Persimmon has been considered to have therapeutic values for various diseases in Korea. Dried persimmon has been applied to wounded parts for anti-inflammatory and analgesic activities. Anti-coagulant fraction from Persimmon stem was purified through gel filtration, phenyl Sepharose, DEAE-Sephadex and additional gel filtration column chromatographies. Its molecular weight was estimated to be 130,000 ∼ 180,000. By element analysis, its main components were C, H, and O. The anti -coagulant was heat- stable and completely inhibited after periodate oxidation, indicating that it was a complex carbohydrate.
The physico-chemical properties of kapok fibers were altered via the combination processes of chlorite-periodate oxidation, in order to assess their efficacy as a heavy metal adsorbent. The chemically-oxidized kapok fibers were found to harbor a certain amount of polysaccharides, together with lowered lignin content. This alteration in lignin characteristics was clearly confirmed via FTIR and NBO yield. Moreover, chemically oxidized kapok fibers retained their hollow tube shape, although some changes were noted. The chemically oxidized kapok fibers evidenced elevated ability to adsorb heavy metal ions with the best fit for the Langmuir adsorption isotherm model. Three cycles of adsorption-desorption were conducted with in-between regeneration steps. Our experimental results indicated that chemically oxidized kapok fibers possessed excellent adsorption characteristics, and the modified kapok fibers could be completely regenerated with almost equimolar diluted sodium hydroxide. Pb, Cu, Cd and Zn ions evidenced adsorption rates of 93.55%, 91.83%, 89.75%, and 92.85% on the chemically oxidized kapok fibers. The regeneration efficiency showed 73.58% of Pb, 71.55% of Cu, 66.87% of Cd, and 75.00% of Zn for 3rd cycle with 0.0125N NaOH.
The adduct between phenylglyoxal and guanine was prepared. The structure of the adduct is similar to the glyoxal-guanine adduct. Aldehyde group of phenylglyoxal is added to 1-N of guanine base, and keto group is added to $N^2$ of guanine base. The structure of the adduct was determined by mass spectrometry, nuclear magnetic resonance spectroscopy and periodate oxidation. Periodate oxidation produced $N^2$-benzoyl-guanine from the adduct. On the basis of these results, ittvhas been assigned the structure I. The adduct is stable in alkaline solution: It does not dissociate into phenylglyoxal and guanine even after 2 hours heating at$60^{\circ}C$ at pH 12. The adduct is soluble in acidic ethanol, and is slightly soluble in neutral or alkaline water. It has a lower $A_{280}/A_{260}$ ratio at pH 1 compared to that of guanine.
The physicochernical properties of Korean buckwheat starches were investigated. The results were as follows; 1. Water binding capacity of kangwon hull buckwheat starch was 106.55% and that of Kangwon rice buckwheat was 99.35%. 2. The pattern of change in swelling power of hull buckwheat starch for increasing temperature started to increase at 60$^{\circ}C$ and increased rapidly from 80$^{\circ}C$, and that of rice buckwheat increased slowly from 60$^{\circ}C$ to 90$^{\circ}C$. 3. The ranges of gelatinization temp. of hull buckwheat and rice buckwheat starches were 70~75$^{\circ}C$ and 75~85$^{\circ}C$, respectively. 4. The blue value of hull buckwheat starch and rice buckwheat starch were 6.25 and 0.62, respectively. 5. The alkali number of hull buckwheat starch and rice buckwheat starch were 1.28 and 3.68 respectively. 6. The amylose content of hull buckwheat and rice buckwheat starch were 32.26% and 38.09%. 7. Periodate oxidation of hull buckwheat starch resulted that amylose had me average molecular weight of 103, 004, degree of polymerizatlon of 572 and amylopectin had me degree of branching of 7.64, glucose unit per segment of 13.09, and periodate oxidation of rice buck wheat starch resulted mat amylose had me average molecular weight of 125, 654, degree of polymerization of 698 and amylopectin had degree of branching of 6.59, glucose unit per segment of 15.16.
In order to evaluate the mechanism of electrolytic oxidation of iodate and to determine the optimum conditions for the electrolysis, studies were made using the cells without diaphragm and the lead peroxide anode. Results are summarized as followings: 1) Current density vs. anode potential curve by lead peroxide electrode had the different limiting current densities from platinum electrode and was more positive than platinum electrode. 2) Additions of potassium bichromate in the electrolyte contribute to maintain high current efficiency. 3) In the acid and alkaline regions, the current efficiencies decreased by reduction of iodate and discharge of hydroxyl ion, so maximum current efficiency was shown at pH 7. 4) Higher current density lowered the current efficiency in the region of 60-80% conversion of iodate. 5) Influence of the conversion on current efficiency in the region of 60-80% conversion of iodate.
A simple, selective and highly sensitive flow injection catalytic method was presented for determination of ruthenium based on its catalytic effect on the oxidation of pyronin B by periodate in pH=1.0. The reaction rate is controlled specrophotometricaly by monitoring the dye absorbance at 555 nm. The optimized conditions make it possible to determine ruthenium in the ranges of 0.1-10.0 ng/mL (r2=0.9982) and 10.0-50.0 ng/mL (r2=0.9934) with a detection limit of 0.04 ng/mL and a sample rate of 30±5 samples/h. Relative standard deviation for the results of five replicate measurements does not exceed 1.44%. The proposed method has been successfully applied for quantitation of ultra trace amounts of ruthenium in some environmental and biological samples.
Water-soluble crude polysaccharide (CAP-0) obtained from the Capsici Fructus(the fruits of Capsium annuum L.) showed a potent anti-complementary activity. The anti-complementary activity did not change by pronase digestion of CAP-0, but decreased by the periodate oxidation. CAP-0 was fractionated into four polysaccharide fractions, CAP-1,2,3 and 4, by the addition of cetyltrimethylammonium bromide. CAP-1, showing the highest anti-complementary activity, was refractioned by anion-exchange chromatography to give three major fractions(CAP-1-III, IV and V). CAP-1-III was shown to have low anti-complementary activity, but CAP-1-IV and V had high activity. CAP-1-III and IV were purified on Sephadex G-100 to give each two fractions(CAP-1-IIIa and IIIb, CAP-1-IVa and IVb), respectively. From the results of gel filtration and electrophoresis, these four fractions and CAP-1-V were found to be homogeneous polysaccharides. High molecular polysaccharies(M.W. CAP-1-IIIa 70,000, IVa 195,000, V 140,000) showed relatively higher anti-complementary activity than low molecular polysaccharides(CAP-1-IIIb and IVb).
The anticoagulant polysaccharide was screened from the ediable mushrooms. Among them, alkali extract of Ganoderma lucidum showed the highest activity in aPTT. A crude polysaccharide fraction (GL-I) was prepared from the 1N NaOH solution extract of Ganoderma lucidum followed by methanol-reflux, precipitation with ethanol, dialysis and lyophilization. GL-I inhibited the intrinsic pathway in blood coagulation pathway and exhibited concentration dependent anticoagulation effects. The anticoagulant activity of GL-I was decreased greatly by periodate oxidation, but was not changed by pronase digestion. These suggest that carbohydrate moiety may be related to the anticoagulant activity. GL-I consisted of glucose, galactose, fucose, xylose, mannose, arabinose in a molar ratio of 19.3:3.0:2.3:1.3:1.0:0.3. GL-I was partially purified on the DEAE-Toyopearl 650C(GL-IalongrightarrowGL-If) and on the Sephadex G-100(GL-Ic-ilongrightarrowGL-Ic-II).
${\beta}-Fructofuranosidase$ was immobilized covalently on the oxidized microbial wall of a Penicillium spp. 'PS-8', which is totally different from the conventional whole cell immobilization in concept. The immobilization of ${\beta}-fructofuranosidase$ by a series of treatments; oxidation of microbial cells with sodium metaperiodate, enzyme loading on the oxidized cells, extrusion, and crosslinking induced by glutaradehyde, were carried out. The final product had a good mechanical strength and showed 26% of the applied enzyme activity. The specific activity was 750 units per g of the dry cell product. The immobilized enzyme showed the kinetic parameters as follows; optimum pH at 5, optimum temperature at $55^{\circ}C$, activation energy of 19 kJ $mol^{-1}$, and apparent Km of 55 mM.
The enzyme-linked immunosorbent assay (ELISA)-inhibition test using a Clonorchis sinensis species-specific mouse monoclonal antibody (MAb) , CsHyb 0605-23, showed increased specificity over the conventional ELISA used for serodiagnosis of clonorchiasis. To characterize the corresponding antigen further, the MAb was tested against polysaccharide, protein and glycolipid fractions obtained from a crude extract of C. slnensis adult worms, using chloroform, methanol and phenol extractions. Only the polysaccharide fraction was recognized by the mb among those fractions. Mild oxidation of the antigen with sodium periodate showed decreased reactivity against the MAb. We concluded that the antigen and antigenic determinants recognized by the MAb are carbohydrates.
