  • The present study examines the inhibitow effect of progesterone (P) on luteinizing hormone $(LH)\beta$ subunit gene expression in anterior pituitary of ovariectomized, estradiol-treated adult rats. A single injection of P (1mg) further decreased the estradiol-Induced decrease in $LH\beta$ mRNA levels in ovariectomTzed rats in a time-dependent manner. p suppressed UIP mRNA levels at lower doses (0.1 and 1mg), but increased $LH\beta$ mRNA levels 81 a high dose (toms). The inhibitor action of P on $Uf\beta$ mRNA was restored when Ru486, a P receptor antagonist, was administered 1h before P treatment. These data clearly indicate that P inhibits gene expression of $LH\beta$ in the rift pituitary in a swersic manner with estrogen.

개의 뇌하수체 의존성 부신 피질 기능 항진증에서의 selegiline 적용 (Use of selegiline in 3 Cases of Canine Pituitary-dependent Hyperadrenocorticism)

  • Pituitary-dependent hyperadrenocorticism (PDH) was diagnosed with history taking, physical examination, complete blood count, serum chemistry profiles, abdominal radiology, ultrasonography and adrenal function tests in 3 dogs. Their clinical signs were polyuria, polydipsia, polyphagia, bilateral symmetrical truncal alopecia and secondary infection in skin or urinary tract. Especially one dog showed severe clinical signs such as calcinosis cutis and delayed wound healing. These 3 dogs were diagnosed as PDH, and treated with selegiline 1-2 mg/kg /ay sid PO. 2 dogs with clinical signs of PDH were disappeared and improved, but 1 dog with severe illness progressed gradually despite of selegiline and mitotane application, and eventually died.

  • Ginseng effectively regulates the immune response and the hormonal changes due to stress, thus maintaining homeostasis. In addition to suppressing the occurrence of psychological diseases such as anxiety and depression, ginseng also prevents stress-associated physiological diseases. Recent findings have revealed that ginseng is involved in adjusting the hypothalamic-pituitary-adrenal axis and controlling hormones, thus producing beneficial effects on the heart and brain, and in cases of bone diseases, as well as alleviating erectile dysfunction. Recent studies have highlighted the potential use of ginseng in the prevention and treatment of chronic inflammatory diseases such as diabetes, rheumatoid arthritis, and allergic asthma. However, the mechanism underlying the effects of ginseng on these stress-related diseases has not been completely established. In this review, we focus on the disease pathways caused by stress in order to determine how ginseng acts to improve health. Central to our discussion is how this effective and stable therapeutic agent alleviates the anxiety and depression caused by stress and ameliorates inflammatory diseases.

Neuroprotective roles of pituitary adenylate cyclase-activating polypeptide in neurodegenerative diseases

  • Pituitary adenylate cyclase-activating polypeptide (PACAP) is a pleiotropic bioactive peptide that was first isolated from an ovine hypothalamus in 1989. PACAP belongs to the secretin/glucagon/vasoactive intestinal polypeptide (VIP) superfamily. PACAP is widely distributed in the central and peripheral nervous systems and acts as a neurotransmitter, neuromodulator, and neurotrophic factor via three major receptors (PAC1, VPAC1, and VPAC2). Recent studies have shown a neuroprotective role of PACAP using in vitro and in vivo models. In this review, we briefly summarize the current findings on the neurotrophic and neuroprotective effects of PACAP in different brain injury models, such as cerebral ischemia, Parkinson's disease (PD), and Alzheimer's disease (AD). This review will provide information for the future development of therapeutic strategies in treatment of these neurodegenerative diseases.

뇌하수체 선종의 방사선 치료 결과 (Radiation Therapy of Pituitary Tumors)

  • Radiation treatment results were analyzed in a retrospective analysis of 47 patients with pituitary adenoma treated with radiation alone or combined with surgery from 1974 through 1987 at the Department of Therapeutic Radiology of Kyung Hee University. The 5-year overall survival rates for all patients was $80.4\%$ Radiation therapy was effective for improving visual symptoms and headache, but could not normalize amenorrhea and galactorrhea. There was no difference of survival rate between radiation alone and combination with surgery. Prognostic factors such as age, sex, disease type, visual field, headache and surgical treatment were statistically no significant in survival rates of these patients.

  • Encephaloceles are known as rare craniofacial developmental anomalies which are herniation of cranial contents through a skull defect. Sphenoidal meningoencephalocele represents an important cause of reversible visual loss. We report a rare case of pituitary adenoma accompanying sphenoidal meningoencephalocele with clival extension. Although the definitive diagnosis of cystic lesions in the sellar region before surgery is difficult, accurate diagnosis of these lesions is important to determine the type of treatment and predict prognostic outcome.

Pituitary Pars Intermedia Dysfunction with Laminitis in a Horse

  • Pituitary pars intermedia dysfunction (PPID), often referred to as equine Cushing's disease, is a common endocrine disorder often diagnosed in older horses. A 13-year-old 460 kg Warmblood gelding showed clinical signs suggestive of PPID, including hypertrichosis, fat redistribution, polyuria and polydipsia (PU/PD), and weight loss. Physical examination, complete blood cell count, and serum chemistry results were normal. However, dexamethasone suppression and plasma adrenocorticotropic hormone (ACTH) level tests confirmed PPID. Three months after the confirmed diagnosis, the horse was referred again with symptoms of laminitis. Radiography and venography were performed to evaluate the laminitis severity level. However, the foot condition continued to worsen, and the horse was eventually euthanized. The purpose of this case report is to describe clinical signs and diagnosis of PPID with laminitis.

Modulation of Pituitary Somatostatin Receptor Subtype (sst1-5) mRNA Levels by Growth Hormone (GH)-Releasing Hormone in Purified Somatotropes

  • We have previously reported that expression of the somatostatin receptor subtypes, sst1-5, is differentially regulated by growth hormone (GH)-releasing hormone (GHRH) and forskolin (FSK), in vitro. GHRH binds to membrane receptors selectively located on pituitary somatotropes, activates adenylyl cyclase (AC) and increases sst1 and sst2 and decreases sst5 mRNA levels, without significantly altering the expression of sst3 and sst4. In contrast FSK directly activates AC in all pituitary cell types and increases sst1 and sst2 mRNA levels and decreases sst3, sst4 and sst5 expression. Two explanations could account for these differential effects: 1) GHRH inhibits sst3 and sst4 expression in somatotropes, but this inhibitory effect is masked by expression of these receptors in unresponsive pituitary cell types, and 2) FSK inhibits sst3 and sst4 expression levels in pituitary cell types other than somatotropes. To differentiate between these two possibilities, somatotropes were sequentially labeled with monkey anti-rat GH antiserum, biotinylated goat anti-human IgG, and streptavidin-PE and subsequently purified by fluorescent-activated cell sorting (FACS). The resultant cell population consisted of 95% somatotropes, as determined by GH immunohistochemistry using a primary GH antiserum different from that used for FACS sorting. Purified somatotropes were cultured for 3 days and treated for 4 h with vehicle, GHRH (10 nM) or FSK ($10{\mu}M$). Total RNA was isolated by column extraction and specific receptor mRNA levels were determined by semi-quantitative multiplex RT-PCR. Under basal conditions, the relative expression levels of the various somatostatin receptor subtypes were sst2>sst5>sst3=sst1> sst4. GHRH treatment increased sst1 and sst2 mRNA levels and decreased sst3, sst4 and sst5 mRNA levels in purified somatotropes, comparable to the effects of FSK on purified somatotropes and mixed pituitary cell cultures. Taken together, these results demonstrate that GHRH acutely modulates the expression of all somatostatin receptor subtypes within GH-producing cells and its actions are likely mediated by activation of AC.

  • 어류의 번식은 뇌에서 분비되는 다양한 신경호르몬과 뇌하수체에서 분비되는 생식소 자극 호르몬에 의해 조절된다. 극동산 뱀장어(Anguilla japonica)의 번식도 이 호르몬들의 작용에 의해 조절되지만 성 성숙 시 신경호르몬이 뇌하수체 호르몬을 조절하는 방법은 완전히 밝혀지지 않았다. 이전 연구에 의하면 progesterone (P4), melatonin 및 serotonin (5-HT) 등과 같은 신경호르몬이 일부 어류의 번식 과정 조절에 관여하는 것으로 밝혀졌다. 본 연구에서는 뱀장어의 뇌하수체를 초대 배양하였고, 안정화된 뇌하수체 세포에 P4, 17β-estradiol (E2), melatonin 및 5-HT를 처리하였다. 이후 처리된 호르몬의 작용이 뇌하수체 세포에서 번식 관련 호르몬인 FSHβ, LHβ, GH 및 SL mRNA 발현에 어떤 영향을 미치는지 조사하였다. 본 연구를 수행한 결과, P4는 뇌하수체 세포에서 FSHβ와 LHβ 발현을 증가시켰고, melatonin은 FSHβ와 LHβ 뿐만 아니라 GH와 SL의 발현을 증가시켰다. 하지만 5-HT는 이 유전자의 mRNA 발현에 유의한 영향을 미치지 않았다. 이상의 결과는 P4 또는 melatonin이 뱀장어의 초기 성 성숙에 중요한 역할을 할 수 있음을 의미한다.

  • Computation of three dimensional dose distribution using CT image and RT plan was applied to a case of pituitary adenoma. Algorithm was based on two dimensional Tissue Maximun Ratio model extended to the third dimension. The resulting isodose curve of transeverse, coronal and sagittal section was demonstrated. This RT plan allows computation of dose distribution in any arbitarily defined plane in addition to conventional cross sectional view.
