  • Objectives : This study was performed to investigate the effects of Perilla frutescens var. acuta extracts on growth inhibition of Streptococcus pyogenes KCTC 3208. Methods : Measured growth inhibition of Streptococcus pyogenes KCTC 3208 by concentration of the Perilla frutescens var. acuta. extract and collection periods of the Perilla frutescens var. acuta. and picking region of the Perilla frutescens var. acuta. Results : The Perilla frutescens var. acuta. extract is effective on retarding growth of Streptococcus pyogenes KCTC 3208. At 3% the growth of Streptococcus pyogenes KCTC 3208 was suddenly retarded. The stem of Perilla frutescens var. acuta. extract in a sunny place on June is more effective then its leaf. Conclusions : These results suggest that Perilla frutescens var. acuta. extract is effective in growth inhibition of Streptococcus pyogenes KCTC 3208.

  • This study investigated the proper dyeing conditions, fastness and functionality of cotton fabrics dyed with Perilla frutescens var. acuta extract. Repeat dyeing, combination dyeing of persimmon juice and fermentation dyeing were conducted as dyeing conditions. It was confirmed that the ΔE and the K/S value of cotton fabrics dyed with Perilla frutescens var. acuta extracts depending on repeat dyeing and combination dyeing slightly increased. Furthermore, the fastness to washing of persimmon combination dyeing and fermentation dyeing was very good. The fastness to rubbing was shown to be above grade 4 in all methods, and the fastness to light was not as good as grade 3 or lower. The persimmon juice dyeing, Perilla frutescens var. acuta extract repeat dyeing 4 times and combination fermentation dyeing showed very good antimicrobial abilities. Thereafter, additional studies are needed to improve the fastness to light and alternatives to further improve the dyeing properties.

  • The natural dyeing of cotton fabric with Perilla frutescens var. acuta extract was investigated. The proper extraction temperature and time were $100^{\circ}C$ and 120 minutes. The proper temperature, time and colorant powder concentration for the dyeing of cotton fabric with Perilla frutescens var. acuta powder were $90^{\circ}C$, 60 minutes and 15%(o.w.b.), respectively. In various mordanted methods, the K/S values of simultaneous-mordanted methods were higher than those of pre-and post-mordanted methods. And the Cu-and Fe mordant showed higher K/S values than those of other mordants. Light colorfasness of mordanted cotton fabric was poor, but the other colorfasnesses were shown to be good. The cotton fabric mordanted with Cu mordant was showed effective bacterial reduction.

  • Antimicrobial activity to the extracts of Perilla frutescens Briton var. acuta Kudo was investigated against various foodborne pathogenes or food poisioning microorganisms(Aspergillus flavus KCTC 6143 and KCTC 6961, Aspergillus niger ATCC 4695, Listeria monocytogenes ATCC 15313, Staphylococcus aureus 196E ATCC 13565, Escherichia coli O157:H7 ATCC 43895, Salmonella typhimurium ATCC 13311 and Yersinia enterocolitica). The ethanol extract of Perilla frutescens Briton var. acuta Kudo was very stable over heat at $121^{\circ}C$ for 15 min. In concentration of $1000\mu\textrm{g}$/mL into culture broth(TSB), the ethanol extract of Perilla frutescens Briton var. acuta Kudo showed the strongest antimicrobial activities against Listeria monocytogenes, followed by Staphylococcus aureus 196E, Salmonella typhimurium. Gram negative bacteria(Escherichia coli O157:H7, Salmonella 쇼phimurium, Yersinia enterocolitica) were less sensitive than Cram positive bacteria but the growth of Escherichia coli O157:H7 and Yersinia exterocolitica were inhibited with increasing concentrations of the extract in culture broth.

  • Objectives : This study aimed to establish cosmeceutical activities of Perilla frutescens var. acuta. Methods : Folium Perillae, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect have been investigated in the cosmeceutical activity measurement of function experiment. Results : Both water and ethanol extracts of Perilla frutescens var. acuta showed relatively high electron donating ability of more than 70% at over 500ppm. Also, SOD like activity increased in a dose dependent matter of more than 80% at 5,000ppm, while tyrosinase showed insufficient inhibitory rate. Xanthine oxidase showed a meaningful inhibitory effect of 72.4% in water extract and 55.3% at 1,000ppm in ethanol extract. The addition of $Fe^{2+}ion\;and\;Cu^{2+}ion$ showed relatively high oxidizing inhibitory effect of 57% and 41%, respectively, at 500ppm in water extract and, when $Fe^{2+}$ ion was added in ethanol extract, the effect of 64% at 1,000ppm was achieved. Also, higher oxidizing inhibitory effect was shown against $Fe^{2+}$ rather than $Cu^{2+}$ in both water and ethanol extracts. For human cancer cells, relatively high growth inhibition ability was shown against melanoma G361 in both water and ethanol extracts. Conclusion : From the above results, it was confirmed that Perilla frutescens var. acuta could be used in functional cosmetics.

Anti-Amyloidogenic Effects of Triterpenoids Isolated from Perilla Leaves (자소엽에서 분리된 트리테르페노이드의 베타-아밀로이드 응집 억제 효과)

  • Perilla frutescens Britton var. acuta Kudo, an annual plant primarily cultivated in China, Japan, and Korea, has been used as a traditional medicine to treat inflammatory diseases, depression, and many anxiety-related disorders. Previously, we reported the inhibitory effects of n-hexane layer of P. frutescens var. acuta extract against beta-amyloid (Aβ) aggregation, and the isolation of asarone derivatives as active constituents from n-hexane layer. In this study, dichloromethane layer of P. frutescens var. acuta was applied to bioassay-guided isolation methods accompanied with Thioflavin T (Th T) fluorescence assay to investigate the inhibitory effect on Aβ aggregation and disaggregation. As the results, three triterpenoids including ursolic acid (1), tormentic acid (2) and corosolic acid (3) were isolated. All compounds reduced Aβ aggregation and increased disaggregation of preformed Aβ aggregates in a dose-dependent manner. However, the inhibitory effect of three compounds on Aβ aggregation was not correlated with antioxidant activity, which was measured by DPPH assay. Taken together, these results suggest that the triterpenoid derivatives from P. frutescens have the potential to be developed as good therapeutics or preventatives for AD.

  • The effects of monazite and fixing agents on cotton fabric dyed with Perilla frutescens var. acuta extract were investigated. The proper monazite treatment concentration, temperature and time were 10%(o.w.b.), $50^{\circ}C$ and 60minutes. By various fixing agent treatment, $FeSO_4$ showed a relatively high K/S value and the order of K/S value decreased as follows, cation surface active agents, soybean and NaCl. And the monazite and $FeSO_4$ fixing agent showed higher anion emissity than those of untreated cotton and other fixing agents. The cotton fabrics showed improved color fastness by monazite and fixing agents treatments with the exception of light fastness. And the cotton fabrics fixed with fixing agents were showed effective bacterial reduction with the exception of NaCl.

  • In the present study, we investigated the effect of the water extract of Perilla frutescens var. acuta (Labiatae; WEPF) on the mast cell-mediated allergic reactions. WEPF was anally administered to mice for high and fast absorption. WEPF inhibited compound 48/80-induced systemic allergic reaction. WEPF attenuated immunoglobulin E (IgE)-mediated local allergic reaction. In addition, WEPF decreased the gene expression of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated HMC-1 cells. These results indicate that WEPF inhibits mast cell-mediated allergic reactions in vivo and in vitro.

  • Nine triterpenes were isolated from the petroleum ether and MeOH extract of Perilla frutescens var. acuta leaves. Their structures were determined to be arjunic acid (1), maslinic acid (2), oleanolic acid (3), euscaphic acid (4), tormentic acid (5), 3-O-trans-p-coumaroyltormentic acid (6), 28-formyloxy-$3{\beta}$-hydroxy-urs-12-ene (7), ursolic acid (8), and corosolic acid (9) by spectroscopic methods. The compounds 1, 2, 4, 6, and 7 were isolated for the first time from this plant and the Genus Labiatae. The isolated compounds (1-9) were tested for cytotoxicity against four human tumor cell lines (A549, SK-OV-3, SK-MEL-2, and HCT-15) in vitro using a Sulforhodamin B bioassay.

  • The purpose of this study was to investigate the antioxidative and anti-inflammatory activities of ethanol extracts from Perilla frutescens var. acuta by varying the concentration of ethanol at 30, 50, 70, and 90% to utilize the effective extract of Perilla frutescens as a cosmetic and pharmaceutical material. In the DPPH antioxidant activity test, the 70% ethanol extract showed the highest activity with an IC50 of 680.98ppm. ABTS showed a high activity in the 50% ethanol extract and the 70% ethanol extract with an IC50 of 646.94 and 661.94 ppm, respectively. Each ethanol extract showed antioxidant activity at a certain concentration (100-10000 ppm), but did not show any significant relationship with the ethanol extract concentration. In RAW 264.7 macrophages induced by LPS, each of the ethanol extracts showed reduced NO production in all extracts, and more than 50% ethanol extract (10000ppm) inhibited nitric oxide formation by 85% or more. In particular, the 70% ethanol extract showed 90% or more nitric oxide production inhibition. In addition, the MTT assay showed no cytotoxicity at all concentrations (1250-10000 ppm) of each extract. In this study, the ethanol extract of Perilla frutescens var. acuta has antioxidant activity and anti-inflammatory activity that is dependent on the concentration at each extraction concentration.
