감나무 잎에서 분리한 Phialocephala piceae에 대한 보고 (A New Report on Phialocephala piceae Isolated from Leaf of Diospyros kaki in Korea)

  • 경상북도 상주시에 재식된 감나무 잎의 표면에서 국내 미기록종인 곰팡이를 분리하여 형태학적 및 계통학적 분석을 수행하였다. 분리된 곰팡이는 Phialocephala piceae의 형태적 특징인 암갈색 균총과 굵고 짧은 분생자경 유사조직을 관찰할 수 있었다. Phialocephala속의 동정에 사용되는 internal transcribed spacer 영역과 RNA polymerase II largest subunit 유전자를 이용한 계통학적 분석을 통해 해당균이 P. piceae임이 확인되었고, 이를 통해 최초로 국내에 P. Piceae가 존재함을 보고하였다.

  • A unrecorded hyphomycete species of Phialocephala was isolated for the first time during the investigation of fungal community in the soil samples collected from different regions of Korea. The fungal isolate was identified as Phialocephala lagerbergii, based on the morphological characteristics and phylogenetic analysis of the ribosomal DNA sequence. In addition, cultural and micro-morphological features were described in detail.

  • A dark septate endophytic fungal strain 13E043 was isolated from the needle leaves of Thuja koraiensis from a forest in Korea. Morphological characteristics of conidia and phialids. Along with a molecular phylogenetic analysis based on the internal transcribed spacer region of rDNA, the isolate was identified as Phialocephala piceae (also known as Phaeomollisia piceae). This is the first report of a dark septate endophyte isolated from the foliage of conifer trees in Korea.

감자난초 뿌리에서 분리한 내생균의 동정 (Notes on Endophytic Fungi Isolated from Roots of Oreorchis patens in Korea)

  • 본 연구에서는 강원도 함백산에서 감자난초(Oreorchis patens)의 뿌리를 채집하여 내생균을 순수분리하였고, 분리된 균주의 형태학적 특징 및 분생포자 등을 관찰하였다. 또한 진균 특이적 primer인 ITS1F와 ITS4를 이용하여 internal transcribed spacer rDNA 영역의 염기서열을 분석하고, primer LR0R과 LR16을 이용하여 large subunit rDNA 영역의 염기서열 분석하여 분자적으로 균을 동정하였다. 그 결과 Thelonectria veuillotiana, Phialocephala humicola 등의 2종의 국내 미기록 내생균을 동정하였고, 이를 보고하고자 한다.

  • 본 연구는 전라북도 진도에서 자란(Bletilla striata), 강원도 정선군 함백산에서 감자난초(Oreorchis patens)와 은대난초(Cephalanthera longibracteata)의 뿌리에서 내생균을 분리하고 이들의 형태 및 분자생물학적 방법을 통해 동정하였다. 각 균주의 형태학적 특징을 분석하기 위해 포자의 모양과 특징을 관찰하였고, 분자생물학적 동정을 위해 ITS1F와 ITS4 primer를 이용하여 internal transcribed spacer (ITS) 및large subunit (LSU) rDNA영역을 분석하였다. 그 결과 Phialocephala bamuru, Coniochaeta mutabilis, Phialophora foetens, Calonectria Canadensis, Neonectria ramulariae 5종의 국내 미기록 내생균이 동정되어 이를 보고하고자 한다.

Occurrence of Sporendocladia bactrospora on Quercus variabilis in Korea

  • A survey to assess the diversity of wound-associated Ophiostomatales and Microascales, the so-called ophiostomatoid fungi, on Korean native trees, was undertaken in 2017. Wounds were artificially created, and a fungus resembling a species of Sporendocladia was consistently isolated from the exposed cambium and inner bark of artificially induced wounds on Quercus variabilis. Morphological examination and DNA sequence comparisons based on the internal transcribed spacer (ITS) and 5.8S regions of the rDNA confirmed the identity of the fungus as Sporendocladia bactrospora. This is the first report on S. bactrospora occurring on Q. variabilis in Korea.

Pyrosequencing and Taxonomic Composition of the Fungal Community from Soil of Tricholoma matsutake in Gyeongju

  • Tricholoma matsutake is an ectomycorrhizal fungus that has a symbiotic relationship with the root of Pinus densiflora. Soil microbial communities greatly affect the growth of T. matsutake, however, few studies have examined the characteristics of these communities. In the present study, we analyzed soil fungal communities from Gyeongju and Yeongdeok using metagenomic pyrosequencing to investigate differences in fungal species diversity, richness, and taxonomic composition between the soil under T. matsutake fruiting bodies (Sample 2) and soil where the fairy ring of T. matsutake was no longer present (Sample 1). The same spot was investigated three times at intervals of four months to observe changes in the community. In the samples from Yeongdeok, the number of valid reads was lower than that at Gyeongju. The operational taxonomic units of most Sample 2 groups were less than those of Sample 1 groups, indicating that fungal diversity was low in the T. matsutake-dominant soil. The soil under the T. matsutake fruiting bodies was dominated by more than 51% T. matsutake. From fall to the following spring, the ratio of T. matsutake decreased. Basidiomycota was the dominant phylum in most samples. G-F1-2, G-F2-2, and Y-F1-2 had the genera Tricholoma, Umbelopsis, Oidiodendron, Sagenomella, Cladophialophora, and Phialocephala in common. G-F1-1, G-F2-1, and Y-F1-1 had 10 genera including Umbelopsis and Sagenomella in common. From fall to the following spring, the amount of phyla Basidiomycota and Mucoromycota gradually decreased but that of phylum Ascomycota increased. We suggest that the genus Umbelopsis is positively related to T. matsutake.
