Composition of Crude Polysaccharides in Some Phezllinus Species (수 종 상황버섯의 조다당 분획물 조성)

  • Various Phellinus species (Hymenochaetaceae) including P. linteus, P. baumii, P. pini, and P. igniarius of domestic and foreign products, were examined by gas chromatography to validate their composition of crude polysaccharides. The crude polysaccharide fraction, which is known to be one of the physiologically active fractions, were prepared by a cold ethanol precipitation method and the yield of the crude polysaccharide fractions of P. linteus, P. baumii, P. pini, and P. igniarius were 15.0%, 5.1%, 6.3% and 5.7%, respectively. The total monosaccharide amount of the crude polysaccharide fraction of P. linteus was 148.713 mg/g (dry weight of mushroom) while those of P. baumii, P. pini and P. igniarius were 29.022 mg/g, 29.222 mg/g and 1.546 mg/g, respectively. Although the major monosaccharide of all Phellinus mushrooms was glucose, there were remarkable differences in the absolute quantity of each monosaccharide. P. linteus included plenty of monosaccharides except mannose and ribose in comparison with P. baumii and P. pini. The absolute quantities of monosaccharides of P. baumii and P. pini were nearly similar except arabinose. The total monosaccharides of P. igniarius were minimal, but the portions of galactose and mannose in P. igniarius were greater than in the other species.

PCR Based Detection of Phellinus linteus using Specific Primers Generated from Universal Rice Primer(URP) Derived PCR Polymorphic Band

  • This study was carried out to develop specific primers for PCR detection of Phellinus linteus. Diverse genomes of 15 Phellinus spp. including five Phellinus linteus isolates were fingerprinted by Primer Universal rice primer(URP)1F. The URP-PCR pattern differentiated P. linteus isolates from other phellinus spp. A polymorphic band(2.8 kb), which is unique for P. linteus isolates, was isolated and sequenced. Twenty four-oligonucleotide primer pairs were designed based on information of DNA sequence. The primer set(PLSPF2/PLSPR1) amplified single band(2.2 kb) of expected size with genomic DNA from seven Phellinus linteus, but not with that of other Phellinus species tested. The primers could be used identically in both DNA samples from mycelium and fruit bodies. This specific primers could offer a useful tool for detecting and identifying P. linteus rapidly.

Molecular Identification of Two Strains of Phellinus sp. by Internal Transcribed Spacer Sequence Analysis

  • Two species of cultivated Phellinus sp. were identified as P. baumii by internal transcribed spacer (ITS) sequence analysis. The fruit bodies of the examined strains were similar to those of naturally occurring strains, having a bracket-like form, yellow-to-orange color, and poroid hymenial surfaces. The DNA sequences of ITS region of both strains showed a homology of 99% with ITS1 to ITS2 sequences of P. (Inonotus) baumii strain PB0806.

Phylogeny of Phellinus and Related Genera Inferred from Combined Data of ITS and Mitochondrial SSU rDNA Sequences

  • To elucidate phylogenetic relationships of Phellinus and its related genera, nuclear internal transcribed spacer and mitochondrial small subunit ribosomal DNA sequences from 65 strains were determined and compared. The combined dataset of two sequences increased informative characters and led to the production of trees with higher levels of resolution. Phylogenetic analysis of the combined dataset revealed thirteen evolutionary lineages and several unresolved species that were together subdivided into two large clusters consisting of oligonucleate species and binucleate species. These results coincided with previous cytological, morphological, and molecular studies. It is newly recognized that the Phellinus linteus complex forms a sister clade to Inonotus, and that Fulvifomes is somehow related to Inocutis. The Phellinus linteus complex of dimitic perennial taxa made an independent clade from Inonotus and suggested that hyphal miticity and fruitbody permanence had enough phylogenetic significance to keep the complex within the traditional genus Phellinus. Taxa lacking setae were clustered into Fulvifomes, Phylloporia, Inocutis, and Fomitiporia, and the first three were closely related sister groups, but Fomitiporia was a genus distantly related to them. Several taxa with branched setae were shown among distantly related genera. Molecular evidence indicated that the ancestral nuclear type could be a binucleate feature, and that there might be parallel gains of branched setae and parallel losses of setae in the Hymenochaetales.

Molecular Detection of Phellinus linteus and P. baumii by PCR Specific Primer

  • Specific primer sets based on ribosomal DNA(rDNA) internal transcribed specer(ITS) sequences were designed for rapid detection of Phellinus linteus and P. baumii. Polymerase chain reaction(PCR) with these primers produced unique bands for each Phellinus species. The annealing temperature range is from $40^{\circ}C\;to\;55^{\circ}C$. The length of PCR products(P. linteus and P. baumii) using designed combinative primer sets of PL1F, PL2R, PB1F, PB2R, ITS5F and ITS4R, were from 520 by to 730 bp. Fifteen strains of Phellinus species including P. linteus, P. baumii, P. weirianus, P. johnsonianus, P. rhabarberinus, P. pini, P. gilvus, P. igniarius, P. nigricans and P. laevigatus were examined in this study. Five strains, including two isolated strains of P. linteus(MPNU 7001 and MPNU 7002), and two isolated strains of P. baumii(MPNU 7004 and MPNU 7005) were shown to have about 520 bp (PL1F-PL2R), 700 bp (TTS5F-PL2R) and 600 bp (PB1F-ITS4R) -sized PCR single bands respectively. This molecular genetic technique provided a useful method for rapid detection and identification of P. linteus and P. baumii.

PCR-Based Sensitive Detection of Wood-Decaying Fungus Phellinus linteus by Specific Primer from rDNA ITS Regions

  • Based on the rDNA ITS sequences data, specific primer set for PCR detection of wood-decaying fungus Phellinus linteus was designed. The length of PCR products using designed primer set(SHF and SHR) was about 540 bp. Among 11 species, 17 isolates of Phellinus spp. including Phellinus linteus, P. pomaceus, P. spiculosus, P. baumi, P. pini, P. igniarius, P. gilvus, P. biscuspidatus, P. weirii, P. johnsonianus, P. robutus, and P. igniarius, seven isolates of Phellinus linteus showed about 540 bp-sized single band. This molecular technique could offer a useful tool for detecting and identifying Phellinus linteus.

Antioxidative Effects of Phellinus linteus Extract (상황버섯 추출물의 항산화 효과)

  • Reactive oxygen species(ROS) or free radical-mediated oxidative stress plays an important role in the pathophysiologic process of disease state. This study investigated antioxidative effects of Phellinus linteus extract on the generation of 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, superoxide anion radical(${O_2}^-$), hydroxyl radical(${\cdot}OH$), nitric oxide(NO), and peroxynitrite($ONOO^-$) radical and free radical-mediated protein oxidation under in vitro assay systems. This results showed that Phellinus linteus extract effectively inhibited the generation of free radicals in the all assay system with dose-dependent manner and also significantly reduced the protein oxidative level. Thus, the present study indicates that Phellinus linteus extract possesses a potent antioxidant activity and plays a beneficial role against free radical-induced oxidative injury.

  • Anti-oxidative effect of Phellinus linteus (P. linteus) and red ginseng extracts on DNA damage induced by reactive oxygen species (ROS) were investigated in this study. P. linteus (PLE) and red ginseng extracts (RGE) inhibited the breaking of E. coli ColE1 plasmid DNA strands as well as nuclear DNA of rat hepatocytes damaged by oxidative stress. In addition, a reaction mixture of PLE and RGE showed synergistic inhibitory effect against DNA damage. These results suggest that PLE and RGE have a cellular defensive effect against DNA damage induced by ROS.

Effect of the Evaporating Extract from Liquor of Fermentation Using Mycelium of Phellinus linteus on the Expression of Inflammatory Proteins and the Generation of Reactive Oxygen Species (상황버섯 균사체를 이용한 발효주의 추출물이 염증관련 단백질의 발현과 세포내 활성산소 생성에 미치는 영향)

  • Phellinus linteus is known as a medicinal mushroom, which has the pharmaceutical activity on tumors and inflammatory diseases in traditional Oriental medicine. However, despite extensive pharmacological studies on P. linteus, the liquor of fermentation using mycelium of P. linteus(LFMP) has not been investigated. In the present study, it was examined the effect of the evaporating extract from LFMP(E-LFMP) on the expression of inflammatory proteins and the generation of reactive oxygen species in human hepatocarcinoma HepG2 cells. E-LFMP inhibited acetaldehyde-induced morphological change in HepG2 cells. Also, E-LFMP inhibits expression of inflammatory proteins including cyclooxygenase(COX)-1 and COX-2 through suppressing nuclear translocation of nuclear factor κB(NF-κB) and degradation of inhibitory κBα(IκBα). In addition, E-LFMP inhibits generation of reactive oxygen species(ROS) by hydrogen peroxide(H₂O₂) in HepG2 cells. These results suggest that LFMP has the pharmaceutical, especially anti-inflammatory, activity similar to P. linteus mushroom.

Notes on the Korean Rotten Wood Fungi(I) (한국산부후균의 기재(I))

  • Many rotten wood fungi were collected at Mt.Moak Provincial Park(Chonlabuk-Do), Mt.Manduck(Chonlabuk-Do) and Byunsan Pennisula National Park from April, 1995 to October, 1995. These higher fungi were identified. According to the results, genera of Hypoderma, Piloderma and Cylidrobasidium are newly to korea. Following species are newly to Korea : Polyporus tuberaster, Phellinus ferruginosus, Corticium bomycinum, Phlebia rufa, Hyphoderma puberum, Piloderma byssinum, Cylinddrobasidium evolens and Peniophora pini.
