β-lapachone에 의한 A549 인체폐암세포의 apoptosis 유도와 cyclooxygenase-2 활성 저하 (β-Lapachone-Induced Apoptosis is Associated with Inhibition of Cyclooxygenase-2 Activity in Human Lung Cancer A549 Cells)

  • ${\beta}$-lapachone은 남미에 자생하는 lapacho 나무(Tabeuia avellanedae)의 수액에 함유된 quinone계열의 일종으로 많은 인체암세포에서 apoptosis를 유발하는 것으로 알려져 있다. 본 연구는 A549 인체폐암세포를 대상으로 ${\beta}$-lapachone에 의한 apoptosis 유발 과정에서 나타나는 또 다른 현상들을 조사하기 위하여 실시되었다. ${\beta}$-lapachone이 처리된 A549 세포는 처리 농도의 증가에 따라 생존율이 감소되었으며, 이는 apoptosis 유발과 연관이 있음을 MTT assay와 flow cytometry 분석을 통하여 확인하였다. ${\beta}$-lapachone에 의한 A549 세포의 증식억제는 종양억제유전자 p53과 cyclin-dependent kinase 저해제인 p21의 발현을 전사 및 번역 수준에서 증가시켰으며, p53 단백질의 인산화 증가와 연관성이 있었다. 또한 ${\beta}$-lapachone은 caspase-3과 -9를 활성화시켰으며, 활성화된 caspase-3의 기질 단백질들[poly(ADP-ribose) polymerase, ${\beta}$-catenin 및 phospholipase C-$\gamma$1]의 단편화를 유도하였다. 아울러 ${\beta}$-lapachone은 cyclooxygenase (COX)-2의 mRNA 및 단백질의 발현을 억제하였으나 COX-1의 발현에는 영향을 미치지 않았으며, ${\beta}$-lapachone에 의한 COX-2의 발현억제는 prostaglandin E2의 생성 저하에 관련이 있었다. 본 연구의 결과는 ${\beta}$-lapachone의 항암활성 기전의 이해와 더불어 ${\beta}$-lapachone이 폐암세포에서 강력한 항암활성이 있음을 보여 주는 것이다.

Protein Profiles Associated with Anoikis Resistance of Metastatic MDA-MB-231 Breast Cancer Cells

  • Resistance to anoikis, a cell-detachment induced apoptosis, is one of the malignant phenotypes which support tumor metastasis. Molecular mechanisms underlying the establishment of this phenotype require further investigation. This study aimed at exploring protein expression profiles associated with anoikis resistance of a metastatic breast cancer cell. Cell survival of suspension cultures of non-metastatic MCF-7 and metastatic MDA-MB-231 cells were compared with their adherent cultures. Trypan blue exclusion assays demonstrated a significantly higher percentage of viable cells in MDA-MB-231 than MCF-7 cell cultures, consistent with analysis of annexin V-7-AAD stained cells indicating that MDA-MB-231 possess anti-apoptotic ability 1.7 fold higher than MCF-7 cells. GeLC-MS/MS analysis of protein lysates of MDA-MB-231 and MCF-7 cells grown under both culture conditions identified 925 proteins which are differentially expressed, 54 of which were expressed only in suspended and adherent MDA-MB-231 but not in MCF-7 cells. These proteins have been implicated in various cellular processes, including DNA replication and repair, transcription, translation, protein modification, cytoskeleton, transport and cell signaling. Analysis based on the STITCH database predicted the interaction of phospholipases, PLC and PLD, and 14-3-3 beta/alpha, YWHAB, with the intrinsic and extrinsic apoptotic signaling network, suggesting putative roles in controlling anti-anoikis ability. MDA-MB-231 cells grown in the presence of inhibitors of phospholipase C, U73122, and phospholipase D, FIPI, demonstrated reduced ability to survive in suspension culture, indicating functional roles of PLC and PLD in the process of anti-anoikis. Our study identified intracellular mediators potentially associated with establishment of anoikis resistance of metastatic cells. These proteins require further clarification as prognostic and therapeutic targets for advanced breast cancer.

The inhibitory activity of ginsenoside Rp4 in adenosine diphosphate-induced platelet aggregation

  • Background: Korean ginseng, Panax ginseng Meyer, has been used as a traditional oriental medicine to treat illness and promote health for several thousand years. Ginsenosides are the main constituents for the pharmacological effects of P. ginseng. Since several ginsenosides, including ginsenoside (G)-Rg3 and G-Rp1, have reported antiplatelet activity, here we investigate the ability of G-Rp4 to modulate adenosine diphosphate (ADP)-induced platelet aggregation. The ginsenoside Rp4, a similar chemical structure of G-Rp1, was prepared from G-Rg1 by chemical modification. Methods: To examine the effects of G-Rp4 on platelet activation, we performed several experiments, including antiplatelet ability, the modulation of intracellular calcium concentration, and P-selectin expression. In addition, we examined the activation of integrin ${\alpha}IIb{\beta}_3$ and the phosphorylation of signaling molecules using fibrinogen binding assay and immunoblotting in rat washed platelets. Results: G-Rp4 inhibited ADP-induced platelet aggregation in a dose-dependent manner. We found that G-Rp4 decreased calcium mobilization and P-selectin expression in ADP-activated platelets. Moreover, fibrinogen binding to integrin ${\alpha}IIb{\beta}_3$ by ADP was attenuated in G-Rp4-treated platelets. G-Rp4 significantly attenuated phosphorylation of extracellular signal-regulated protein kinases 1 and 2, p38, and c-Jun N-terminal kinase, as well as protein kinase B, phosphatidylinositol 3-kinase, and phospholipase C-${\gamma}$ phosphorylations. Conclusion: G-Rp4 significantly inhibited ADP-induced platelet aggregation and this is mediated via modulating the intracellular signaling molecules. These results indicate that G-Rp4 could be a potential candidate as a therapeutic agent against platelet-related cardiovascular diseases.

The Signal Transduciton of Ginsenosides, Active Ingredients of Panax ginseng, in Xenopus oocyte: A Model System for Ginseng Study

  • Recently, we have provided evidence that ginsenosides, the active components of Panax ginseng, utilize pertussis toxin (PTX)-insensitive $G{\alpha}_{q/11}-phospholipase\;C-{\beta}3(PLC-{\beta}3)$ signal transduction pathway for the enhancement of $Ca^{2+}-activated\;Cl^{-}$ current in the Xenopus oocyte (British J. Pharmacol. 132, 641-647, 2001; JBC 276, 48797-48802, 2001). Other investigators have shown that stimulation of receptors linked to $G{\alpha}-PLC$ pathway inhibits the activity of G proteincoupled inwardly rectifying $K^+$ (GIRK) channel. In the present study, we sought to determine whether ginsenosides influenced the activity of GIRK 1 and GIRK 4 (GIRK 1/4) channels expressed in the Xenopus oocyte, and if so, the underlying signal transduction mechanism. In oocyte injected with GIRK 1/4 channel cRNAs, bath-applied ginsenosides inhibited high potassium (HK) solution-elicited GIRK current $(EC_{50}:4.9{\pm}4.3\;{\mu}g/ml).$ Pretreatment of the oocyte with PTX reduced the HK solution-elicited GIRK current by $49\%,$ but it did not alter the inhibitory ginsenoside effect on GIRK current. Prior intraoocyte injection of cRNA(s) coding $G{\alpha}_q,\;G{\alpha}_{11}\;or\;G{\alpha}_q/G{\alpha}_{11},\;but\;not\;G{\alpha}_{i2}\;or\;G{\alpha}_{oA}$ attenuated the inhibitory ginsenoside effect. Injection of cRNAs coding $G{\beta}_{1{\gamma}2}$ also attenuated the ginsenoside effect. Similarly, injection of the cRNAs coding regulators of G protein signaling 1, 2 and 4 (RGS1, RGS2 and RGS4), which interact with $G{\alpha}_i\;and/or\;G{\alpha}_{q/11}$ and stimulates the hydrolysis of GTP to GDP in active GTP-bound $G{\alpha}$ subunit, resulted in a significant reduction of ginsenoside effect on GIRK current. Preincubation of GIRK channel-expressing oocyte in PLC inhibitor (U73122) or protein kinase C (PKC) inhibitor (staurosporine or chelerythrine) blocked the inhibitory ginsenoside effect on GIRK current. On the other hand, intraoocyte injection of BAPTA, a free $Ca^{2+}$ chelator, had no significant effect on the ginsenoside action. Taken together, these results suggest that ginsenosides inhibit the activity of GIRK 1/4 channel expressed in the Xenopus oocyte through a PTX-insensitive and $G{\alpha}_{q/11}$-,PLC-and PKC-mediated signal transduction pathway.

육울탕(六鬱湯)에 의한 인체자궁경부암세포의 증식억제에 관한 연구 (Induction of Apoptosis by Yukwool-tang in Human Cervical Carcinoma HeLa Cells)

  • Yukwool-tang (YWT) is a traditional Chinese medicine, which has been used for patients suffering from a uterine disease in Oriental medicine. In the present study, it was examined the biochemical mechanisms of apoptosis by YWT in human cervical carcinoma HeLa cells. It was found that YWT could inhibit the cell growth of HeLa cells in a dose-dependent manner, which was associated with apoptotic cell death such as formation of apoptotic bodies and DNA fragmentation. Flow cytometry analysis confirmed that YWT treatment increased populations of apoptotic-sub-G1 phase of the cell cycle. We observed the p53-independent induction of p21 proteins, down-regulation of anti apoptotic Bcl-2 expression and proteolytic activation of caspase-3 in YWT-treated HeLa cells. YWT treatment also concomitant degradation and/or inhibition of poly (ADP-ribose) polymerase (PARP), phospholipase C-1 ($PLC{\gamma}1$), ${\beta}-catenin$ and DNA fragmentation factor 45/inhibitor of caspase-activated DNase (DFF45/ICAD). Taken together, these findings partially provide novel insights into the possible molecular mechanism of the anti-cancer activity of YWT.

The Inhibitory Effect and Mechanism of Luteolin 7-Glucoside on Rat Aortic Vascular Smooth Muscle Cell Proliferation

  • The abnormal proliferation of aortic vascular smooth muscle cells (VSMCs) plays a central role in the pathogenesis of atherosclerosis and restenosis after angioplasty and possibly also in the development of hypertension. The present study was designed to examine the inhibitory effects and the mechanism of luteolin 7-glucoside (L7G) on the platelet-derived growth factor (PDGF)-BB-induced proliferation of VSMCs. L7G significantly inhibited the PDGF-BB-induced proliferation and the DNA synthesis of the VSMCs in a concentration-dependent manner. Pre-incubation of the VSMCs with L7G significantly inhibited the PDGF-BB-induced extracellular signal-regulated kinase 1/2 (ERK1/2), Akt and the phospholipase C $(PLC)-{\gamma}1$ activation. However, L7G had almost no affect on the phosphorylation of $PDGF-{\beta}$ receptor tyrosine kinase, which was induced by PDGF-BB. These results suggest that L7G inhibits the PDGF-BB-induced proliferation of VSMCs via the blocking of $(PLC)-{\gamma}1$, Akt, and ERK1/2 phosphorylation.

방사선조사후 백서소장점막에서 발생하는 신호전달체계에 관한 연구 (The Signal Transduction Mechanisms on the Intestinal Mucosa of Rat Following Irradiation)

  • 목적 : 최근 신호전달체계에서 중요한 효소로 알려진 phosphollpase C(PLC) 동위효소들의 발현이 조직의 종류와 발달과정에 따라 특이한 양상을 보이고 $PLC-{\gamma}1$은 세포의 성장, 분화 및 증식에 중추적 역할을 하는 것으로 알려져 있다 방사선 조사 후 세포내 신호전달에 관한 연구도 최근 활발하여 소장 점막의 재생에 $PLC-{\gamma}$ 및 ras 암 유전자단백이 관여하고, 조직 손상에 protein kinase C(PKC)가 관여하는 등 연구들이 보고되었으나 이들의 연구가 단편적이며 아직 확실히 밝혀진 바가 없다. 본 연구는 백서의 소장에 방사선을 조사하여 PLC 동위효소, epidermal growth factor receptor(EGFR), ras 암유전자단백, 및 PKC와 같이 신호전달체계에 관여하는 물질들의 발현을 시간적으로 관찰하여 방사선에 의한 소장 조직의 손상 및 재생 기전을 밝히고자 하였다. 대상 및 방벌 실험동물로 암.수 구별없이 생후 4-5개월, 체중 250-3009의 백서(Spraque-Dawley) 60마리를 대상으로 하여 실험군으로 전신에 8Gy의 방사선을 조사하고 방사선 조사 후 1일, 3일, 5일, 7일,14일에 각각 10마리씩 희생시켜 소장을 적출하여 사용하였고, 정상대조군은 각 시기별로 2마리씩 사용하였다. 적출된 소장의 반은 즉시 얼려 PLC의 면역블로팅 및 phosphoinositide(PI) 가수분해 활성도 측정에 사용하였고, 나머지 반은 포르말린에 고정한 다음 파라핀에 포매하여 조직병리검색과 면역조직화학염색에 사용하였다. EGFH, ras 암유전자단백, PLC, PKC의 발현은 면역조직화학염색법으로 관찰하였다. 점막세포의 재생여부는 광학현미경상의 유사분열 수와 proliferating ceil nuclear antigen(PCNA) kit를 이용한 증식세포핵 수로 확인하였다. PLC는 $PLC-{\beta},\;-{\gamma},\;-{\delta}$ 발현을 모두 검색하였고, 각각 면역블로팅과 Pl 가수분해 활성도 측정으로 확인하였다. 결과 : 1) 조직병리학 소견상 방사선 조사에 의한 소장의 조직손상은 1일부터 관찰되어 3일까지 심하였고 재생은 3일과 5일에 현저하였다. 2) 면역조직화학염색 결과 $PLC-{\gamma}1$의 발현은 재생을 보이는 3일과 5일에 발현되었으며 5일째의 점막에서 가장 강하게 나타났다. $PLC-{\delta}1$은 방사선 조사 후 손상을 보이는 1일과 3일의 점막에서 강한 발현을 나타내었다. $PLC-{\beta}1$은 모든 실험군에서 발현되지 않았다. 면역블로팅 결과도 면역조직화학염색과 일치하는 결과를 보였다. 3) $PLC-{\gamma}1$의 활성도를 보기 위한 PI 가수분해 활성도 측정결과는 3일과 5일에 현저히 높은 수치를 보여 방사선 조사후 소장점막의 재생과정의 중요한 신호전달과정이 PLC-1이 관여하는 PI 가수분해에 의해 이뤄짐을 알 수 있었다. 4) ras 암유전자단백의 발현온 재생이 시작되는 3일부터 나타나 7일까지 지속되었고, EGFR 도 재생시기인 3일과 5일에 가장 강하게 나타났고 그 후 점차 감소하는 추세를 보였다. 한편 PKC는 발현이 미약했으나 증식지수가 높은 점막에 3일과 5일에 발현이 관찰되었다. 결론 : 방사선조사에 의한 소장점막조직의 손상 및 재생과정의 신호전달기전에 PLC의 신호전달체계에 관여하는 효소가 중요한 역할을 하며 특히 $PLC-{\gamma}1$과 ras암유전자단백, EGFR, PKC는 방사선조사 후 공장점막세포의 재생기전에 주로 발현되어 재생과정과 관련된 신호전달기전에 관여함을 나타내었다. $PLC-{\delta}$은 방사선 조사 후 세포의 손상시기에 강한 발현을 보여 특히 손상과정과 관련된 신호전달기전에 관여함을 추측할 수 있었다. $PLC-{\beta}1$의 발현은 모든 실험군에서 음성인 소견을 보여 방사선조사 후 소장점막의 세포손상 및 재생과정에서 $PLC-{\beta}1$과 관련된 신호전달체계는 관여하지 않음을 알 수 었었다. 그러나, 이러한 신호전달체계의 기전을 구체적으로 밝히기 위해서는 추후 지속적인 연구가 필요하다고 사료된다.

방사선 조사후 백서 공장 점막의 재생과정에서 5-fluorouracil 투여가 phospholipsse C 와 ras 암유전자단백의 발현에 미치는 영향 (The Influence of 5-Fluorouracil Administration Mode on the Expression of Phospholipase C and Ras Oncoprotein Associated with Regeneration of Rat Intestinal Mucosa Following Radiation)

  • Purpose : Phospholipase C(PLC) isozymes play significant roles in transmembrane signal transduction. PLC-${\gamma}1$ acts as the intracellular effector in signal transduction for cellular proliferation and differentiation. Ras oncoprotein is also involved in cell growth. We determined the biological significance of PLC and ras oncoprotein in regeneration following radiation and the effect of different modes of administration of 5-FU. Materials and Methods : To determine the effect of the administration mode of 5-FU on the regeneration of intestinal mucosa of rats following radiation, we compared the expression of PLC and ras oncoprotein in six groups. Group I had no treatment. Group II received radiation(8 Gy) only. Group III received radiation(8 Gy) and 5-FU(150mg/kg) continuous intravenous (iv) infusion for 12 hours. Group IV received radiation(8 Gy) and 5-FU(750mg/kg) iv bolus injection. Group V received only 5-FU(150mg/kg) continuous iv infusion for 12 hours, Group VI received only 5-FU (150mg/kg) iv bolus injection. Through immunoblotting and immunohistochemistry, we examined the expression of PLC and ras oncoprotein in rat jejunum at 96 hours after radiation or 5-FU administration and at 120 hours after radiation and 5-FU adminstration. We also investigated the histological findings using hematoxylin and eosin stain. Results : In the immunohistochemistry study, PLC-${\gamma}1$ expression was the highest in group III followed by groups II and VI in that order and was weakly positive in groups V and VI. PLC-${\gamma}1$ was hardly detected in the control group. The expression of ras oncoprotein was the same as the PLC-${\gamma}1$ expression for all groups. These results were confirmed by the histological findings regarding the mucosal regeneration. In the immunoblotting analysis, PLC-${\gamma}1$ expression was the highest in group III followed by group IV and II in that order. This difference between the immunoblotting and immunohistochemistry study was due to the high expression of PLC-${\gamma}1$ on the damaged surface epithelium rather than to its expression in the regeneration region as observed in the immunohistochemistry study for group IV. The expression of PLC-${\delta}1$ was positive only in group V and VI, which received both radiation and 5-FU, and the expression of PLC-${\beta}1$ was negligible for all groups. Conclusion : These results suggest that PLC-${\gamma}1$ mediated signal transduetion and ras oncoprotein may have a significant role in mucosal regeneration after radiation, and that continuous iv infusion of 5-FU may induce active regeneration in intestinal mucosa following radiation. In addition, the expression of PLC-${\delta}1$ in combined group of radiation and 5-FU implies that PLC-${\delta}1$ may be involved in signal transduction mediated by concerted action between radiation and 5-FU.

PI3K, Akt, p38을 포함한 인산화단백질에 대한 Cordycepin의 억제효과 (The Inhibitory Effects of Cordycepin on Phosphoproteins including PI3K, Akt, and p38)

  • 진균 속에 속하는 종인 Cordyceps는 중국의 전통약제로서, 그 유효성분인 cordycepin이 혈소판 응집에 관여한다는 보고가 있지만 phosphoprotein 조절에 관련된 연구는 미흡하다. 본 연구에서는, cordycepin이 fibrinogen binding에 관여한다고 알려진 PI3k/Akt와 $TXA_2$ 분비 및 과립방출에 관여한다고 알려진 p38와 같은 phosphoprotein의 인산화를 어떻게 조절하며 혈소판응집을 억제시키는지 규명하고자 하였다. 그 결과, cordycepin가 $261.1{\mu}M$$IC_{50}$으로 collagen이 유도한 혈소판 응집을 강력하게 억제하였고, PI3K와 Akt의 인산화를 감소시키며 ${\alpha}IIb/{\beta}_3$에 대한 fibrinogen 결합을 농도의존적으로 억제하였다. 또한, cordycepin은 collagen이 촉진시킨 p38의 인산화를 억제함으로써, 과립방출의 지표인 ATP 과 serotonin의 방출을 억제하였고 COX-1과 TXAS의 활성 및 $PLC-{\gamma}_2$ 인산화에 대한 영향없이 $TXA_2$ 생성량을 감소시켰다. 따라서, cordycepin은 PI3K/Akt, p38와 같은 phosphoprotein의 인산화를 억제함으로써 혈소판 응집억제를 나타내는 항혈전 치료 및 예방약물로서 유용한 가치가 있다고 여겨진다.

약침용 봉독성분 melittin의 영향에 의한 인체 폐암세포의 apoptosis 유도 (Melittin-induced Aapoptosis is Associated with Inhibition of COX-2 and hTERT Expression in Human Lung Carcinoma A549 Cells)

  • Objective : To investigate the possible molecular mechanism(s) of melittin as a candidate of anti-cancer drug, we examined the effects of the compound on the growth of human lung carcinoma cell line A549. Methods: MTT, morphological changes, DAPI staining, Western blot, RT-PCR and in vitro prostaglandin E2 (PGE2) accumulation assays were performed. Results: The anti-proliferative effect by melittin treatment in A549 cells was associated with morphological changes such as membrane shrinking and cell rounding up. Melittin induced apoptotic cell death in a concentration-dependent manner, which was associated with inhibition or degradation of apoptotic target proteins such as ${\beta}$-catenin, poly(ADP-ribose) polymerase(PARP) and phospholipase $C-{\gamma}1(PLC-{\gamma}1)$. Melittin treatment inhibited the expression of cyclooxygenase-2(COX-2) and accumulation of PGE2 in aconcentration-dependent fashion. In addition, Melittin treatment induced the down-regulation of telomerase reverse transcriptase(hTERT) and proto-oncogene c-myc expression of A549 cells. Conclusions: Taken together, these findings suggest that melittin-induced inhibition of human lung cancer cell proliferation is associated with the induction of apoptotic cell death via regulation of several major growth regulatory gene products, and melittin may have therapeutic potential in human lung cancer.
