Induction of Ovulation by LHRH-a and Pimozide in the Bullhead, Pseudobagrus fulvidraco (LHRH-a 와 pimozide에 의한 동자개의 배란유도)

  • Effects of intraperitoneal administration of LHRH-a (luteinizing hormone releasing hormone analogue), pimozide and HCG (human chorionic gonadortropin) were studied on ovulation in the bullhead, Pseudobagrus. fulvidraco. Ovulatin was induced in 71.4, 80 and 100% by injection of HCG 5,000, 10,000 and 20,000 IU/kg of body weight, respectively. Also, the majority of fish injected with LHRH-a in the presence or absence of pimozide were ovulated. The ovulating rates in fish injected with LHRH-a alone showed 50, 75 and 100% at a dose 200, 300 and $400^{\mu}$g/kg, respectively. Pimozide enhanced the activity of LHRH-a at $300^{\mu}$g/kg plus pimozide at 1 mg/kg. Pimozide alone was ineffective in inducing ovulation of P. fulvidraco. There were no significant differences in gonadosomatic index (GSI) and pseudo-GSI values among the groups treated with HCG, LHRH-a and LHRH-a plus pimozide (P>0.05). The latency time groups treated with HCG was 17~22 hours, and the group injected with LHRH-a alone and/or in combination with pimozide was ovulated within 23~29 hours.

  • Ovulation of maturing female yellow puffer, Takifugu obscrus, was induced by using single injection of human chorionic gonadotropin (HCG) or gonadotropin releasing hormone-analogue (GnRH-A) $des-Gly^{10}[D-Ala^6]$ GnRH-ethylamide plus pimozide. The response was evaluated using the fertilization and embryo-formation rate after insemination and the gonadotropin (GTH) level in blood plasma using radioimmunoassay. In the fertilization and embryo-formation, maximal effects were recorded by using 1,000 IU/kg HCG or $10\;{mu}g/kg$ GnRH-A plus 5 mg/kr pimozide. Pimozide (1, 5 mg/kg) or GnRH-A treatment alone was not effective in elevation of GTH level, however combinations of these treatments were particularly effective. Injection of dopamine blocked the rapid elevation of plasma GTH levels of blood. These data suggest that yellow puffer secrete GnRH and gonadotropin-releasing-inhibiting factor during the spawning or the other period.

A Case of Recurrent Ventricular Tachycardia after Pimozide and Haloperidol Overdose (Pimozide와 Haloperidol 과량 복용 후 반복적으로 발생한 심실 빈맥 1례)

  • Pimozide and haloperidol are typical antipsychotics. They share a similarity in pharmacotherapeutic and adverse effect profiles. Cardiovascular effects may be seen as alterations in heart rate, blood pressure, and cardiac conduction. Conduction disturbances may occur ranging from asymptomatic prolongation of the QT interval to fatal ventricular arrhythmia. So in the case of anti psychotics overdose, the patient must be carefully monitored by continuous electrocardiography (ECG). We experienced a 34-year-old woman of schizophrenia with recurrent ventricular tachycardia after pimozide and haloperidol overdose. Initially she was slightly drowsy, however her ECG showed normal sinus rhythm. After 6 hours on emergency department entrance, her ECG monitoring showed ventricular tachycardia and we successfully defibrillated. There were five times events of ventricular arrhythmia during the in-hospital stay. She was discharged 5 days later without any other complications.

Utilization of Hypothalamic Hormones for Maturational Induction in Seawater Chum Salmon, Oncorhynchus keta (해면연어의 성숙을 유도하기 위한 시상하부호르몬의 이용)

  • Freshwater-adapted maturing chum salmon (Oncorhynchus keta) involuntarily captured by stationary nets in Yang-yang seashore areas were transferred to freshwater in an outdoor raceway tank at Yeongdong Inland Fisheries Research Institute, NFRDI, Yang-yang, Gangwon, Korea and kept over 1 day until the start of the experiments. The freshwater-adapted females were single-injected intraperitoneally with gonadotropin-releasing hormone analogue, (GnRH-a: $70\;{\mu}g/kg$ body weight, BW) alone or combined with a dopamine receptor antagonist, pimozide($700\;{\mu}g/kg$ BW). Although gonadosomatic indices [GSI, (gonad weight/BW)${\times}100$] did not show significant changes in both 2004 and 2005, GSI of GnRH-a-injected fish during the 2005 trial slightly increased on the 5th and 7th days post-injection compared to those of vehicle treated fish. Hepatosomatic indices [HSI, (liver weight/BW)${\times}100$] of fish injected with GnRH-a alone and combined with pimozide decreased significantly on the 7th day post-injection in 2004(P<0.05). In 2005 trials, HSI was significantly reduced in GnRH-a treated fish on the 7th day post-injection (P<0.05). Pimozide-injected fish showed a pattern with increase of GSI and decrease of HSI, without significant differences. Taken together, these results suggest that at least in part hypothalamic hormones and dopamine receptor antagonist may induce sexual maturation in freshwater-adapted maturing chum salmon. It remains to evaluate these preliminary results by further researches.

Induction of Ovulation by Hormone Treatment in Ussurian Bullhead Leiocassis ussuriensis

  • The effects of ovulation induction in ussurian bullhead, Leiocassis ussuriensis, were investigated by treating ussurian bullhead with hCG, LHRHa, GnRHa, ovaprim, and pimozide. hCG was injected to ussurian bullhead at 0.75% NaCl, 5,000, 10,000, 20,000, and 30,000 IU, respectively. The ovulation inducement rates were 100% in 20,000 and 30,000 IU. Fertilization rates were 82.7% and 79.8%. Hatching rates were 59.4% and 57.2%. Ovulation time was between 16-19 hr The concentrations of LHRHa injected were 0.75 NaCl, 50, 100, 200, 300, and $300{\mu}g/kg$. The ovulation inducement rates were 100% in 300 and $400{\mu}g/kg$. Fertilization and hatching rates were 84.9% and 68.4% at $200{\mu}g/kg$. The times to ovulation were between 23 hr and 34 hr. Ovaprim of 0.75% NaCl, 1.0, 1.5, 2.0, 2.5 and 3.0 ml/kg were injected to the abdominal cavity. The ovulation inducement rate was highest at 2.0 and 3.0 ml/kg to 92% and ovulation time was between 27-38 hr. LHRHa concentrations of 0.75% NaCl, 50, 100, 200, 300 and $400{\mu}g$ were injected with pimozide ($1,000{\mu}g$). Ovulation inducement rate was 100% from 200 to 400 IU with pimozide. Ovulation time was 22-36 h. Fertilization and hatching rates were 88.9% and 70.4% in $200{\mu}g/kg$ with pimozide.

Induced Ovulation in the Mandarin Fish, Siniperca scherzeri by Sex-Maturation Hormones (성성숙 호르몬 처리에 의한 쏘가리의 배란 유도)

  • Ovulation of maturing femal mandarin fish, Siniperca scherzeri was induced using single injection of human chorionic gonadotropin (HCG) or gonadotropin releasing hormone-analogue (GnRH-a), GnRH-a plus prostaglandin F2 (PG$F_2$) or GnRH-a plus pimozide. The response was evaluated by fertilization, embryo-formation and hatching rate after insemination. Those rates were generally higher in GnRH-a group than in HCG group. The higher hatching rat of above 89% was achived using a dosage of 5,000 IU/kg HCG plus 10 ${\mu}$g/kg GnRH-a, 10${\mu}$g/kg GnRH-a plus 500 ng/kg PGF2, and 10 ug/kg GnRH-a plus 1-5 mg/kg pimozide. Ovulation was induced in all female injected with sex-maturation hormones and stimulator, but blocked in female injected with HCG plus GnRH-a plus dopamine combination, and GnRH-a plus PGF2 plus indometacin combination. These results show that the mandarin fish in spawning period secrete a sex-mutruation assosiated hormones and gonadotropin-releasing -inhibiting factor(GRIF).

Effects of Neuroleptics on the Opioid Receptor Binding in the Mouse Striatum (마우스 선조체에서 Opioid 수용체 결합에 대한 Neuroleptics의 영향)

  • Our purpose was to gain insight into a possible modulatory role for ${\mu},\;{\delta},\;and\;{\kappa}$ opioid receptors by neuroleptics (chlorpromazine, thioridazine, haloperidol, sulpiride, and pimozide) in chronic morphine 5 mg/kg and 20 mg/kg treated mouse striatum. We attempted quantitative receptor assays using highly specific radioligands, $[^3H]\;DAGO\;([D-Ala^2,\;N-Mephe^4,\;Glycol^5]\;enkephalin)$, $[^3H]DPDPE\;([D-Pen^2,\;D-Pen^5]\;enkephalin)$ and $[^3H]\;DPN(diprenorphine)$ to measure the binding affinity in the experimental groups. The decrease of $[^3H]DAGO$ binding was potentiated by sulpiride and pimozide in the chronic morphine treatment (5 mg/kg and 20 mg/kg). The decrease of $[^3H]DPDPE$ binding was inhibited by chlorpromazine, thioridazine, haloperidol, sulpiride, and pimozide in chronic morphine treatment (5 mg/kg and 20 mg/kg). The decrease of $[^3H]\;DPN$ binding was significantly inhibited by chlorpromazine, thioridazine, sulpiride, and pimozide in chronic morphine 20 mg/kg treatment. $[^3H]\;DPN$ binding on the neuroleptics was antagonized by naloxone pretreatment in chronic morphine 20 mg/kg treatment. These findings suggest that neuroleptics influence opposing tonically active on the ${\delta},\;and\;{\kappa}$ opioid receptor compared with ${\mu}$ opioid receptor in the chronic morphine treated mouse striatum.

  • This study aims to investigate how ovulation induction of red spotted grouper, Epinephelus akaara varies when treated with different hormonal substances such as ovaprim, pimozide, LHRHa, and HCG. As for LHRHa, we injected it at four different concentrations: 50 ㎍kg-1, 100 ㎍kg-1, 150 ㎍kg-1, and 200 ㎍kg-1. All hormonal substances including LHRHa were injected into the back muscles. Buoyant, embryonic survival, fertilization, and hatching rate were the highest in LHRHa and LHRHa + pimozide group. When LHRHa was injected at different concentrations, buoyancy and fertilization rate were the highest in 100 ㎍kg-1 group. Embryonic survival and hatching rate were the highest in 150 ㎍kg-1. Based on these experimental results, we concluded that LHRHa at the concentration of 100 ㎍kg-1 is the most efficient for the ovulation induction in this fish species.

Effect of HCG, LHRHa, Ovaprim and Pimozide on Ovulation Induction in Long Snout Bullhead Leiocassis longirotris (HCG, LHRHa, Ovaprim and Pimozide 처리에 따른 종어 Leiocassis longirotris의 배란유도)

  • The main objective of this study is to investigate the effects of ovulation induction by treating HCG, LHRHa, GnRHa, ovaprim and pimozidein in long snout bullhead, L. longriostris. All hormons were injected into the muscles of back. Concentration of LHRHa to injection were 20, 50, 100, 150 and 200 ${\mu}g/kg$ and same concentration of LHRHa was injected after 24 hour. The ovulation induction rate was 100% and fertilization and hatching rates were 68.4, 45.2, 58.4 and 33.6% in 50 and 100 ${\mu}g/kg$. The times to ovulation were between 28 and 44 h. HCG was injected in long snout bullhead at 5,000, 10,000, 15,000, 20,000 and 25,000 IU/kg. The ovulation induction rate was 50% in 15,000 and 20,000 IU/kg. Fertlilzation and hatching rates were 55.2, 45.6, 52.8 and 44.8%. Ovulation time was between 72~80 h. HCG concentration of 500, 1,000, 2,000 and 4,000 IU/kg were injected with $50{\mu}g$ of LHRHa. Ovulation and hatching rates in 2,000 IU/kg were 75 and 35%. Ovulation time was 28~48 h. Ovaprim of 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 mL/kg were injected to the abdominal cavity. The ovulation induction rate was highest at 2.5 mL/kg to 50% and ovulation time was between 66~86 h. LHRHa concentration of 50, 100, 200, 300 and 400 ${\mu}g/kg$ was injected with pimozide (1,000 ${\mu}g/kg$). Ovulation induction rate was 87% at 100 and 200 IU/kg with pimozide. Ovulation time was between 66~86 h.

Ovulation Induction Effect of Sevenband Grouper, Epinephelus septemfasciatus by Treating Hormones (호르몬 처리에 의한 능성어 Epinephelus septemfasciatus의 배란유도 효과)

  • This study aims to investigate how ovulation induction of sevenband grouper, Epinephelus septemfasciatus varies when treated with different hormonal substances such as ovaprim, pimozide, LHRHa, and HCG. As for LHRHa, we injected it in different concentrations: $50{\mu}g/kg$, $100{\mu}g/kg$, $150{\mu}g/kg$, and $200{\mu}g/kg$. All hormonal substances including LHRHa were injected into back muscles. As a result, ovulation occurred in all sample groups. The sample group injected with HCG released the largest amount of eggs, and the dimeter of eggs and oil globules were also significantly bigger than those of others. Fertilization rate, embryonic survival rate, and hatching rate were the highest in a LHRHa ($100{\mu}g/kg$) +pimozide ($1,000{\mu}g/kg$) group and buoyant rate in a LHRHa group. Ovulation rate was 100% when LHRHa was injected in different concentrations. The largest amount of eggs was released in $200{\mu}g/kg$. The diameter of eggs and oil globules were the biggest in $200{\mu}g/kg$ and the smallest in $50{\mu}g/kg$. Fertilization rate showed no significant difference through all different levels of concentration: 94.6-95.3%. Buoyant rate was the highest in $100{\mu}g/kg$ and embryonic survival rate and hatching rate in $150{\mu}g/kg$. Based on these experimental results, we concluded that LHRHa in $100{\mu}g/kg$ is the most efficient for the ovulation induction of sevenband grouper.
