Liquid Crystal Based Optical Sensor for Imaging Trypsin Activity at Interfaces Between Aqueous Phases and Thermotropic Liquid Crystals

  • In this study, we developed a liquid crystal (LC)-based optical sensor for monitoring enzymatic activity through orientational changes in liquid crystals (LCs) coupled to the properties of a poly-${\small{L}}$-lysine (PLL)-based polymeric membrane. We prepared a PLL-based polymeric membrane at the planar interface between the thermotropic liquid crystal and aqueous phases. The PLL-based polymeric membrane was obtained by contacting the PLL solution with water immiscible LCs, 4-cyano-4'-pentyl-biphenyl (5CB) doped with adipoyl chloride. We then investigated the membrane properties by examining the permeability of the membrane to phospholipids, 1,2-didodecanoyl-rac-glycero-3-phosphocholine (DLPC). The permeability of the membrane to transport phospholipids was monitored through the orientational transition of 5CB in contact with the dispersions of DLPC. Since trypsin can enzymatically catalyze the hydrolysis of PLL, we incubated an aqueous trypsin solution with the membrane for 2 h at room temperature to cause an increase in the permeability of the polymeric membrane to DLPC. As a result, a bright to dark optical shift of LCs was observed, which implied that an enzymatic reaction between trypsin and PLL-based membrane occurred. Two control experiments using chymotrypsin and bovine serum albumin (BSA) revealed no sign of improved permeability based on the orientational transition of LCs.

Plasma Phospholipids, including Plasmalogens, after Consumption of Diets Enriched in Long-chain n-3 Fatty Acids

  • The level of long-chain n-3 fatty acids in chicken and pork can be increased by changing the diet of the animals. Increased levels of these essential fatty acids improve cardiovascular health in humans. The purpose of this study was to study the effects of the consumption of pork and chicken enriched in docosahexaenoic acid (DHA) on plasma lipids. The consumption of these products decreased the levels of two cardiovascular risk factors, LDL-cholesterol and triacylglycerols, in the plasma of female college students. The effect on LDL-cholesterol differed from that of fish oil, which does not affect the level of LDL-cholesterol. The proportions of DHA in the triacylglycerols and the glycerophospholipids were increased markedly. The greatest changes in the glycerophospholipids were in the ether types of the ethanolamine glycerophospholipids. Dietary DHA appears to be incorporated preferentially into the plasma ethanolamine plasmalogens, which can act as antioxidants. This agrees with our hypothesis that DHA stimulated the transcription of the genes for peroxisomal enzymes that are required for plasmalogen synthesis.

Enhanced Ex Vivo Buccal Transport of Propranolol: Evaluation of Phospholipids as Permeation Enhancers

  • The aim of the present study was to evaluate the effects of two phospholipid permeation enhancers, lysophosphatidylcholine (LPC) and didecanoylphosphatidylcholine (DDPC), along with a fusidic acid derivative, sodium taurodihydrofusidate (STDHF) and ethanol (EtOH) on the buccal transport of propranolol hydrochloride (PPL) using an ex vivo buccal diffusion model. The permeation rate of [$^3 H$]PPL as measured by steady-state fluxes increased with increasing EtOH concentration. A significant flux enhancement (P<0.05) was achieved by EtOH at 20 and 30 %v/v concentrations. At a 0.5 %w/v permeation enhancer concentration, the buccal permeation of [$^3 H$]PPL was significantly enhanced by all the enhancers studied (i.e., LPC, DDPC and STDHF) compared to the control (phosphate-buffered saline pH 7.4, PBS). LPC and DDPC displayed a greater degree of permeation enhancement compared with STDHF and EtOH-PBS mixtures with an enhancement ratio of 3.2 and 2.9 for LPC and DDPC, respectively compared with 2.0 and 1.5 for STDHF and EtOH:PBS 30:70 %v/v mixture, respectively. There was no significant difference between LPC and DDPC for the flux values and apparent permeability coefficients of [$^3$H]PPL. These results suggest that phospholipids are suitable as permeation enhancers for the buccal delivery of drugs.

The Penetration of n-Alkanols into Model Membranes of cholesterol Plus Phospholipids Extracted from Brain Membranes (n-Alkanols가 인지질들로서 제제한 인공세포막에서의 침투정도)

  • 소의 신선한 대뇌피질로부터 synaptosomal plasma membrane vesicles(SPMV)를 분리한 후 이 SPMV로부터 추출한 모든 인지질들로서 제제한 인공세포막(SPMVPL)에서의 n-alkanols 침투 정도를 형광 probe를 이용한 형광 소광법을 통하여 검색하였다. n-alkanols는 SPMVPL 외부 단층(outer monolayer)의 표면에 주로 분포하되 그 탄소수에 비례하여 소수성 부위에 분포되는 양이 증가되는 경향을 나타내었다. methanol, ethanol, 1-propano, 1-butanol, 1-pentanol, 1-hexanol, 1-heptanol, 1-octanol, 1-nonanol 및 1-decanol은 SPMVPL 외부 단층의 표면(친수성 부위)에 분포되는 것이 소수성 부위에 분포되는 것에 비하여 각각 432.4, 208.9. 125.6, 88.2, 19.3, 7.9, 2.6, 1.0, 0.42, 1.36배가 되었다. 1-decanol은 $C_{10}$인데도 불구하고 $C_8$인 1-octanol에 비하여 적은 양이 소수성 부위에 침투 분포된다는 것이 확인되었다. n-alkanols의 침투에 대하여 저자등이 이미 보고한 바 있는 SPMV 및 SPMVTL(cholesterol+phospholipids)의 경우보다도 본 연구에서의 SPMVPL의 경우가 현저하게 많은 양이 소수성 부위로 침투 분포된다는 것도 확인되었다.

  • Environmental technology (ET)는 이미 세계적으로 가장 중요한 기술로 주목받고 있으며 친환경적이고 생체적합한 상품에 대한 수요가 계속해서 증가하고 있다. 섬유시장에서도, 항균성, 피부노화 방지, 아토피 방지, 보습제와 같이 수많은 다기능성 친환경 제품들이 계속해서 개발되고 소개되어지고 있다. 특히, 피부 보호를 위한 생체적합 보습 가공제로 쓰이는 squalene, collage, chitosan, hyaluronic acid과 같은 물질들은 cosmetics, medical 시장에서 널리 쓰이고 있다. 그러나 섬유시장에서의 이러한 물질들은 섬유에 적용시켰을 때 만족스럽지 못한 기능으로 인해 그리 좋지 않은 평가를 받고 있다. 천연소재를 이용해 스킨케어 제품 제조 시 지속적인 안정성의 문제와 제품의 기능성, 내구성이 만족스럽지 못한 결과를 보인다. 이러한 문제를 해결하고자 binder처리를 시도하였지만 가공제의 스스로의 기능이 현저하게 저하되는 문제로 인해 널리 쓰이진 않았다. 또한 안정성을 증진시키고자 마이크로캡슐을 개발하였지만 섬유의 터치감이 거칠어지고 가격적인 면에서 만족스럽지 못한 문제점이 있다. 본 연구에서는 새로운 phospholipid 화합물 2-methacryloyl oxyethyl phosphoryl choline (MPCE)를 이용하여 polyester에 적용시킨 후 다양한 조건들을 살펴보았다. MPCE처리가 된 섬유는 흡습성, 대전방지성, 항균성, 피부자극이 없는 것과 같은 생체적합한 기능을 보여주었다.

Lipid and Fatty Acid Compositions of Korean Corns (한국산(韓國産) 옥수수의 지질성분(脂質成分)에 관(關)한 연구(硏究))

  • The lipid compositions of corns produced in Korea were analyzed. Free and bound lipids of the corn kernels were fractionated, quantitated and compared by silicic acid column, thin layer and gas liquid chromatography. Corn kernels contained 5.02% total lipids, which is consisted of 4.09% free lipids and 0.93% bound lipids. Free lipids comprised of 89.61% neutral lipids, 3.75% glycolipids and 6.40% phospholipids, while bound lipids contained 14.26% neutral lipids, 46.06% glycolipids and 37.18% phospholipids. In the neutral lipids of free lipids, triglycerides were predominant (67.68%) and minor components such as esterified sterols, free sterols, free fatty acids, 1,3-diglycerides, 1,2-diglycerides were present. But in the neutral lipids of bound lipids, esterified sterols were not present and the contents of triglycerides were lower (47.68%) and free fatty acids were higher than those of free lipids. Among the phospholipids in free and bound lipids, phosphatidyl choline and phosphatidyl serines and phosphatidyl inositols were also present as minor components. The major fatty acids in the three lipid classes were linoleic, oleic and palmitic acids.

Physical Property and Stability of Liposome Prepared from Egg Yolk Phospholipids at Various Storage Conditions (난황 유래 인지철로 합성한 리포좀의 물리적 특성과 안정성)

  • Liposomes were prepared from egg yolk phospholipids to study their physical properties and stability at various storage conditions. Under storage conditions at different pH levels, the particle sizes of liposomes increased at the range of pH 1-2, and the absolute values of $\xi$-potentials were reduced at the range of pH 1-4. The leakage of sulforhodamine B (SRB), a fluorescent dye which is encapsulated in the liposome, increased greatly at pH 2-4. At different storage temperatures, the particle size of liposomes increased from the 10 days of storage at $4^{\circ}C$ and the 40 days at 20 and $35^{\circ}C$. The $\xi$-potentials decreased slightly later during storage under 4, 20 and $35^{\circ}C$. At the storage temperature of $50^{\circ}C$, the leakage of SRB was the greatest. Therefore, we concluded that the pH conditions lower than pH 6 and high temperature of $50^{\circ}C$ are not conducive to storing liposomes. The results obtained here may prove helpful in developing liposome-based encapsulation and diagnostic reagents.

Quantitative Fractionation of Total Lipids and Their Fatty Acid Composition in Korean Yam Tubers. (한국산 마지질의 분획 정량과 지방산조성)

  • To investigate the lipid components of Korean yams(Dioscorea batatas and Dioscorea aimadoimo), silicic acid column, thin layer and gas chromatographies were applied. Total crude lipid contents in D. batatas and D. aimadoimo were 1.70% and 1.31%, and total purified lipid contents were 1.35% and 0.99% on a dry weight basis. Lipids were fractionated and quantitated by silicic acid column chromatography. The contents of neutral lipids, glycolipids and phospholipids were 43.33, 39.60 and 17.07% in D. batatas and 43.94, 34.74 and 21.32% in D. aimadoimo. Neutral lipids were the most abundant, in which triglycerides and sterol ester were found as the major components. Glycolipids were mainly composed of monogalactosyl and trigalactosyl diglycerides. Phosphatidyl choline and phosphatidyl ethanolamine were the major components of phospholipid. Fatty acids of the total lipids were mainly linoleic and palmitic acids. The fatty acid compositions of neutral lipids, glycolipids and phospholipids were the same tendency as that of total lipids. However, the content of linolenic acid in glycolipids was higher than in neutral lipids and phospholipids when fractionated.

The Effect of 1-Propanol on the Rotational Mobility of n-(9-Anthroyloxy) stearic acid in Outer Monolayers of Neuronal and Model Membranes

  • The aim of this study was to provide a basis for the molecular mechanism underlying the pharmacological action of ethanol. We studied the effects of 1-propanol on the location of n-(9-anthroyloxy)palmitic acid or stearic acid (n-AS) within the phospholipids of synaptosomal plasma membrane vesicles (SPMV). The SPMV were isolated from the bovine cerebral cortex and liposomes of total lipids (SPMVTL) and phospholipids (SPMVPL). 1-Propanol increased the rotational mobility of inner hydrocarbons, while decreasing the mobility of membrane interface, in native and model membranes. The degree of rotational mobility varied with the number of carbon atoms at positions 16, 12, 9, 6 and 2 in the aliphatic chain of phospholipids in the neuronal and model membranes. The sensitivity of increasing or decreasing rotational mobility of hydrocarbon interior or surface by 1-propanol varied with the neuronal and model membranes in the following order: SPMV, SPMVPL and SPMVTL.
