An Antioxidant Hispidin from the Mycelial Cultures of Phellinus linteus

  • In the course of screening for reactive oxygen species scavengers from natural products, an antioxidant was isolated from the mycelial culture broth of Phellinus linteus and identified as hispidin. The hispidin content was reached its maximum level at 12 days after onset of inoculation. About 2.5 mg/mL of hispidin was produced by P linteus in a yeast-malt medium (pH 5.8, $25^{\circ}C$). Hispidin inhibited 22.6 and 56.8% of the super oxide anion radical, 79.4 and 95.3% of the hydroxyl radical, and 28.1 and 85.5% of the DPPH radical at 0.1 and 1.0 mM, respectively. The positive control ${\alpha}-tocopherol$ scavenged 25.6 and 60.3%, 74.6 and 96.3%, and 32.7 and 77.5% of each radical, respectively, at the same concentrations. However, hispidin showed no significant activity on the hydrogen peroxide radical.

  • Many higher fungi were collected at Korea from 1976 to 1998. They were identified and surveyed on resources with many reference books. According to the results, fungal fungi were 40 families, 90 general and 215 species. Among them , anti-cancer resources used in Korea were Ganoderma lucidum, Phellinus linteus, Agaricus brazei and Cordyceps militaris. Three species exception Agaricus brazei were distributed in Korea. All these are cultivated in Korea.

Protective Effect of an Ethanol Extract Mixture of Curcuma longae Radix, Phellinus linteus, and Scutellariae Radix on Oxidative Neuronal Damage (Curcuma longae Radix, Phellinus linteus 및 Scutellariae Radix 혼합추출물의 산화성 신경세포손상 보호효과)

  • Previous work demonstrated that an ethanol extract (HS0608) of a mixture of three medicinal plants of Curcuma longae radix, Phellinus linteus, and Scutellariae radix markedly inhibits $A{\beta}$ (25-35)-induced neurotoxicity. The present study was performed to further verify the neuroprotective effect of HS0608 on oxidative and ischemic cerebral injury using cultured rat cortical neurons and rats. Exposure of cultured cortical neurons to $100\;{\mu}M$ hydrogen peroxide ($H_2O_2$) induced neuronal apoptotic death. At $10-100{\mu}g/ml$, HS0608 inhibited neuronal death, elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), and generation of reactive oxygen species (ROS) induced by $H_2O_2$ in primary cultures of rat cortical neurons. In vivo, HS0608 prevented cerebral ischemic injury induced by 2-h middle cerebral artery occlusion (MCAO) and 24-h reperfusion. The ischemic infarct and edema were significantly reduced in rats that received HS0608 (200 mg/kg). These results suggest that the anti-oxidative properties of HS0608 may be responsible for its neuroprotective effect against focal cerebral ischemic injury and that HS0608 may have a therapeutic role in neurodegenerative diseases such as stroke.

  • Phellinus linteus (Berk. & M.A. Curtis) Teng, commonly referred to as Sangwhang in Korea, is a well-known species of the genus Phellinus, which attracts great attention due to its phamarcological values. P. linteus has been reported to produce anti-tumor, anti-angiogenic, anti-mutagenic and immunomodulatory activities in vivo and in vitro. However, despite extensive biochemical studies on P. linteus, the wine produced by P. linteus fermentation (WPLF) has poorly investigated. In the present study, it was compared the in vitro cytotoxic effects of WPLF with ethanol as positive control. WPLF as well as ethanol induced the inhibition of cell proliferation and morphological changes in both HepG2 and A549 cells in a concentration-dependent manner, however, WPLG treatment has less cytotoxic effects than ethanol treatment. These cytotoxic effects were associated with the induction of apoptotic cell death, but, WPLG treatment has less apoptotisis inducing effects than ethanol treatment.

  • The present study investigated an ethanol extract (HS0608) of a mixture of three medicinal plants of Curcuma longae radix, Phellinus linteus, and Scutellariae radix for possible neuroprotective effects on neurotoxicity induced by amyloid $\beta$ protein ($A{\beta}$) (25-35) in cultured rat cortical neurons and antidementia activity in mice. Exposure of cultured cortical neurons to $10\;{\mu}M$ $A{\beta}$ (25-35) for 36 h induced neuronal apoptotic death. At $1-50\;{\mu}g/m{\ell}$, HS0608 inhibited neuronal death, elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), and generation of reactive oxygen species (ROS) induced by $A{\beta}$ (25-35) in primary cultures of rat cortical neurons. Memory loss induced by intracerebroventricular injection of ICR mice with 15 nmol $A{\beta}$ (25-35) was inhibited by chronic treatment with HS0608 (25, 50 and 100 mg/kg, p.o. for 7 days) as measured by a passive avoidance test. From these results, we suggest that the antidementia effect of HS0608 is due to its neuroprotective effect against $A{\beta}$ (25-35)-induced neurotoxicity and that HS0608 may have a therapeutic role in preventing the progression of Alzheimer's disease.

Profit Analysis in Using Small Diameter Log of Quercus species (참나무류 소경재 활용용도에 따른 수익성 분석)

  • This study aims to compare the profitability of using small diameter logs of Quercus species in terms of net profit (ratio), net income (ratio). The small logs of Quercus species could be used for the bed logs for shiitake mushroom and Phellinus linteus, sawdust, charcoal, and wood chip. The data of cost and revenue are collected by a face-to-face survey method from the associated factories. Our results show that the profitability for Phellinus linteus production is highest because its price has been formed high due to low market competition mainly resulted from capital service cost and production technique requirement. The next is followed by Shiitake mushroom, sawdust, wood chip, and charcoal. The study also reveals that profitability for long returning periods of investment is higher than that of short returning periods.

Liquid Culture of Basidiomycetes on Natural Media (천연물을 이용한 담자균의 균사체 배양)

  • This study was designed to develop functional drink from jujube extract through a simple submerged culture of three basidiomycetes species. The optimum Brix and pH of the jujube extract for culturing the Ganoderma lucidum appeared to be 5 Brix and pH 4. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Coriolus versicolor appeared to be 5 Brix and pH 5. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Phellinus igniarius appeared to be 3 Brix and pH 5. For the maximum mycelial production eighteen days of culture period was required for Phellinus igniarius. The antitumor activity of the polysaccharides extracted from the fermented drinks was demonstrated through the tumor cell line experiments. The $IC_{50}$ values of the jujube drinks fermented with Ganoderma lucidum and Phellinus igniarius against stomach cancer cell line appeared to be one fourth that of the jujube drink which was not fermented with basidiomycetes.

Optimal Conditions of Mycelial Growth and Exopolysaccharide Production in Submerged Culture of Phellinus baumii (Phellinus baumii으로부터 세포외 다당체 생산의 최적화)

  • The polysaccharide isolated from Phellinus species has been known as a folk remedy, including antitumor and immune-stimulating activities. However, there are lacks of knowledge about mycelial growth and exopolysaccharide (EH) production in its submerged culture. We investigated the optimal conditions on mycelial growth and EPS production in Phellinus baumii. The optimal temperature and initial pH for mycelial growth and EPS production in shake flask culture of P. baumii were proved to be 3$0^{\circ}C$ and pH 5.0, respectively. In case of carbon source, cellobiose and maltose were highly efficient for mycelial growth and fructose and mannitol were also relatively favorable for EPS production. Yeast extract was the most suitable nitrogen source for mycelial growth and EPS production. The composition of optimal culture medium was determined to be fructose 20 g/L, yeast extract 20 g/L, and $CaCl_2$ 0.55 g/L, respectively. Under the optimal culture condition, the maximum mycelial biomass and EPS achieved in a 5-L stirred-tank fermenter were 17.43 g/L and 3.6 g/L, respectively. It was found that the EPS was a glycoprotein onsisted of mainly arginine (14.1%) and glycine (12.0 %) in protein moiety and mainly mannose (48.7%) and arabinose (38.4%) in carbohydrate moiety.

A herbological study on the plants of Polyporaceae in Korea (한국산 구멍쟁이버섯과 다공균과(多孔菌科)에 관한 본초학적(本草學的)연구)

  • Objective : For the purpose of developing Korean herbology of the plants to Polyporaceae in Korea, the literatures of the successive generations have been thoroughly investigated to prepare this article. Methods : The examined herbological books and research paper which published domestically and abroad. Results : 1. Polyporacease plants (raised in Korea) are devided into 31 classes with 59 species. Among them, 26 species in 19 classes were found serviceable, which indicates 44 % of all Polyporacease plants. 2. Among those 59 species, $Coriolus$ $Phellinus$ $Trametes$ includes 5 species. $Ganoderma$, a medicinal plant, includes 3 species. 3. Dividing the serviceable plants of Polyporacease, the fruit body family had the largest number; 25. 4. Out of plants Polyporacease, neutral taste plants was 17 species and sweet taste was 14 species. 5. Only 8 species of Polyporaceae plants were found to enterd into the spleen and heart. 6. 11 species were found to prevent cancer, 7 species were found to dispel wind-dampness and Alleviate edem antittusive and antasth matic. 7. There were no toxic species in the Plyporaceae. Conclusion : There were totaled to 31 genera and 59 species in Polyporaceae in Korea and among them medicinal plants are 19genera, 26 species, some 44% in total.
