Muscle Type Change and Vascularization Effect of Semen Persicae in MCAo rats (도인(桃仁)이 중대뇌동맥 폐색 유발 흰쥐의 근육형태 변화와 혈관신생에 미치는 영향)

  • Objectives : This study was performed to investigate the effects of Semen Persicae(SP) on muscle type change and vascularization effect by measuring expression of Myosin heavy chain (MHC) and Vascular endothelial growth factor (VEGF) protein in the Middle cerebral artery occlusion(MCAo) rats. Methods : The middle cerebral artery was occluded, SP extraction was administrated for 4 days. The effects of SP on muscle type change and vascularization were measured. Results : 1. VEGF protein expressions in the Semen Persicae oral administration group of MCAo group were increased compared to the control group. 2. There were no significant difference between the Semen Persicae oral administration of MCAo group and the control group in MHC isoform (Type I, Type IIa) expression change. Conclusions : The present study demonstrates the effect of Semen Persicae in the vascularizing after ischemia, but has no significant effect in musle type change and the improvement of musle atrophy.

  • In order to study effects of ${\ulcorner}Persicae\;Semen{\lrcorner}$ on the hyperlipidemia rats induced by Triton WR, we have made hyperlipidemia rats by administering Triton WR-1339 for 3 days, then have administered. The results were summerized as follows: 1. The total cholesterol, triglyceride and phospholipid by hypercholesteremic rats injected by Triton WR-1339 of the sample group were decreased as compared with those of the control group, 2. The HDL-cholesterol by hypercholesteremic rats injected by Trion WR -1339 were increased as compeared those of control group. According to the above results, Persicae Semen showed inhibition on increase of serum total cholesterol, triglyceride and phosholipid contents as well as inhibition on decrease of HDL-cholesterol contents in hyperlipidemia.

Hepatoprotective and antifibrotic effects of extracts from Persicae Semen (도인 추출물의 간보호 및 항섬유화 효과)

  • This study was carried out to investigate the liver protective effects of extracts from Persicae Semen (WT-003, WT-005, WT-006). The acute hepatic injury was induced by intraperitoneal injection of alpha-naphtylisothiocyanate (75 mg/kg, p.o.) and treated with WT-006 (100 mg/kg/day, 200 mg/kg/day, 400 mg/kg/day). The experimental hepatic fibrosis was induced by bile duct ligation/scission(BDL/S), duration of 4 weeks and treated with WT-003, WT-005 or WT-006 (200 mg/kg/days for 4 week). In acute liver injury, WT-006 (200 or 400 mg/kg) lowered serum alanine transferase(ALT) and aspartate transferase(AST) significantly. In fibrotic rats, WT-006 treatment inhibited the hydroxyproline deposition in liver and lowered serum AST, ALT and ALP, significantly. These results suggest WT-006 extract, which does not contain amygdalin, from Persicae Semen have liver protective and antifibrotic effects in rats.

  • Tonin(Persicae Semen) is the herb medicine that contains amygdalin as a major ingredient. Amygdalin in water is decomposed into benzaldehyde, HCN, and glucose by emulsin, a hydrolysis enzyme in tonin. A useful and practical method for the optimum extraction condition of amygdalin without enzymatic hydrolysis is required. The extraction yield of amygdalin of natural formula to nin was 0.1 % from crude powders, 1.4% from small pieces, 3.5% from half pieces and 2.4% from whole pieces. The extraction yield of amygdalin of outer shell-eliminated to nin was 0.3% from crude powders, 1.4% from small pieces, and 3.5% from half pieces and whole pieces respectively. The extraction yield of amygdalin was most high when using the size larger than half.

  • Objectives : This study were carried out to investigate the anti-oxidative activities of $\gamma$-SPE(gamma irradiated shell of Persicae semen extract with ethanol). Methods : Anti-oxidative was measured by DPPH free radical scavenging activities, nitric oxide radical scavenging activities, SOD-like activities, xanthine oxidase radical scavenging activities, nitrite scavenging activities and tyrosinase inhibition activities. Results : Effects of free radial scavenging was increased by irradiation. Tyrosinase inhibition activities was decreased by $\gamma$-SPE. Conclusions : Results suggested that $\gamma$-SPE have a potential as a new anti-oxidative material.

Cosmeceutical activities and Anti-inflammatory effects of Shell from Persicae semen (도인(桃仁) 껍질의 화장품약리활성 및 항염 효과)

  • Objectives : In this paper, we tested the applicability of shell from Persicae semen in cosmetics through cosmeceutical activities including anti-oxidant, tyrosinase inhibition and anti-inflammatory effects. Methods : Persicae semen, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect have been investigated in the cosmeceutical activity measurement of function experiment. Results : In the electron donating ability test, 1,000ppm of EPS (ethanol extract of shell from Persicae semen) showed an effect of 87%. SOD-like activities showed 93% at the 10,000ppm of WPS (water extract of shell from Persicae semen). In the xanthine oxidase inhibition test, 1,000ppm of BHA showed an effect of 27%, while EPS showed an effect of 62%. We were able to get an effect of 95% from EPS at 10,000 ppm in the tyrosinase inhibition test. In the anti-inflammatory test, the EPS inhibited the generation of nitric oxide. In the case of the EPS, there were no signs of cytotoxicity against raw 264.7 and anti-inflammatory effects could be identified when the manifestation of iNOS was decreased. Conclusion : Therefore, the EPS has potential as an effective raw materials for cosmetic.

Study on Quality Changes Caused by Rancidity and Methods to Reduce Rancidity for Domestically Distributed Herbal Medicines (유통 한약재의 산패에 따른 품질변화 및 산패 저감화 연구)

  • Rancidity changes were examined for 6 herbal medicines, namely Persicae Semen, Armeniacae Semen, Lini Semen, Trichosanthis Semen, Arecae Semen, Myristicae Semen known to have relatively high fat content. In order to reduce rancidity of herbal medicines, samples were stored at 3 different conditions of room, refrigerating and freezing temperatures, and the rancidity was measured for 10 months with every 2 month interval. Fat content was extracted by using ethyl ether, and acid values and peroxide values, which are generally accepted indicators of fat rancidity, were measured. When storing Persicae Semen, Lini Semen and Arecae Semen at room temperature, the acid values increased as the storage period increased, and it was higher than when stored in refrigeration or freezing. The measurement of peroxide value showed more significantly higher initial degree of rancidity when Persicae Semen, Trichosanthis Semen, Arecae Semen and Myristicae Semen were stored at room temperature. It was observed that storing herbal medicines in refrigeration or freezing inhibited their rancidity compared to storing them at room temperature. To investigate the quality changes according to rancidity, the analysis of aflatoxins and indicator components showed that aflatoxins B1 and B2 were detected in Armeniacae Semen, Arecae Semen and Myristicae Semen, and the amount of amygdalin was well maintained within the specification standard.

Neuroprotective Effect of Semen Persicae in Middle Cerebral Artery Occlusion Rats (도인(桃仁)의 중대뇌동맥 폐색 유발 흰쥐에서 신경보호 효과)

  • Objectives: This study was performed to investigate the effects of Semen Persicae (SP) on infarct volume, COX-2 protein expression in the middle cerebral artery occlusion (MCAo) rats. Methods: Twenty-eight rats were randomly assigned to four groups (MCAo experimental group, MCAo control group, sham experimental group, control group). The middle cerebral artery (MCA) was occluded in the MCAo group by proximal focal cerebral ischemia rat model, while the MCA was not occluded in the sham group. SP extraction was administrated for 4 days to each experimental group. Neuroprotective effects were investigated by measurement of brain damage using 2, 3, 5-triphenyltetrazolium chloride staining and analysis of COX-2 protein expression by western blotting. Results: The occurrence of infarct volume in the SP oral administration group decreased compared to the control group. COX-2 protein expression in the SP oral administration group decreased compared to the control group. Conclusions: The present study demonstrates the effect of SP in reducing infarct volume and decreasing COX-2 expression.

The HDL-cholesterol by hypercholesteremic rats injected by Trion WR -1339 of the herb-Acupuncture group were increased as compeared those of control group, especialy hypercholesteremic rats injected by Triton WR-1339 of the herb-Acupuncture+Acupuncture group were some more increased as compared with those of the control group. According to the above results, Persicae Semen showed inhibition on decrease of serum total cholesterol, triglyceride and phosholipid contents as well as inhibition on increase of HDL-cholesterol contents in hyperlipidemia.

In vitro Antiinflammatory Activity of Amygdalin in Murine Macrophage Raw 264.7 Cells (Amygdalin의 Murine Macrophage Raw 264.7 세포에서 in vitro 항염효과)

  • In the present study, anti-inflammatory activity of amygdalin isolated from persicae Semen have been evaluated on lipopolysaccharide (LPS)-induced release of nitric oxide (NO), prostaglandin $E_2\;(PGE_2)$ and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) by the macrophage RAW 264.7 cells. Amygdalin significantly inhibited generation of NO and $TNF-{\alpha}$ on LPS-stimulated RAW264.7 cells in a concentration-dependent manner. Consistent with these observations, the expression of inducible NO synthase (iNOS) enzyme was also inhibited by amygdalin in a concentration-dependent manner. However, amygdalin did not show any influence on the synthesis of $PGE_2$ and the expression of COX-2. Thus, this study suggests that amygdalin-mediated inhibition of iNOS expression, and $TNF-{\alpha}$ release may be one of the mechanisms responsible for the anti-inflammatory effects of Persicae Semen.
