Extraction yield and nutraceuticals of mushroom Pholiota species (비늘버섯류의 추출수율 및 영양 약리학적 특성)

  • This study was performed to obtain basal data for development of new functional food by using mushroom Pholiota spp.. Among several kinds of extracts, water extracts of Pholiota adiposa ASI 24027 showed the highest solid yield of 58.7%. Crude protein of Pholiota adiposa ASI 24015 and ${\beta}$-glucan contents of Pholiota adiposa ASI 24005 were 25.0% and 0.66%, respectively. K and Mg were also contained plentifully in the fruiting body of Pholiota adiposa ASI 24004 (3,649 mg and 138 mg/100g dried fruiting body, respectively). Amino acid and total fatty acid contents were similar between almost all of Pholiota spp..

  • The genus Pholiota (Strophariaceae, Basidiomycota) is made up of wood-rotting saprotrophic mushrooms characterized by a yellow or brown pileus with scales and/or slimy, and by a brownish smooth spore with a germ pore. However, these features are not enough to distinguish its species, or separate the genus Pholiota from other brown-spored wood-rotting genera such as Hypholoma and Stropharia. Although internal transcribed spacer (ITS) sequencebased identification has improved identification accuracy for species of Pholiota, most Pholiota species in Korea are reported based on morphological features. To evaluate the taxonomy of Pholiota species, we investigated 62 specimens collected from 1999 to 2019 in Korea using ITS sequence analysis and morphological observation. Twelve of the 16 recorded Pholiota species in Korea were identified. While eight species were clearly separated, the ITS analysis did not distinguish three in the Pholiota adiposa complex. Therefore, further investigation is required to distinguish these three species. ITS sequences deposited in GenBank confirm that P. highlandensis exists in Korea. The presence of the other four Pholiota species could not be confirmed through specimens or sequence information in GenBank. A taxonomic key and the ITS sequence data for Korean Pholiota species are included and can be good baselines for further research on Pholiota taxonomy and diversity.

  • To elucidate the composition of phytosterols in the fruit body of Pholiota spp. 6 species(P. adiposa, P. aurivella, P. highlandensis, P. nameko, P. squarrosa and Pholiota sp.) were analyzed with Gas chromatography(GC). Pholiota spp. were contained campesterol, stigmasterol, and $\beta$-sitosterol as the major phytosterols.

Pholiota adiposa and its Related Species Collected from the Wild Forestry (야생에서 채집된 검은비늘버섯(Pholiota adiposa)균에 관한 연구)

  • Five basidiocarps of Pholiota species have been collected from the areas of BubJu Temple for last two years, and identified to those of P. adiposa or Pholiota species. The taxonomy of these basidiocarps with the morphological aspects was compared with the analysis obtained from the polymorphisms of PCR-RAPD bands made after reacted with the random primers. The polymorphic variations were observed within the species of the basidiocarps, but not between genomic DNA's of the mycelia obtained and the basidiocarps. Several different bands made from the primers (28 and 36) in PCR-RAPD reactions were identified within the genus of Pholiota and speculated to be specific for the individual basidiocarp of P. adiposa collected. The primers employed here were considered to be very useful for distinguishing the individual isolates or basidiocarps collected from the fields. Also, the basidiospores were obtained from the sporeprints of the above basidiocarps as a simple agar and confirmed with observations of clamp connection under microscopes. The matings of them indicated the 'tetrapolar' type, being different from the 'bipolar' type reported by Japanese basidiocarps of P. adiposa or P. nameko. Based on our work, the edible fungi collected were speculated to be a new breeding resource for those of Pholiota commercialized in Japan.

The Fruit-body Formation and Properties of Pholiota sp. (비늘버섯속균(Pholiota sp.)의 특징과 자실체 형성)

  • Pholiota sp. were collected from the forest of Kangwon province (Mt. Samak, Mt. Odae and Kangwon National University Forest Experimentation) from the early of August to the end of October 1997. Among these fungi, Poliota adiposa was selected for culture experiment because it is suitable for edible mushroom. The Optimal temperature for the mycelial growth of Pholiota adiposa was in the range of $28^{\circ}C$ while that of fruit body formation was $15{\pm}1^{\circ}C$. Brown sugar as carbon source and soybean flour as nitrogen source were good for mycelial growth in commercial liquid culture. The fruit bodies of Pholiota adiposa were artifitially produced on plastic bottle including poplar sawdust and rice bran (4:1).

  • Cultivation of Pholiota adiposa on sawdust media and its mixed substrates with various agricultural by-products was attempted. The composition of each substrates were compared from the viewpoint of cultivation possibilities with the production of Pholiota adiposa. Populus deltoides showed the best production of the mushroom with 795 g, followed by Robinia pseudo-acacia and Quercus acutissima. The mixed sawdust media with 70% of Robinia pseudo-acacia and 30% of Populus deltoides showed the best production of the mushroom with 265 g followed by the mixture of Robinia pseudo-acacia and Populus deltoides with 50% each with 228 g, while the addition of Quercus acutissima showed the negative effect for the production of Pholiota adiposa. Thus, Robinia pseudo-acacia was considered to be an effective amendment for the production of the mushroom. In addition, various kinds of agricultural by-products showed additive effect for the production of the mushroom, too. Therefore, we could concluded that the sawdust of Robinia pseudo-acacia and agricultural by-products can be used to cultivate Pholiota adiposa.

  • Among the higher fungi which wore collected in the Geong Gi Province and Seoul Korea, nine species, i.e., Mycena haematopoda, Amanita griseofarinosa, Pholiota spectabilis. Pholiota vermiflua, Boletus erythroporus, Russula alutacea, Russula chameleontina, Russula xcampelina and Polyporus sulphureus, were found to be unrecorded species in Korea. The morphological, histological and microchemical characteristics of these fungi were reported.

Studies on Antitumor Components of Wild Pholiota squarrosa (Fr.) Quel. (야생 비늘 버섯의 항암 성분에 관한 연구)

  • In order to investigate the antitumor components of Korean wild higher fungi, the carpo-phores of Pholiota squarrosa collected in Kaung Nung area were extracted with water by refluxing for eight hours at 80-90.deg. C. The extracts were dialized for six days against distilled water and lyophilized to produce crude polysaccharide powders. They were found to have the antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Pholiota squarrosa was 78.7% in the dose of 20mg/kg/day for the period of ten days. The tumor in two of the ten mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The total polysaccharide was 42% by Anthrone method and the protein was 55% by Lowry-Folin method in the antitumor component of Pholiota squarrosa.

  • Present experiments were conducted to determine the possibility of cultivation of 9 edible wild species selected among the higher fungi growing in Korea. In the investigation on the mycelial growth according t6o the different media, the mycelial growth of Coprinus comatus was fast on the CSA medium, when malt extract was added to the basal medium PSA, the mycelial of Lapista nuda and Auricularia auricula-judae was fast in growth and density. In the spawning, the mycelial growth of Pholiota squarrosa on the oak tree's sawdust, Pleurotus cornucopiae on the broad-leaves' sawdust, and Coprinus comatus on the compost was respectably fast and also it shown to be possibility of artificial cultivation owing to their carporphore budding when Coprinus comatus and Lepiota alborubescens cultivated on the rice straw, Auricularia auriculajudae and Pleurotus cornucopiae on the sawdust of the popla and Pholiota squarrosaon the sawdust of the oak tree.

Phylogenetic relationships in different strains of Pholiota species based on PCR polymorphism (PCR 다형성 분석에 의한 비늘버섯 속 계통의 유연관계 분석)

  • Pholiota species were collected from different geographical regions of the world. Genetic diversity and phylogenetic relationships were analyzed by rDNA-ITS sequences and RAPD polymorphism. The sizes of rDNA-ITS PCR amplicons of Pholiota spp. varied from 233~271, 158~223 and 174~219 bp, respectively. A phylogenetic tree was constructed on the ITS region sequences and Pholiota strains were classified into 8 clusters. Twenty strains in seven Pholiota spp. were classified into seven clusters by RAPD polymorphism using 15 arbitrary primers. Our experimental results suggested that rDN-ITS and RAPD analysis are useful tool for classifying Pholiota spp. and strains.
