Photo-induced inter-protein interaction changes in the time domain; a blue light sensor protein PixD

  • For understanding molecular mechanisms of photochemical reactions, in particular reactions of proteins with biological functions, it is important to elucidate both the initial reactions from the photoexcited states and the series of subsequent chemical reactions, e.g., conformation, intermolecular interactions (hydrogen bonding, hydrophobic interactions), and inter-protein interactions (oligomer formation, dissociation reactions). Although time-resolved detection of such dynamics is essential, these dynamics have been very difficult to track by traditional spectroscopic techniques. Here, relatively new approaches for probing the dynamics of protein photochemical reactions using time-resolved transient grating (TG) are reviewed. By using this method, a variety of spectrally silent dynamics can be detected and such data provide a valuable description about the reaction scheme. Herein, a blue light sensor protein TePixD is the exemplar. The initial photochemistry for TePixD occurs around the chromophore and is detected readily by light absorption, but subsequent reactions are spectrally silent. The TG experiments revealed conformational changes and changes in inter-protein interactions, which are essential for TePixD function. The TG experiments also showed the importance of fluctuations of the intermediates as the driving force of the reaction. This technique is complementary to optical absorption detection methods. The TG signal contains a variety of unique information, which is difficult to obtain by other methods. The advantages and methods for signal analyses are described in detail in this review.

Photochemical Reflectance Index (PRI) Mapping using Drone-based Hyperspectral Image for Evaluation of Crop Stress and its Application to Multispectral Imagery (작물 스트레스 평가를 위한 드론 초분광 영상 기반 광화학반사지수 산출 및 다중분광 영상에의 적용)

  • The detection of crop stress is an important issue for the accurate assessment of yield decline. The photochemical reflectance index (PRI) was developed as a remotely sensed indicator of light use efficiency (LUE). The PRI has been tested in crop stress detection and a number of studies demonstrated the feasibility of using it. However, only few studies have focused on the use of PRI from remote sensing imagery. The monitoring of PRI using drone and satellite is made difficult by the low spectral resolution image captures. In order to estimate PRI from multispectral sensor, we propose a band fusion method using adjacent bands. The method is applied to the drone-based hyperspectral and multispectral imagery and estimated PRI explain 79% of the original PRI. And time series analyses showed that two PRI data (drone-based and SRS sensor) had very similar temporal variations. From these results, PRI from multispectral imagery using band fusion can be used as a new method for evaluation of crop stress.

Fabrication of C2H2 Gas Sensors Based on Ag/ZnO-rGO Hybrid Nanostructures and Their Characteristics (Ag/ZnO-rGO 하이브리드 나노구조 기반 C2H2 가스센서의 제작과 그 특성)

  • In this work, pure hierarchical ZnO structure was prepared using a simple hydrothermal method, and Ag nanoparticles doped hierarchical ZnO structure was synthesized uniformly through photochemical route. The reduced graphene oxide (rGO) has been synthesized by typical Hummer's method and reduced by hydrazine. Prepared Ag/ZnO nanostructures are uniformly dispersed on the surface of rGO sheets using ultrasonication process. The synthesized samples were characterized by SEM, TEM, EDS, XRD and PL spectra. The average size of prepared ZnO microspheres was around $2{\sim}3{\mu}m$ and showed highly uniform. The average size of doped-Ag nanoparticles was 50 nm and decorated into ZnO/rGO network. The $C_2H_2$ gas sensing properties of as-prepared products were investigated using resistivity-type gas sensor. Ag/ZnO-rGO based sensors exhibited good performances for $C_2H_2$ gas in comparison with the Ag/ZnO. The $C_2H_2$ sensor based on Ag/ZnO-rGO had linear response property from 3~1000 ppm of $C_2H_2$ concentration at working temperature of $200^{\circ}C$. The response values with 100 ppm $C_2H_2$ at $200^{\circ}C$ were 22% and 78% for Ag/ZnO and Ag/ZnO-rGO, respectively. In additions, the sensor still shows high sensitivity and quick response/recovery to $C_2H_2$ under high relative humidity conditions. Moreover, the device shows excellent selectivity towards to $C_2H_2$ gas at optimal working temperature of $200^{\circ}C$.

Fabrication of CuO/ZnO Nano-heterostructure by Photochemical Method and Their H2S Gas Sensing Properties

  • This study reports the H2S gas sensing properties of CuO / ZnO nano-hetero structure bundle and the investigation of gas sensing mechanism. The 1-Dimensional ZnO nano-structure was synthesized by hydrothermal method and CuO / ZnO nano-heterostructures were prepared by photo chemical reaction. Scanning electron microscopy (SEM) and X-ray diffraction (XRD) spectra confirmed a well-crystalline ZnO of hexagonal structure. In order to improve the H2S gas sensing properties, simple type of gas sensor was fabricated with ZnO nano-heterostructures, which were prepared by photo-chemical deposition of CuO on the ZnO nanorods bundle. The furnace type gas sensing system was used to characterize sensing properties with diluted H2S gas (50 ppm) balanced air at various operating temperature up to 500$^{\circ}C$. The H2S gas response of ZnO nanorods bundle sensor increased with increasing temperature, which is thought to be due to chemical reaction of nanorods with gas molecules. Through analysis of X-ray photoelectron spectroscopy (XPS), the sensing mechanism of ZnO nanorods bundle sensor was explained by well-known surface reaction between ZnO surface atoms and hydrogen sulfide. However at high sensing temperature, chemical conversion of ZnO nanorods becomes a dominant sensing mechanism in current system. Photo-chemically fabricated CuO/ZnO heteronanostructures show higher gas response and higher current level than ZnO nanorods bundle. The gas sensing mechanism of the heteronanostructure can be explained by the chemical conversion of sensing material through the reaction with H2S gas.

Comparison of Chromotropic Acid Method and DNPH-HPLC Method for the Measurement of Formaldehyde in the Air (공기 중 포름알데히드 측정을 위한 크로모트로핀산법과 DNPH/HPLC방법의 비교.평가)

  • Formaldehyde is known to have adverse health effects, and is also found to be a product of atmospheric. photochemical reaction%. Several methods have been employed for the measurement of formaldehyde. Among them, two common methods are comparatively evaluated in this study, i.e. the chromotropic acid method and the HPLC method coupled with 2,4- dinitrophenylhydrazine (DNPH) derivatization of carbonyl compounds. Method evaluation and validation were performed with respect to repeatability, linearity, collection efficiency, and recovery efficiency. Comparison of formaldehyde concentrations measured simultaneously in the field using the two (n=20) and 35.8%, respectively. Overall, the measured levels by chromotropic acid method appeared to ice significantly higher than those by DNPH-HPLC method, indicating a positive interference due to the other aldehydes collected together with formaldehyde.

  • This paper describes the fabrication and characteristics of $C_2H_2$ gas sensor based on Ag-doped hierarchical ZnO nanostructures. The gas sensing properties of as-prepared products were investigated using resistivity-type gas sensors. 5 at% Ag-doped ZnO based sensors exhibited good performances for $C_2H_2$ gas in comparison with the un-doped one. The sensor based on Ag-doped hierarchical ZnO structures had linear response property from 5~1000 ppm of $C_2H_2$ concentration at working temperature of $200^{\circ}C$. The response values with 100 ppm $C_2H_2$ at $200^{\circ}C$ were 10% and 75% for pure and 5 at% Ag-doped hierarchical ZnO nanostructures, respectively. Moreover, the device showed excellent selectivity towards to $C_2H_2$ gas at optimal working temperature of $200^{\circ}C$.

Synthesis of Pt@TiO2 Nano-composite via Photochemical Reduction Method (광화학 환원방법을 이용한 Pt@TiO2 나노 복합체 합성)

  • Pt has been widely used as catalyst for fuel cell and exhausted gas clean systems due to its high catalytic activity. Recently, there have been researches on fabricating composite materials of Pt as a method of reducing the amount of Pt due to its high price. One of the approaches for saving Pt used as catalyst is a core shell structure consisting of Pt layer on the core of the non-noble metal. In this study, the synthesis of Pt shell was conducted on the surface of $TiO_2$ particle, a non-noble material, by applying ultraviolet (UV) irradiation. Anatase $TiO_2$ particles with the average size of 20~30 nm were immersed in the ethanol dissolved with Pt precursor of $H_2PtCl_6{\cdot}6H_2O$ and exposed to UV irradiation with the wavelength of 365 nm. It was confirmed that Pt nano-particles were formed on the surface of $TiO_2$ particles by photochemical reduction of Pt ion from the solution. The morphology of the synthesized Pt@$TiO_2$ nano-composite was examined by TEM (Transmission Electron Microscopy).

  • Pusan is the largest coastal city with a population of about four mi18ion in Korea. Because of increased and confused traffic, photochemical air pollution become a major urban environmental problem recently. The photo-chemical air pollution weather forecasting method preciser than existing air pollution forecast method has been developed to forecast ozone episode days with meteorological conditions using the data measured at 7 air quality continuous monitoring stations from lune to September using 2 years (1994, 1995). The method developed in present study showed higher percentage correct and skill score than existing air pollution forecasting in KMA ( Korea Meteorological Administration).

  • There are several methods for the hydrogen production such as steam reforming of natural gas, photochemical method, biological method, electrolysis and thermochemical method, etc. These days it has been widely studied for the hydrogen production method having low hydrogen production cost and high efficiency. In this paper, patents in the hydrogen production by steam reforming of natural gas were gathered and analyzed. The search range was limited in the open patents of USA(US), European Union(EP), Japan(JP), and Korea(KR) from 1996 to 2006. Patents were gathered by using key-words searching and extracted by filtering criteria. The trends of the patents was analyzed by the years, countries, companies, and technologies.

Photochemical Index Analysis on the Influence of LED Illumination Color Temperature on Donarium Cherry (도시야간조명의 LED 색온도별 겹벚나무의 광화학적 생리지표 해석)

  • In this study, the efficiency of photosynthetic electron transfer according to LED color temperature was verified in order to find a way to efficiently grow trees under night illumination. The experiment was carried out with White treatment, Warmwhite treatment, and non-treatment with donarium cherry. The study uses to a method for analyzing and evaluating the color temperature of an LED light source by photochemical analysis. We found that all treatments 115 DAT of maximum fluorescence amount(P) had the lowest. In the treatment using white light and the Warm-white light, the T amount of florescence of the late stage during the transition of the J-I level was increased, and the photosystem I electron transfer efficiency was decreased. Therefore, the electron transport efficiency of $RE1_O/CS$ and RE1o/RC were reduced. Especially, compared to Warmwhite, the light intensity increased greatly in the white-light treatment, The $PI_{TOTALABS}$ of 7 DAT was the highest value, but it was decreased to the lowest value on 115 DAT. This study has shown that the white treatment was low in electron transfer efficiency and soundness. Warmwhite-light treatments showed lower stress.
