Use of hot water, combination of hot water and phosphite, and 1-MCP as post-harvest treatments for passion fruit (Passiflora edulis f. flavicarpa) reduces anthracnose and does not alter fruit quality

  • This research aimed to evaluate the effectiveness of hot water ($43-53^{\circ}C{\cdot}5min^{-1}$; $47^{\circ}C{\cdot}2-6min^{-1}$), 1-methylcyclopropene (1-MCP) at $50-300nL\;L^{-1}$ and a combination of hot water ($47/49^{\circ}C{\cdot}5min^{-1}$) and phosphite $40%\;P_2O_5+20%\;K_2O$;$40%\;P_2O_5+10%\;Zn$) in anthracnose control and the effect on fruit quality [fresh weight loss (FWL-%); pH, total soluble solids ($TSS-^{\circ}Brix$), and titratable acidity (TA = % citric acid (CA)] of passion fruit ( Passiflora edulis f. flavicarpa ) at the postharvest stage. When the fruits were in the stage of 0% dehydration and fully yellow peels, they were disinfested and inoculated with Colletotrichum gloeosporioides. They were then subjected to the above mentioned treatments; this was followed by incubation for 120 h. The diameter of the disease lesions was monitored daily. After the incubation, a physico-chemical analysis was performed. Hot-water treatment resulted in disease reduction at 47 and $49^{\circ}C$ for 4 and 5 min. The combination of hot-water treatment at $47^{\circ}C$ (4 or 5 min) and application of the phosphite of K or Zn significantly reduced disease severity in fruits. The 1-MCP treatment reduced anthracnose severity in passion fruit mainly at $200nL\;L^{-1}{\cdot} 24h^{-1} $. None of the treatments significantly changed the physico-chemical characteristics of the fruit [FWL (2.6-4.1%); pH (3.2-3.5), TSS ($8.9-10.9^{\circ}Brix$), and TA (1.8-2.5% CA)].

  • For the development of higher-sizing performance of paper, a sizing agent using recycled PET was synthesised. Polyester resin was extracted from wasted PET by subcritical hydrolysis and finally modified to synthetic sizing agent by mixing water-disperse PET with triphenyl phosphite(TPP). The modified PET was considered as an internal sizing agent in different wet-end papermaking conditions. The optimum condition in sizing efficiency was obtained in initial pH of 6.5 in case of rosin+alum system, and 7.5 in case of rosin+alum+PET system, respectively, and in addition amount of PET 3%. The sizing efficiency was also closely related according to the fiber properties of wet-end slurry, such as virgin fibres from UKP and recycled fibres from KOCC. The application of modified PET was good in strength improvement of paper, specially in tensile strength.

  • 7- Bromomethylbenz[a]anthracene is a known mutagen and carcinogen. The two major DNA adducts produced by this carcinogen, i.e., $N^2$-(benz[a]anthracen-7-yl methyl)-2'-deoxyguanosine (2, b[a]$a^2$G) and $N^6$-(benz[a]anthracen-7-ylmethyl)-2'-deoxyadenosine (4, b[a]$a^6$/A), as wel 1 as the simpler benzylated analogs,$N^2$-benzyl-2'deoxyguanosine (1, $bn^2$G) and $N^6$-benzyl-2'-deoxyadenosine (3, $bn^6$/A), were prepared by direct aralkylation of 2'-deoxyguanosine and 2'-deoxyadenosine. To determine the site-specific mutagenicity of these bulky exocyclic amino-substituted adducts, the suitably protected nucleosides were incorporated into 16-base oligodeoxyribonucleotides in place of a normal guanine or adenine residues which respectively are part of the ATG initiation codon for the lac Z' \alpha-complementation gene by using an in situ activation approach and automated phosphite triester synthetic methods. The base composition and the incorporation of the bulky adducts into synthetic oligonucleotides were characterized after purification of the modified oligonucleotides by enzymatic digestion and HPLC analysis.

Paper Disk를 지지체로 이용한 Oligodeoxyribonucleotides 화학적 합성 (Chemical Synthesis of Oligodeoxyribonucleotides Using Paper Disk as a Support)

  • Whatmann chromatography paper(3MM)을 지지체로 사용하여 phosphite-triester법에 의해 미량의 염기가 부착된 5'-d(GAATTCCGGCCA)와 5'-d(GGAGCTGTC)의 두 oligodeoxyribonucleotide를 합성 하였다. Trityl yield 분석에 의하면 76.1${\%}$와 86.5${\%}$의 평균 coupling 수율을 각각 얻었다.두 oligodeoxyribonucleotide들을 HPLC를 이용하여 분리 정제할 수 있었고 snake venom phosphodiesterase와 bacterial alkaline phosphatase에 의해 분리 정량화할 수 있었으며 이때의 염기의 수는 합성을 시도한 염기수와 일치했다. 이처럼 paper를 지지체로 이용한 oligodeoxyribonucleotide의 합성은 값싸고 동시에 많은 염기가 부착된 미량의 DNA를 빠르고 짧은 기간에 수행할 수 있는 장점이 있다.

  • A simple efficient and neat synthesis of ${\alpha}$-hydroxy phosphonates has been accomplished from aromatic/heterocyclic aldehydes and diethyl phosphite using $KHSO_4$ as a catalyst under solvent-free conditions at ambient temperature. These compounds are characterized by spectral and analytical data and tested for their anti-oxidant activity.

  • 합성 DNA는 분자 생물학의 모든 분야의 연구에 사용될 수 있으며 앞으로도 그 이용도는 더욱 증가할 것으로 전망된다. 합성 기술의 발달로 생리활성을 가진 인조 유전자를 제조하는 것이 가능해졌으며 또한 다른 방법으로는 어려운 유전자의 분리와 cloning, site specific mutagenesis, 질병의 진단, 유전자의 구조 및 기능의 연구등 수많은 분야의 연구에 이용되고 있다. 본 고에서는 현재 여러 군데 분자 생물학 연구실에서 성공적으로 사용되고 있는 자동 합성기에 의한 phosphite합성법의 기초 이론과 합성된 oligonucleotide의 정제법에 대하여 간단히 서술하고 그 응용 방법에 대하여 논하고자 한다.

폴리락트산/폴리카프로락톤 블렌드의 결정화 거동 (Crystallization Behavior of Poly(lactic acid) / Poly($\varepsilon$-caprolactone) Blends)

  • 폴리락트산/폴리카프로락톤 (PLA/PCL) 블렌드의 조성비에 따른 상용성 및 결정화 거동을 살펴보았다. 특히 triphenyl phosphite (TPP)가 반응 상용화제로써 이들 블렌드의 상용성과 결정화 거동에 미치는 영향을 중점적으로 고찰하였다. PLA/PCL 블렌드의 열적 특성을 살펴보아 이들 블렌드가 비상용성 블렌드임을 확인하였으며 블렌드의 조성비에 따라 상용성 정도가 달라졌다. 블렌드에 TPP를 첨가하는 경우 TPP가 반응 상용화제로 작용하여 블렌드의 상용성을 증가시키며 상용화제 함량 증가에 따라 상용성이 우수해졌다. PLA에 PCL을 첨가하는 경우, 블렌드 조성비에 따른 비상용 특성에 의한 계면 형성이 구정 형성에 필요한 기핵 발현 정도를 변화시켜 결정화 속도가 달라졌다. TPP는 이들 블렌드의 상용성의 개선과 동시에 구정 성장을 촉진하여 PLA의 결정화 속도를 증가시킴을 확인하였다.
