RAW 264.7 세포에서 Euryale ferox Salisbury 추출물의 항산화기전을 통한 산화적 스트레스.염증반응 억제효과 규명 (Suppressive Effect of Euryale ferox Salisbury Extracts on Inflammatory Response in LPS-stimulated RAW 264.7 Cells through the Antioxidative Mechanism)

  • The stems and branchs of Euryale ferox Salisbury (EF), are used in Chinese herbal medicine for latent-heat-clearing, antipyretic, detoxicant and anti-inflammatory ailments. This plant is used worldwide for the treatment of many types of inflammatory disease including respiratory infections, diabetes mellitus, rheumatoid arthritis and play an important role in the immune reaction. Topical natural antioxidants are a useful strategy for the prevention of oxidative stress mediated inflammatory disease. Plants produce significant amounts of antioxidants to prevent the oxidative stress caused by photons and oxygen, therefore they represent a potential source of new compounds with antioxidant activity. This study was designed to evaluate whether EFEA (ethylacetate fraction of EF) may ameliorate oxidative stress and inflammatory status through the antioxidative mechanism in LPS-stimulated RAW 264.7 murine macrophage cell line. Treatment of RAW 264.7 cells with EFEA significantly reduced LPS-stimulated inflammatory response in a dose-dependent manner. In conclusion, the EF extracts have anti-inflammatory effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and chronic inflammatory disease.

  • There is currently increased interest in the identification of antioxidant compounds that are pharmacologically potent and have low or no side effects. Plants produce significant amounts of antioxidants to prevent the oxidative stress caused by photons and oxygen, therefore they represent a potential source of new compounds with antioxidant activity. FARFARE FLOS has been frequently used on the respiratory system including bronchitis, phthisis. In this study, the antioxidant activity of extract from FF was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on Cu2+-induced human LDL oxidation. The FF extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation. And this study was designed to evaluate whether FFEA may ameliorate oxidative stress and inflammatory status through the antioxidative mechanism in LPS-stimulated RAW 264.7 murine macrophage cell line. Treatment of RAW 264.7 cells with FFEA significantly reduced LPS-stimulated inflammatory response in a dose-dependent manner. In conclusion, the FF extracts have anti-oxidative and anti-inflammatory effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.

미국이민 한국인의 스트레스 반응 양상과 생활사건과의 상관 연구 (Correlation Study between Stress Responses and Life Events as a Stressor)

  • Koreans are one of the fastest growing immigrant groups in America. Stress responses and stressors among this large cultural minority has been rarely been studied by nursing researchers. Adjusting to life in foreign country produces a great deal of stress. Differences in culture, language, expectations and social behavior can lead to misunderstandings between health care providers and clients. These misunderstandings are not well accounted for in health assessment. This study investigated the relationship between life events or / and daily activities as a stressor and the symptoms of stress among a sample of Korean immigrants in America. The symptoms of stress scale (SOS) was used to identify stress responses and open-ended questions were used to identify life events and daily activities considered by the respondents to be stressful. A simple random sample of 283 subjects was selected from the Directory of the Korean Society of Chicago, New York, Los Angeles, Philadelphia and Seattle. Demographically, the subjects ranged in age from 20 to 69 years, and the percentage of women and men was approximately 50% each. Almost ninety percent of the subjects were highly educated, 17% owners of business, 19% white collar professionals, 14% employed in sales or as skilled /unskilled labor, 27% as housewives and students and 3% had no occupation. The total group SOS mean was 0.8042 ; the SOS men for man was 0.7371, and for women was 0.8713. The stress response of this subject group was high, -the stress response of women higher than that for men. In an earlier study(June, 1992) with another sample, the total mean SOS score was similar to this one. The main stressful life events or / and daily activities were, in order, economic problems (N=97), interpersonal problems (N=68), children care problems (N=258), health problems (N=49), communication problems (N=42), family problems (N=38), worry about future career (N=36), and religious problems (N=25). There was a significant difference in the SOS means between the group that expressed life events or / and daily activities to be stressful and the group that did not. Interpersonal relationships and economic and family problems were stressors for those who complained about peripheral manifestations. cardiopulmonary symptoms, central-neurological symptoms, gastrointestinal symptoms, muscle ten-sion, habitual patterns, depression, anxiety, emotional irritability and cognitive disorganization. In summary, interpersonal relationships and economic and family problems influenced stress response manifestations. Income, the number of people in the family, the year of immigration. the level of education, and marital status were related to physiological and psychosocial stress responses.

실외 온난화 처리에 따른 낙엽송 묘목의 생리 및 생장 반응 (Effect of Experimental Warming on Physiological and Growth Responses of Larix kaempferi Seedlings)

  • Seedling stage is particularly important for tree survival and is easily influenced by warming. Therefore, air temperature being increased due to climate change may affect physiological traits and growth of seedlings. This study was conducted to investigate the physiological and growth responses of Larix kaempferi seedlings to open-field experimental warming. 1-year-old and 2-year-old L. kaempferi seedlings were warmed with infrared lamps since April 2015 and April 2014, respectively. The seedlings in the warmed plots were warmed to maintain the air temperature to be $3^{\circ}C$ higher than that of the control plots. Physiological responses (stomatal conductance, transpiration rate, net photosynthetic rate and total chlorophyll content) and growth responses (root collar diameter (RCD), height and biomass) to experimental warming were measured. Physiological and growth responses varied with the seedling ages. For 2-year-old L. kaempferi seedlings, stomatal conductance, transpiration rate and net photosynthetic rate decreased following the warming treatment, whereas there were no changes for 1-year-old L. kaempferi seedlings. Meanwhile, total chlorophyll content was higher in warmed plots regardless of the seedling ages. Net photosynthetic rate linked with stomatal conductance also decreased due to the drought stress and decrease of photosynthetic efficiency. In response to warming, RCD, height and biomass did not show significant differences between the treatments. It seems that the growth responses were not affected as much as physiological responses were, since the physiological responses were not consistent, nor the warming treatment period was enough to have significant results. In addition, multifactorial experiments considering the impact of decreased soil moisture resulting from elevated temperatures is needed to explicate the impacts of a wide range of possible climate change scenarios.

Changing Salinity Affects Hematological and Histological Response in Hybrids and Hybrid Triploids between River Puffer, Takifugu obscurus and Tiger Puffer, T. rubripes

  • We investigated the physiological responses of the river puffer, Takifugu obscurus, the tiger puffer, T. rubripes, a hybrids of the two (river puffer${\times}$tiger puffer) and a hybrid triploids to acute changes of salinity from 30 psu to 0 psu and from 0 psu to 30 psu. The blood and plasma factors of each species were elevated for 48, 72, or 96 hrs, and thereafter decreased due to hyper-osmoregulation and hypo-osmoregulation. In hyper-osmoregulation and hypo-osmoregulation, the cortisol concentration of river puffer, hybrids, hybrid triploids and tiger puffer increased for 12 or 48 hrs, and decreased thereafter. Chloride cells in the gill filaments of each species increased with increasing salinity, and melano-macrophages in the kidney tissue of each species increased with decreasing salinity. In conclusion, the hematological and stress responses of the hybrids were between those of the river puffer and tiger puffer, and the hematological responses of the hybrid triploids were higher than those of the other groups. The stress response of the hybrids was more sensitive than that of the hybrid triploids. In all groups, the histological responses of kidney in hyper-osmoregulation were more sensitive than those in hypo-osmoregulation.

Development and Ergonomic Evaluation of Spring and Autumn Working Clothes for Livestock Farming Workers

  • Objective:In this study, we designed working clothes for livestock farmers to wear in spring and autumn to improve their work efficiency, conducted a physiological test on their performance, and evaluated their comfort. Background: In recent years, livestock farming in Korea has expanded, yet farmers' safety and sanitation levels remain low in hazardous environments that include organic dust, toxic gas, and heat stress, as well as the risk of accidents. Furthermore, most livestock farmers wear ordinary or dust-resistant clothes that are unsuitable for rearing livestock and compromise their safety and health. Thus, it is important to design specialized working clothes for livestock farmers that are comfortable and that minimize their health and safety risks. Method: To this end, we examined the literature on livestock (poultry, swine, and cattle) farmers' safety and sanitation issues, designed appropriate working clothes, and tested them in terms of sensory feel, physiological response, and subjective comfort. Results: The respondents expressed satisfaction with the new working clothes. The results of a physiological test showed a decline in temperature and humidity inside the clothes, a lower pulse rate, and a lower oxygen intake compared to the measurements taken when famers wore their previous working clothes. This indicates a fall in heat stress and fatigue, which was mostly consistent with the results of the assessment of subjective comfort. Conclusion: The results of the analysis show an improvement in the comfort of the new working clothes compared to the dust-resistant clothes that are widely worn. Based on this study, the new working clothes need to be further tested and evaluated to improve the design. Application: This study is expected to contribute to designing better working clothes for livestock farmers.

고주파 처리에 따른 파밤나방(Spodoptera exigua)의 생리적 스트레스의 생화학적 분석 (Biochemical Analysis of Physiological Stress Induced by High Frequency Sound Treatment in the Beet Armyworm, Spodoptera exigua)

  • 고주파 처리는 파밤나방(Spodoptera exigua)의 생리변화를 유발시켜 섭식행동, 발육 및 면역반응의 변화를 초래한다. 본 연구는 이러한 고주파의 영향을 파밤나방의 생화학적 변화를 통해 분석했다. 고주파(5,000 Hz, 95 dB) 처리는 중장 상피세포의 단백질 합성과 분비를 억제시켰다. 또한 이 고주파 처리는 중장의 인지질분해(phospholipase $A_2$) 소화효소의 활성을 현격하게 억제시켰다. 고주파 처리는 세 종류의 열충격단백질과 지질운송단백질(apolipophorin III)의 유전자 발현을 변동시켰고 이러한 변화는 중장 조직에서 뚜렷했다. 혈림프 혈장에 존재하는 지질 및 유리당의 함량이 고주파 처리에 의해 현격하게 증가했다. 이러한 결과는 고주파 처리가 파밤나방의 체내 생화학적 변화를 유발시켜 생리적 교란을 유도하는 스트레스로 작용한다는 것을 제시하고 있다.

Effect of Oral Administration of DiakurTM (a Glucose and Electrolytes Additive) on Growth and Some Physiological Responses in Broilers Reared in a High Temperature Environment

  • An experiment was conducted to determine effects of oral administration of $Diakur^{TM}$ (an additive of glucose and electrolytes for young calves) on growth performance and some physiological responses in male broilers reared in a high temperature. A 2 by 3 factorial arrangement test of 2 temperatures (24 and $36^{\circ}C$) and 3 levels of oral administration of the glucose and electrolytes additive, $Diakur^{TM}$, (0, 150 and 300 mg/day/100 gBW) were applied in the experiment. Male broiler chicks (2 weeks of age) were assigned to six groups and received dietary and temperature treatments for 7 days. The additive of glucose and electrolytes was suspended with water and intubated into crop twice a day (08:00 and 17:00). Oral administration of the additive prevented decreases in food intake and growth rates in broilers due to exposure of the hot environment. Oral administration of the additive also improved a lowered electrolyte ($Na^+$ + $K^+$ - $Cl^-$) balance in plasma, low mitogenic response of blood mononuclear cell and an increase in glucose concentration due to exposure to the high environmental temperature. Oral administration of the additive increased rectal temperature regardless of environmental temperatures. On the other hand, blood pH, $pCO_2$ and $HCO_3$ - concentration, and plasma creatine kinase activity were not affected by the oral administration. The results suggested that oral administration of the glucose and electrolytes additive, $Diakur^{TM}$ during heat stress did not only prevent decrease in growth performance, but also normalized some physiological and immunological responses in male broilers.

Different Physiological Response to Salt in Salt Tolerant Rice Mutants Induced by Gamma-Mutagenesis

  • When plants undergo stress, Reactive oxygen species (ROS) which remove bad elements such as mildew and virus is activated in plant body. However, if ROS is excessively increased, plant will be harmed itself by destruction of cell and signal system and phenomenon of lipid peroxidation. In order to identify content of lipid peroxidation and activity of some enzymes scavenging ROS, phenotypical and physiological analysis was performed with two mutant lines, Till-II-877 and Till-II-894, comparing with cv. Dongan (WT). In phenotype analysis, two mutant lines give to well-conditioned growth better than WT in since 5 days after salt treatment. In enzyme activities, there was a modest difference in the content of catalase (CAT) and peroxidase (POD) between Till-II-877 and Till-II-894, two mutant lines showed high levels in CAT contents than WT. However, they express low levels in POD contents. In MDA analysis, the content of Till-II-877 was higher than that of WT, but Till-II-894 was lower. This result indicates that two mutants have different mechanism against salt stress.

Autophagy Dysregulation and Obesity-Associated Pathologies

  • Autophagy is one of the major degradative mechanisms that can eliminate excessive nutrients, toxic protein aggregates, damaged organelles and invading microorganisms. In response to obesity and obesity-associated lipotoxic, proteotoxic and oxidative stresses, autophagy plays an essential role in maintaining physiological homeostasis. However, obesity and its associated stress insults can often interfere with the autophagic process through various mechanisms, which result in further aggravation of obesity-related metabolic pathologies in multiple metabolic organs. Paradoxically, inhibition of autophagy, within specific contexts, indirectly produces beneficial effects that can alleviate several detrimental consequences of obesity. In this minireview, we will provide a brief discussion about our current understanding of the impact of obesity on autophagy and the role of autophagy dysregulation in modulating obesity-associated pathological outcomes.
