  • 1. 원유와 시유 40점에서 내열성균으로 생각되는 330균주의 세균이 분리되었다. 2. 현미경적 관찰, 생리적시험, 단백질분해성과 지방분해성으로 보아 Bacillus stearothermophilus 125균주, Bacillus coagulans 69균주, Bacillus subtilis 57균주, Bacillus cereus 76균주, Lactobacillus thermophilus 3균주로 동정되었다. 3. 이들 반이상의 균주들은 skim milk배지에서 $85^{\circ}C$, 15분간 처리에도 생존할 수 있었다.

  • Dextrinogenic and saccharogenic amylase activities of Rhizopus isolated from various substrates collected throughout South Korea are measured, and their amylase activities are surveyed from taxnomical, ecological, and physiological viewpoints. Among the 151 strains of Rhizopus, strain number 49 exhibiting most prominent saccharogenic amylase activity is selected as a best strain, and strain number 17 is selected as a best strain for dextrinogenic amylase activity. Amylase activities of Rhizopus are so variable in different strains even in the same species. In general, however, R. japonicus, R. liquefaciens, and R. shanghaiensis show high saccharogenic amylase activities, and R.chiuniang, R.achlamydosporus, and R.bahrnensis show high dextrinogenic amylase activities, while R.nigricans exhibits lower activities for both amylase. The strains isolated from kneaded cereals, such as "meju" and "kockja", exhibited very strong dextrinogenic and saccharogenic amylase activities as compared with the strains isolated from other sunstrates. The mean value of dextrinogenic amylase activities of the strains from southern inland region and saccharogenic amylase activities of the strains from middle coast region are, more or less, higher than those of the strains from the other regions. In the culture at 30.deg.C, those strains which exhibit better mycelial and air mycelial growth than sporangial formation show very high dextrinogenic and saccharogenic amylase activities. amylase activities.

The Effect of Rotating Magnetic Field on Enterotoxin Genes Expression in Staphylococcus Aureus Strains

  • Staphylococcus aureus cultures exposed to rotating magnetic field (RMF) were studied in order to analyse the possible induced changes in staphylococcal enterotoxin genes (se) expression. Liquid cultures of S. aureus strains carrying different se were exposed to the RMF of magnetic frequency 50 Hz and magnetic induction 34 mT for 10 h at $37^{\circ}C$. Three time points of bacterial growth cycle were considered for RNA extractions. Gene expression analyses were evaluated using real-time quantitative PCR method. The present study confirmed, that the RMF can stimulate the growth rate of S. aureus cultures in comparison to the unexposed controls, while the stimulation is not strain dependent. The studies have also shown, that the RMF, depending on the exposure time but regardless the bacterial strain, can influence on the expression of various se. In general, except for sea, as a result of bacterial exposure to the RMF through subsequent growth phases, the expression of se decreased, reaching the values below results recorded for unexposed controls. In the case of sea expression remained at a lower level as compared to the control, regardless the time of exposition.

Isolation and characterization of new Methanosarcina mazei strains KOR-3, -4, -5, and -6 from an anaerobic digester using pig slurry

  • Objective: An experiment was conducted to isolate and identify new methanogens in Korea from an anaerobic digester that uses pig slurry. Methods: An anaerobic digestate sample was collected from an anaerobic digester using pig slurry. Pre-reduced media were used for the growth and isolation of methanogens. Growth temperature range, pH range, NaCl concentration range, substrate utilization, and antibiotic tolerance were investigated to determine the physiological characteristics of isolated methanogens. The isolates were also examined microscopically for their morphology and Gram-stained. Polymerase chain reaction of 16S rRNA and mcrA gene-based amplicons was used for identification purpose. Results: Four strains, designated KOR-3, -4, -5, and -6, were isolated and were non-motile, irregular coccoid, and 0.5 to $1.5{\mu}m$ in diameter. Moreover, the cell walls of isolated strains were Gram-negative. KOR-3 and KOR-4 strains used acetate for methane production but did not use $H_2+CO_2$, formate, or methanol as a growth substrate KOR-5 and KOR-6 strains utilized acetate, methanol, and trimethylamine for methanogenesis but did not use $H_2+CO_2$ or formate as a growth substrate. The optimum temperature and pH for growth of four strains were $39^{\circ}C$ and 6.8 to 7.2, respectively. The optimum concentration of NaCl for growth of KOR-3, KOR-5, and KOR-6 were 1.0% (w/v). The optimum NaCl concentration for KOR-4 was 0.5% (w/v). All of the strains tolerated ampicillin, penicillin G, kanamycin, streptomycin, and tetracycline; however, chloramphenicol inhibited cell growth. Phylogenetic analysis of 16S rRNA and mcrA genes demonstrated that strains KOR-3, -4, -5, and -6 are related to Methanosarcina mazei (M. mazei, 99% sequence similarity). Conclusion: On the basis of physiological and phylogenetic characteristics, strains KOR-3, -4, -5, and -6 are proposed to be new strains within the genus Methanosarcina, named M. mazei KOR-3, -4, -5, and -6.

토양 특성에 따른 다양한 희소방선균의 분리 (Isolation of Rare Actinomycetes on Various Types of Soil)

  • Actinomycetes occur in a wide range of environments and many more actinomycetes remain to be detected in the natural environment. In isolation stages, selection of the environment as a source of useful isolates is important. Two hundred and eighteen strains were isolated on Bennet's agar and 346 strains were on humic acid-vitamin agar from five each paddy field, field, forest, grass land, riverside soil samples. These isolates were identified to the genus level based on morphological and physiological characteristics. Among them, 386 strains were Streptomyces, 49 strain were Nocardia, 35 strains were Microbispora and Micromonospora each, 15 strains were Nocardiopsis, 13 strains were Actinomadura, 10 strains were Streptosporangium, and the others were isolated rarely. According to soil type, Nocardia, Micromonospora, Microbispora and Streptosporangium were dominant in paddy field, Microbispora Nocardia, Nocardioides and Micromonospora were dominant in field, Nocardia, Micromonospora, Microbispora and Actinomadura were dominant in grass land, Nocardia, Micromonospora and Microbispora were dominant in forest, Nocardia, Microbispora and Micromonospora were dominant in riverside. Generally, Nocardia, Micromonospora, Microbispora and Actinomadura were isolated in all kinds of soils, Streptosporangium were paddy field, Dactylosporangium were forest, Nocardiopsis were field, forest and riverside.

Comprehensive Analysis of Proteomic Differences between Escherichia coli K-12 and B Strains Using Multiplexed Isobaric Tandem Mass Tag (TMT) Labeling

  • The Escherichia coli K-12 and B strains are among the most frequently used bacterial hosts for scientific research and biotechnological applications. However, omics analyses have revealed that E. coli K-12 and B exhibit notably different genotypic and phenotypic attributes, even though they were derived from the same ancestor. In a previous study, we identified a limited number of proteins from the two strains using two-dimensional gel electrophoresis and tandem mass spectrometry (MS/MS). In this study, an in-depth analysis of the physiological behavior of the E. coli K-12 and B strains at the proteomic level was performed using six-plex isobaric tandem mass tag-based quantitative MS. Additionally, the best lysis buffer for increasing the efficiency of protein extraction was selected from three tested buffers prior to the quantitative proteomic analysis. This study identifies the largest number of proteins in the two E. coli strains reported to date and is the first to show the dynamics of these proteins. Notable differences in proteins associated with key cellular properties, including some metabolic pathways, the biosynthesis and degradation of amino acids, membrane integrity, cellular tolerance, and motility, were found between the two representative strains. Compared with previous studies, these proteomic results provide a more holistic view of the overall state of E. coli cells based on a single proteomic study and reveal significant insights into why the two strains show distinct phenotypes. Additionally, the resulting data provide in-depth information that will help fine-tune processes in the future.

내열성 세균에 관한 연구 1 (Studies on Thermal Resistance Bacteria. (Part. 1))

  • The purpose of this paper is to study on the morphological and physiological nature generally observed for the identifications, and the four strains of thermal resistanting aerobic bacteria isolated from swelled cans and the different soils collected from the wide area in Korea. The results obtained in the light of the Bergey's Manual for the identification of the bacteria, have been shown that the four strains of bacteria are pertained to Bacillus subtilis. The optimum temperature, pH and the thermal resistance (2.5 min. at $121^{\circ}C$) of the bacteria have been measured.

자연 동굴 토양 방선균의 속 다양성 분포

  • Total 276 soil actinomycete strains were isolated from 46 soil samples collected at domestic natural caves; the Kosu, Chundong, and Nodong caves at Chungbook province, the Kossi cave at Kangwon province, the Sungruye cave at Kyungbook province, the Hyupjae, Ssangyong, and Manjang caves at Cheju province. All of these isolates were identified to the genus level based on morphological and physiological characteristics. As the result, 52.5% of those isolates were Streptomyces, 16.3% were Micromonospora, 22.8% were Nocardioform group, 1.1% were Actinomadura, 0.3% were Nocardiopsis, 0.3% were Streptosporangium, 0.3% were Nocardioides, 1.4% were Kineosporia, 4.7% were the others. Streptomycete strains were the most abundant, but were relatively less comparing to general distribution pattern. Nocardioform and Micromonospora strains were quite abundant, and other rare actinomycete groups were somewhat abundant comparing to general distribution pattern previously reported. Especially Nocardioform strains were highly abundant at almost of the natural caves.

Phylogenetic Analysis of Trichaptum Based on the RFLP of PCR-Amplified DNAs

  • To infer phylogenetic relationships between species of Trichaptum (Polyporaceae), RFLP analyses of PCR-amplified DNAs were accomplished. Regions coding for ITSs of nuclear SSU rRNA genes and for mitochondrial SSU rRNA genes from thirteen strains of four Trichaptum species (T. abietinum, T. biforme, T. fusco-violaceum, and T. laricinum) were amplified and digested with eight restriction enzymes. All the fragmentation patterns were characterized and coded as 0/1 for the absence/presence of fragments. A phylogenetic tree based on the combined data sets was constructed using the Dollo parsimony method. While every two strains of T. abietinum, T. biforme, T. fusco-violaceum, and T. laricinum formed an independent group, the other strains of T. abietimum and T. fusco-violaceum made mixed groupings among compared strains. It is inferred that T. abietinum and T. fusco-violaceum have more variations, possibly geographic or physiological ones, than other species in the genus.

돼지감자를 이용한 고농도 알코올발효 균주의 탐색 (Screenig and Indentification of Wild Strains for the Production of High Concentration of Alcohol from Jerusalem artichoke Tubers)

  • Yeast screening for effective production of alcohol from Jerusalem artichoke tubers as an alternative energy source was performed. Inulin assimilative strains with high alcohol tolera- nce were isolated from wild sources and cultured in the liquid media of Jerusalem artichoke powder varying its concentraion from 15 to 30%. As a result, four strains of 2,445 isolates showing the inulin assimilation were selected as alcohol fermentative and alcohol tolerant yeasts. These strains were assignated to be Kluyveromyces marxianus F043 and Kluyveromyces sp. F173, E040, and F334, respectively, by their cultural and physiological characteristics. The F043 strain produced ethanol of 98.1 g/l in the 25% Jerusalem artichoke medium for 3 days.
