Photostability of Anthocyanin Extracted from Purple-Fleshed Sweet Potato (자색고구마에서 추출한 anthocyanin의 광안정성)

  • Effect of EDTA, gallic acid, phosphoric acid, propyl gallate and sodium ascorbate on the photostability of anthocyanin extracted from purple-fleshed sweet potato were investigated by measuring the absorbance at 530 nm with a spectrophotometer. White light of 20,000 lux was used to illuminate the pigment and the temperature was $20^{\circ}C$. EDTA and sodium ascorbate were more effective in improving the photostability of the pigment when added at least 100 or 1,000 ppm, respectively.

  • To minimize the use of animals in toxicity testing, and to reduce the cost in vivo test, more rational test method was described which determines, in the same animal, photoxic and photoallergic potential of a substance, and is daptable to routine testing. The other purpose of this study was to investigate the usefulness of in vivo alternatives ; photostability and spectrophotometric carbonyl assay. In this modified photosensitization model, animal numbers and resting periods, the number and method of topical application were simplified. Two positive photoreactive agents, Benzocaine and 6-methyl coumarine, showed a similar photoallergic potential to that of Ichikawa's method. Two sunscreens, Octyl methoxy cinnamate, Butyl methoxyl dibenzoyl methane, hardly showed photoallergic potentials. The photostability test could be used in the step of prescreening of photosensitization potential because most of the photoreactive agents represented the reduction of more than 20% in the absorbance. And photoreactive agents have a high potential of photosensitization in the sddessment of spectrophotometric carbonyl level although two sunscreens have a low possibility of photosensitization. Therefore this method was assumed as a valuable in vivo alternatives in the respect even in the very low concentrations which phototoxicity test using almonella showed no phototoxic potential.

  • Jawarishe Jalinoos (JJ) is an orally used formulation available in semisolid dosage form, prepared with powdered plant materials mixed in honey or sugar syrup. It has many admirable pharmacological effects and used in Unani medicine to treat various acute and chronic disorders since ancient times. The ICH Harmonised Tripartite Guideline stated that photostability testing should be an essential part of stability testing to confirm that light exposure does not result in an unacceptable change in drugs substance and finished products. To date, the effect of light on JJ is not studied, in this study photostability evaluation of JJ was carried out. The test sample was manufactured with genuine ingredients in the in-door pharmacy of the National Institute of Unani Medicine. JJ was packed in two transparent polyethylene terephthalate airtight containers. The first sample was analysed at zero-day and the second sample was placed in a stability chamber subjected to light challenge with an overall illumination of 1.2 million lux hours combined with near ultraviolet energy of 200-watt hours per square meter by using option 2, along with 30±2℃ temperature and relative humidity 70±5%. Analysis of both finished products showed no considerable changes in organoleptic characters. Less than 5% variation was observed in physicochemical parameters. HPTLC fingerprinting showed justifiable variation. Microbial load and specific counts were within the limit prescribed by WHO. As no unacceptable changes were noted in JJ subjecting to light challenge, it is concluded that JJ is a photostable Unani compound formulation.

Photostability Evaluation of a New Sunscreen Agent, Methoxycinnamidopropyl Polysilsesquioxane (신규 자외선차단제인 Methoxycinnamidopropyl Polysilsesquioxane의 광안정성 평가)

  • The new sunscreen agent, methoxycinnamidopropyl polysilsesquioxane, is manufactured as polymeric particles with an organic/inorganic hybrid composition. We have already reported the manufacturing method, physical properties, and sunprotection effects of methoxycinnamidopropyl polysilsesquioxane. In this study, we evaluated the photochemical properties and photostabilities of methoxycinnamidopropyl polysilsesquioxane that has the same functional group as a typical organic sunscreen agent, ethylhexyl methoxycinnamate (EHMC). Using the correlation of UV absorbance and fluorescence, we studied photostabilizers to enhance the photostability of methoxycinnamidopropyl polysilsesquioxane. Finally, we confirmed that octocrylene, ethylhexyl methoxycrylene, and bis-ethylhexyloxyphenol methoxyphenyl triazine were good photostabilizers for methoxycinnamidopropyl polysilsesquioxane.

Fluorescent Magnetic Silica Nanotubes with High Photostability Prepared by the Conventional Reverse Micro-Emulsion Method

  • Magnetic fluorescent silica nanotubes were fabricated using reverse micro-emulsions coupled with conventional sol-gel methods. Anodic aluminum oxide templates were used to separate spatially the magnetic and the fluorescent moieties on individual nanotubes and so prevent quenching of the fluorescence. C18 and fluorescent layers were deposited sequentially on silica. Magnetism was then obtained by the introduction of pre-made magnetic nanoparticles inside the nanotubes. The photo- and chemical stabilities of nanotubes were demonstrated through dye release and photobleaching tests. The produced nanotubes did not show fluorescence quenching upon the addition of the nanoparticles, an advantage over conventional spherical fluorescent magnetic nanoparticles. High photostability of nanotubes, magnetism and biocompatiblily make them potentially useful in bioanalysis.

Synthesis of Bis(dithiobenzil) Metal Complex and Its Photostability (Bis(dithiobenzil) 금속 화합물의 합성 및 광안정성)

  • Bis(dithiobenzil) metal complex, used as functional NIR absorbing dye and photostabilizer, was synthesized using bezoin and anisoin as intermediate compounds. And squarylium, a charge generation material, was synthesized to find its photostability effect. The structure of the product was determined by $^1H-NMR$ and FT-IR and the thermal property was analyzed by DSC and TGA. Optical property and photostability were determined by UV-Vis-NIR spectroscopy. High absorbance was obtained in the NIR range and maximum absorbing wavelength was shifted depending on the nature and position of substituent in the bis(dithiobenzil) metal complex. The photofading effect of squarylium decreased by the addition of bis(dithiobenzil) metal complex.

Phototoxicity studies of LB20304a (LB20304a의 광독성시험)

  • The phototoxic potentials of LB20304a a new quinolone compound being developed by LG Chemical Ltd, and reference compounds (Ciprofloxacin; CPFX, Enoxacin; ENX and Lomefloxacin; LFLX) were compared in a murine model. in addition photostability of these compunds was studied after irradiation with long-wave UV light(UVA, 0, 0.3 1 or 3 Joule/$Cm^2$) When hairless mice(9 to 11 weeks old 19-26g) were orally administered with different dose levels of test compunds and exposed to UVA(40J/$cm^2$) inflammatory reactions were observed in a dose dependent manner. Among the compounds tested, LB20304a demonstrated the least phototoxic effects and showed no inflammatroy lesions at a dose level of 100mg.kg (Low dose). ENX and LFLX demonstrated much greater phototoxic reactions while CPFX showed similar or slightly greater phototoxic reactions compared to LB20304a. Similar to the in-vivo results the solutions of LB20304a and CPFX irradiated with UVA demonstrated reduced spectral changes compared to those of ENX and LFLX. In conclusion these data suggest that phototoxic potencies of the quinolones tested were; LFLX > ENX > CPFX $\geq$ LB20304a. No phototoxic dose of LB20304a in mice was 100 mg/kg.
