Complete Genome Sequencing and Infectious cDNA Clone Construction of Soybean Mosaic Virus Isolated from Shanxi

  • Soybean mosaic virus (SMV) is the predominant viral pathogen that affects the yield and quality of soybean. The natural host range for SMV is very narrow, and generally limited to Leguminosae. However, we found that SMV can naturally infect Pinellia ternata and Atractylodes macrocephala. In order to clarify the molecular mechanisms underlying the cross-family infection of SMV, we used double-stranded RNA extraction, rapid amplification of cDNA ends polymerase chain reaction and Gibson assembly techniques to carry out SMV full-length genome amplification from susceptible soybeans and constructed an infectious cDNA clone for SMV. The genome of the SMV Shanxi isolate (SMV-SX) consists of 9,587 nt and encodes a polyprotein consisting of 3,067 aa. SMV-SX and SMV-XFQ008 had the highest nucleotide and amino acid sequence identities of 97.03% and 98.50%, respectively. A phylogenetic tree indicated that SMV-SX and SMV-XFQ018 were clustered together, sharing the closest relationship. We then constructed a pSMV-SX infectious cDNA clone by Gibson assembly technology and used this clone to inoculate soybean and Ailanthus altissima; the symptoms of these hosts were similar to those caused by the virus isolated from natural infected plant tissue. This method of construction not only makes up for the time-consuming and laborious defect of traditional methods used to construct infectious cDNA clones, but also avoids the toxicity of the Potyvirus special sequence to Escherichia coli, thus providing a useful cloning strategy for the construction of infectious cDNA clones for other viruses and laying down a foundation for the further investigation of SMV cross-family infection mechanisms.

Status of reports of adverse events related to botanical herbal medicines with toxic precautions officially managed by Korean government: A descriptive analysis from WHO VigiAccess (WHO VigiAccess에 수록된 식물성 독성주의한약재 관련 이상사례 보고 현황)

  • Objectives: This study was aimed to review the global status of adverse event (AE) reports and the characteristics of the reported AEs of plants managed as herbal medicines (HMs) with toxic precautions in Korea. Methods: This is a cross-sectional quantitative study that analyzed information available through VigiAccess, a website that provides summarized statistical information from the WHO's global AE database to the public. VigiAccess was searched in 8 Jan, 2024. Information on the total number of reports, number of reports by year and continent, and the age and gender of patients were obtained, and the types of frequently reported AEs were also reviewed. Results: Data on the status of report submissions were obtained for a total of 9 HMs including Aconitum ciliare, Aconitum carmichaeli, Arisaema japonicum, Pinellia ternata, Euphorbiae Lathyridis, Croton tiglium, Strychni Ignatii, Strychnons nux-vomica, and Linum usitatissimum. The number of reports per HM was from 1 to 137. The most commonly reported type of AEs were gastrointestinal disorders in most of the HMs, followed by neurological disorders. Serious adverse events were reported only in Strychni Ignatii, Strychnons nux-vomica, and Linum usitatissimum, including one case of death. Conclusions: This study shows the status of reported AEs of botanicals considered as HMs with toxic precautions in Korea based on real world data. However, when interpreting the findings of this study, readers should consider the significant limitations of this study mainly because of the characteristics of the data source.

Effects of Several Medicinal Plants Extract on Survival Rate, Chlorophyll Contents and Photosynthetic Electron Transport Activity of Liverwort Photoautotrophic Cultured Cell (약용식물 추출액이 우산이끼 자가관양배양세포의 생존율, 엽록소함량 및 광합성전자전달 활성에 미치는 영향)

  • The effects of allelochemicals from medicinal plants have been studied as photo-synthetic inhibitor for photoautotrophic(PA) cultured cells. The extracts from 9 plant species were used for measuring the physiological effects on the liverwort cultured cell in following areas; germination inhibition, chlorophyll contents, hill activity, cell viability, photosynthetic oxygen evolution,and protein pattern changes on SDS PAGE. Germination inhibitions were detected in all plant after treating with 10% extract. Especially, treatment with 10% extract from Pulsatilla koreana and Aconitum carmichael inhibited germinations completely. Chlorophyll fornation was inhibited completely by treating PA cells with extract of Pulsatilla koreana, whose effect was similar to that of DCMU 10-3M, inhibitor for photosynthetic electron trans-fer. The treatment with extract from Pulsatilla koreana on PA cell showed the highest hill activity and the lowest cell viability among extracts studied. Oxygen releasing has been decreased down to 14-77% after treating with extracts from Pinellia ternata, Araliacont inentaila, Pulsatilla koreana and Vitex rotundifolia. Especially, 60$\mu$l of Pulsatilla koreana extract into 2ml mixture of PA cell inhibit-ed oxygen release up to 50%. Protein bands on SDS-PAGE, 14kD, 31kD, 41kD, 53kD, and 73kD, were not detected after treating Pulsatilla koreana extract on PA cells.

Plant Regeneration and in vitro Tuber Enlargement from Callus in Pinellia ternata(Thunb.) Breit (반하(半夏) 캘러스로부터 식물체(植物體) 재생(再生)과 기내(器內) 괴경(塊莖) 생장(生長) 유도(誘導))

  • This study carried out to induce plant regeneration and callus formation from leaflet in MS medium with 2.4-D, NAL and IAA for in vitro growth of tuber, Kinetin and BA were used for plant regeneration. $NH_4NO_3$ and $KNO_3$ as a nitrogen source and $(NH_4)_2SO_4$, as a sulfate source were tested for in vitro growth of tuber. The resutls ars as fellows : 1. For the callus formation from leaflet and differentiation potency of organ, 2.4-D was more effective than IAA in MS medium under $26^{\circ}C$ and light condition of 8 hours a day. 2. For the plant regeneration from callus, MS medium with $2.0\;mg/{\ell}$ BA was most effective under $26^{\circ}C$ and light condition of $16{\sim}24$ hours a day. 3. For the in vitro growth of tuber, $KNO_3$ by $3.0g/{\ell}$ in MS medium was effective. This condition enhanced the growth of tuber 2.5 times compared with that in MS medium with $2.0\;mg/{\ell}$ BA.

In Fluence Chemicals From Artemisis argyi on the Growth of Selected Species of Plants and Microorganisms (황해쑥에 함유된 화학물질이 다른 식물과 미생물의 생장에 미치는 영향)

  • To investigate phytotoxic substances in Artemisia argyi, the donor plant, and their biological activities, seed germination and seedling growth of receptor plants such as Arundinella hirta, Echinochloa crus-galli, Rumex crispus and Lactuca sativa were examined at different concentrations of aqueous extracts of the donor plant. Germination of four receptor species was inhibited by the extracts, while seedling growth was decreased to a lesser degree than in the germintion test. Germination, seedling growth and dry weight growth of Achyranthes japonica grown in pot were proportionally inhibited by the extracts. Volatile substances emitted from A, argi plant caused slight inhibition in the germination and seedling growth of the receptor species. Essential oil of the plant extracted by Karlsruker's apparatus inhibited growth of microorganisms and callus growth of Pinellia ternata and Oryza sativa. The GC /MS method was employed for analysis and identification of allelochemicals from A. argyi leaves. Sixty-one chemical substances such as a-pinene, camphene, 1. 8-cineol, etc. were identified from essential oil of A. argyi. The results of this experiment on seed germination, seedling growth, microorganism culture and tissue culture indicated that naturally occurring chemical substances from A. argyi would be responsible for the growth inhibition of plants studied.

Translational Study for "Treatise on Bundon Disease in Chapter 8 of the Essence of the Synopsis of the Golden Chamber" ("금궤요략심전.분돈기병맥증치제팔(奔豚氣病脈證治第八)"에 대한 번역연구)

  • After translating the contents of "Treatise on Bundon Disease in Chapter 8 of the Essence of the Synopsis of the Golden Chamber" and it was compared with the contents of "The Lecture of Synopsis of Prescriptions of the Golden Chamber" and speculated to review the characteristics of the comments of Ui(尤怡) and the following conclusions were made. As the causes of bundon(奔豚) disease, which means gas rushing like a running piggy from the lower abdomen to the throat, they could be categorized into the cases belongs to miscellaneous diseases (雜病) and into the cases of suffering from cold diseases(傷寒). If it was diagnosed as miscellaneous disease, terror and fear(驚恐) could be regarded as its cause and if it was diagnosed as cold disease, insufficiency of heart Gi(心氣) due to the excessive sweating and the invasion of exterior coldness could be regarded as its cause. As the pathogenesis of this disease, the pathogenic factors of liver and kidney could be found. Since the liver and Soyang meridian(少陽經) are subjected to the exterior and interior relation in case of the pathogenic factor of liver, alternate attacks of chills and fever(寒熱往來) could be characteristic for the disease. If this disease was caused by the pathogenic factor of kidney, there are two cases. One is the palpitation under the umbilicus(欲作奔豚: 臍下動悸) developed by the lack of heart Gi(心氣) and the other is gas rushing like a running piggy from the lower abdomen to the throat which could be developed by the exterior cold disease which invade into the interior and reach into the kidney when the heart Gi(心氣) was insufficient. In its treatment, bundon disease caused by the pathogenic factor of liver could be treated with Bundontang(奔豚湯), and the understanding of Ui(尤怡) on Pinellia ternata Breitenbach(半夏) having the effect of perspiration in the prescription is quite unique concept. In case of bundon disease caused by the pathogenic factor of kidney, treatment is focused to treat exterior coldness by applying moxibustion(灸) and Gyejigagyetang(桂技加桂湯), and in case of the palpitation under the umbilicus(欲作奔豚: 臍下動悸), its pathogenic factor of kidney is controlled with Cinnamomum louerii Nees(桂技), Polia cocos Walf and the spleen Gi(脾氣) is cared by using Glycyrrhiza uralensis Fischer et. De Candolle(甘草) and Zizyphus jujuba Mill(大棗). Especially, Ui(尤怡) emphasized Cinnamomum louerii Nees(桂技) as the medication to treat the pathogenic factor of kidney.

  • The study was conducted to screen paraquat-tolerant plant species among crops and weeds, using the response of plant like leaf disc discoloration, visual injury and dry weight in the presence of paraquat. Mechanism of paraquat-tolerance was investigated in strains of soybean through evaluating activities of superoxide dismutase and peroxidase and the multiplication of callus derived from soybean cotyledon. In crops, Kwanggyo has been selected as a paraquat-tolerant variety among soybean cultivars tested, and Hood as a susceptible one. In weeds, Polygonum aviculare, Chenopodium album and Pinellia ternata were evaluated as the paraquat resistant species, providing the possibility for the donor plant species for paraquat resistance. Activity of superoxide dismutase known to detoxify paraquat was markedly greater in Kwanggyo, a paraquat-tolerant cultivar than in Hood, a susceptible one. In addition, the similar response like superoxide dismutase was observed in peroxidese activity. The greater inhibition of callus multiplication was determined in Hood, a susceptible one than a tolerant one, Kwnggyo. Based on all the informations, it is strongly proposed that paraquat tolerance in soybean is due to destruction of $O_2^-$ by elevated concentration of superoxide dismutase in the tolerant cultivar.

Effect of Hot Water Extracts from Medicinal Plants on the Mutagenicity of Indirect Mutagens (간접변이원의 돌연변이원성에 대한 생약재 열수 추출물의 효과)

  • For screening antimutagenic effects, the effects of 95 medicinal plants on the mutagenicity of aflatoxin $B_1$ $(AFB_1)$ and benzo(a)pyrene [B(a)P] were investigated using the SOS chromotest with Escherichia coli PQ37. The mutagenicity induced by $AFB_1$ or B(a)P was reduced over 26% by 2 kinds and 8 kinds of medicinal plant, respectively. Eight plants (Bupleurum falcatum, Corydalis ternata, Gasfrodia elata, Ostericum koreanum, Pinellia ternatia, Poncirus trifoliata, Prunus armeniaca and Rehmannia glutinosa) were also shown to have inhibitory effects on both $AFB_1$ and B(a)P. The mutagenicity induced by $AFB_1$ or B(a)P was increased over 20% by 46 kinds and 2 kinds, respectively, and 8 medicinal plants (Chrysanthemum indicum, Cinnamomum cassia, Cyperus rotundus, Morus bombycis, Patrinia scabiosaefolia, Petasites japonicus, Polygonum multiflorium, Thyja orientalis) increased significantly the mutagenicity of both mutagens. However the 8 plants themself did not show the mutagenicity in SOS Chromotest with S-9 mix alone. This result suggests that the above 8 plants may have the co-mutagenic activities. In two bacterial mutation system, SOS Chromotest and Ames test, the mutagenic or antimutagenic activities of some medicinal plants wire similar except Ostricum koreanum, Eugenia caryophyllata and Scutellaria baicalensis.

Effects of OMC-2010 constituents on cytokine productions in mouse spleen cells (OMC-2010 구성약재가 마우스의 비장세포 cytokine 생성에 미치는 영향)

  • Objective : We recently reported that OMC-2010 has an immuno-modulatory effects via inhibiting tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-5. However, we did not find out which constituents play an important role in immuno-modulatory effect of OMC-2010. Thus, this study was performed to estimate the effects of constituents of OMC-2010 on cytokine production in mouse spleen cells, then ultimately reach to find out effective constituents regulating splenic cytokine production. Methods : Mouse spleen cells were pre-treated with water and ethanol extract of constituents of OMC-2010 such as Rehmannia glutinosa (RG), Pinellia ternata (PT), Citrus unshiu Markovich (CUM), Glycyrrhiza uralensis (GU), Platycodon grandiflorum (PG), Schisandra chinensis (SC). After 1 h, the cells were stimulated with lipopolysaccharide (LPS, 1 ${\mu}g/ml$) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokine productions. Results : The water extract of RG extract significantly inhibited the LPS-induced inTNF-${\alpha}$ and IL-5 mRNA expressions, but the water extract of PT, CUM, GU, PG, and SC did not. The ethanol extract of RG, PT, and SC significantly inhibited the LPS-induced TNF-${\alpha}$, and IL-5 mRNA expressions, but the ethanol extract of CUM, GU, and PG did not. Conclusions : Theses results could suggest that the water extract of RG and the ethanol extract of RG, PT, and SC inhibited the expression of TNF-${\alpha}$ and IL-5, which means that the possible candidate of OMC-2010 water extract's action might be RG, and ethanol extract's action might be RG, PR, and SC.

Effects of OMC-2010 Constituents Extract on the Ovalbumin-Induced Allergic Asthma in Mice (OMC-2010 구성약재 배합추출물 투여가 Ovalbumin으로 유도한 마우스 알레르기성 기관지 천식에 미치는 영향)

  • Objectives : We recently have reported that constituents of OMC-2010 have an immuno-modulatory effects via inhibiting tumor necrosis factor (TNF)-alpha and interleukin (IL)-5. In this study, based on previous data, we investigated the effects of combinations with each OMC constituents on splenocyte cytotoxicity, cytokine productions, and ovalbumin (OVA) induced experimental allergic asthma. Methods : Mouse splenocytes were pre-treated with ethanol extract of constituents of Rehmannia glutinosa (RG), Pinellia ternata (PT), Schisandra chinensis (SC). We made 4 combinations using RG, PT, and SC (A;1:1:1, B;2:1:1, C;1:2:1, D;1:1:2). The cells were pretreated with A, B, C, or D for 1 h, then stimulated with lipopolysaccharide (LPS, $1{\mu}g/ml$) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokine productions. Then using effective combination from RG, PR and SC, we administrated the combination orally, then challenged with OVA to induce asthma. Then we analyzed the airway hyper-reactivity (AHR), lung histology and lung TNF-${\alpha}$ and IL-5 mRNA. Results : A. B. C. and D did not showed significant cytotoxicity on splenocytes. Pre-treatment of A inhibited the expression of TNF-${\alpha}$ and IL-5 significantly, but not B, C, and D. In experimental asthma, administration of A significantly inhibited the increase of AHR, lung damage, TNF-${\alpha}$ and IL-5 expression. Conclusions : Theses results could suggest that inhibitory effects of the ideal combination with RG, PT and SC (1:1:1) could be applied to treatment of asthma and study of asthma mechanisms.
