Antagonists of Both D1 and D2 Mammalian Dopamine Receptors Block the Effects of Dopamine on Helix aspersa Neurons

  • Dopamine mediates inhibitory responses in Helix aspersa neurons from the right parietal lobe ("F-lobe") of the circumoesophageal ganglia. The effects appeared as a dose-dependent hyperpolarization of the plasma membrane and a decrease in the occurrence of spontaneous action potentials. The average hyperpolarization with 5 ${\mu}m$ dopamine was -12 mV (${\pm}1.5$mV, S.D., n=12). Dopamine also modulated the currents 'responsible for shaping the action potentials in these neurons. When dopamine was added and action potentials were triggered by an injection of current, the initial depolarization was slowed, the amplitude and the duration of action potentials were decreased, and the after-hyperpolarization was more pronounced. The amplitude and the duration of action potential were reduced about 15 mV and about 13% by 5 ${\mu}m$ dopamine, respectively. The effects of dopamine on the resting membrane potentials and the action potentials of Helix neurons were dose-dependent in the concentration range 0.1 ${\mu}m$ to 50 ${\mu}m$. In order to show 1) that the effects of dopamine were mediated by dopamine receptors rather than by direct action on ionic channels and 2) which type of dopamine receptor might be responsible for the various effects, we assayed the ability of mammalian dopamine receptor antagonists, SCH-23390 (antagonist of D1 receptor) and spiperone (antagonist of D2 receptor), to block the dopamine-dependent changes. The D1 and D2 antagonists partially inhibited the dopamine-dependent hyperpolarization and the decrease in action potential amplitude. They both completely blocked the decrease in action potential duration and the increase in action potential after-hyperpolarization. The dopamine-induced slowdown of the depolarization in the initial phase of the action potentials was less effected by SCH-23390 and spiperone. From the results we suggest 1) that Helix F-lobe neurons may have a single type of dopamine receptor that binds both SCH-23390 and spiperone and 2) that the dopamine receptor of Helix F-lobe neurons may be homologous with and primitive to the family of mammalian dopamine receptors.

CORRELATIONS BETWEEN HIPPOCAMPAL THETA RHYTHM AND INTRACELLULAR CHARACTERISTICS OF PYRAMIDAL NEURONS (해마 theta 리듬과 pyramidal neuron의 세포내 특성과의 상관관계)

  • Electrophysiological phenomena of pyramidal cells in the CA1 area of the dorsal hippocampus were recorded from and filled with neurobiotin in anesthetized rats. The electropharmacological properties of membrane as well as the cellular-synaptic generation of rhythmic slow activity (theta) were examined. The intracellular response characteristics of these pyramidal cells were distinctly different from responses of interneurons. Pyramidal cells had a high resting membrane potential, a low input resistance, and a large amplitude action potential. A afterhyperpolarization was followed a single action potential. Most of pyramidal cells did not display a spontaneous firing. Pyramidal cells displayed weak inward rectification and anodal break excitation. The slope of the frequency-current relation was 53.4 Hz/nA for the first interspike interval and 15.9 Hz/nA for the last intervals, suggesting the presence of spike frequency adaptation. Neurobiotin-filled neurons showed pyramidal morphology. Cells were generally bipolar dendritc processes ramifying in stratum lacunosum-moleculare, radiatum, and oriens. Commissural stimulation discharged pyramidal cells, followed by excitatory and inhibitory postsynaptic potentials (EPSPs and IPSPs). The frequency of theta-related membrane potential oscillation was voltage-independent in pyramidal neurons. At strong depolarization levels (less than 30 mV) pyramidal cells emitted sodium spike oscillation, phase-locked to theta. The observations provide direct evidence that theta-related rhythmic hyperpolarization of principal cells is brought by the rhythmically discharging interneurons. Furthermore, the findings in which interneurons were also paced by rhythmic inhibitory postsynaptic potentials during theta suggest that they were periodically hyperpolarized by their GABAergic septal afferents.

Effects of Thyroid Hormone in vitro on the Electrical Activity of the Rabbit Heart Cell (토끼 심장세포의 전기적 활동에 대한 갑상선 호르몬의 효과)

  • To verify the direct effects of the thyroid hormone ($T_3$) on the rabbit heart, $T_3$-Tyrode solution in vitro was perfused on the normal atrial muscles and enzymatically isolated ventricular myocytes of the rabbit. All the experimental procedures were conducted at $35^{\circ}C$ and the same procedures were repeated after Ca. 120 minutes from the beginning of $T_3$-Tyrode perfusion. Compared to the state between the normal Tyrode solution and $T_3$-Tyrode solution, results were observed on the same cells by electrophysiological methods (conventional intracellular recording and whole cell patch clamping) as soon as possible. The results obtained were as follows : 1. Action potential duration (APD) on the left atrial muscle was reduced under the perfusion of $T_3$-Tyrode. 2. Absolute refractory Period was shortened by $T_3$-Tryrode perfusion. (117 msec./114 msec., 90 msec./78 msec.) 3. Maximal Ca currents ($i_{Ca}$) were decreased in single: ventricular myocytes under the $T_3$-Tyrode (2.98 nA) than under the normal Tyrode (6.65 nA) 4. On I-V relation, reversal potential was shifted to lower membrane potential and membrane potential showing maximal $i_{Ca}$was lowered from +10mV to -20mV by $T_3$ effect. 5. Above results were likely to explain that tachycardia in the hyperthyroid state was caused in part by the reduced repolarization phase and the reduced refractory period due to the decrease of the Ca current.

Characteristics of Formation of Chlorination Disinfection By-Products in Extracellular Organic Matter of Various Algal Species (다양한 조류종들의 세포외 유기물질에서의 염소 소독부산물 생성 특성)

  • Formation of disinfection by-products (DBPs) including trihalomethans (THMs) and haloacetic acids (HAAs) from chlorination of six different species (Chlorella vulgaris, Scenedesmus sp., Anabaena cylindrical, Microcystis aeruginosa, Asterionella formosa and Aulacoseira sp.) of algal extracellular organic matter (EOM). The EOM characteristics evaluation of six algal species reaching at the stationary phase in the growth curve showed most of its SUVA254 showed below 1 and this means hydrophilic organic matter is much higher than hydrophobic organic matter. Chloroform formation potential (CFFP), dichloroacetic acid formation potential (DCAAFP) and trichloroacetic acid formation potential (TCAAFP) were mainly composed of THMFP and HAAFP in the EOM of various algal species. In the case of THMFP/DOC and HAAFP/DOC values, EOM of blue-green algae has appeared highest and EOM of green algae and diatom in order. THMFP/DOC was higher than HAAFP/DOC in EOM of blue-green algae. In comparison of formation potential by unit DOC composed of HAAFP in algal species EOM, DCAAFP/DOC was 1.5 times to 7.5 time higher than TCAAFP/DOC in the EOM of blue-green algae, while DCAAFP/DOC was found to be relatively high compared to TCAAFP/DOC in the EOM of green algae and diatom.

Electrochemical Behavior of Li-B Alloy Anode - Liquid Cadmium Cathode (LCC) System for Electrodeposition of Nd in LiCl-KCl

  • The performance of Li-B alloy as anode for molten salt electrolysis was firstly investigated. The crystalline phase of the prepared Li-B alloy was identified as $Li_7B_6$. The potential profile of Li-B alloy anode was monitored during the electrodeposition of $Nd^{3+}$ onto an LCC (liquid cadmium cathode) in molten LiCl-KCl salt at $500^{\circ}C$. The potential of Li-B alloy was increased from -2.0 V to -1.4 V vs. Ag/AgCl by increasing the applied current from 10 to $50mA{\cdot}cm^{-2}$. It was found that not only the anodic dissolution of Li to $Li^+$ but also the dissolution of the atomic lithium ($Li^0$) into the LiCl-KCl eutectic salt was observed, following the concomitant reduction of $Nd^{3+}$ by the $Li^0$ in Li-B alloy. It was expected that the direct reduction could be restrained by maintaining the anode potential higher that the deposition potential of neodymium.

  • Research and development for the technology, that is about maintenance and accidents prevention of underground power distribution line, are demanding. The precise detection of leakage point of underground power line is very important, because it is difficult to detect the exact location of a fault in underground power line and to repair faults. When earth electric potential is measured to detect underground electric leakage point after transmitting AC electric pulse wave to underground power line, it must be measured in a specific half period of AC pulse wave because the distribution of the electric earth potential varies with the polarity of the transmitted wave. In this paper we proposed the measurement of half period modulated earth potential as a method to detect a underground leakage point. And We compared the proposed method with other methods. Through experiments we verified that the proposed method can be implemented and operated properly.

Analyzing the Potential of Offset Credits in the Korean Emission Trading Scheme Focusing on Clean Development Mechanism Projects (CDM사업을 대상으로 한 국내 온실가스 상쇄배출권의 잠재량 산정 및 정책 제언)

  • The purpose of this study is to analyze the potential quantity of Korean Offset Credits (KOC) resulting from Certified Emission Reductions (CER) in 98 domestic Clean Development Mechanism (CDM) projects that were registered with the United Nations Framework Convention on Climate Change (UNFCCC) as of the end of 2016. Our results show that the total amount of potential KOC is 62,774 kt CO2eq. The potential KOC is only 23.4% of the total CER Issuance. During the first phase, this will be 3.2% of the allocated volume. This is because many projects are related to Renewable Portfolio Standard (RPS), HFC-23, and adipic acid N2O. There is a strong bias in some sectors and projects which could act as market distortion factors. Therefore, it is necessary to expand the target CDM project and activate non CDM offset projects. RPS projects bring fundamental changes to the energy sector, and it is worth reconsidering their acceptability. A wide variety of policy incentives are needed to address strong biases toward certain sectors and projects. The offset scheme has the advantage of allowing entities to reduce their GHG emissions cost effectively through a market mechanism as well as enabling more entities to participate in GHG reduction efforts both directly and indirectly. In contrast, having an inadequate offset scheme range and size might decrease the effort on GHG reduction or concentrate available resources on specific projects. As such, it is of paramount importance to design and operate the offset scheme in such a way that it reflects the situation of the country.

  • $G{\alpha}_q$-coupled receptor stimulation was implied in the activation process of transient receptor potential canonical (TRPC)1/4 and TRPC1/5 heterotetrameric channels. The inactivation occurs due to phosphatidylinositol 4,5-biphosphate ($PI(4,5)P_2$) depletion. When $PI(4,5)P_2$ depletion was induced by muscarinic stimulation or inositol polyphosphate 5-phosphatase (Inp54p), however, the inactivation by muscarinic stimulation was greater compared to that by Inp54p. The aim of this study was to investigate the complete inactivation mechanism of the heteromeric channels upon $G{\alpha}_q$-phospholipase $C{\beta}$ ($G{\alpha}_q-PLC{\beta}$) activation. We evaluated the activity of heteromeric channels with electrophysiological recording in HEK293 cells expressing TRPC channels. TRPC1/4 and TRPC1/5 heteromers undergo further inhibition in $PLC{\beta}$ activation and calcium/protein kinase C (PKC) signaling. Nevertheless, the key factors differ. For TRPC1/4, the inactivation process was facilitated by $Ca^{2+}$ release from the endoplasmic reticulum, and for TRPC1/5, activation of PKC was concerned mostly. We conclude that the subsequent increase in cytoplasmic $Ca^{2+}$ due to $Ca^{2+}$ release from the endoplasmic reticulum and activation of PKC resulted in a second phase of channel inhibition following $PI(4,5)P_2$ depletion.

Effect of Solvents on Phase Behavior and Flux Removal Efficiency in Alkyl Ethoxylates Nonionic Surfactant Based Cleaners (Alkyl Ethoxylates계 비이온 계면활성제를 주체로 한 세정제에서 용제에 따른 상거동과 플럭스 제거 효능)

  • In this work, the effect of additives such as solvent, sodium dodecyl sulfate and NaCl on microemulsion phase behavior and flux removal efficiency in systems containing commercial alkyl ethoxylates nonionic surfactant was investigated. The addition of a n-hydrocarbon as a solvent produced on O/W (Oil/Water) microemulsion phase over a wider range of temperature and cosurfactant to surfactant ratios. Especially, the addition of n-hexadecane to the surfactant system, which was the most hydrophobic solvent among the solvents used in this study, produced a microemulsion phase over a wide range of temperatures and promoted formation of a microemulsion phase at lower temperatures. The candidate for cleaner samples, prepared from phase behavior experiments, showed excellent removal efficiency for abietic acid at $40^{\circ}C$. These data suggested the potential applicability of hydrocarbons to actual cleaner formulations.

Analysis on the Electrode Kinetic Parameters at the Pd/LiOH Electrolyte Interface using the Phase-shift Method (위상이동 방법에 의한 Pd/LiOH 전해질 계면의 전극속도론적 패러미터 해설)

  • The electrode kinetic parameters at the Pd/0.5 M LiOH electrolyte interface have been qualitatively studied using the phase-shift method. The phase shift $(\phi)$ depends on both the cathode potential (E>0) and frequency (f), and $\theta$ is inversely proportional to the fractional surface coverage $\theta$. At an intermediate frequency (10 Hz), the phase-shift profile $(\phi\;vs.\;E)$ can be related to the fractional surface coverage $(\theta\;vs.\;E)$. The phase-shift method can be used to estimate or plot the Frumkin adsorption isotherm. The rate (r) of change of the free energy of adsorption with $({\theta})$ is 22.3 kJ/mol. The equilibrium constant (K) for adsorption and the standard free energy $({\Delta}G_{\theta})$ of the adsorbed hydrogen atom $(H_{ads})$ are $3.7\times10^{-3}{\Delta}G_{\theta}>-8.4kJ/mol$, respectively. For 1$0.38>\theta>0$, the energy liberation or the exothermic reaction for hydrogen adsorption at the Pd cathode can be occurred. The electrode kinetic parameters $(r,\;K,\;{\Delta}G_{\theta}$ depend on the fractional surface coverage $({\theta})$ or the phase shift $(\phi)$.
