In maize, immature embryos (IEs) are highly regenerative explants most suitable for producing high frequencies of plantlet regeneration in vitro. Apart from media, explants, and hormones, genotypic variation also influences in vitro characters to a great extent. In the present study, IEs were used to study the distinctive effect of variation of size/stage and hormones in different genotypes on five in vitro characters viz., frequency of callus induction, growth rate of total callus, frequency of E. callus induction, and volume and number of regenerated plantlets. LS medium with different concentrations of 2,4-D (0.5, 1.5, 2.5, 4.0 and 5.0 mg/L) were used to study the former four in vitro characters, and medium with 6-benzylaminopurine and kinetin (0.5 mg/L, each) was used for plantlet regeneration. IEs of 1.0, 1.5, 2.0, 2.5 and 3.0 mm in size were isolated from four inbred lines viz., NM74C, NM81A, NM5883 and NM5884. Two-way ANOVA revealed that explant size and genotypes, as well as hormonal concentrations showed significant effects on in vitro characters. Two millimeter IEs were found to be suitable for in vitro cultures. LS medium with 1.5 mg/L 2,4-D and LS with BAP and Kn (0.5 mg/L, each) were found to be the best hormonal concentrations for callus induction, maintenance, and regeneration, respectively. Among the four genotypes, NM81A and NM5883 yielded more non-embryogenic and Type I E. calli. In contrast, NM74C and NM5884 yielded more highly regenerative Type II calli. Inbred line NM5884 was found to be the best among these four genotypes.

Gibberellins (GAs) are essential phytohormones for plant growth that influence developmental processes and crop yields. Recent functional genomic analyses of model plants have yielded good characterizations of the canonical GA signaling pathways and related genes. Although Panax ginseng has long been considered to have economic and medicinal importance, functional genomic studies of the GA signaling pathways in this crucial perennial herb plant have been rarely conducted. Here, we identified and performed functional analysis of the GA signaling-related genes, including PgGID1s, PgSLY1s, and PgRGAs. We confirmed that the physiological role of GA signaling components in P. ginseng was evolutionarily conserved. In addition, the important functional domains and amino acid residues for protein interactions among active GA, GID1, SCFSLY1, and RGA were also functionally conserved. Prediction and comparison of crystallographic structural similarities between PgGID1s and AtGID1a supported their function as GA receptors. Moreover, the subcellular localization and GA-dependent promotion of DELLA degradation in P. ginseng was similar to the canonical GA signaling pathways in other plants. Finally, we found that overexpression of PgRGA2 and PgSLY1-1 was sufficient to complement the GA-related phenotypes of atgid1a/c double- and rga quintuple-mutants, respectively. This critical information for these GA signaling genes has the potential to facilitate future genetic engineering and breeding of P. ginseng for increased crop yield and production of useful substances.
Background: Promising specific growth regulators are employed in the tissue cultures of various bamboo species. Specific natural hardening mixtures support the acclimatization and adaptation of bamboo under protected cultivation. Results: The growth regulators like 2, 4-Dichlorophenoxyacetic acid (2, 4-D), Naphthaleneacetic Acid (NAA), Thidiazuron (TDZ), 6-Benzylaminopurine (BAP), Kinetin, Gelrite, Benzyl Adenine (BA), Indole Butyric Acid (IBA), Coumarin, Putrescine, Gibberellic acid (GA3), Indole Acetic Acid (IAA) has been widely used for callus induction, root regeneration and imposing plant regeneration in various species of bamboo such as Bambusa spp. and Dendrocalamus spp. Different combinations of growth regulators and phytohormones have been used for regenerating some of the major bamboo species. Natural hardening materials such as cocopeat, vermicompost, perlite, cow dung, farmyard manure, compost, soil, garden soil, and humus soil have been recommended for the acclimatization and adaptation of bamboo species. Standard combinations of growth regulators and hardening mixtures have imposed tissue culture, acclimatization, and adaptation in major bamboo species. Conclusions: Bamboo contributes to soil fertility improvement and stabilization of the environment. Bamboo species are also involved in managing the biogeochemical cycle and have immense potential for carbon sequestration and human use. This paper aims to review the various growth regulators, natural mixtures, and defined media involved in regenerating major bamboo species through in vitro propagation. In addition, the ecological benefits of safeguarding the environment are also briefly discussed.
Background: Panax ginseng, one of the valuable perennial medicinal plants, stores numerous pharmacological substrates in its storage roots. Given its perennial growth habit, organ regeneration occurs each year, and cambium stem cell activity is necessary for secondary growth and storage root formation. Cytokinin (CK) is a phytohormone involved in the maintenance of meristematic cells for the development of storage organs; however, its physiological role in storage-root secondary growth remains unknown. Methods: Exogenous CK was repeatedly applied to P. ginseng, and morphological and histological changes were observed. RNA-seq analysis was used to elucidate the transcriptional network of CK that regulates P. ginseng growth and development. The HISTIDINE KINASE 3 (PgHK3) and RESPONSE REGULATOR 2 (PgRR2) genes were cloned in P. ginseng and functionally analyzed in Arabidopsis as a two-component system involved in CK signaling. Results: Phenotypic and histological analyses showed that CK increased cambium activity and dormant axillary bud formation in P. ginseng, thus promoting storage-root secondary growth and bud formation. The evolutionarily conserved two-component signaling pathways in P. ginseng were sufficient to restore CK signaling in the Arabidopsis ahk2/3 double mutant and rescue its growth defects. Finally, RNA-seq analysis of CK-treated P. ginseng roots revealed that plant-type cell wall biogenesis-related genes are tightly connected with mitotic cell division, cytokinesis, and auxin signaling to regulate CK-mediated P. ginseng development. Conclusion: Overall, we identified the CK signaling-related two-component systems and their physiological role in P. ginseng. This scientific information has the potential to significantly improve the field-cultivation and biotechnology-based breeding of ginseng.
멜론 흰가루병(Podosphaera xanthii)은 멜론 생산량에 영향을 미치는 중요한 병해 중 하나로 알려져 있다. 작물 육종에 있어서 흰가루병을 포함한 병저항성 계통 육성은 병저항성 관련 유전자들의 유전양식 및 그 유전자들을 조절하는 식물호르몬에 대한 정보는 매우 중요하다. 식물에 있어 흰가루병균 저항성에 관여한다고 알려진 Mildew Resistance Locus O (MLO) 유전자를 멜론 database인 ‘Melonomics’으로부터 14개 동정하여 CmMLO1~14 (ucumis elo MLO)로 표기하였다. 동정된 14개의 CmMLO유전자들의 아미노산 서열을 비교한 결과, 9개의 CmMLO 유전자들은 흰가루병균에 대한 감수성 관련 아미노산 cysteine과 proline이 잘 보존되어 있었지만, 나머지 CmMLO 유전자들은 다른 아미노산 서열을 가지고 있었다. 멜론 흰가루병의 7 race에 대하여 이병성을 나타내는 멜론 계통 ‘SCNU1154’에 멜론 흰가루병균(P. xanthii)을 접종하고, 식물호르몬(metyl jasmonate와 salicylic acid) 처리한 후 qPCR을 통해 CmMLO 유전자들의 상대적인 발현양을 분석한 결과, 멜론 흰가루병 7 race에 대하여 14개의 CmMLO 유전자들 중 3개의 유전자들에서 발현이 증가하였고, 7개의 유전자들은 발현이 감소하였으며, 4개의 CmMLO 유전자들은 race 특이적으로 발현양이 증가 혹은 감소하였다. 또한 14개의 CmMLO 유전자들의 발현양은 methyl jasmonate와 salicylic acid를 처리하였을 때 다양한 발현 양상을 나타내었다. 11개의 CmMLO 유전자들은 salicylic acid 처리하였을 때 발현양이 증가하였으며, 7개의 유전자들은 methyl jasmonate 처리하였을 때 발현양이 증가하였다. 이와 같이, 스트레스에 반응을 보이는 CmMLO 유전자들은 멜론 흰가루병 저항성 계통 육성을 위한 유용한 정보가 될 것으로 기대된다. 본 논문에서 벼 ascorbate peroxidase (OsAPx1) 유전자의 발현 분석을 Northern과 Western 분석을 통하여 유묘 에서는 뿌리, 정단분열조직(shoot apical meristem, SAM), 잎 보다는 잎집에서 더 많이 발현되는 것을 확인하였다. 성숙된 조직에서는 OsAPx1 유전자가 잎을 제외하고는 뿌리, 줄기, 꽃에서 강하게 발현되었다. 또한 이 OsAPx1 유전자는 벼 곰팡이 병원균인 벼 도열병 및 세균성 병원균인 흰빛잎마름병에도 반응하였고 특히 흥미있게도 OsAPx1 유전자는 식물호르몬에 대해서 서로 다르게 발현 양상을 보였다. 이 유전자는 자스몬산(JA)에 대해서는 강한 발현 을 보였지만 반대로 살리실산(SA) 및 ABA와 같이 처리된 세포에서는 강한 발현 억제를 보였다. 이는 이 유전자가 JA에는 반응하지만 SA와 ABA하고는 서로 길항작용을 하는 것으로 보인다. 근연관계분석을 통하여 OsAPx1유전 자가 애기장대의 AtAPx1 와 거의 유사하여 AtAPx1 결손 라인을 가지고 표현형 조사를 실시하였다. 그 결과, 외부에서 H2O2를 처리하였을 때에 O2- 와 H2O2의 축적이 wild type과 비교하여 AtAPx1 결손 라인에서는 현저히 높았다. 따라서 본 연구를, 통하여 OsAPx1 유전자는 벼에서 산화 환원 균형 유지를 통하여 다양한 세포 분화발달 및 병원균 방어에도 관여하며 이 유전자의 발현은 JA의 신호전달에 의해서 매개되는 것으로 예상이 된다.
작물 원형질체를 배양하면서 세포의 생존과 노화 그리고 고사의 상태를 대사적으로 정확히 판단이 가능한 marker물질을 확인하고 간편한 측정법을 확립하기 위하여 일련의 연구를 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 담배와 벼의 원형질체를 나출하여 MS와KM-8P를 기본배지로 하여 2,4-K와 CM을 첨가하였던 바, ABA는 경시적으로 감소하였으며 CM첨가에 의해서 더 많이 감소하였다. GA$_3$, IAA, zeatin은 반대로 증가하는 경향을 나타내었다. 특히 ABA의 감소율과 zeatin의 증가율이 가장 크게 나타났다. 2. Osmoticum(mannitol)을 적정 농도 이상으로 처리하였을 때 ABA는 일시적으로 증가되었다가 감소되었고, 다른 성장 호르몬은 모두 증가폭이 농도가 증가될수록 둔화되었다. 3. Supermine 10mM과 NaCl을 처리하였을 때 담배와 벼에서 각 호르몬의 증감은 서로 반대로 나타났다. 특히 담배는 NaCl의 stress가 크게 나타났다. 그리고 배양 후반기에는 원형질체의 분열이 중지되었다. 4. 원형질체의 배지 성분을 제거하고 32$^{\circ}C$로 고온 배양에 의해서 인위적으로 노화를 촉진시켰을 때 세포의 활력은 48시간 이후에 크게 감소되었다. 세포의 활력 상실과 반비례적으로 ABA는 배양 10일까지 크게 증가되었다. 이와 반대로 성장 호르몬들은 모두 증가되지 않고 감소가 이루어졌다. 특히 zeatin의 감소가 크게 나타났다. 5. 이상의 결과에 따라서 식물 호르몬은 원형질체가 처한 환경 요인이나 부여된 어떤 조건에 따라서 민감하게 변동하고 있음을 확인하였다.
나무의 유전 공학적 연구를 위해서는 목본 고유의 유전자 및 프로모터 연구가 필수적이다. 우리는 포플러(P. alba ${\times}$ P. glandulosa)의 Pagns-LTP 유전자의 867 bp 프로모터를 분리하였고, ${\beta}$-glucuronidase (GUS) reporter 유전자를 이용한 프로모터의 형질전환 포플러를 제작하여 특성 분석하였다. Pagns-LTP 유전자는 어린뿌리에서 강하게 발현되었고 어린잎에서는 약하게 발현되었으며, 그밖에 다른 조직에서는 발현되지 않았다. 또한, 프로모터의 활성은 뿌리와 어린잎에서 한정되었으며 어린뿌리의 세포 전체에서 강한 활성을 나타내었다. 이에 포플러 ns-LTP 프로모터 내의 cis-element를 조사하고 현사시나무에서 Pagns-LTP 프로모터를 분리한 후 활성을 분석하였다. 프로모터 내의 cis-element를 분석한 결과, 조직 특이적 발현과 호르몬 및 스트레스에 반응하는 다양한 cis-element가 존재함을 확인하였다. 이를 통해 포플러의 ns-LTP는 생장뿐만 아니라, 스트레스에도 관여할 것이라고 추측할 수 있었다. 본 연구는 목본의 유전자 기능 분석 및 다양한 응용 연구를 위해 유용하게 이용될 수 있는 도구로서의 가능성을 제시하였다.
본 실험은 기내 배양된 홍경천으로부터 캘러스를 유도하고 가장 잘 자라는 생장조건을 찾기 위해 수행되었다. 실험 결과를 종합해 보면 캘러스의 증식을 위한 배양조건은 최적배지를 조사한 결과,2B$_{5}$배지에서 생장율이 가장 좋았으며, 탄소원으로는 sucrose를 3%로 첨가했을 때 생장율이 가장 높았다. 질소원으로써는 49.6mM KNO$_3$와 2.0mM (NH$_4$)$_2$SO$_4$를 첨가했을 때 생장율이 가장 좋았으며, 인산의 농도는 2.16mM에서 높은 생장율을 보였다. 암배양과 광배양을 비교했을 때 암배양의 경우가 생장율이 높았으며,pH6에서 세포의 생장율이 가장 좋은 조건임을 확인되었다. 기내에서 배양된 캘러스와 자연산 뿌리는 TLC로 분석하여 salidroside를 확인하였으며, HPLC로 salidroside를 정량한 결과, 자연상태 홍경천의 뿌리는 건중량 당 0.17%으로 나타났으며 5% sucrose처리구에서 0.41%의 생산성을 보여, 자연산 홍경천의 뿌리에 비하여 캘러스에서 salidroside의 함량이 높은 것으로 나타났다.
