Development of carotenoid production process using perenniporia fraxinea (아까시재목버섯 유래 카르테노이드 계열 항산화 물질 생산)

  • In this study, we analyzed the effect of media on the production of carotenoids and mycelia by using Perenniporia fraxinea. Malt extract-based medium with less than 0.1% peptone stimulated the production of carotenoids, and the one with more than 0.2% peptone inhibited its production. P. fraxinea grown in medium without malt extract did not produce carotenoids, although a small amount of peptone was added to the medium.After carotenoid production, the culture broth was separated using simple centrifugation and the supernatant was harvested as a carotenoid solution. Ethanol was used to extract carotenoids from mycelia. Carotenoid solution separated or extracted from the culture solution showed DPPH radical scavenging activity. The antioxidant carotenoids produced by P. fraxinea are derived from natural products, have no toxicity and side effects, and exhibit excellent antioxidant effects; therefore, they can be effectively used to remove oxides produced by active oxygen.

Effect of Perenniporia fraxinea on Eliminating Urushiol from Rhus verniciflua Stokes Stem Bark (아까시재목버섯이 옻나무 껍질의 urushiol 제거에 미치는 영향)

  • This study evaluated the raw materials for spawning Perenniporia fraxinea, to eliminate urushiol. The growth rates of spawns on grains of millet, brown rice, and wheat were 4.92±0.05, 2.20±0.03, and 1.93±0.03 mm/day, respectively, and the laccase activity was 0.86±0.02, 0.04±0.01, and 0.01±0.00 U/mL, respectively. These observations revealed millet as the most appropriate grain for spawn production in terms of growth rate and enzyme activity. Inoculation of lacquer tree (Rhus verniciflua Stokes) stem bark with millet spawns of P. fraxinea resulted in a reduction of its urushiol contents, up to 86.6% on the third day and up to 98.5% on the seventh day. The optimal period of cultivation to eliminate urushiol was three days with 68% of residual flavonoids and 42% of phenolic components. When compared to the product cultivated from liquid spawn, the millet spawn reduced the cultivation period from 10 days to 3 days for eliminating urushiol.

Evaluation of biological activities of extracts of Korean wild mushrooms (국내 수집 야생버섯류 추출물의 생리활성 비교)

  • The aim of the study was to obtain the extracts of various native wild mushrooms and select the useful resources though biological activity evaluation. The anti-oxidant potential, nitrite scavenging activity, and ��-glucan content of wild mushrooms collected from Eumseong and Bonghwa in Korea were investigated. Based on the results of this study, Ganoderma lingzhi (OK1362) showed the highest DPPH radical scavenging activity (73.2%), ferric reducing anti-oxidant power (0.134), reducing power (0.155), nitrite scavenging activity (53.6%), total polyphenol content (28.9 mg GAE/g), flavonoid content (10.0 mg QE/g), and ��-glucan content (25.2%) when compared to other wild mushrooms sampled in this study. In addition, it was confirmed that Perenniporia fraxinea (OK1360), Amanita sp. (OK1398), and Russula sp. (OK1406) had relatively high anti-oxidant and nitrite scavenging potentials. In conclusion, our results can provide fundamental data for extracting beneficial compounds from wild mushrooms.
