Effects of growth characteristics of 'Nongjingo'(Lentinula edodes) according to relative humidity with sawdust cultivation (표고 톱밥재배 시 상대습도가 자실체 생육에 미치는 영향)

  • This study was investigated the growth characteristics of 'Nongjingo' cultivar(Lentinula edodes) according to relative humidity(RH). The color difference of the pileus showed the highest L(Lightness, L) value in RH65 and the a(Redness, a) value in RH95. b(Yellowness, b) values were similar in all treatments. The hardness of pileus is highest at RH95. As the relative humidity increased, the length of pileus and stipe tended to increase. The diameter and thickness of pileus were high at RH95%. The diameter of stipe could not see the big difference in the three treatment groups. This study growth characteristics and yield were increased at higher relative humidity, but quality was decreased. Therefore, must adjust the relative humidity to produce high quality mushrooms.

Studies on Inorganic Composition and Immunopotentiating Activity of Ganoderma lucidum in Korea (한국산(韓國産) 영지(靈芝)의 무기(無機) 성분(成分) 및 면역(免疫) 증강(增强) 작용(作用)에 관한 연구(硏究))

  • To determine contents of inorganic elements of Ganoderma lucidum, the horn-shaped carpophores and the pileus of Ganoderma lucidum were incinerated and analyzed by inductively coupled plasma atomic emission spectrophotometry. The ash contents of the pileus and the horn-shaped carpophore were 1.48% and 1.40%, respectively. The pileus contained calcium, magnesium, sodium, manganese, iron, zinc and germanium in that order. The horn-shaped carpophore contained magnesium, calcium, zinc, manganese, iron, copper and germanium in that order. To examine the protein-bound polysaccharide from Ganoderma lucidum for immunopotentiating activity, its fruit bodies were extracted with hot water. Purification of the extract was carried out by acetone precipitation and dialysis. The fraction obtained during the purification procedure consisted of a polysaccharide moiety (51%) and a protein moiety (5%). When the compound was administered intraperitoneally to the mice at a dose of 50mg/kg, it enhanced the accumulation of the peritoneal exudate cells, macrophage and polymorphonuclear leucocytes, thereby indicating immunopotentiation.

  • This study was carried out to investigate optimum wind velocity for growth of Pleurotus ostreatus. In the chamber experiments, the divergence of pileus was $63^{\circ}$ in 0.3m/s, and it showed the tendency that the more the air velocity was increased, the more the divergence became small. And the ratio of commercial yields was 96% in the air velocity of 0.3m/s. In the cultivation room experiments, the divergence of pileus was $64^{\circ}$ in 0.3m/s, and it was the tendency that the more the air velocity was fast, the more the divergence became larger. And the commercial yields was similar to the trend of the chamber experiment as 94.8% in 0.3m/s, and the quality characteristics of the fruit-body showed the uniform quality in comparison with other treatment because the standard deviation of the size of the pileus, etc. was the lowest in 0.3m/s like the chamber experiment.

The study of quality standard of Pleurotus eryngii (큰느타리버섯의 품질기준에 관한 연구)

  • In order to make the objective quality standard of Pleurotus eryngii, we statistically analysed fruit body collected by the type of market with our own mushroom research lab's quality standard and suggested the quality standard of Pleurotus eryngii by grade. Namely, premium(extra large), first(large), second(middle), and third grade(small) fruit body were 120~130, 110~120 below, 80~110below, and 80mm and below, respectively in height. The diameters of stipe of each grades were 40~45, 30~40below, 25~30below, 25mm below, respectively. The standard of the diameter of pileus indicated ratio of the diameter of pileus/the diameter of stipe instead of the diameter of pileus only because a quality is depend on a ratio of pileus/stipe. The premium and first grade of a ratio of pileus/stipe were 1.6 below and 1.7and below respectively. The weights of premium(extra large), first(large), second(middle), and third grade(small) fruit body were 95~120, 70~95below, 50~70below, 50g below, respectively.

Antioxidant and tyrosinase inhibitory activity of Hypsizygus marmoreus (brown cultivar) methanol extracts (갈색 느티만가닥버섯 메탄올 추출물의 항산화 및 tyrosinase 저해 효과)

  • The objective of this study was to evaluate antioxidant effect and tyrosinase inhibitory activity of methanol extracts from Hypsizygus marmoreus. The Hypsizygus marmoreus was divided into two parts (pileus and stipe) and extracted with methanol. Total polyphenolics and flavonoids in the methanol extracts were measured by spectrophotometric methods and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities have been determined for antioxidant activities. The total polyphenolics and flavonoids contents of methanol extract of the pileus were higher than methanol extract of the stipes. The total polyphenolics contents in methanol extracts of the pileus and stipes were 8.7 ug/mg and 5.6 ug/mg, respectively. The total flavonoids contents in methanol extracts of the pileus and stipes were 2.8 ug/mg and 1.4 ug/mg, respectively. The tyrosinase inhibitory activity was proportional to concentration of methanol extract. The tyrosinase inhibitory activity of the methanol extract (200 mg/ml) of pileus (66.9%) and stipe (57.97%) was lower than those of positive control 2% arbutin. The DPPH radical scavenging activity of the methanol extract (20 mg/ml) of pileus and stipes was 52.55% and 30.35%, respectively. Moreover, the effects of methanol extarcts on cell proliferation of B16BL6 mouse melanoma cells were investigated using WST-1 assay (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) and B16BL6 mouse melanoma cells treated with methanol extract of 200-2,000 ug/ml were higher proliferation rate than those of 0.04% adenosine.

An Action Spectrum for Light-induced Growth rate in Pleurotus ostreatus (느타리버섯 생장에 미치는 광 감응성 작용 스펙트럼)

  • The action spectrum for light-induced growth of fruit body in Pleurotus ostreatus has been studied by irradiation at various wavelengths. Effective wavelengths were distributed from near ultraviolet to blue region of spectrum. The most effect of light was observed in the region between 340 to 520 nanometers. The growth weigth obtained from which has been irradiated 144 hours in the region of 340-520 nm increases 74%, on the contrary the observed growth weight has been observed in the longer wavelengths than 620 nm. The pileus size was promoted 30$\times$32% in the region of 340-500 nm, and retarded 22$\times$19% in the dark. the strip length was rearded 12% in the region of 340-500 nm, and promoted 38% in the dark. The color thickness for pileus is getting light in longer wavelengths.

Determination of Glucan Contents in the Fruiting Bodies and Mycelia of Lentinula edodes Cultivars

  • Shiitake mushrooms (Lentinula edodes) containing ${\beta}$-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The objective of this study was to determine the ${\beta}$-glucan content in different sections of the fruiting bodies and mycelia of ten shiitake mushroom cultivars. The measured ${\beta}$-glucan content ranged from $20.06{\pm}1.76%$ to $44.21{\pm}0.13%$ in the pileus sections, and from $29.74{\pm}1.40%$ to $56.47{\pm}4.72%$ in the stipe sections. The results of this study indicate that the variance in ${\beta}$-glucan content dependent on the shiitake cultivar, and that the ${\beta}$-glucan content is higher in the stipe than in the pileus.

  • This study was focused on finding a relationship between fruit-body growth and relative humidity in Neolentinus lepideus cultivation. The higher the relative humidity, the moisture content of pileus and stipe were also tends to be increased. The day of formation of primordia and development of fruit body was shortest at the relative humidity 95%. The yield and commercial yield were higher in the relative humidity 95% than in the other treatment. Individual weight of fruit body was the highest at the relative humidity 75% and 85%. Crack indices of pileus was the strongest at the relative humidity 65%.

Effect of Temperature on the Growth and Microstructure of Fruit Body in the Basidiomycetes, Pleurotus ostreatus (느타리버섯 자실체의 생육 및 미세구조에 미치는 온도의 영향)

  • To elucidate the effect of temperature on the fruit body growth and structure of pleurotus ostreatus cultivar Chunchu No.2, microstructures were observed with scanning- and transmission electron microscope. Pileocystidia were well developed on the surface of pileus in the fruit body cultivated at $7^{\circ}C$. As the increasing temperature for fruit body development, thickness of pileus, thickness of stipes and length of stipes shown thin, thick and long, respectively. Color of pileus was also changes to whitish grown under high temperature. Physical structure shown as hardness and gumminess of stipes grown at high temperature, fruit body were soften than that of low temperature. Microstructures of fruit body grown at high temperature shown fast-discharge of basidiospore and sytoms aging. vacuolation in the cell observed at mycelium grown under high temperature. Optimum temperature for vegetative growth of fruit bodies were $15^{\circ}C$, and shown regular arrangement of mycelium on the surface of stipes.
