Production of Recombinant Rotavirus Capsid Protein VP7 from Stably Transformed Drosophila melanogaster S2 Cells

  • Stably transformed Drosophila melanogaster 52 cells producing recombinant VP7 were obtained, and recombinant VP7 expression was confirmed by Western blot analysis. The molecular weight of recombinant VP7 expressed in 52 cells was approximately 35.5 kDa, and 75% of the total VP7 produced was present in the medium. Recombinant VP7 contained N-linked glycosylated oligosaccharides. Aprotinin, leupeptin, and polyvinylpyrrolidone did not have any noticeable effect on recombinant VP7 production; however, DMSO and sodium butyrate increased its production by 120% and 60%, respectively.

Immunocytolocalization of Cell Wall Peroxidase and Other Wall Antigens from Maize Seedlings

  • Immunocytochemistry utilizes the specificity of the antigen-antibody reaction to localize specific antigens in cells or cellular organelles. Here we report the use of monoclonal antibodies, in conjunction with gold-labeled second antibodies to study the ultrastructural localization and tissue distribution of the Mr 98, 000 anionic peroxidase and other wall antigens. The antibody specific for this wall peroxidase, mWP3, labeled mainly the cell wall area. At the tissue level, the Mr 98, 000 peroxidase is located predominantly in the leaf mesophyll, internal coleoptile and sieve elements, but not in the root, as assayed with these procedures. The coleoptile walls were less heavily stained than the walls of leaf mesophyll cells. At the subcellular level, it is localized mainly in intercellular regions of the cell walls. A similar staining pattern was revealed by mWP19, one of anti-$\beta$ glucosidase antibody, though it looked less heavily stained than one with mWP3. In order to serve as a control wall staining using IgM monoclonal antibodies, mWP18 was used. Most of the label is localized over wall regions of cells of the young leaf mesophyll and coleoptile.

Atractylodes japonica Rhizome Inhibits Cell Proliferation and Induces Apoptosis in vitro

  • Antiproliferative activity of the ethanol extract of Atractylodes japonica rhizomes (AJEX) was investigated using methyl thiazolyl tetrazolium (MTT) assays with various cancer cell lines (HL-60, MCF-7, SK-Br-3, MDA-MB-453, HepG2, Hep3B, PC-3, LNCaP, MKN 28, MKN 45, and HT-29 cells). Gastric carcinoma cell lines were the most responsive in terms of cell proliferation. The $IC_{50}$ of MKN 28 and MKN 45 cells were 35.98 and 27.57 ${\mu}g/mL$, respectively. Moreover, gastric carcinoma cells exposed to AJEX underwent apoptosis, as determined by Annexin V binding assay. Compared to respective control level, exposure to the AJEX at each $IC_{50}$ concentration resulted in a remarkable increase in the shift of cell populations. Present results suggest that AJEX possess potential anticancer properties.

Effects of $Ca^{2+}$ and Polyamine on Callose Contents in Carrot Suspension Cultured Cells (당근 현탁배양세포에서 $Ca^{2+}$과 Polyamine이 Callose 함량에 미치는 영향)

  • The effects of Ca2+ on polyamines on callose contents of carrot suspension cultured cells were studied. The regeneration process of the cell wall of carrot protoplast observed through the electron microscope. Treatment of the carrot suspension cultured cells with Ca2+ and polyamines resulted in considerable increase on callose contents at 0.1 mM of Ca2+ and polyamines, particulary spermidine. Poly-L-lysine and poly-L-ornithine increased about 30% and 100% of callose contents than that of the control respectively, whereas verapamil and flunarizine markedly decreased the callose contents. These effects of Ca2+ of free ion rather than as Ca2+-calmodulin complex. During the cultivation of the protoplast, the regeneration of the cell wall was somewhat observed on the 4th day, however, it was inhibited by verapamil. These results suggested that the promotive action of Ca2+ and polyamines were manifested in the callose contents and the regeneraton of the cell wall.

Structure, Origin, Development and Senescence of Collecters in Nerium indicum Mill. (N. odorum Soland., Apocynaceae)

  • Nerium indicum has adaxial colleters on each petiole, bract, bracteole, calyx and corolla. The colleters are finger-shaped sometimes lobed structures with tall heads on short stalks. The petiolar colleters are more abundant than those on the bract, bracteole, calyx and corolla but their structure is similar. Meristematic activity in a small group of protodermal and hypodermal cells at the base of the developing leaf gives rise to the colleter that further differentiates by frequent anticlinal and periclinal divisions. Many of the colleters are vascullarized, sometimes a layer of redially elongated cells is present beneath the epithelial cells. Senescence of a colleter begins when the leaves have either completed their growth or are still expanding. In senescent colleters, the central cell cytoplasm and nuclei are considerably reduced. Thin layer chromatography of the freshly harvested exudate of colleter shows the presence of rhamnose, glucose and arabinose sugars.

  • The objective of this work was to accomplish the isolation, molecular identification and characterizing the physiology of the causal agent of the algal spot in mango trees. For this purpose, the pathogen growth was assessed in different culture media, with subsequent observation and measurements of the filamentous cells. The molecular identification was made using mycelium obtained from leaf lesions and pure algae colonies grown in culture medium. Descriptions based on DNA sequencing indicated that the algae is Cephaleuros virescens. The algae must be isolated primarily in liquid medium for further pricking into agar medium. The highest mycelial growth average in Petri dishes occurred when the algae were grown in Trebouxia and BBM. Trebouxia enabled larger cells in the filamentous cells when compared to other culture media.

  • The K+ and Na+ contents in the guard cells of Agave which had a characteristic of CAM plant were measured by using "Rolling technique". That results were correspond with the change of the stomatal aperture width. That is to say, stomatal movement of Agave is due to the change of K+and Na+ concentration in the guard cells. As Agave which was used in this experiment showed two peaks of which one was at 3 hour and the other was at 24 hour in stomatal aperture width, it was seemed that Agave had both characteristics of CAM and $C_3$pattern.

Inhibitory Effects of the Extracts from Eleutherococcus senticosus Maxim. on Histamine-release from Rat's Mast Cell

  • Eleutherococcus senticosus Maxim. has been successfully used as an oriental medicine for various diseases including allergic disorders. Histamine is a major factor on various allergic responses and it is reported that histamine was released from mast cells by sensitization of allergens. In this study, ethanol extracts from E. senticosus Maxim. were prepared and the composition was analyzed by high performance liquid chromatography. The eleutheroside B as a primary effective component of E. senticosus was contained approximately 225 mg/kg in root bark extracts. The extracts were found to significantly inhibit compound 48/80-induced histamine release form mast cells in dose dependent manner. However the extracts had low cytotoxicity on the mast cells with MTT assay. These results showed that E. senticosus Maxim. extracts may be the effective materials on inflammatory disorders.

Regulation of Immunological Effect of Rubia cordifolia Extract and Associated MAPKs Pathway in RBL-2H3 Cell-line

  • Rubia cordifolia has been used to treat diseases for many years in China and India. Although the biological properties and major compounds of R. cordifolia have been extensively studied, the underlying mechanisms of its biological effects remain elusive. In terms of immunological effects, anti-inflammation effect of macrophage (Raw 264.7) simply has been reported. In this study, R. cordifolia was extracted in 70% ethanol and the extract did not affect to macrophage (Raw 264.7) pro-inflammation and T cell (Molt-4). However, in mast cell (RBL-2H3), it showed inhibition of degranulation. The inducing inhibitory effect on degranulation was related to concentration dependent variation in phosphorylation of ERK-1/2 and upregulating the JNK phosphorylation in RBL-2H3 cells. Based on these data, we concluded that R. cordifolia newly have anti-allergenic effects in RBL-2H3 and might be used as a therapeutic agent to treat or prevent allergic diseases such as asthma and atopic dermatitis.

Identification of 4-Demethylsterols from Suspension Cultured Cells of Marchantia polymorpha L. (Marchantia polymopha(우산이끼) 현택배양 세포내으 4-methylster이들의 동정)

  • Five kinds of 4-demethylsterol were isolated from suspension cultured cells of a liverworth, Marchantia polymorpha. Four 4-demethylsterols among them were analyzed by a capillary gas chromatography-massspectrometry and 500 MHz 1H-NMR, and characterized to be avenasterol, 24-methylene-cholesterol, stigmasterol, and campesterol. And the fifth 4-demethylsterol was characterized to be sitosterol by a capillary gas chromatography-mass spectrometry. The concentration of the 4-demethylsterols in the cell decreased in order of avenasterol>stigmasterol>24-methylene-cholesterol>campesterol>sitosterol. When carbon skeleton and the oxidation state was compared with those of brassinosteroids, these 4-demethylsterols may be potent biosynthetic procusors of brassinosteroids in the cells.
