  • Production of shikonin derivatives through cell suspension culture of Lithospermum erythrorhizon was investigated. Optimal concentrations of IAA and kinetin on the growth of cell suspension were 0.2 and 0.1 ppm respectively. Pigment content was markedly increased when aluminum oxide was added to the production medium and its optimal concentration was 1.5g/70ml medium. The most effective concentration of IAA was 0.5 ppm and the production of pigment did not depend on the kinetin concentration.

Characteristics of Red Pigment from Marine Bacterium Utilizing Colloidal Chitin

  • Studies on extraction of red pigment was performed to provide the basic information for the utilization of red pigment as s new source of natural food colorant. A bacterium isolated from marine resources were carried out the test for excretion of red pigment. One strain of a marine bacterium, KSR-97 showed a high production of red pigment on the medium of colloidal chitin, peptone-yeast extract with minerals. In physicochemical and sensory properties in aqueous solution of red pigment was investigated at various condition of pH, temperature, concentration of ethanol and stability of storage. Potent antioxidative of red pigment was separated by thin layer chromatograpy, silica gel chromatography and reverse high performance liquid chromatography using ODS hypersil column.

Pilot Plant Scale Extraction and Concentration of Purple-Fleshed Sweet Potato Anthocyanin Pigment (자색고구마 anthocynin 색소의 대량추출 및 농축)

  • Performance of pilot plant scale extraction and concentration of purple-fleshed sweet potato anthocyanin pigment was tested and the characteristics of pigment extracts and concentrates were investigated. Fifty kilograms of purple-fleshed sweet potato was extracted with 500 L of 1% citric acid in 20% ethanol. As a whole, extraction pattern of the large scale extraction was similar to that of the laboratory scale extraction. The extracted pigment solution was filtered twice with a bag filter and a winding type microfilter and the filtrate was concentrated by a large scale vacuum evaporator at $40^{\circ}C$ and 600 mmHg vac. The mean values of total optical density (TOD) of the extract and the concentrate were 6.53 and 120.45, respectively. Browning index (BI) and Degradation index (DI) of extract were 5.86 and 1.55 and those of concentrate were 5.89 and 1.56, respectively, which indicated that the pigments were not changed or degraded through the extraction and concentration process.

Antioxidant Activity and Cell Differentiation Effects of Monascus purpureus Pigment on Osteoblast-like MC3T3-E1 Cells (홍국색소의 항산화 활성 및 조골세포 분화에 미치는 영향)

  • The purpose of this study was to investigate antioxidant activity and cell differentiation effects of Monascus purpureus pigment on osteoblast-like MC3T3-E1 cell. In order to examine the antioxidant activities of Monascus purpureus pigment, DPPH radical scavenging, ABTS radical scavenging and SOD-like activities were investigated. DPPH radical and ABTS radical scavenging activities of Monascus purpureus pigment were increased in a dose-dependent manner, and maximum activity were 94% and 99% at a concentration of 1,000 ㎍/ml, respectively. Additionally, SOD-like activity of Monascus purpureus pigment showed 62% at a concentration of 1,000 ㎍/ml. MC3T3-E1 cells did not show cytotoxicity in the concentration range of Monascus purpureus pigment 1~100 ㎍/ml. The ALP activity was increased by addition of Monascus purpureus pigment, and the maximum activity was 124% as compared with control. In addition, nodule formation, a late differentiation factor for bone formation, was increased by adding Monascus purpureus pigment compared to control. These results suggest that Monascus purpureus pigment is expected to be a natural source for developing functional materials to prevent bone-related diseases by osteoblast differentiation.

  • Artists have been used animal glues for a long time as binding and sizing materials of paintings in many countries including Korea. Since animal glues as a binding material lose their own adhesive strength by aging, pigment particles or pigment layers are easily detached from painting surface. Restaurators prefer to use animal glues for consolidation of pigment particles or pigment layers because artists use animal glues as a binder for their painting works. It is widely known that animal glues have different characteristics such as viscosity, stability on water etc. But there are still a few requirements to consider their stabilities for the restoration of paintings. Therefore, this study was aimed to evaluate of stability and efficiency of animal glue applications. The efficiency of animal glue application was decreased with increasing the concentration of animal glue solution. This result means the simple tendency, but does not representative data for other animal glues made from same raw materials. In most cases, the surface and mechanical characteristics of animal glues are dependent on some factors, like viscosity and concentration of animal glue solution.

Determination of the Conditions for Anthocyanin Extraction from Purple-Flashed Sweet Potato (자색고구마 Anthocyanin 색소의 추출조건 결정)

  • To establish the optimum conditions for the extraction of anthocyanin pigment from purple-fleshed sweet potato, a suitable extraction solvent with the optimum citric acid concentration for acidification of the solvent, and the optimum extraction time and temperature were determined. Twenty percent ethanol solution acidified with citric acid was found to be a good solvent for the extraction of the pigment from purple-fleshed sweet potato. About 10 hour extraction at room temperature was appropriate for the extraction. pH of the extract was below 3 when more than 0.7% citric acid was added. The higher the concentration of citric acid added was, the higher the total optical density (TOD) of the extract was. However, the increase in TOD of the extract was insignificant when more than 1% of citric acid was added. Therefore, addition of 1% citric acid was determined for acidification of the extracting solvent. Though the initial rate of the pigment extraction increased as the extracting temperature increased, extraction at higher temperatures of 60 or 8$0^{\circ}C$ for an extended time caused a decrease in the extraction yield due to degradation of the pigment. The optimum extraction temperature for the anthocyanin pigment from purple-fleshed sweet potato with the solvent used was determined as 4$0^{\circ}C$.

Extractive Fermentation of Red Pigment Using Monascus sp. JlOl (Monascus sp. J101을 이용한 적색색소의 Extractive Fermentation)

  • The characteristcs of monascus fermentation using a hyperpigment-producing mutant, Monascus sp. J101, were analyzed, and the extractive fermentations employing permeabilizing agents and resin were carried out to increase the productivity of red pigment. And the kinetic analysis was also carried out in case of the monascus fermentation using Amberlite XAD-7. The extracellular content of the red pigment produced by Monascus sp. J101 was about 17% of the total, and the production of pigment was regulated by its own product. The cell growth reached a stationary phase at 48 hours ofter inoculation, whereas the pigment production continued up to 100 hours, which showed the pattern of a mixed growth-associated type. During the fermentation, various permeabilizing agents were added to the culture medium and their effects on pigment production were examined. By adding 0.05% Triton X-100 at 48 hours of cultivation, about an 18% increase in pigment production was accomplished as compared to the control, 12% ethyle acetate and 15% for 0.05% deoxycholate, respectively. When a nonionic adsorbent, Amberlite XAD-7 was added to the culture medium at a concentration of 12.0% at 48 hours of cultivation, the pigment production was enhanced by about 48.9% as compared to the control.

Production of Red Pigment from Marine Bacterium Utilizing Colloidal Chitin. (Colloidal Chitin을 자화하는 해양세균으로부터 적색색소의 생산)

  • This study is that of providing a fairly practical practival guide to the use of natural pigment in food industry. A strain isolated from marine resources was carried out the production of red pigment. The pigment showed UV absorption maxima at 520 and 550 nm. The color intensity in aqueous solution was fairly stable in the ranges of pH 5~8. The strain KS-97 produced a maximum yield of red pigment at$ 25^{\circ}C$ for 72 hrs with pH 7.0. The strain KS-97 was iden-tified a Bacillus sp. based on morphological and biochemical characterization such as a rod from, motility, spore for-mation, Gram positive and catalase production. The production of red pigment indicated that the strain Ks-97 utilized at thigh concentration of colloidal chitin as carbon sources obtained maximum yield of red pigment at $25^{\circ}C$ for 72 hrs. The highest production of red pigment was observed with cultivation in medium containing 20% colloi-dal chitin, 2.5g polypeptone, 2.5g yeast extract, 1.0g $KH_2$$PO_4$, 0.01g $MgSO_4$.$6H_2$O, 0.01g $ZnSO_4$, 0.01 g $MnSO_4$(per 1).

  • Temporal and spatial variability of phytoplankton pigment concentrations around the Korea Peninsula was described, using the monthly mean composite images of the SeaWiFS (Sea-Viewing Wide Field-of-View Sensor). The high pigment concentrations appear in the spring and fall in the East Sea The spring bloom in the southern regions (in April) occurs one month in advance in comparison with tint in the northern regions (in May). In summer season, the pigment concentrations are low all over the region in the East Sea And the high pigment concentrations exist yearly around warm stream along the coast of the East Sea, and in the coast of the West Sea and South Sea In particular, the high pigment concentrations linking near the mouth q the Yangze River to coast of South Sea in Korea appear during August to December.

Exopolysaccharide Production by Aureobasidium pullulans - Appearance of Melanin Pigment - (Aureobasidium pullulans 에 의한 Exopolysaccharide 생산 - 멜라닌 색소의 출현에 관한 연구 -)

  • In exopolysaccharide fermentation by Aureobasidium pulluans, the effects culture conditions (concentration of nitrogen, potassium phosphate, dissolved oxygen, and initial pH) on the production of exopolysaccharide and the appearance of melanin pigment were investigated. The results are as follows. (1) The specific growth rate and the specific production rate of exopolysaccharide were inhibited by substrate when the carbon source concentration higher than $50g\;/\;{\ell}$ and the cell growth increased with increases of nitrogen source but exopolysaccharide production decreased with the nitrogen concentration when it become greater than $1\;g\;/\;{\ell}$. (2) The maximum cell growth and the maximum exopolysaccharide production were obtained at initial pH values of 3.0 and 7.5 respectively. As the initial pH increased, the yeast-like cells increased and the increased of dissolved oxygen increased the cell growth and exopolysaccharide production. (3) As the concentration of dissolved oxygen is increased or the concentration of nitrogen source is decreased, the period of melanin pigment appearance becomes shorter and the melanin pigment never appeared when the initial pH was less than 3.0 or the potassium phosphate was not added.
