Production and Characteristics of Hongkuk-ju using Monascus anka (Monascus anka를 이용한 홍국주의 제조 및 특성)

  • To reproduce the brewing process of Hongkuk-ju and to identify the functional properties of it, Hongkuk-ju was brewed using different additions of Hongkuk (100%, 90%, 70%, 50%) and Nuruk (0%, 10%, 30%, 50%). The quality elements, including pH, total acidity, reducing sugar content, alcohol content and pigments (yellow, red, monacolin K and citrinin), were measured. The pH values of Hongkuk-ju showed a slight difference (pH 4.08~4.58) right after the $1^{st}$ stage mash; further, the pH on all groups (H1, H2, H3 and H4) in the terminal of the $2^{nd}$ stage mash (9 days in fermentation) were similar, ranging approximately at pH 3.70. The total acidity change did not show a difference directly the $1^{st}$ stage mash (nearby 0.2 %); however, it began to show a slight difference at the terminal of the $2^{nd}$ stage mash between the range of 0.69~0.76%. The residual reducing sugar of the content was decreased with the increased Nuruk content. The alcohol concentrations of the treatment brew with Nuruk ranging from 12.3% to 13.7% were higher than Hongkuk on its own. The yellow and red pigment contents of Hongkuk-ju ranged from 7.2~8.8 O.D. units (yellow pigment) and from 4.4~5.1 O.D. units (red pigment). The production of monacolin K and citrinin was the highest (9.48 mg/kg and 10.14 mg/kg) when the treatment solely brewed Hongkuk. The concentration of Nuruk and the preparation of the seed mash from it were critical factors compared to the treatment of rice in brewing Hongkuk-ju.

Simulated Radiances of the OSMI over the Oceans

  • Prior to launch, simulated radiances of the Ocean Scanning Multispectral Imager (OSMI) will be very useful to guess the real imagery of OSMI and to check the data processing system for OSMI. The data processing system for OSMI which is one sensor of Korea Mult i - Purpose Satellite (KOMPSAT) scheduled for launch in 1999 is being developed based on the SeaWiFS Data Analysis System (SeaDAS). Such a simulation should include the spectral bands, orbital and scanning characteristics of the OSMI and KOMPSAT spacecraft. The simulation is also very helpful for finding and preparing for problem areas before launch. This paper describes a method to create simulated radiances of the OSMI over the oceans. Our method for constructing a simulated OSMI imagery is to propagate a KOMPSAT orbit over a field of Coastal Zone Color Scanner (CZCS) pigment values and to use the values and atmospheric components to calculate total radiances. A modified Brouwer - Lyddane model with drag was used for the realistic orbit prediction, the CZCS pigment data were used to compute water - leaving radiances, and a variety of radiative transfer models were used to calculate atmospheric contributions to total radiances detected by OSMI. Imagery of the simulated OSMI total radiances for 6 nominal bands was obtained. As expected, water - leaving radiances were only a small fraction of total radiances and sun glint contaminations were observed near the solar declination. Therefore, atmospheric correction is very important in the calculation of pigment concentration from total radiances. Because the imagery near the sun's glitter pattern is virtually useless and must be discarded, more advanced mission planning will be required.

An Effective Defensive Response in Thai Aromatic Rice Varieties(Oryza sativa L. spp. indica) to Salinity

  • Rice is one of the world's staple crops and is a major source of carbohydrate. Rice is exported from several countries, providing a major source of income. There are many documents reporting that rice is a salt-sensitive crop in its developmental stages. The objective of this investigation is to evaluate the effective salt-tolerance defense mechanisms in aromatic rice varieties. Pathumthani 1(PT1), Jasmine(KDML105), and Homjan(HJ) aromatic rice varieties were chosen as plant materials. Rice seedlings photoautotrophically grown in-vitro were treated with 0, 85, 171, 256, 342, and 427 mM NaCl in the media. Data, including sodium ion$(Na^+)$ and potassium ion$(K^+)$ accumulation, osmolarity, chlorophyll pigment concentration, and the fresh and dry weights of seedlings were collected after salt-treatment for 5 days. $Na^+$ in salt-stressed seedlings gradually accumulated, while $K^+$ decreased, especially in the 342-427 mM NaCl salt treatments. The $Na^+$ accumulation in both salt-stressed root and leaf tissues was positively related to osmolarity, leading to chlorophyll degradation. In the case of the different rice varieties, the results showed that the HJ variety was identified as being salt-tolerant, maintaining root and shoot osmolarities as well as pigment stabilization when exposed to salt stress or $Na^+$ enrichment in the cells. On the other hand, PT1 and KDML105 varieties were classified as salt-sensitive, determined by chlorophyll degradation using Hierarchical cluster analysis. In conclusion, the HJ-salt tolerant variety should be further utilized as a parental line or genetic resource in breeding programs because of the osmoregulation defensive response to salt-stress.

  • Detection of red tides by satellite remote sensing can be done either by detecting enhanced level of chlorophyll pigment or by detecting changes in the spectral composition of pixels. Using chlorophyll concentration, however, is not effective currently due to the facts: 1) Chlorophyll-a is a universal pigment of phytoplankton, and 2) no accurate algorithm for chlorophyll in case 2 water is available yet. Here, red band algorithm, classification and PCA (Principal Component Analysis) techniques were applied for detecting patches of Cochlodinium polykrikoides red tides which occurred in Korean waters in 1995. This dinoflagellate species appears dark red due to the characteristic pigments absorbing lights in the blue and green wavelength most effectively. In the satellite image, the brightness of red tide pixels in all the three visible bands were low making the detection difficult. Red band algorithm is not good for detecting the red tide because of reflectance of suspended sediments. For supervised classification, selecting training area was difficult, while unsupervised classification was not effective in delineating the patches from surrounding pixels. On the other hand, PCA gave a good qualitative discrimination on the distribution compared with actual observation.

Studies on the Pseudomonas aeruginosa Isolated from Infected Patient (감염환자에서 분리한 녹농균의 특성에 관한 연구)

  • Seventy-two strains of Pseudomonas aeruginosa isolated from the patients were tested for pigment production, exoenzyme production and antimicrobial susceptibility. In the pigment production test, 23.6% of total 72 strains produced both pyocyanine and pyoverdin. Pyoverdin and pyomelanin producing strains were in 9.7%, and 5.5% produced pyoverdin and pyorubin. Strains producing of all of exoenzyme, protease, elastase and lecithinase were in 5.6%. The most common type of exoenzyme production was both protease and elastase producing. Protease producing strain were 23.6%, Among the 72 strains, 50% produced protease. As the result of antimicrobial susceptibility in the isolated 20 strain, most strain were resistant to sulfamethoxazole(90%), but sensitive to other tested antibiotics more than 60%. The MIC50 and MIC90 level of tested antibiotics to 70 strains were 128 $\mu\textrm{g}$/$m\ell$, 512 $\mu\textrm{g}$/$m\ell$ for KM, 8,256 $\mu\textrm{g}$/$m\ell$ for GM, 8, 128 $\mu\textrm{g}$/$m\ell$ for CPZ, and 8, 64 $\mu\textrm{g}$/$m\ell$ for PIPC respectively.

  • Seashore joining with land and sea, which is typical habitat for marine macroalgae, is classified two types of shore as soft- and hard-bottom shore according to topographical (geological) and ecological features. We compared two of Ulva pertusa Kjellman from two contrasting habitats, sandy (soft-bottom, Haenam) and rocky shore (hard-bottom, Hadong) in terms of chlorophyll-a fluorescence and its parameters, and various photosynthetic pigment and nutrient content in the tissue of those. Both of habitats were different in the light environment such as light attenuation coefficient and even in nutrient concentration of ambient seawater. Electron transport rate (ETR) of Ulva from sandy shore was higher than from rocky shore. The range of photosynthetic pigment content in the tissue of U. pertusa was significantly much more in from sandy shore, and also nitrogen and phosphorus content were significantly higher except for carbon content. However, there were no significant differences in the ratio of among photosynthetic pigments, and N:P ratio was similar between each other, even though significantly different. Our result implied on the reason of why most of green tides in the worldwide concentrated and frequently occurred at sites with sandy, muddy and silty bottoms, being classified as soft-bottom shore.

Effects of Cosmetic Pigments on the Bactericidal Activities of Parabens (파라벤류의 방부력에 대한 화장품용 안료의 영향)

  • In this study, we evaluate the anti-microbiological activity of paraben in eye shadows that are composed of pigments and oil binders using various analytical methods and microbiological tests. Paraben does not show the microbiological activity properly when it was used with Nylon SP$^{(R)}$ 10, Talc RF SSA$^{(R)}$, OMC Talc AS$^{(R)}$ and $BaSO_4$. In the test of fungi, Nylon SP$^{(R)}$ 10 causes the decrease of microbiological activity regardless of the type of oil binders. The pigment of Mango violet also causes the decrease of microbiological activity when ester oil binder was used. Regardless of the type of oil binder, samples containing nylon SP 10, 0.15% of methyl paraben and 0.05% of propyl paraben had not been able to maintain microbiological activity only if the concentration of parabens were increased. Trace amounts of metal ions present in pigments reduced the activity of preservatives by inactivation of hydroxyl group of paraben. It is thought that swollen nylon SP 10 in ester oil increase the absorption or interaction of parabens and swollen nylon powder causes the inactivation of paraben.

Browning Pattern and Pigment of Glucose/Glycine Model Systems (글루코스-글리신 혼합용액의 갈색화 패턴 및 색소)

  • Browning pattern was developed in aqueous solutions of glucose/glycine mixture under controlled conditions. Browning pattern was definitely influenced by pH of medium and concentration of reactants. Filter paper disks were immersed in diluted solutions of glucose/glycine system and fried in cooking oil. Concentrations of reactants only affected browning pattern of fried filter paper disks and pH effect was obscured at high temperatures. Amorphous brown precipitate was obtained from the lowest pH medium of glucose/glycine system. An attempt was made to characterize the brown pigment produced in the present model system.

Monitoring of Climatological Variability Using EOS and OSMl Data

  • Dramatic changes in the patterns of satellite-derived pigment concentrations, sea-level height anomaly, sea surface temperature anomaly, and zonal wind anomaly are observed during the 1997-1998 El Ni$\bar{n}$o. By some measures, the 1997-1998 El Ni$\bar{n}$o was the strongest one of the 20$^{th}$ century. A very strong El Ni$\bar{n}$o developed during 1997 and matured late in the year. A dramatic recovery occurred in mid-1998 and led to La Nina condition. The largest spatial extent of the phytoplankton bloom was fellowed recovery from El Ni$\bar{n}$o over the equatorial Pacific. The evolution towards a warm episode (El Ni$\bar{n}$o) started from spring of 2002 and continued during January 2003, while equatorial SSTA remained greater than +1$^{\circ}C$ in the central equatorial Pacific. The OSMI (Ocean Scanning Multispectral Imager) data are used for detection of dramatic changes in the patterns of pigment concentration during next El Ni$\bar{n}$o.

Biocidal Activity of Metal Nanoparticles Synthesized by Fusarium solani against Multidrug-Resistant Bacteria and Mycotoxigenic Fungi

  • Antibiotic resistance by pathogenic bacteria and fungi is one of the most serious global public health problems in the 21st century, directly affecting human health and lifestyle. Pseudomonas aeruginosa and Staphylococcus aureus with strong resistance to the common antibiotics have been isolated from Intensive Care Unit patients at Zagazig Hospital. Thus, in this study we assessed the biocidal activity of nanoparticles of silver, copper and zinc synthesized by Fusarium solani KJ 623702 against these multidrug resistant-bacteria. The synthesized Metal Nano-particles (MNPs) were characterized by UV-Vis spectroscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and Zeta potential. The Fourier transform infrared spectroscopy (FTIR) result showed the presence of different functional groups such as carboxyl, amino and thiol, ester and peptide bonds in addition to glycosidic bonds that might stabilize the dispersity of MNPs from aggregation. The antimicrobial potential of MNPs by F. solani against the multidrug-resistant (MDR) P. aeruginosa and S. aureus in addition to the mycotoxigenic Aspergillus awamori, A. fumigatus and F. oxysporum was investigated, based on the visual growth by diameter of inhibition zone. Among the synthesized MNPs, the spherical AgNPs (13.70 nm) displayed significant effect against P. aeruginosa (Zone of Inhibition 22.4 mm and Minimum Inhibitory Concentration 21.33 ㎍/ml), while ZINC oxide Nano-Particles were the most effective against F. oxysporum (ZOI, 18.5 mm and MIC 24.7 ㎍/ml). Transmission Electron Microscope micrographs of AgNP-treated P. aeruginosa showed cracks and pits in the cell wall, with internalization of NPs. Production of pyocyanin pigment was significantly inhibited by AgNPs in a concentration-dependent manner, and at 5-20 ㎍ of AgNPs/ml, the pigment production was reduced by about 15-100%, respectively.
