Shoot induction and regeneration using internodal transverse thin cell layer culture in Sesamum indicum L.

  • An efficient protocol for shoot regeneration was developed for sesame (Sesamum indicum L.) internodes using the transverse thin cell layer (tTCL) culture method. The frequency of shoot regeneration and the number of adventitious buds produced from regenerated shoots depend significantly on explant age, thickness of the tTCL sections, and the phytohormones supplemented to the culture medium. A combination of 6-benzyladenine (2.0 $mg\;l^{-1}$) and a-naphthaleneacetic acid (0.5 $mg\;l^{-1}$) was found to be the best phytohormone combination for shoot bud induction, with the maximum number of shoots obtained when the tTCL sections were 0.5-1.0 mm thick and derived from 4- to 6-week-old seedlings of sesame. Well-developed shoots were rooted on MS medium without phytohormones, and 80% of the regenerated plantlets were successfully established in soil.

Large-scale Culture of Plant Cell and Tissue by Bioreactor System

  • Large-scale cultures of plant cell, tissue, and organ have been achieved by using BTBB. When different sized BTBBs (5 L, 20 L, 100 L, 300 L, and 500 L) were tested for the culture of yew cells (Taxus cuspidata Sieb. et Zucc.), cell growth increment reached to 94.5% in SCV after 24 days of culture with 30% of inoculation cell density. However, there were some variations in the production of taxol and its derivatives among the BTBBs of different size. Approximate 4 ㎎/l of taxol and 84 ㎎/l of total taxanes were obtained by using a 500L BTBB after 6 weeks of culture. With a 20L BTBB, about 20,000 cuttings of virus-free potatoes (cv. Dejima) could be obtained by inoculating 128 explants and maintaining 8 weeks under 16 hr light illumination. The frequency of ex vitro rooting of the cuttings revealed as more than 99% under 30% shade. By incorporating two-stage culture process consisting of multiple bulblet formation in solid medium and bulblet development in liquid medium, mass propagation of lily through bioreactor seemed to be possible. In the case of 'Marcopolo', the growth of mini-bulblets in BTBB was nearly 10 folds faster than that of the solid medium. Time course study revealed that maximum MAR yield of ginseng (Panax ginseng C. A. Meyer) in a 5 L and 20 L BTBB after 8 weeks of culture was 500 g and 2.2 ㎏, respectively. By cutting the MAR once and/or twice during the culture, the yield of root biomass could be increased more than 50% in fresh weight at the time of harvest. With initial inoculum of 500 g of sliced MAR in a 500 L BTBB, 74.8 ㎏ of adventitious root mass was obtained after 8 weeks of culture. The average content of total ginseng saponin obtained from small-scale and/or pilotscale BTBBs was approximately 1% per gram dry weight. Based on our results, we suggest that large-scale cultures of plant cell, tissue, and organ using BTBB system should be quite a feasible approach when compared with conventional method of tissue culture.

  • Single cells obtained from suspension culture of mature embryo-derived callus in wheat(Triticum aestivum L. cv Jang Kwang) were cultured to regenrated into the plantlet. Cell clusters and embryogenic calluses were efficiently developed from when the single cells clutured on the MS medium supplemented with 10${\mu}{\textrm}{m}$ 2,4-D. Upon transfer to hormone-free MS medium containing 10 mg/I AgNO3, embryogenic calluses gave rise to shoots, probably through somatic embryogenesis.

Effects of Phosphate, Precursor and Exogenous Berberine on the Production of Alkaloid in Plant Cell Cultures

  • The effects of phosphate concentration in the medium, feeding of biosynthetic precursor, and the addition of exogenous berberine on cell growth and berberine production were studied in cell suspension cultures of Thalictrum rugosum. The depletion of phosphate in the medium enhanced the specific productivity up to twofold with significant release of berberine into the medium. Extracellular berberine was 19% of the total in the culture without phosphate while it was 2-5% of total berberine in the culture with even low amounts of phosphate. Precursor feeding was not effective in enhancing alkaloid formation. Initial presence of exogenous berberine did not have much effect on cell growth and alkaloid production. It was found that the cells have the capacity to take up large quantities of berberine. When $500{\;}mg{\cdot}l^{-1}$ of berberine was added exogenously at the beginning, 81% of total berberine was found in the cells.

  • Corydalis remota Fish. ex Max. (Papaveraceae) is a well known medicinal plant being used as analgesics or anticonvulsive in oriental medicine. As the alkaloid content is known to vary depending on the environmental factors, the technology of plant tissue culture can be adopted as source of Corydalis-alkaloids. The present study describes an establishment of tissue cultures of Corydalis which produce alkaloids consistently. Callus were induced from immature seeds of Corydalis remota by placing the seeds on MS static media containing NAA(0.25, 1.0 and 4.0 mg/l, respectively). The combined treatment of NAA(1.0 mg/l) with cytokinin(BAP 0.5 mg/l) improved the induction of callus. TLC scanning data followed by sequential extraction and purification revealed that the induced callus contains a significant amount of alkaloids. Cell suspension cultures were established by transferring the induced callus into the liquid media with the same condition of plant growth regulators as the callus culture.

Effect of Nitrogen Source on the Cell Growth and Production of Secondary Metabolites in Bioreactor Cultures of Eleutherococcus senticosus (가시오갈피 생물반응기 배양에서 질소급원이 세포생장과 이차대사 생산에 미치는 영향)

  • The effects of inorganic nitrogen sources such as KNO$_3$ and NH$_4$ NO$_3$ on cell growth and production of chlorogenic acid and eleutheroside E derivative were investigated in 5L bioreactor cultures of Eleutherococcus senticosus. The cell growth in the 1/2MS medium containing 15mMKNO$_{3}$. The fresh weight of cells harvested from bioreactor was affected by the concentration ratio of NO$_3$$^{[-10]}$ and NH$_4$$^{+}$ in culture medium. At the viewpoint of secondary metabolite production, the production of chlorogenic acid was affected by the concentration of NH$_4$$^{+}$ in the culture medium, but not by the total concentration of nitrogen sources in the culture medium. Futhermore, eleutheroside E derivative production was also affected by the concentration ratio of NO$_3$$^{[-10]}$ and NH$_4$$^{+}$ in the culture medium. Base on those results, it is suggested that cell growth and production of secondary metabolite(chlorogenic acid and eleutheroside E derivative) could be manipulated by controlling the total concentration of nitrogen sources and the concentration ratio of NO$_3$$^{[-10]}$ and NH$_4$$^{+}$ in the culture medium. medium.

Effects of Sucrose Concentration on the Production of hGM-CSF in Transgenic Plant Cell Suspension Culture (형질전환된 식물세포에서 Sucrose 농도가 hGM-CSF 생산에 및 미치는 영향)

  • The effects of sucrose concentration on the secretion of hGM-CSF, total protein and protease into the medium were investigated in transgenic tobacco cells. The dry cell weight (11.22 g/L), hGM-CSF (181.53 $\mu\textrm{g}$/L) and total protein (66.8 mg/L) were detected as highest at 30 g/L sucrose and protease activity (2660 U/L) was highest at 120 g/L sucrose after 5-day culture. However after 10-day culture, the maximum dry cell weight (28.36 g/L) was found at 60 g/L sucrose while the maximum hGM-CSF (95 $\mu\textrm{g}$/L) was appeared at 150 g/L sucrose. The total protein and protease activity was secreted as 52.28mg/L and 3430 U/L, respectively in the same culture.

  • Plant cell culture can be divide into two classes non-organic culture and organic culture. Non-organic culture such as suspension culture has many researches, however organic culture about recombinant protein production has little researches. Recombinant protein produced through organ culture is quite stable and it can make proteins by itself without any grow regulators. Therefore organ culture is much easier than other methods. In this research, we used transformed tobacco seed. At first we germinated the seed then separated stems and leaves from the grown plant. And raised in liquid medium by in vitro vegetative reproduction. Continuing most suitable conditions, we compared the Quantities of recombinant protein from intra cellular with from extra cellular. And adding some permeabilizing agents (Pluronic F-68, Triton X-100, DMSO, PEG8000), we increased the productivity of the recombinant protein.

Selection of Cell Source and the Effect of pH and MS Macronutrients on Biomass Production in Cell Cultures of Tongkat Ali (Eurycoma longifolia Jack)

  • Callus and cell suspension cultures of Eurycoma longifolia Jack were initiated from leaves of different trees. The leaf explant of tree Eu9 produced the most calli and also induced high cell biomass in the cell suspension culture. Optimum production of cell biomass could be initiated in proliferating culture medium with a pH of 5.75 prior to autoclaving. The effects of macronutrient inorganic salts of Murashige and Skoog (MS) liquid medium supplemented with X on production of cell biomass of Eurycoma longifolia were also investigated. The highest cell biomass was produced in MS medium containing macronutrients of $21\;mM\;NH_4NO_3,\;12.25\;mM\;KNO_3,\;3.00\;mM\;CaCl_2.2H_2O,\;0.575\;mM\;MgSO_4.7H_2O$, and $1.83\;mM\;KH_2PO_4$. A new medium labeled as TAM was formulated for the production of Eurycoma longifolia cell biomass in the cell suspension culture.
