Post-Infectional Biochemical Changes in Mulberry Due to Xanthomonas campestris pv. mori Induced Bacterial Leaf Spot

  • Post-infectional biochemical changes due to Xanthomonas campestris pv. mori (Xcm) infection in five elite mulberry varieties viz., $S_1$, $S_{1635}$, $V_1$, RF $S_{175}$ and JRH was studied under inoculated condition. It was revealed that total soluble sugar and protein content was significantly declined in all the varieties due to X. campestris infection. Total phenol content was at par prior to inoculation in all varieties, but it was significantly increased in $S_1$, RF $S_{175}$, $S_{1635}$ and JRH 7 days after inoculation. The correlation coefficient (r) between total soluble sugar and total phenol content was found positive (r = 0.825) and statistically significant. Similarly, correlation coefficient (r) between total soluble protein and phenol content was found positive (r = 0.897) and statistically significant. The present study indicates that X. campestris infected leaves are nutritionally inferior in quality and the duration of phenol production in a mulberry variety play decisive role on disease resistance.nce.

Protective Effects of SAPP, a Novel Herbal Complex, in Acute Hepatotoxic Mouse Model

  • The protective effect of SAPP, an extract from a novel herbal complex, on acute liver injury was investigated using mouse animal model in this study. The content of total phenol in SAPP was increased at dose dependent manner. Consistent with the content of total phenol, SAPP showed the significant anti-oxidative effects on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) method. Acute liver injury was induced by D-galactosamine (D-GalN) in mouse. Treatment with SAPP significantly reduced the level of alanine transaminase (ALT) and aspartate transaminase (AST) in serum. Histological observation revealed that whereas D-GalN treated mouse showed vacuolization of hepatocytes, sinusoidal dilation and congestion, loss of cell boundaries and ballooning degeneration, loss of architecture and cell necrosis, treatment with SAPP improved D-GalN-induced liver injury. These results suggest that SAPP shows protective effects against D-GalN-induced hepatotoxicity in vivo acute mouse model.

Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.

Pseudomonas putida F1과 Burkholderia cepacia G4에 의한 BTEX, trichloroethylene 분해 (Degradation of BTEX and Trichloroethylene by Pseudomonas putida F1 and Burkholderia cepacia G4)

  • Two cometabolic trichloroethylene (TC) degraders, Pseudomonas putida F1 and Burkholderia (Pseudomonas) cepacia G4, were found to catabolize phenol, benzene, toluene, and ethylbenzene as carbon and energy sources. Resting cells of P. putida F1 and B. cepacia G4 grown in the presence of toluene and phenol, respectively, were able to degrade not only benzene, toluene and ethylenzene but also TCE and p-xylene. However, these two strains grown in the absence of toluene or phenol did not degrade TCE and p-xylene. Therefore, it was tentatively concluded that cometabolic degradation of TC and p-xylene was mediated by toluene dioxygenase (P. putida F1) or toluene-2-monooxygenase (B. cepacia G4). Maximal degradation rates of BTEX and TCE by toluene- and phenol-induced resting cells of P. putida F1 and B. cepacia G4 were appeared to be 4-530 nmol/(min$.$mg cell protein) when a single compound was solely served as a target substrate. In case of double substrates, the benzene degradation rate by P. putida F1 in the presence of toluene was decreased up to one seventh of that for the single substrate. TCE degradation rate was also linearly decreased as toluene concentration increased. On the other hand, toluene degradation rate was enhanced by benzene and TCE. For B. cepacia G4, degradation rates of TCE and toluene increased 4 times in the presence of 50 ${\mu}$M phenol. From these results, it was concluded that a degradation rate of a compound in the presence of another cosubstrate(s) could not be predicted by simply generalizing antagonistic or synergistic interactions between substrates.

식용 어패류 조직중의 glutathione S-transferase 활성과 화학물질 오염에 의한 변화 (Glutathione-S-transferase Activity and its Changes to Chemical Pollution in Edible Shells and Fishes)

  • 본 연구는 식용으로 사용되는 어패류의 화학오염지표로서 glutathione S-transferase (GST)의 활성을 사용 할 수 있는 가를 검토하기 위하여 수행하였다 어류의 간췌장 및 패류의 소화선에서 기초적 GST 효소활성은 시험한 동물종에 따라 차이를 보였다. 시험한 어패류 중 큰이랑 피조개에서의 활성이 가장 높았으며 메기 및 홍합이 그 다음이었다. 백합 및 이스라엘 잉어에서는 낮은 기초적 활성을 보여 주었다. 큰이랑 피조개를 전형적인 PAH물질인 3-methylcholanthrene에 1주일간 노출 시켰을 경우 GST의 활성은 약 30% 감소하였으며 노출중단 2주경에는 회복되었다. 다른 대부분의 동물종에서는 GST의 활성이 3-MC에 의하여 증가하였다. 홍합의 경우 기초 활성의 약 200%수준으로 증가하여 노출중단 후에도 1주일간 지속되다가 서서히 감소하였다. 이스라엘 잉어에서도 홍합과 유사한 반응이 관찰되었다. Phenobarbital은 홍합 및 이스라엘 잉어에서 GST활성을 증가시켰다. Clobibrate, butylated hydroxyanisole 및 oxolinic acid 등은 효소활성의 변화를 유발하지 아니하였다. 한편 phenol은 이스라엘 잉어에서 활성을 감소시켰다. 이 결과를 종합하면 식용 어패류의 정상적 GST활성은 동물종에 따라 큰 차이가 나며 화학물질 오염에 따른 변화도 증가, 감소 및 불변으로 다양한 것으로 관찰되었다. 따라서 이 효소의 활성을 측정함으로서 PAH나 phenol과 같은 환경 오염물질에 의한 오염정도를 추정할 수 있는 지표로의 사용이 가능하리라고 본다.

Plant Terpene-Induced Expression of Multiple Aromatic Ring Hydroxylation Oxygenase Genes in Rhodococcus sp. Strain T104

  • Recent studies have shown that some of the PCB (polychlorinated biphenyl)-degraders are able to effectively degrade PCB in the presence of monoterpenes, which act as inducers for the degradation pathway. Rhodococcus sp. T104, an effective PCB degrader, has been shown to induce the degradation pathway by utilizing limonenes, cymenes, carvones, and pinenes as sole carbon sources which can be found in the natural environment. Moreover, the strain T104 proved to possess three separate oxidation pathways of limonene, biphenyl, and phenol. Of these three, the limonene can also induce the biphenyl degradation pathway. In this work, we report the presence of three distinct genes for aromatic oxygenase, which are putatively involved in the degradation of aromatic substrates including biphenyl, limonene, and phenol, through PCR amplification and denaturing gradient gel electrophoresis (DGGE). The genes were differentially expressed and well induced by limonene, cymene, and plant extract A compared to biphenyl and/or glucose. This indicates that substrate specificity must be taken into account when biodegradation of the target compounds are facilitated by the plant natural substrates.

고종시 감껍질의 추출조건에 따른 항산화 활성 (Changes in the Antioxidant Potential of Persimmon Peel Extracts Prepared by Different Extraction Methods)

  • Background: Astringent persimmon (Diospyros kaki Thunb. cv. Kojongsi) peels are by-products of dried persimmons. This study aimed to evaluate the antioxidant activities of Kojongsi persimmon peel (KPP) extracts prepared by 15 different extraction methods: 5 heating durations (0.5 - 2.5 h) at 3 heating temperatures (50, 70, and $90^{\circ}C$). Methods and Results: An increase in heating temperature increased the antioxidant effect of KPP extracts. Those prepared by heating at 1 h had the highest total phenol content, regardless of the heating temperature. In addition, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and cell-protective effects against $H_2O_2-induced$ oxidative stress were dependent on the total phenol contents of the extract. However, the KPP-induced increased in catalase expression was dependent on heating temperature and duration. Conclusions: These results suggest that extraction by heating at $90^{\circ}C$ for 1 h may enhance KPP's antioxidant effects, which mainly involve non-enzymatic antioxidant systems.

Biocontrol Activity of Aspergillus terreus ANU-301 against Two Distinct Plant Diseases, Tomato Fusarium Wilt and Potato Soft Rot

  • To screen antagonistic fungi against plant pathogens, dual culture assay (DCA) and culture filtrate assay (CFA) were performed with unknown soil-born fungi. Among the different fungi isolated and screened from the soil, fungal isolate ANU-301 successfully inhibited growth of different plant pathogenic fungi, Colletotrichum acutatum, Alternaria alternata, and Fusarium oxysporum, in DCA and CFA. Morphological characteristics and rDNA internal transcribed spacer sequence analysis identified ANU-301 as Aspergillus terreus. Inoculation of tomato plants with Fusarium oxysporum f. sp. lycopersici (FOL) induced severe wilting symptom; however, co-inoculation with ANU-301 significantly enhanced resistance of tomato plants against FOL. In addition, culture filtrate (CF) of ANU-301 not only showed bacterial growth inhibition activity against Dickeya chrysanthemi (Dc), but also demonstrated protective effect in potato tuber against soft rot disease. Gas chromatography-tandem mass spectrometry analysis of CF of ANU-301 identified 2,4-bis(1-methyl-1-phenylethyl)-phenol (MPP) as the most abundant compound. MPP inhibited growth of Dc, but not of FOL, in a dose-dependent manner, and protected potato tuber from the soft rot disease induced by Dc. In conclusion, Aspergillus terreus ANU-301 could be used and further tested as a potential biological control agent.

Enhanced Onion Resistance against Stemphylium Leaf Blight Disease, Caused by Stemphylium vesicarium, by Di-potassium Phosphate and Benzothiadiazole Treatments

  • In this study, we investigated the induced defense response and protective effects against Stemphylium vesicarium by application of benzothiadiazole ($Bion^{(R)}$) and di-potassium phosphate salt $(K_2HPO_4)$ to onion. Onion leaves were sprayed with $Bion^{(R)}$ and $K_2HPO_4$, then inoculated 2 days later with a virulent strain of S. vesicarium under greenhouse conditions. Disease severity and activities of peroxidase (PO), polyphenoloxidase, phenylalanine ammonia-lyase (PAL) and phenol contents were evaluated in the treated leaf tissues. Reduction in the disease severity was observed in plants treated with $Bion^{(R)}$ and $K_2HPO_4$. Onion plants treated with $Bion^{(R)}$ and $K_2HPO_4$ and inoculated with the pathogen showed significantly higher PAL activity, PO activity, and phenol contents than inoculated water-treated plants 2 days after the treatment. In conclusion, the results of this study provide evidence that application of simple non-toxic chemical solutions as di-potassium phosphate and $Bion^{(R)}$ can control Stemphylium leaf blight of onion.

(E)-2-Methoxy-4-(3-(4-Methoxyphenyl)Prop-1-en-1-yl)Phenol Induces Apoptosis in HeLa Cervical Cancer Cells via the Extrinsic Apoptotic Pathway

  • (E)-2-Methoxy-4-(3-(4-methoxyphenyl)prop-1-en-1-yl)phenol (MMPP), derived from butenal, is a recently synthesized Maillard reaction product. Owing to its novelty, little is known about the function of MMPP. In this study, we elucidated the effects of MMPP on apoptosis in cervical cancer by using the HeLa cervical cancer cell line, which is widely used in cancer research. We observed that MMPP was cytotoxic to HeLa cells and induced activation of caspase-3, -8, and -9, without affecting the expression of the viral oncogenes E6 and E7. In particular, the expression of the death receptors DR5 and FAS was significantly increased by MMPP treatment. There were no significant alterations of mitochondrial intrinsic factors. Taking all these results together, our findings show that MMPP primarily induces apoptosis in HeLa cervical cancer cells via the extrinsic apoptotic signaling pathway, accompanied by an enhanced expression of death receptors.
