이 시스템은 국내 건설현장에서 근무하고 있는 외국인 근로자들이 안전교육을 잘 받을 수 있도록 지원해 줄 수 있을 것이다. 또한 현장의 안전관리자로 하여금 외국인 근로자들을 잘 교육하고 관리할 수 있도록 지원해 줄 것이다.

Effects of Soils Containing Arbuscular Mycorrhizas on Plant Growth and Their Colonization

  • Four arbuscular mycorrhizal fungal(AMF) inocula collected from three arable sites in Korea were used to determine plant growth, mycorrhizal root colonization rate and spore production in three different host plant species; Sorghum bicolor, Allium fistulosum, Tagetes patula. Growth of plant treated with AMF differed from those without AMF. Different AMF inocula showed significantly different root colonization rates and spore production of AMF on the wild plants, A. fistulosum and T. patula, but did not on the cultivated plant, S. bicolor. Results suggested that indigenous mycorrhizal fungal community would be important factors in mycorrhizal symbiosis, and play important roles in the plant succession.
