  • Background: Endophytic fungi play an important role in balancing the ecosystem and boosting host growth. In the present study, we investigated the endophytic fungal diversity of healthy Panax notoginseng and evaluated its potential antimicrobial activity against five major phytopathogens causing root-rot of P. notoginseng. Methods: A culture-dependent technique, combining morphological and molecular methods, was used to analyze endophytic fungal diversity. A double-layer agar technique was used to challenge the phytopathogens of P. notoginseng. Results: A total of 89 fungi were obtained from the roots, stems, leaves, and seeds of P. notoginseng, and 41 isolates representing different morphotypes were selected for taxonomic characterization. The fungal isolates belonged to Ascomycota (96.6%) and Zygomycota (3.4%). All isolates were classified to 23 genera and an unknown taxon belonging to Sordariomycetes. The number of isolates obtained from different tissues ranged from 12 to 42 for leaves and roots, respectively. The selected endophytic fungal isolates were challenged by the root-rot pathogens Alternaria panax, Fusarium oxysporum, Fusarium solani, Phoma herbarum, and Mycocentrospora acerina. Twenty-six of the 41 isolates (63.4%) exhibited activity against at least one of the pathogens tested. Conclusion: Our results suggested that P. notoginseng harbors diversified endophytic fungi that would provide a basis for the identification of new bioactive compounds, and for effective biocontrol of notoginseng root rot.

AMOVA (Analysis of molecular variance)

  • Endophytic fungi occupy inner plant tissues, which results in various interactions between the fungus and host. Studies on endophytic fungi have been conducted in Korea for over 30 years. This paper summarizes the published results of those studies. The endophytic fungi of approximately 132 plant species in Korea have been studied since the 1990s, resulting in over 118 publications. The host plants featured in these studies comprised 3 species of mosses, 34 species of woody plants, and 95 species of herbaceous plants. At the family level, the most studied plants were members of the Poaceae family, covering 18 species. Regionally, these studies were conducted throughout Korea, but over half of the studies were conducted in Gyeongsangbuk-do, Gangwon-do, and Chungcheongnam-do. Relatively few studies have been conducted in a metropolis such as Seoul. We confirmed 5 phyla, 16 classes, 49 orders, 135 families, 305 genera, and 855 taxa of endophytic fungi, excluding Incertae sedis, whose relationship with others are unknown. Most of the endophytic fungi belonged to Ascomycota (93.2%), and a few belonged to Basidiomycota (3.6%). Since the diversity of endophytic fungi differs depending on the host plant, plant tissue, and distribution region, future studies should be conducted on multiple host plants and in various regions. Future studies on endophytic fungi are expected to broaden, including genomics and taxonomic and ecological studies of secondary metabolites.

  • The characteristics of 124 Phytolacca americana present areas and 88 surrounding absent areas over the country were investigated and compared from July 2016 to Sept 2017. The Phytolacca americana was confirmed as having been nationally distributed and is usually found in roadsides at an altitude of 380 m (26%), forest edges (24%), forests (19%), sunny spots (60%), dry soils (53%), sandy loams (31%) and sand+gravel soils (22%). 321 taxons consisting of 79 families, 209 genera, 289 species, 2 subspecies, 26 varieties and 4 cultivars and 286 taxons consisting of 76 families, 195 genera, 256 species, 2 subspecies, 24 varieties and 4 cultivars were found in the Phytolacca americana present areas and the absent areas, respectively. The relative net contribution degree was found highest in the Phytolacca americana (100) followed by Robinia pseudoacacia (29.4), Humulus japonicus (8.8) and Artemisia princeps (6.0) in the present areas, while it was found highest in Robinia pseudoacacia (100) followed by Humulus japonicus (44.4), Artemisia princeps (35.9), Pueraria lobata (18.6) and Setaria viridis (12.3) in the absent areas. The Raunkier's life form in the present/absent areas was found highest in the therophytes (34%/34%), followed by hemicryptophytes (17%/18%), megaphanerophytes and mesophanerophytes (18%/17%), nanophanerophytes (11%/11%) and geophytes (9%/11%). There was no significant difference in the species diversity between the present (2.141) and the absent (2.124) areas. However, the allelopathic agent and light blocked-out by the Phytolacca americana could possibly inhibit germination and the development of other species; especially, where a difference in species diversity was found high in forests, compared to other habitats (p=0.0605). Accordingly, it is suggested to review possible disturbances of native species habitats in shady spots including forest gaps, although the effect of Phytolacca americana on the ecological system in the suburban open and disturbed areas is not large.

  • The study investigated species diversity, relative abundance, and decline of flying insects and plants within a fragmented forest in the Federal University of Technology Akure (FUTA), Ondo State, Nigeria. It is known that habitat fragmentation can reduce biodiversity. Thus, it is important to perform comprehensive assessments to understand implications of the habitat fragmentation for flora and fauna. Species richness and abundance of flying insects and plants across fragmented forest patches were quantified using field surveys and taxonomic identification. This study revealed shifts in species diversity, with fragmented areas exhibiting reduced biodiversity compared to contiguous forest ecosystems. Flying insects crucial for ecosystem functioning and pollination services demonstrated decreased species richness and relative abundance within fragmented habitats. This decline was attributed to habitat loss, altered microclimates, and limited movement pathways known to hinder insect dispersal. Similarly, plant species richness and abundance showed decline in fragmented forest due to disrupted mutualistic interactions with pollinators, altered nutrient cycling, and increased competition among plant species. This study underscores the importance of maintaining intact forest habitats to sustain healthy ecosystems and preserve biodiversity. Effective conservation strategies should focus on habitat connectivity, reforestation efforts, and protection of essential ecological corridors to mitigate effects of fragmentation. In conclusion, this investigation provides empirical evidence for effects of habitat fragmentation on flying insects and plants in a forest ecosystem in FUTA Akure, Nigeria. Findings emphasize an urgency of adopting conservation measures to safeguard these invaluable components of biodiversity and ecosystem stability in the face of ongoing habitat loss and fragmentation.
