유산간균 Phage에 대한 Sodium Pyrophosphate의 용균억제작용 (Inhibition of Phage Lytic Activity in Lactobacilli Cells by Sodium Pyrophosphate)

  • CaC $l_2$를 제거한 MRT 액체배지에 phage가 MOI =$10^{0}$ 이상으로 감염된 경우에는 0. 15% sodium pyrophosphate 농도에서 유산균이 용균되었으나. 0.175% 이상의 농도에서는 유산균이 정상적으로 증식함을 알 수 있었다. 그러므로 phage 감염 후 유산균의 증식은 sodium pyrophosphate농도와 phage 감염수에 의해 결정된다고 사료된다. 이 때에 sodium pyrophosphate의 phage 증식억제 농도에서 phage불활성화는 나타나지 않았다. 0.15% sodium pyrophosphate를 첨가한 배지에서는 MOI=$10^{-1}$ 이하로 phage가 감염되면 유산균이 증식하여 배양 24시간 후 약 $10^{8-9}$/$m\ell$의 균수를 나타냈으며, 무첨가 배지에서는 MOI =$10^{-3}$ 이상일 때 유산균이 거의 사멸하였고 $10^{-4}$ 일 때는 약 1$\times$$10^{5}$ /$m\ell$로 용균됨을 나타내었다.다.

Phage typing of Staphylococcus aureus Isolates from Poultry Meat in Spain

  • Phage typing is currently used for typing of Staphylococcus aureus strains beyond the species level in epidemiological studies. A total of 168 Staphylococcus aureus isolates from chicken meat and chicken by-products were phage-typed using the international bacteriophage set for typing Staphylococcus aureus of human origin. One hundred and forty-eight (88.79%) strains were phage-typeable (at least one phage produced 20 or more plaques of lysis). Lysis by phages of group Ⅲ was the mast frequent with 99 (58.93%) sensitive strains. This fact coincides with results of other authors. Twenty-nine different phage patterns were observed and three (95, 75/84 and 6/1030/W57) were most common. One hundred and thirty-two (89.19% of typeable strains) skewed these or indistinguishable (only one phage reaction difference) patterns. Twenty-six out of seventy chicken samples (37.14%) harboured more than one phage type of Staphylococcus aureus. This fact emphasizes the convenience of subtyping several Staphylococcus aureus isolates from the same sample in epidemiological studies. 80% of sausages and hamburgers contained the same Staphylococcus aureus phage types, which wore not found in any of the other food types. This fact suggests a cross contamination during the processing of these foods. Phages 6, 75, 84, 1030 and W57 skewed the greatest activity. None of the Staphylococcus aureus strains were sensitive to phages 47, 81 and 94.

  • The RK-temperate phage which infected with Bacillus cereus was isolated and the characters were investigated. The induction of RK-temperate phage from host bacterium attained by ultraviolet light irradiation (15W, 30cm, 30-120sec) and mitomycin C treatment (0.2-2 ug/ml). The host range of RK-temperate phage was not revealed with lysogenic and related strains of B. cereus. But B. cereus(PS) 352 which obtained by N-nitrosoguanidine treatment(1,000.$\mu$g/ml) to phage infected with host bacteria was sensitive bacteria of RK-temperate phage. RK-temperate phage was stabilized at the condition of nutrient broth (pH 7-8), Tris-buffer (pH 7-8) and ammonium buffer (pH 8-9) and Sorensen's phosphate buffer (pH 6-7), but unstabilized at other salt solutions and pH range. Also, thermostability was to 45.deg.C but unstabilized at above 50.deg.C. At RK-temperate phage, the measurment values of head, neck, mid tail and end tail were 59nm, 9*16nm, 10*189nm, and 10*14nm respectively. The morphology of head was regular polyhedron, and the end tail was coneate form. On the one hand, the number of capsid protein layer of tail were consist of 4, 35, and 1 at neck, mid tail, and end tail, respectively. RK-temperate phage was identified with DNA phage and G+C contents were 38.63. The latent time of RK-temperate phage was 30 minutes and the burst size was 70-80. And the host bacteria was lysed in case of multi-infection, above moi 1.

  • Lysogenic conversion of Staphylococcus aureus to loss of ${\beta}-hemolysin$ production by serological group F phages is always associated with gain in staphylokinase production. In this study, the new phages belonging to serotype F were detected during the course of isolation of phages from Staph aureus of bovine origin and some characteristics of the new phages isolated were investigated. The new phages, ${\phi}470$ and ${\phi}499$, isolated from Staph aureus producing ${\beta}-hemolysin$ and staphylokinase(${\beta}^+\;K^+$) were found to convert ${\beta}^+\;K^+$ strain to ${\beta}^+K\;^+$, Staph aureus strains lysogenized by this serotype F single-converting phage ${\phi}470$ or ${\phi}499$ could be again lysogenized with serotype F double-converting phage ${\phi}506$. The frequency of lysogenization of indicator strains by serotype F single-converting phage was 100%, whereas the frequency for serotype F double-converting phage ${\phi}506$ varied from 4.2% to 97.6% according to the indicator strains. The indicator strain lysogenized with phage ${\phi}470$ was resistant to phage ${\phi}499$, and vice versa, but not to phage ${\phi}506$. Therefore, phage ${\phi}470$ and ${\phi}499$ were shown to be identical by immunity test.

Phage Assembly Using APTES-Conjugation of Major Coat p8 Protein for Possible Scaffolds

  • Filamentous phages have been in the limelight as a new type of nanomaterial. In this study, genetically and chemically modified fd phage was used to generate a biomimetic phage self-assembly product. Positively charged fd phage (p8-SSG) was engineered by conjugating 3-aminopropyltriethoxysilane (APTES) to hydroxyl groups of two serine amino acid residues introduced at the N-terminus of major coat protein, p8. In particular, formation of a phage network was controlled by changing mixed ratios between wild type fd phage and APTES conjugated fd-SSG phage. Assembled phages showed unique bundle and network like structures. The bacteriophage based self-assembly approach illustrated in this study might contribute to the design of three dimensional microporous structures. In this work, we demonstrated that the positively charged APTES conjugated fd-SSG phages can assemble into microstructures when they are exposed to negatively charged wild-type fd phages through electrostatic interaction. In summary, since we can control the phage self-assembly process in order to obtain bundle or network like structures and since they can be functionalized by means of chemical or genetic modifications, bacteriophages are good candidates for use as bio-compatible scaffolds. Such new type of phage-based artificial 3D architectures can be applied in tuning of cellular structures and functions for tissue engineering studies.

Characterization of Phage-Resistant Strains Derived from Pseudomonas tolaasii 6264, which Causes Brown Blotch Disease

  • Pseudomonas tolaasii 6264 is a representative strain that causes bacterial blotch disease on the cultivated oyster mushroom, Pleurotus ostreatus. Bacteriophages are able to sterilize the pathogenic P. tolaasii strains, and therefore, they can be applied in creating disease-free mushroom cultivation farms, through a method known as "phage therapy". For successful phage therapy, the characterization of phage-resistant strains is necessary, since they are frequently induced from the original pathogenic bacteria in the presence of phages. When 10 different phages were incubated with P. tolaasii 6264, their corresponding phage-resistant strains were obtained. In this study, changes in pathogenic, genetic, and biochemical characteristics as well as the acquired phage resistance of these strains were investigated. In the phylogenetic analyses, all phage-resistant strains were identical to the original parent strain based on the sequence comparison of 16S rRNA genes. When various phage-resistant strains were examined by three different methods, pitting test, white line test, and hemolytic activity, they were divided into three groups: strains showing all positive results in three tests, two positive in the first two tests, and all negative. Nevertheless, all phage-resistant strains showed that their pathogenic activities were reduced or completely lost.

Use of Antibody Displayed Phage for the Detection of Dextran Using a Dipstick Assay and Transmission Electron Micrograph

  • An antibody displayed phage collection (SBAE-2R), screened from a human synthetic phage antibody library (Fab 21ox), was used for the determination of dextran. The dextran-binding affinity was determined by serologically specific transmission electron microscopy (TEM) and a paper dipstick assay. The phage collection was distributed over the dextrancoated grids with 39$\pm$25 phages/$\mu$m$^2$ on the grids. Phages were not seen on dextran-coated grids exposed to the Fab 2lox phage library. The phage collection (SBAE-2R) produced 54$\pm$3 color normalized intensity (N.I.) from 125 ppm to 1,000 ppm of dextran and 5$\pm$1 (N.I.) for 63 ppm of dextran in a paper dipstick assay. This research extends the analytical options for dextran analysis by antibody displayed phage with a minimum of equipment usage.

Genome Analysis of Phage SMSAP5 as Candidate of Biocontrol for Staphylococcus aureus

  • In this study, we reported the morphogenetic analysis and genome sequence by genomic analysis of the newly isolated staphylococcal phage SMSAP5 from soil of slaughterhouses for cattle. Based on transmission electron microscopy evident morphology, phage SMSAP5 belonged to the Siphoviridae family. Phage SMSAP5 had a double-stranded DNA genome with a length of 45,552 bp and 33 % G+C content. Bioinformatics analysis of the phage genome revealed 43 open reading frames. A blastn search revealed that its nucleotide sequence shared a high degree of similarity with that of the Staphylococcus phage tp310-2. In conclusion, this study is the first report to show the morphological features and the complete genome sequence of the phage SMSAP5 from soil of slaughterhouses for cattle.

공장배수계에 존재하는 유산간균 Virulent Phage에 관한 연구 (Studies on Lactobacillus Virulent Phage in Plantdrainage)

  • 한국야쿠르트 제조공장의 배수중에 존재하는 L. casei 숙주로 하는 Lactobacillus phage J$_1$계의 분포를 조사한 결과는 다음과 같다. 1. 공장의 배수중의 J$_1$계 phage에는 Type I, II, III, IV가 존재하고 있었으며 분포순위로 보면 Type IV>Type II>Type I>Type III 이었 다. 2. 공장배수중의 phage는 역성비누를 격포하여 완전히 제거되었다. 3. 각종 소독제의 Phage에 대한 살균핵과는 염소수, 역성비누, 크레졸이 가장 강력한 효력을 나타내었고, isopropyl alcohol, ethyl alcohol등은 살균력이 미약하였다.

Ex12 helper phage improves the quality of a phage-displayed antibody library by ameliorating the adverse effect of clonal variations

  • The quality of a phage-displayed antibody library deteriorates with clonal variations, which are caused by differentially expressed Escherichia coli antibody genes. Using the human Fab SP114 against the pyruvate dehydrogenase complex-E2 (PDCE2), we created four E. coli TOP10F' clones with a pCMTG phagemid encoding Fab-pIII (pCMTG-Fab), Fd ($V_H+C_{H1}$)-pIII (pCMTG-Fd), or light chain (L) (pCMTG-L), or the vector only (pCMTG-${\Delta}Fab$) to investigate the effect of clonal variations in a defined manner. Compared to the others, the E. coli clone with pCMTG-Fab was growth retarded in liquid culture, but efficiently produced phage progenies by Ex12 helper phage superinfection. Our results suggest that an antibody library must be cultured for a short duration before helper phage superinfection, and that the Ex12 helper phage helped to alleviate the detrimental effect of clonal variation, at least in part, by preferentially increasing functional phage antibodies during phage amplification.
