Effects of Pretense Inhibitors and Antioxidants on In Vitro Survival of Porcine Primordial Germ Cells

  • One of the problems associated with in vitro culture of primordial gern cells (PGCs) is the large loss of cells during the initial period of culture. This study characterized the initial loss and determined the effectiveness of two classes of apoptosis inhibitors, protease inhibitors and antioxidants, on the ability of the porcine PGCs to survive in culture. Results from electron microscopic analysis and in situ DNA fragmentation assay indicated that porcine PGCs rapidly undergo apoptosis when placed in culture. Additionally, \ulcorner2-macroglobulin, a protease inhibitor and cytokine carrier, and N-acetylcysteine, an antioxidant, increased the survival of PGCs in vitro. While other protease inhibitors tested did not affect survival of PGCs, all antioxidants tested improved survival of PGCs (p<0.05). Further results indicated that the beneficial effect of the antioxidants was critical only during the initial period of culture. Finally, it was determined that in short-term culture, in the absence of feeder layer, antioxidants could partially replace the effect(s) of growth factors and reduce apoptosis. Collectively, these results indicate that the addition of \ulcorner2-macroglobulin and antioxidatns can increase the number of PGCs in vitro by suppressing apoptosis.

Denaturing Gradient Gel Electrophoresis Analysis of Bacterial Populations in 5-Stage Biological Nutrient Removal Process with Step Feed System for Wastewater Treatment

  • Changes in the bacterial populations of a 5-stage biological nutrient removal (BNR) process, with a step feed system for wastewater treatment, were monitored by denaturing gradient gel electrophoresis (DGGE) of PCR-amplified 16S ribosomal DNA fragments. DGGE analysis indicated seasonal community changes were observed, however, community profiles of the total bacteria of each reactor showed only minor differences in the samples obtained from the same season. The number of major bands was higher in the summer samples, and decreased during the winter period, indicating that the microbial community structure became simpler at low temperatures. Since the nitrogen and phosphate removal efficiencies were highly maintained throughout the winter operation period, the bacteria which still remaining in the winter sample can be considered important, playing a key role in the present 5-stage BNR sludge. The prominent DGGE bands were excised, and sequenced to gain insight into the identities of the predominant bacterial populations present, and most were found to not be closely related to previously characterized bacteria. These data suggest the importance of culture-independent methods for the quality control of wastewater treatment.

Foreign and Domestic Influence of the First Half of the Twentieth-Century American Furniture

  • Throughout the twentieth century, American furniture was subject to a number of influences-both domestic and foreign. Especially, in the first half of the twentieth century, there were strong foreign influences compare to the later half of the century. Therefore, this study aimed for discussing foreign influences and for addressing issues such as attitudes toward materials, ornamentation, and technology. As a result, this study suggests how American furniture becomes instinctive moving away from strong foreign influences from the second half of the century. The twentieth century was a period of rapid and dynamic change for American furniture design. In many ways, the designs throughout the century were reflective of the social, political, and economic culture of the time. With the birth of the modern movement in Europe and a strong influx of foreign immigrants, American design in the first half of the century was characterized by a reliance on European and foreign influences. However, after the two world wars, strong domestic influences came into play. The two world wars provided the essential catalysts for change: new materials, developing technology, and changes in life style and consumer values. A historical examination of design trends and individual designers illustrates how American furniture design evolved during this period.

Changes of haemolymph proteins in Pieris rapae L. during the cuticle formation and hardening process (배추흰나비의 큐티클 形成과 硬化에 따른 혈림프 단백질의 變化)

  • Changes and possible origin of haemolymph proteins during the cuticle formation and hardening are determined by means of acrylamide gel electrophoresis and immunodiffusion. The results by acrylamide gel electrophoresis showed at least 19 protein bands in the haemolymph and 13 fractions in the fat body with relatively constant pattern during the period of cuticle formation and hardening. Both haemolymph and fat body proteins are generally characterized by the presence of three to four heavy stained bands and several thin bands near the top region of the gel. At least over five haemolymph proteins are constantly present during this period. Immunodiffusion tests show that of total eight to nine pupal haemolymph proteins two proteins were already detected in the fat body before pupation and other two proteins were also found in the fat body immediately after pupation, suggesting fat body as possible source of these two haemolymph proteins.

  • The purpose of this study was to search the possibility to utilize materials in creative designs through examining the relationship between design materials and form by analyzing form by materials in the process of designing. In order to achieve this purpose, the objects to analyze designs were limited to chairs and lighting fixtures, and the period of analysis was also limited on 20th century. In this study, it was understood that changes of design materials and forms characterized by years. The period, when materials were used, was similar by design items, but the forms were different though they were same materials, and the same combinations of materials also changed whole forms. Also, it was found that design materials were affecting the change of forms if it is considered that whenever design materials were characteristically changed, forms were also peculiarly changed. Therefore, the result of this study might be utilized more effectively by design items and by structural elements even in each items, are arranged, connecting them with the trend of design.

Study on Interior Characteristics of Modern Department Stores in Korea (한국 근대백화점 실내공간의 표현 특성에 관한 연구)

  • This paper has focused on comprehending the interior designs and characteristics of modern department stores in Korea by looking into the birth and changes of those built under the Japanese Forced Occupation Period. The first modem department stores including Mitsukoshi, Georgia and Hwashin Department Stores began to appear in Korea under the Japanese Colonial Rule. They considered a variety of architectural elements like construction planning, shop arrangement, path of customer movement and interior design. The interior of modem department stores aimed to pursue the so-called modernization movement, but failed to fully contain the unique characteristics of the times in that particular period. Modem department stores took on new western styles prevailed in Korea and were characterized by the eclecticism in which various architectural forms exist together. In other words, their interiors implied a sign of the times in the confusing and corrupt world then. Especially those modern department stores were designed, repaired and extended by foreign designers who were ignorant of the history of Korean architecture. What is worse, their further remodelling practices made big mistakes by considering only functional and economical aspects and giving a back seat to the architectural and historical value of existing modern department stores. Therefore, we should make more efforts to study and conserve the fundamentals of modem architectures like modem department stores in order to have better understanding of the aesthetic value of indoor space and facilitating smooth communication between past and present.

The Characteristics of the Aerosol Number Concentration Observed in Seoul and Anmyondo During an Yellow Sand Phenomenon (황사시 서울과 안면도의 대기 중 에어로졸 수농도 특성)

  • The size-resolved number concentrations of aerosols ranging from 0.3 to 25 $mu extrm{m}$ were observed in Seoul and Anmyondo in the west coast of Korea during an Yellow Sand phenomenon in April 1998. Number size distributions of aerosols observed in both places are characterized by decrease in small particles of diameter less than 1${\mu}{\textrm}{m}$ and increase in large size between 1.35 and 10${\mu}{\textrm}{m}$ in heavy dust period. For particles in this size range, there was a good correlation between number concentrations observed in both places during the Yellow Sand episode. On the other hand, the number of small particles less than 0.82${\mu}{\textrm}{m}$ decreased, but the correlation between these particles in both places was enhanced during more intense dust period. The number of coarse particle larger than 10 ${\mu}{\textrm}{m}$ showed a distince diurnal variation without a significant change in amplitude, which was more visible in Seoul. It suggests that these coarse particles were more affected by local sources. Form these results, it was range of 1~10${\mu}{\textrm}{m}$ originated possibly from the dust source regions and the source signature depended on the degree of dust intensity.

  • The purpose of this study is to interpret the articles revision of the 2009 ISC(Cargo) compared to the 1982. The result of this study is summarized as followings: 1) the 'clause' attached to article in 1982 ISC(Cargo) was deleted in the revision of 2009 ISC(Cargo). 2) 2009 ISC(Cargo) is characterized as the marine insurance firms' acceptance of new environment change, limitation in causation and subject-matter insured, expansion of insurance period, and limited revision. 3) The assured has a large range of choice in 2009 ISC(Cargo) even though both 1982 ISqCargo) and 2009 ISC(Cargo) would be existed further. 4) There are few studies which have a focus on the impact of the relationship between responsibility of the insurer and opportunity of the assured on rate of premium. In the future, the studies on clauses, relationship among clauses, relationship between clause and rate of premium are needed in the 2009 ICC(Air), 2009 IWC(Air Cargo), 2009 ISC(Air Cargo), 2009 IWC(sending by post) et al.

Corrosion Behaviour of DH36 Steel Used for Oil Platform in Splash Zones

  • The splash zone is the most corrosive area of the marine environment, and the corrosion of steel structures exposed in this area is a serious concern. DH36 steel is one of most commonly used steels for offshore oil platforms in China, and its corrosion behaviour in splash zones was studied in this paper. Polarization curves were obtained from the corroded steel exposed in this area while the morphologies and rusts of the rust steel were characterized using scanning electron microscopy and X-ray diffraction. Double rust layers were formed in the splash zone. The inner layer contained magnetite and fine flaky lepidocrocite, and the outer layer was composed of accumulated flaky lepidocrocite and a small amount of goethite. In the wet period, the iron dissolved and reacted with lepidocrocite, and magnetite appeared, while the magnetite was oxidized to lepidocrocite again during the dry period. Electrochemical reduction and chemical oxidization cycled in intermittent wetting and drying periods, and magnetite and lepidocrocite were involved in the reduction reaction, leading to serious corrosion.

Change of Flavor Compounds of Pickled Garlic with Different Pickling Treatments (처리 조건을 달리한 마늘 장아찌의 향기 성분의 변화)

  • Raw whole garlic was pickled in two different ways, in soy sauce and in brine, to test the change of flavor compounds with aging period. The changes of pH, acidity, hardness and flavors were measured, accompanied with sensory evaluation. The pH of whole garlic in soy sauce and in brine was decreased as the aging period increased. The acidity was increased as pH decreased. Hardness tended to decrease as the aging time increased. For whole garlic in soy sauce and in brine, trans propenyl methyl disulfide, allyl methyl trisulfide, diallyl trisulfide, dimethyl trisulfide, allyl- 2,3-epoxypropyl sulfide, and 2-methyl-l,3-dithiane were increased as the aging proceeded but allyl methyl disulfide and diallyl disulfide tended to decrease. Hardness through the sensory evaluation decreased as the aging time increased. When the overall quality was compared between whole garlic in soy sauce and in brine after 60 days, the latter could be better estimated. Pickled garlic increased the flavor compounds such as trans propenyl methyl disulfide, allyl methyl trisulfide, diallyl trisulfide, dimethyl trisulfide, allyl-2,3-epoxypropyl sulfide, and 2-methyl-l,3-dithiane as the aging proceeded. Therefore, the above flavor compounds are considered as the major ingredients of the characterized flavor of pickled garlic.
