  • 벼 흰빛잎마름균을 이용하여 Streptomycin 계약제인 Agrepto에 대한 내성형질의 선발효과를 조사하여 다음과 같은 결과를 얻었다. 1) 사용된 약제 Agrepto는 $105^{\circ}C$에서 20분간의 온도처리에서 약제의 안정성을 보였다. 2) 서로 다른 지역균주인 75-6 및 75-9 묘주는 선발영제농도 10ug/ml 및 100ug/ml에서 상이한 내성력을 나타내는 내성균이 선발되었다. 3) 저농도인 10ug/ml 및 100ug/ml의 선발농도에서도 10,000ug/ml에서 생장이 가능한 내성균이 선발될 수 있었다. 4) 전세대 100ug/ml에서 선발하고 재차 3,000ug/ml에서 선발된 선발균주는 처리농도 100ug/ml에서 무처리와 동일한 생장량을 보였다. 5) 자연집단의 균주 75-9를 100ug/ml에서 선발한 이들 분리균주는 내성 정도에 있어서 500ug/ml 이하, 500-1,000ug/ml 및 1,000-3,000ug/ml의 내성을 나타내는 몇단계의 내성력 정도가 상이한 균들이 분포하고 있었다.

  • 벼 흰빛잎마름병의 병원균(Xanthomonas oryzae)을 이용하여 항생제의 일종인 Agrepto에 대한 약제저항성형성을 조사하기 위하여 세대 경과에 따른 약제처리효과를 Optical Density로 간접 측정하여 다음과 같은 결과를 얻었다. 1. Agrepto는 액체배양기에 첨가되었을 때 처리 제1세대 30ppm까지의 낮은 농도 처리에서도 세균증식에 현저한 저지 효과를 나타내었다. 2. 동일농도의 4세대간 처리에서 세대의 경과에 따라 세균 증식량이 현저히 증가하였고 증식율은 10ppm 처리구에서 보다 30ppm 처리구에서 더욱 뚜렷하였다. 3. 전세대 20ppm 처리구의 선발집단의 경우 세대가 경과할수록 더욱 내성의 증가 현상을 나타내었다. 4. 고농도 처리구에서 선발된 세균이 저농도처리구에서 선발된 세균의 경우보다 저항성 형성이 현저히 증가되는 경향을 보였다.

  • 선발에 의하여 얻은 벼 흰빛잎마름병균 Xanthomonas oryzae의 Streptomycin 내성균에 대하여 병원성, 배지내에서의 생장량 및 자외선 감수성에 대한 변이성을 조사하여 다음과 같은 결과를 얻었다. (1) SM 내성균은 Wase-Aikoku-3, Rant Emas-2 황옥 및 Kimmase의 4개 판별품종에 대한 병원성 비교에서 75-6의 내성균주가 황옥에 대하여 감수성 반응이 모균과 달리 중정도의 저항성을 나타내었다. 2) SM-내성균은 정상배지에서 모균보다 약간 높은 생장량을 보였고 100ug/ml Agrepto 첨가 배지에서는 접종후 60시간까지 생장이 조지되었으나 70시간 이후에는 모균의 생장량을 능가하였다. (3) SM-내성균은 254mu 파장의 자외선 조사에서 모균과 동일한 감수성을 나타내어 내성인자는 안정된 것으로 생각되었다.

Identification of hrcC, hrpF, and maA Genes of Xanthomonas campestris pv. glycines 8ra: Roles in Pathogenicity and Inducing Hypersensitive Response on Nonhost Plants

  • Nonpathogenic mutants of Xanthomonas campestris pv. glycines were generated with Omegon-Kim to isolate genes essential for pathogenicity and inducing hypersensitive response (HR). Three nonpathogenic multants and two mutants showing slow symptom development were isolated among 1,000 colonies tested. From two nonpathogenic mutants, 8-13 and 26-13, genes homologous to hrcC and hrpF of X. campestris pv. vesicatoria were identified. The nonpathogenic mutant 8-13 had a mutation in a gene homologous to hrpF of X. campestris pv. vesicatoria and failed to cause HR on pepper plants but still induced HR on tomato leaves. The nonpathogenic mutant 26-13 had an insertional mutation in a gene homologous to hrcC of X. campestris pv. vesicatoria and lost the ability to induce HR on pepper leaves but still caused HR on tomato plants. Unlike other phytopathogenic bacteria, the parent strain and these two mutants of X. campestris pv. glycines did not cause HR on tobacco plants. a cosmid clone, pBL1, that complemented the phenotypes of 8-13 was isolated. From the analysis of restriction enzyme mapping and deletion analyses of pBL1, a 9.0-kb Eco RI fragment restored the phenotypes of 8-13. pBL1 failed to complement the phenotypes of 26-13, indicating that the hrcC gene resides outside of the insert DNA of pBL1. One nonpathogenic mutant, 13-33, had a mutation in a gene homologous to a miaA gene encoding tRNA delta (2)-isopentenylpyrophosphate transferase of Escherichia coli. This indicated that tRNA modifications in X. campestris pv. glycines may be required for expression of genes necessary for pathogenicity. The mutant 13-33 multiplied as well as the parent strain did in the culture medium and in planta, indicating that loss of pathogenicity is not due to the inability of multiplication in vivo.

  • To develope a microbial weed control agent, HCN-producing bacteria were isolated, and their characteristics were investigated. A selected strain of WA15 was identified as Pseudomonas koreensis by morphological, cultural, biochemical and 16S rRNA gene analyses. The conditions for HCN production was investigated by a One-Variable-at-a-Time (OVT) method. The optimal HCN production conditions were tryptone 1%, glycine 0.06%, NaCl 1%, and an initial pH and temperature of 5.0 and $30^{\circ}C$, respectively. The major component for HCN production was glycine. Under optimal conditions, HCN production was about 3 times higher than that of the basal medium. The WA15 strain had physiological activities, such as indoleacetic acid that was associated with the elongation of plant roots and siderophore and ammonification inhibiting fungal growth, and produced hydrolytic enzymes, such as cellulase, pectinase and lipase. The strain was able to inhibit the growth of phytopathogenic fungi, such as Rhizoctonia solani, Botrytis cinerea and Fusarium oxysporum, by the synergistic action of volatile HCN and diffusible antimicrobial compounds. A microscopic observation of R. solani that was teated with the WA15 strain showed morphological abnormalities of fungal mycelia, which could explain the role of the antimicrobial metabolites that were produced by the WA15 strain. The volatile HCN produced by the WA15 strain was also found to have insecticidal activity against termites. Our results indicate that Pseudomonas koreensis WA15 can be applied as a microbial agent for weed control and also as a termite repellent. Furthermore, it could be applied as a microbial termiticidal agent to replace synthetic insecticides.

포도 잿빛곰팡이병의 생물적 방제를 위한 길항세균 선발 (Selection of Antagonistic Bacteria for Biocontrol of Botrytis cinerea Causing Gray Mold on Vitis spp)

  • Botrytis cinerea에 의한 포도 잿빛곰팡이병은 특히 하우스 재배시 큰 피해를 주는 병원 진균이다. Pseudomonas속 세균들은 토양 미생물중 가장 잘 연구되어 있고, 토양 내에서 중요한 역할을 담당하고 있다. 근권토양에서 분리한 형광성 Pseudomonas속 세균 83균주 중 P84균주는 실내 항균력 실험결과 다양한 식물병원진균(Phytophthora capsici, Sclerotium spp., Botryosphaeria dothidea, Fusarium spp.)에 대해 항균효과를 나타내었다. 생리적 실험과 유전적 실험결과 P84균주는 P. putida로 동정되었다. 이 세균의 항균력은 항생물질(2,4-diacetylphloroglucinol)의 생산과 관련되어 있는 것으로 사료되며, 이 세균이 포도 잿빛곰팡이병의 생물적 방제에 이용될 수 있는 가능성이 시사되었다.

Isolation and Partial Characterization of Phytotoxic Mycotoxins Produced by Sclerotinia sp., a Potential Bioherbicide for the Control of White Clover(Trifoliorum repens)

  • Sclerotinia sp. (isolate BWC98-105) causes stem blight and root rot in Leghum sp., and is presently being evaluated as a potential mycoherbicide for the control of Trifoliorium repens. Bioassays have shown that Sclerotinia sp. produces phytotoxic substance which is biologically active against T. repens. Two biologically active compounds, designated as compoundsI and II, were produced in vitro from the culture filtrate of BWC98-105 isolate Sclerotium sp. Compounds I and II were purified by means of liquid-liquid extraction and $C_{18}$ open column chromatography (300 ${\times}$ 30 mm, i.d). To determine the purity, the purified compounds were analyzed by RP-HPLC. The analytical RP-HPLC column was a TOSOH ODS-120T (150 ${\times}$ 4.6 mm i.d, Japan), of which the flow rate was set at 0.7 mL/min using the linear gradient solvent system initiated with 15 % methanol to 85 % methanol for 50 min with monitoring at 254 nm. Under these RP-HPLC conditions, compounds I and II eluted at 3.49 and 4.13 min, respectively. Compound II was found to be most potent and host specific. However, compound I had a unique antibiotic activity against phytopathogenic bacteria like bacterial leaf blight (Xanthomonas oryzae) on rice, where it played a less important role in producing toxicity on T. repens. No toxin activity was detected in the water fraction after partitioning with several organic solvents. However, toxin activity was detected in the ethyl acetate and butanol fractions. In the leaf bioassay using compound II, the disease first appeared within 4-5 h as water soaked rot, which subsequently developed into well-defined blight affecting the whole plant.

  • 지칭개, 구절초 및 산국에서 분리한 9종류의 sesquiterpene lactones에 대한 생리활성 실험의 일환으로, 10종의 세균에 대한 항세균활성과 6종의 식물병원균에 대한 항진균활성을 조사하였다. 항세균 활성실험에서는 지칭개에서 분리한 hemistepsin A와 B 그리고 산국에서 분리한 tulipinolide가 시험균주 전반에 대하여 강한 활성을 보였으며, 항진균 활성실험에서는 hemistepsin B, angeloylcumambrin B, tigloylcumambrin B, 그리고 costunolide가 광범위한 활성을 보였다. 또한, 항세균 활성실험과 항진균 활성실험 모두 비교적 뚜렷한 균주특이성을 나타내었다.

Systemic Induction of the Small Antibacterial Compound in the Leaf Exudate During Benzothiadiazole-elicited Systemic Acquired Resistance in Pepper

  • Plants protect themselves from diverse potential pathogens by induction of the immune systems such as systemic acquired resistance (SAR). Most bacterial plant pathogens thrive in the intercellular space (apoplast) of plant tissues and cause symptoms. The apoplastic leaf exudate (LE) is believed to contain nutrients to provide food resource for phytopathogenic bacteria to survive and to bring harmful phytocompounds to protect plants against bacterial pathogens. In this study, we employed the pepper-Xanthomonas axonopodis system to assess whether apoplastic fluid from LE in pepper affects the fitness of X. axonopodis during the induction of SAR. The LE was extracted from pepper leaves 7 days after soil drench-application of a chemical trigger, benzothiadiazole (BTH). Elicitation of plant immunity was confirmed by significant up-regulation of four genes, CaPR1, CaPR4, CaPR9, and CaCHI2, by BTH treatment. Bacterial fitness was evaluated by measuring growth rate during cultivation with LE from BTH- or water-treated leaves. LE from BTH-treatment significantly inhibited bacterial growth when compared to that from the water-treated control. The antibacterial activity of LE from BTH-treated samples was not affected by heating at $100^{\circ}C$ for 30 min. Although the antibacterial molecules were not precisely identified, the data suggest that small (less than 5 kDa), heat-stable compound(s) that are present in BTH-induced LE directly attenuate bacterial growth during the elicitation of plant immunity.

Antifungal Mechanism and Properties of Antibiotic Substances produced by Bacillus subtilis YB-70 as a Biological Control Agent

  • Antibiotic substances were produced by Bacillus subtilis YB-70, a potential biocontrol agent found to suppress root-rot of eggplant (Solanum melonggena L) caused by Fusarium solani, in a dextrose glutamate medium and isolated by isoelectric precipitation. Partial purification was performed by column chromatography on silica gel with two solvent systems: chloroform-methanol and methanol-chloroform-water as eluting solvents, This active fraction YBS-1 s contained antifungal activity were soluble in ethanol, methanol, and water, but were not soluble in other solvents including acetone, butanol, ethyl ether, dimethylformamide, propanol, and etc. High performance liquid chromatography and thin layer chromatographic separation of YBS-1s showed that they have been composed of three biological active bands that were named YBS-1A, -1B, and -1C. The substances were stable to heat and resistant to protease. YBS-1s were active against a wide range of plant pathogenic fungi but did not inhibit the growth of bacteria and yeasts. They were not only fungicidal but also fungistatic against chlamydospores of F. solani. The $ED_{50}$ values for the chlamydospore germination and the germ-tube growth of F. solani were $O.725\mu\textrm{m}/ml\;and\;O.562\mu\textrm{m}/ml$, respectively. Microscopic observations proved the substances restricted the growth of phytopathogenic fungus F. solani by spore burst followed by dissolving of its germ-tube, and caused abnormal hyphal swelling after application to chlamydospores or growing hyphae. Cultural filtrate of B; subtilis YB-70 also suppressed the development of root-rot of eggplant in pot tests.
