Effects of Prepartum Dietary Carbohydrate Source on Metabolism and Performance of Primiparous Holstein Cows during the Periparturient Period

  • Forty-six Holstein heifers were used in a completely randomized design and assigned to 1 of 2 treatments to evaluate the effects of 2 diets varying in ruminal fermentable carbohydrate sources, namely ground corn (GC) and rolled wheat (RW), on metabolism and performance of primiparous cows in the periparturient period. The heifers were fed diets as a total mixed ration (TMR) with similar energy and crude protein content including i) 18.57% GC, or ii) 18.57% RW from -24.13${\pm}$7.73 d relative to expected calving until calving. After calving, all animals received the same lactation diet until 28 d. Animals were group fed from the beginning of the study to -7 d relative to expected calving, fed individually from d -7 to 7 days in milk (DIM), and again group fed to 28 DIM. The pre-partum diets affected (p<0.05) dry matter intake (DMI), energy intake, energy balance (EB) and urinary pH during the last week pre-partum. There was no effect of pre-partum carbohydrate source on overall plasma concentration of glucose, nonesterified fatty acid (NEFA), $\beta$-hydroxybutyrate (BHBA), albumin, triglyceride (TG), cholesterol, aspartate aminotransferase (AST), insulin, and cortisol during the periparturient period. Cows fed the RW diet during the pre-partum period had greater calcium for the first week (p<0.05) and during 28 d (p = 0.08) of lactation compared with heifers fed the GC diet. Primiparous cows fed the RW diet produced greater milk protein content and yield (p<0.05). Primiparous cows fed the RW diet had lower milk urea nitrogen (MUN) and somatic cell count (SCC) than cows fed the GC diet (p<0.05). The results of this study show that feeding pre-partum diets with a rapidly fermentable source of starch but low energy content can improve animal metabolism and performance and smooth the transition of primiparous Holstein cows from gestation to lactation.

  • 젖소는 분만전후기에 당 및 지방대사를 조절하여 태아와 유선에서 요구하는 영양소를 충족하고자 한다. 이러한 대사적 적응과정에서 중요한 역할을 하는 호르몬으로 insulin을 꼽을 수 있으며, 특히 지방조직에 작용하여 동물체내 필요한 에너지를 충족시키기 위해 지방산의 재배치를 촉진하고 분만전 말기에 insulin resistance를 야기시킨다. 분만전에 혈중 유리 지방산의 농도가 증가됨에 따라 insulin resistance가 발생하며 이것은 사료섭취량의 감소와 밀접한 관계가 있다. Insulin resistance을 완화시키는 물질로 $PPAR{\gamma}$ agoinst인 thiazolidinedione (TZD)는 분만전후기에 발생하는 대사성 질병의 발생을 억제하여 젖소의 건강 및 유생산 증가를 도모할 수 있다고 사료된다.

Hormonal Profiles during Periparturient Period in Single and Twin Fetus Bearing Goats

  • The effect of fetal number (single or twin) on plasma concentrations of progesterone, estradiol $17{\beta}$, cortisol, prolactin, growth hormone, triiodothyronine, thyroxine and insulin around parturition (periparturient period) were studied on ten $Alpine{\times}Beetle$ crossbred goats in their first to third lactation. The hormone profiles were studied on days -20, -15, -10, -5, -4, -3, -2, -1 prior to kidding and on day 0 and +1, +2, +3, +4, +5, +10, +15, +20 days postkidding. Plasma progesterone levels were significantly (p<0.01) higher in twin bearing goats comparison to single bearing goats during all the days of sampling. The decline in progesterone concentration from day 20 to day 1 before kidding was 56% in twin and 42% in single bearing goats. In single bearing goats plasma estradiol $17{\beta}$ was significantly (p<0.01) higher during prekidding days compared to twin bearing goats. The level of estradiol $17{\beta}$ was highest on the day of kidding in both the groups. The plasma prolactin level in twin bearing goats from day 10 to day 1 prepartum was higher as compared to single fetus bearing goats. However there was abrupt increase in prolactin level on the day of kidding in both the groups. The plasma growth hormone levels were significantly (p<0.01) higher in twin compared to single bearing goats. On the day of kidding growth hormone levels were significantly (p<0.01) higher in twin as compared to single bearing goats (1.40 vs. 0.95 ng/ml). In twin bearing goats plasma cortisol values from day 5 till the day of kidding remained elevated and the levels on the day of kidding was significantly highest in both the groups. The levels of triiodothyronine ($T_3$) were significantly higher (p<0.01) during all the periods of sampling in single compared to twin bearing goats. Plasma thyroxine ($T_4$) was significantly (p<0.01) lower in twin compared to single bearing goats. In single bearing goats plasma insulin levels were significantly (p<0.01) higher than twin bearing goats during prepartum period however during post partum period the levels in both the groups remained similar. It can be concluded that number of fetuses is having significant influence on the hormone profile during periparturient period.

Predicted functional analysis of rumen microbiota suggested the underlying mechanisms of the postpartum subacute ruminal acidosis in Holstein cows

  • Background: The relationships between the postpartum subacute ruminal acidosis (SARA) occurrence and predicted bacterial functions during the periparturient period are still not clear in Holstein cows. Objectives: The present study was performed to investigate the alterations of rumen fermentation, bacterial community structure, and predicted bacterial functional pathways in Holstein cows. Methods: Holstein cows were divided into the SARA (n = 6) or non-SARA (n = 4) groups, depending on whether they developed SARA during the first 2 weeks after parturition. Reticulo-ruminal pH was measured continuously during the study period. Reticulo-ruminal fluid samples were collected 3 weeks prepartum, and 2 and 6 weeks postpartum, and blood samples were collected 3 weeks before, 0, 2, 4 and 6 weeks postpartum. Results: The postpartum decline in 7-day mean reticulo-ruminal pH was more severe and longer-lasting in the SARA group compared with the non-SARA group. Changes in predicted functional pathways were identified in the SARA group. A significant upregulation of pathway "PWY-6383" associated with Mycobacteriaceae species was identified at 3 weeks after parturition in the SARA group. Significantly identified pathways involved in denitrification (DENITRIFICATION-PWY and PWY-7084), detoxification of reactive oxygen and nitrogen species (PWY1G-0), and starch degradation (PWY-622) in the SARA group were downregulated. Conclusions: The postpartum SARA occurrence is likely related to the predicted functions of rumen bacterial community rather than the alterations of rumen fermentation or fluid bacterial community structure. Therefore, our result suggests the underlying mechanisms, namely functional adaptation of bacterial community, causing postpartum SARA in Holstein cows during the periparturient period.

Modulation of the Somatotropic Axis in Periparturient Dairy Cows

  • This review focuses on modulation of growth hormone (GH) and its downstream actions on periparturient dairy cows undergoing physiological and metabolic adaptations. During the periparturient period, cows experience a negative energy balance implicating that the feed intake does not meet the total energy demand for the onset of lactation. To regulate this metabolic condition, key hormones of somatotropic axis such as GH, IGF-I and insulin must coordinate adaptations required for the preservation of metabolic homeostasis. The hepatic GHR1A transcript and GHR protein are reduced at parturition, but recovers on postpartum. However, plasma IGF-I concentration remains low even though hepatic abundance of the GHR and IGF-I mRNA return to pre-calving value. This might be caused by alternation in IGFBPs and ALS genes, which consequently affect the plasma IGF-I stability. Plasma insulin level declines in a parallel manner with the decrease in plasma IGF-I after parturition. Increased GH stimulates the lipolytic effects and hepatic glucose synthesis to meet the energy requirement for mammary lactose synthesis, suggesting that GH antagonizes insulin-dependent glucose uptake and attenuates insulin action to decrease gluconeogenesis.

한국재래빈산양의 번식과정에 따른 혈청내 Progesterone과 Estradiol-$17\beta$수준변화에 관한 연구 (Studies on the Changes of Progesterone and Estradiol-$17\beta$ Levels in Serum of Female Korean Native Goats during the Reproductive Stages)

  • This study was conducted to find out the changes of progesterone and estradiol-17$\beta$ levels in the serum of female Korean native goats during the reproductive stages such as the estrous cycle, pregnancy and periparturient period. Nine heads of female Korean native goats of 3 year old in average and weighing 35.7$\pm$1.4 kg were offered for the experiment. Blood samples were taken at 0, 1, 2, 5, 7, 10, 13, 15, 18 and 19 days after onset of estrus, and 1, 30, 60, 90, 120 and 149 days of pregnancy, and -5, -2, -1, 0, +1, +2 and +5 days of periparturient period where minus figures denote the days before paturition. The progesterone, estradiol-17$\beta$ in the serum samples were assayed by radioimmunoassay methods. The results of this study are summarized as follow: 1. The progesterone levels during the estrous cycles reached a peak level of 0.98$\pm$0.60ng/ml at 13 days after onset of estrus and decreased thereafter and were lower than 0.09$\pm$0.02mg/ml on the first day of estrus. 2. The estradioe17$\beta$ levels during the estrous cycles showed a peak level of 15.97$\pm$1.72pg/ml at onest of estrus, and decreased (5.41$\pm$0.51pg/ml-9.09$\pm$1.82pg/ml) during luteal phase. 3. The progesterone levels during the gestation period increased from day 1 and peaked at 90 days after mating and then decreased until 149 days. The peak level was 6.27$\pm$0.23ng/ml at 90 days. 4. The estradiol-17$\beta$ levels during the gestation period showed gradual increase, which were 9.03$\pm$0.88, 32.96$\pm$2.85, 46.03$\pm$2.42, and 54.06$\pm$1.64pg/ml on 30, 60, 90 and 120 dyas after mating respectively. 5. The progesterone levels measured from 5 days before the parturition to 5 days after showed the highest level at the shart of measurement (4.46$\pm$0.31ng/ml) and decreased gradually and bottomed out at one day post-partum and thereafter (0.24$\pm$0.02-0.45$\pm$0.06ng/ml). 6. The estradiol-17$\beta$ levels measured during the same periparturient period as progesterone showed increase to reach the peak level at 1 day before parturition and decreased rapidly thereafter (-5 dyas 69.46$\pm$3.62, -2 days 107.07$\pm$1.91, -1 days 137.83$\pm$7.54, 0 days 50.06$\pm$6.71 and +1 to +5 days 3.21-4.72 pg/ml).

Folic acid supplementation regulates key immunity-associated genes and pathways during the periparturient period in dairy cows

  • Objective: The current research was aimed to profile the transcriptomic picture of the peripheral blood lymphocytes (PBLs) associated with immunity in Chinese Holsteins supplemented orally with coated folic acid during the periparturient period. Methods: The total of 123 perinatal cows were selected for this study and divided into three groups; group A (n = 41, 240 mg/500 kg cow/d), group B (n = 40, 120 mg/500 kg cow/d) and group C (n = 42, 0 mg/cow/d) based on the quantity of folic acid fed. Three samples of PBLs were selected from each folic acid treated group (high, low, and control) and RNA sequencing method was carried out for transcriptomic analysis. Results: The analysis revealed that a higher number of genes and pathways were regulated in response to high and low folic acid supplementation compared to the controls. We reported the novel pathways tumor necrosis factor (TNF) signaling, antigen processing and presentation, Staphylococcus aureus infection and nuclear factor (NF)-kappa B signaling pathways) and the key genes (e.g. C-X-C motif chemokine ligand 10, TNF receptor superfamily member 1A, cluster difference 4, major histocompatibility complex, class II, DQ beta, NF-kappa-B inhibitor alpha, and TNF superfamily 13) having great importance in immunity and anti-inflammation in the periparturient cows in response to coated folic acid treatment. Conclusion: Collectively, our study profiled first-time transcriptomic analysis of bovine lymphocytes and compared the involved cytokines, genes, and pathways between high vs control and low vs control. Our data suggest that the low folic acid supplementation (120 mg/500 kg) could be a good choice to boost appropriate immunity and anti-inflammation as well as might being applied to the health improvement of perinatal dairy cows.

유우의 번식과정에 따른 유즙중의 성호르몬 수준 변화에 관한 연구 (Studies on the Changes of Sex Hormone Concentrations in Milk during the Reproductive Stages of Dairy Cows)

  • The study was carried out to find out the changes of the sex hormone levels in the milk of Holstein cows during the reproductive stages such as the estrous cycle, pregnancy and periparturient period. The FSH, LH, estradiol-17$\beta$ and progesterone from the milk samples were assayed by radioimmunoassay methods. The results of this study were summarized as follows: 1. The levels of progesterone and estradiol-17$\beta$ were similar among inter-quarters, but they were higher in after milking than before milking times, with no statistical significance. 2. The milk progesterone levels during the estrous cycles reached a peak mean level of 3.55$\pm$0.26ng/$m\ell$ at 15 days after estrus and they did not show any differences among the length of estrous cycles. The estradiol-17$\beta$ levels during the estrous cycles showed a peak level of 36.40$\pm$2.38pg/$m\ell$ at estrus, and decreased(17.20$\pm$0.46 pg/$m\ell$ to 18.65$\pm$1.26pg/$m\ell$) at luteal phase. 3. The FSH levels during the estrous cycles ranged from 2.25$\pm$0.23mIU/$m\ell$ to 4.35$\pm$0.24mIU/$m\ell$ showing significant changes. The LH levels during the estrous cycles gradually increased and remained a peak level of 10.90$\pm$0.36mIU/$m\ell$ from 20 to 25 days after estrus. 4. The progesterone levels during the pregnancy were decreased from 30 to 60 days after artificial insemination, and therafter continuously increased until 240 days. The estradiol-17$\beta$ levels during the pregnancy were 24.56$\pm$1.19pg/$m\ell$ at day 30 after artificial inseminaton, and increased rapidly until 180 days. The levles were agagin decreased by 26.17$\pm$3.03pg/$m\ell$ until 210 days and markedly increased by 68.00$\pm$8.70pg/$m\ell$ until 240 days. 5. The prolactin levels during the pregnancy were 31.27$\pm$2.31ng/$m\ell$ and 42.60$\pm$2.37ng/$m\ell$ at day 150 and 240 after artificial insemination respectively. The LH levels during the pregnancy reached a peak of 27.47$\pm$7.90mIU/$m\ell$ at day 30 after artificial insemination, and thereafter gradually decreased. 6. The progesterone levels during the periparturient period reached a peak of 4.61$\pm$0.34ng/$m\ell$ at day 3 prepartum, and thereafter gradually decreased, and showed 2.05$\pm$0.60ng/$m\ell$ at day 7 postpartum. The estradiol-17$\beta$ levels during the periparturient period showed high level from 207.23$\pm$6.04pg/$m\ell$ at day 1 prepartum to 239.90$\pm$13.90pg/$m\ell$ at day 2 prepartum, and thereafter began to decline and reached 51.87$\pm$1.72pg/$m\ell$ at by 7 postpartum. 7. The prolactin levels during the periparturient period showed relatively higher level at the time of parturition. The LH levels during the periparturient period rnage from 6.32$\pm$0.32mIU/$m\ell$ to 13.90$\pm$1.37mIU/$m\ell$ showing significant changes. 8. The progesterone levels(4.6$\pm$0.8ng/$m\ell$) of the pregnant cows were significantly higher than those (1.84$\pm$1.4ng/$m\ell$) of nonpregnant cows. The cows of artificial insemination from 61 to 90 days after parturition showed higher progesterone levels. 9. During 20 to 25 days after artificial insemination, the accuracy of pregnancy diagnosis from milk progesterone levels were 94.4% for nonpregnant cows(<2.3ng/$m\ell$), and 75.0% for pregnant cows( 3.2ng/$m\ell$). The average overall accuracy of pregnancy prediction for nonpregnant and pregnant cows 83.3% 10. The results obtained this study suggest that the understanding of the endocrinological mechanisms by means of milk hormone analysis during the estrous cycle, pregnancy and parturition would give the basic information needed for increasing efficiency of reproduction. This study would not only provide an accurate method of the early pregnancy diagnosis by milk progesterone levels but also contribute to the research of providing the method of detecting of FSH levels in milk, which was difficult in blood serum.

Effects of Monensin on Metabolism and Production in Dairy Saanen Goats in Periparturient Period

  • This trial evaluated the effects of dietary supplementation with monensin sodium on dry matter intake, metabolic parameters and milk yield and milk composition of dairy Saanen goats in the periparturient period. Twelve Saanen pregnant dairy goats were assigned to a treatment and a control group. Saanen goats were fed monensin as its 10% sodium salt in the amount of 33 mg/kg of total dry matter intake during 30 d before parturition till d 42 in milk. Blood samples were collected from each goat at d 30, 15 and 7 before expected kidding time and also in d 1, 7, 13, 19, 21, 28, 35 and 42 postpartum. The serum concentrations of ${\beta}$-Hydroxybutyrate (BHBA), non-esterifed fatty acid (NEFA), calcium, magnesium, inorganic phosphorus, cholesterol, triglyceride, urea, total protein, albumin and glucose and aspartate aminotransferase (AST) activity were determined. The BHBA concentration significantly decreased in goats, which received monensin in comparison to controls in the postpartum period (p = 0.049). The concentration of sodium (Na) was significantly influenced by monensin treatment in the postpartum period (p = 0.048). In addition, the difference in dry matter intake was highly significant between the two groups during the pre-partum period. Controls had more dry matter intake (DMI) than treatment goats (p = 0.0001). Mean 3.5% fat corrected milk production was not influenced by monensin treatment. However, milk fat percentage was significantly decreased in monensin treated goats (p = 0.0017).

Periparturient stocking density affects lying and ruminating behavior and one-week-calf performance of Holstein cows

  • Objective: This study aimed to investigate the effect of stocking density on the behavior, productivity, and metabolism of periparturient Holstein cows as well as calf performance. Methods: A total of 48 periparturient cows were randomly assigned into three groups at 28 days (±3 days) before their expected calving date. The stocking densities of the groups, relative to the standard cubicle and feed bunk number, were i) 80% (13 cows), ii) 100% (16 cows), and iii) 120% (19 cows). Lying and rumination behavior was recorded using electronic data loggers and HR-Tags from d -21 ("d-" means days before calving) until the calving date, d 0. Lying time was assessed to determine the diurnal total hours spent lying per day. Rumination time was averaged in 2 hours interval periods over 24 hours during the experimental period. Results: Cows in the 80% group spent more time lying and ruminating between d -21 and d -7 and tended to ruminate more between d -14 and d 0. Calcium levels tended to be higher for cows in the 80% group, no other observable differences were found in monitored blood parameters. Moreover, 3.5% fat corrected milk and energy corrected milk yields were higher in 80% group in the first month of lactation. No other observable differences were found in the yield and composition of colostrum and milk in the first 10 months of lactation. The growth and performance of calves in the first week of life was not affected by stocking density of the dams. Conclusion: We concluded that lower stocking density may increase lying and ruminating behavior of prepartum Holstein cows. However, this did not translate into improved productivity and metabolism.
