  • The syntheses and properties of polynuclear metal complexes have been reported to develop the easy syntheses and noble photo-characteristics of those complexes for photodynamic diagnosis (PDD) or photodynamic therapy (PDT). We have been focused on the design and synthesis of polynuclear lanthanide dendritic molecule due to long life time of fluorescence. Therefore, we will be presented on the design of home (Eu or Gd) or hetero (Tb or Lu) polynuclear lanthanide dendritic molecule.

  • The preparation of dendritic molecule for photodynamic diagnosis (PDD) or photodynamic therapy (PDT) has been interested on design and synthesis of macromolecule toward a new generation. Herein, the binding site of polyether group is an important role on the construction of macromolecule toward a new generation. Therefore, we will be presented on the preparation of dendritic molecule having the binding site.

Photodynamic Therapy for Methicillin-resistant Staphylococcus aureus Using Various Photosensitizer

  • The aim of this study was to evaluate the photodynamic effect of various photosensitizing agents against methicillin-resistant Staphylococcus aureus (MRSA). MRSA was exposed to light from a 632 urn diode laser (15 J/$cm^2$) in the presence of various photosensitizer, such as photofrin, photogem, radachlorine and ALA. In vivo study was performed using ICR mice. Twenty eight mice had a standard wound ($100\;mm^2$) created on the dorsum, and MRSA was inoculated into the wound region. The four groups were classified as follows: (1) the untreated control group (bacteria alone), (2) the bacteria plus light group (15 J/$cm^2$), (3) the bacteria plus photofrin group (kept in the dark), and (4) the photodynamic therapy (PDT) group (bacteria, photofrin, and light). After photofrin (dose 1 mg/kg) injection, the experimental group was irradiated with 632 urn diode laser (15 J/$cm^2$) for 30 minutes after In vitro results of PDT showed the complete killing of MRSA at the photofrin, radachlorine, and photogem However, ALA-PDT was ineffective on MRSA viability. In vivo results showed that photofrin has therapeutic effect on the wound infection. These results demonstrate that selective lethal photosensitization of MRSA can be achieved using phofrin, photogem and radachlorin. Thus, PDT can inactivate MRSA survival.

  • A simple and efficient synthetic route for making purpurin-18-N-aminoimides is described. The purpurin-18-Naminoimides are obtained by treatment of purpurin-18 methyl ester with various amines. These new compounds have long wavelength absorptions in the range of 706 - 711 nm. In preliminary screening, the purpurin-18-N-aminoimides have shown promising photosensitizing activity for the cancer cell by in vitro study in photodynamic therapy.

  • Photodynamic therapy (PDT) is a cancer treatment modality which utilizes the cytotoxicity of the active singlet oxygen derived from irradiation of a tumor accumulated photosensitizer. As the oxygen in the singlet state radiates an emission of 1270nm wavelength when it decays to the triplet state, detection of the emission helps us to understand the mechanism of PDT or to evaluate photosensitizers. We detected the 1270nm emission from photosensitizers Photofrin and ATX-SI0 in vitro and in vivo by means of high sensitive NIR detectors. We obtained the maximum amount of singlet oxygen at irradiation wavelength of 665-670nm from a HeLa tumor in a nude mouse which is injected with ATX-S10.

Antibacterial Effect of Photodynamic Therapy using Photogem and a 632 nm Diode Laser on $Helicobacter$ $pylori$

  • Eradication of $H.$ $pylori$, usually carried out by using antimicrobial agents, is essential for preventing gastric ulcers and cancers. The $H.$ $pylori$ isolates, however, have continuously grown antimicrobial resistance, which have caused difficulty in treating the bacteria and in turn, photodynamic therapy (PDT) has been found to be effective in inducing deaths of variety of bacteria. After PDT treatment, the number of colony forming units (CFU), the morphologic changes, and flow cytometry were observed. In the PDT group containing 100 and 200 ${\mu}g$/ml photogem, no live $H.$ $pylori$ was observed, while 10 and 50 ${\mu}g$/ml photogem were only partially effective. $H.$ $pylori$ of the PDT group also displayed distortion and shrinkage in morphology. This study demonstrated that photogem-mediated PDT effectively induces deaths of $H.$ $pylori$.

Photodynamic Therapy Using Topically Applied 5-ALA, MAL and CLC for Canine Otitis Externa

  • Canine otitis externa is a common disorder in small animal practice with prevalence up to 20%. In a large percentage of cases, canine otitis externa is a chronic and recurrent disease also associated with drug-resistant bacteria that is difficult to treat with traditional antibiotics. Photodynamic therapy (PDT) is a new strategy to exterminate pathogenic microorganisms such as bacteria and fungi. The purpose of this study was to investigate the effectiveness of photodynamic therapy against canine otitis externa using three photosensitizer (PS); 5-Aminolevulinic acid (5-ALA) and Methyl aminolevulinic acid (MAL) with semiconductor laser diode (SLD, 635nm of wave length), Chlorophyll-lipoid complex (CLC) with light-emitting diode (LED, 660nm of wave length). After PDT, dogs showed improved Otitis Index Score (OTIS) in swelling, exudate, odor, and pain. A result of the cytology test revealed decrease of bacteria and malassezia count in the oil immersion field and colony forming units count. PDT was effective as a bacteriocide of methicillin-resistant Staphylococcus pseudintermedius (MRSP) and a fungicide of Malassezia pachydermatis. MAL and 5-ALA were more effective PS against canine otitis externa than CLC. These results suggest that PDT is a new strategy to exterminate pathogenic microorganisms such as bacteria and fungi. PDT can be considered as a new therapeutic approach for canine recurrent otitis externa and a countermeasure to drug resistance that is a disadvantage of traditional antibiotic and antifungal therapy.

Photodynamic Therapy에 의한 산화적 스트레스 조건에서 Helicobacter pylori의 Fur 단백질의 역할 (The Role of Helicobacter pylori's Fur Protein in the Oxidative Stress Induced by Photodynamic Therapy)

  • Helicobacter pylori균의 ferric uptake regulator (Fur) 단백질의 산화적 손상에 대한 역할을 연구하였다. H. pylori균의 fur 유전자를 제거한 돌연변이체를 만들고 wild type H. pylori균과 돌연변이체 균의 산화적 스트레스에 대한 반응을 비교하였다. 산화적 스트레스는 methylene blue와 660 nm 파장의 빛을 이용하는 광역학적 치료방법으로 유도하였다. 산화적 스트레스를 가한 실험조건에서 wt H. pylori와 돌연변이체의 생존력, DNA 손상의 정도를 비교 검토하였다. 그 결과 fur 유전자가 제거된 돌연변이체의 생균수가 wt에 비해 10,000배 가량 감소한 것을 알 수 있었으며 DNA의 산화적 손상의 marker인 8-hydroxy-2-deoxyguanosine (8-OHdG)의 양도 fur 유전자 제거된 돌연변이에서 wild type에 비해 3배 정도 더 생성됨을 확인하였다. 따라서 본 실험결과 H. pylori균의 fur 유전자가 PDT법으로 유도한 산화적 스트레스에 방어 기작을 하는 것으로 사료된다.

광역동 항암치료를 위한 광과민제 스크리닝 연구 (Screening of Photosensitizer for Photodynamic Therapy)

  • 광역동요법 (photodynamic therapy, PDT)은 악성종양을 치료하기 위해 광과민제와 레이저광을 이용하는 표적지향적 항암치료법이다. 본 연구에서는 PDT에 적용할 수 있는 새로운 광과민제를 스크리닝하기 위해 4종의 phthalocyanine계 광과민제 (silver phthalocyanine, iron (III) phthalocyanine, copper (II) phthalocyanine, nickel (II) phthalocyanine)을 이용하여 폐암과 유방암 세포주에 대한 항암활성 테스트를 수행하였다. 연구결과 phthalocyanine계 광과민제들은 폐암보다 유방암에 높은 활성을 나타냈으며, 그 중 silver phthalocyanine (AgPc)는 정상세포에 대해서는 세포독성이 낮으면서도 물에 용해할 경우 청색증을 유발하지 않아 유방암에 대한 항암제로서의 개발가능성을 확인하였다.
