위암환자에서 혈중 Pepsinogen 검사의 의미 (Clinical Correlation between the Serum Pepsinogen I/II Ratio and Gastric Cancer)

  • 목적: 위암환자의 임상적 특성과 수술 전 혈중 pepsinogen I/II 비율의 결과를 비교 분석하여 위암 발생과 혈중 pepsinogen의 변화와의 관련성 및 혈중 pepsinogen의 위암 진단의 이용 가능성 여부를 밝히는 것이 본 연구의 목적이다. 대상 및 방법: 분당차병원에서 절제술을 받은 103명의 위선암 환자를 대상으로 수술 전 혈중 pepsinogen I과 II를 측정하고, 이 결과를 종양의 임상 및 조직학적 요소와 비교 분석하였다. 결과: Pepsinogen I/II 비율의 평균치는 mucinous type, 종양 주위에 위축이 있는 경우 및 종양이 큰 경우에 감소하였으며 종양주위에 위축이 있는 환자에서도 종양이 클수록 의미 있게 감소하였다. 암세포가 고분화를 보이는 환자가 인환세포암 환자에 비하여 pep I/II이 의미 있게 감소하였으나 종양의 위치,침윤도, 림프절 전이 여부 및 Lauren분류에 따른 종양의 아형은 pep I/II의 변화와 의미 있는 관련을 보이지 않았다. 결론: 본 연구의 결과는 혈중 pepsinogen과 점막위축 정도와의 연관성은 증명하였지만 이 결과만으로 pepsinogen과 위암의 전암 병소의 연관성이나 일차점진의 유용성을 논하기는 부족하다고 생각된다.

Pepsinogen 분비에 대한 분비촉진제 억제제 및 인삼 Saponin의 상호작용 (The Interaction of Ginseng Saponin with Secre Tagogues, Inhibitors and Its Relative Agents on Pepsiogen Secretion in Isolates Rabbit Gastric Glands)

  • The pepsinogen secretion was stimulated by the cholecystokinin, caerulein, isoproterenol, and carbachol, respectively. But it was increased slightly and returned to control level by the combiantions of total saponin with each above the agents. Even though the atropine had the inhibition effect, the pepsinogen secretion was recovered to normal level from depressed condition by the combination of the atropine with total saponin. Propranolol showed the same pattern as atropine, too. On the other hand, the pepsinogen secretion was stimulated by the DBcAMP alone, but decreased to control level by the combination with the total saponin. In the case of DBcGMP, the pepsinogen secretion was decreased by itself, but stimulated the above control level by the combination with total saponin. Histamine alone had little effect on the pepsinogen secretion, but when combinated with total saponin, the pepsinogen secretion was increased. Serotonin alone and with total saponin, had no effect respectively, From the above results, the total saponin may have the normalization action stimulating or decreasing the pepsinogen secretion to the control level.

닭에 있어서 Pepsinogen 과립(顆粒)의 분포(分布), 분필(分泌) 및 생성(生成)에 관한 광학(光學) 및 전자현미경적연구(電子顯微鏡的硏究) (Light and Electron Microscopic Studies on Distribution, Secretion and Formation of Pepsinogen Granules in Chickens)

  • In mammals there are two distinct cellular units of the gastric glands which are responsible for the secretion of acid and pepsin respectively, namely, the parietal cells for acid and the peptic or chief cells for pepsin. On the other hand, the bird does net have separate parietal and chief cells in the glandular stomach. There exist only a single cell type in the asian gastric secretory-glands. In spite of this single cell type, however, variation in pepsin and acid secretion can he seen. Present study was conducted to know distribution, secretion and formation of the pepsinogen granules in chicken and rat stomach which observing by light and electron microscope. 1. In chicken, the pepsinogen granules are distributed in all submucosal gland cells and yet there are no distinction of parietal and chief cells. In rat, the pepsinogen granules are distributed in chief cells which lined the lower two-thirds of the gastric tubles and the parietal cells occupy upper third of the tuble. 2. Carbachol markedly stimulates the secretion of pepsinogen granules in chiken and rat, but Histamine is slightly. 3. After Histamine and Carbachol treatment, the pepsinogen granules are formated continuously and reaccmulated as control after 3 to 4 hours.

Pepsinogen 분비조절물질과 인삼성분의 복합처리가 cAMP의 양에 미치는 영향 (Effect of Ginseng Components with Pepsinogen Secretion Regulatory Agents on cAMP Content in Isolated Rabbit Gastric Glands)

  • On the cAMP content in isolated gastric glands from rabbit stomach, the effect of ginseng components (total saponin, diol saponin, triol saponin) with pepsinogen secretion regulatory agents (cholecystokinin, isoproterenol, carbachol, propranolol, atropine, DECAMP, DBcGMP) were studied in vitro. According to the results, ginseng components may have the effect of stimulation or inhibition on cAMP production, and both dial saponin and triol saponin may be reciprocal effect to pepsinogen secretion regulatory agents. It seemed that the ginseng components may have the normalization action to pepsinogen regulatory agents on cAMP content in isolated rabbit gastric glands.

  • This study was carried out to investigate the effect of red ginseng extract on development of pepsinogen 1 decrased pyloric glands in experimental stomach carcinogenesis in Sprague-Dawley male rats. Sequential quantitative analyses (by ABC immunohistochemical staining) were made of pepsinogen 1 decreased pyloric glands (PDPG) after treating rats first with a single dose (160 mg/kg) N-methyl-N'-nitrosoguanidine (MNNG) and then with N-ethyl-N'-nitro-N-nitrosoguanidine (ENNG) (100ug/ml of drinking water) as a second gastric carcinogen (or promoter).

  • The present paper demonstrates the expressions and amounts of pepsinogen and prochymosin in neonate, young and adult goat's proper gastric glandular regions by the immunochemical and the immunohistochemical analyses with the anti-bovine pepsinogen serum and anti-bovine chymosin serum. Each bovine serum was demonstrated to have reactivities against corresponding goat's antigen by immunochemical analses and enzymatic activities. The anti-pepsinogen was higher in the new born animals than the maternal milk feeding one, suggesting that the maternal milk might control the pepsinogen production in the proper gastric glands. The patterns of prochymosin expression in the goats was similar to that in cattle.

  • In order to observe the time of the first appearance of Bowie positive pepsinogen granules in the gland cells of the chicken proventriculus, histological examination was performed on the chicken embryo proventriculus at 7, 10, 14, 18, 19, 20 and 21 days postincubation. The first appearance of Bowie positive pepsinogen granules in the gland cells of the chicken proventriculus was at 21 days postincubation.

Differences in the Levels of Gastric Cancer Risk Factors Between Nanjing and Minqing Counties, China

  • Objectives: In Fujian Province, China, gastric cancer is one of the leading causes of mortality among all malignant tumors. Nanjing county and Minqing county are located in inland Fujian and have similar general demographics. However, the adjusted mortality rate of gastric cancer in Minqing was found to be much higher than that in Nanjing. We sought to explore factors associated with this increased risk of gastric cancer between the two counties. Methods: We recruited 231 and 224 residents from Nanjing and Minqing, respectively, and analyzed differences between their dietary habits, Helicobacter pylori infection rates, and concentrations of serum pepsinogen I, pepsinogen II, gastrin-17, and ratio of pepsinogen I:II. Results: Subjects in Minqing had more first-degree relatives who had been diagnosed with upper gastrointestinal tumor, more unhealthy dietary habits, a higher Helicobacter pylori positive rate, and greater proportion of abnormal serum gastrin-17 than those in Nanjing did. Conclusions: The factors that differed between these two counties might indicate that residents in Minqing have a higher risk for developing gastric cancer than those in Nanjing do.

Role of Nitric Oxide in Pepsinogen Secretion from Rat Gastric Chief Cells

  • Nitric oxide (NO), a cellular messenger synthesized from L-arginine by NO synthase (NOS, EC.1.14.13.39), is considered to be a regulator of gastric secretion. In the present study, the role of NO in the regulation of exocrine secretion was investigated in rat gastric chief cells. Treatment of chief cells with carba-chol resulted in an increase in the arginine conversion to citrulline, the amount of $NO_{x}$, the release of pepsine-gen, and the level of cGMP Especially, carbachol-stimulated increase of arginine to citrulline transformation, the amount of $NO_{x}$, cGMP level and the release of pepsinogen were partially reduced by the natural NOS inhibitor, $N^{G}$-monomethyl-L-arginine (MMA) and $N^{G}$, $N^{G}$-dimethyl-L-arginine (DMA). Furthermore, MMA- and DMA-induced decrease of pepsinogen secretion showed dose-dependent patters. Activation of NOS is one of the early events in receptor-mediated cascade of reactions in gastric chief cells and NO, not completely, but partially mediates gastric secretion. Agonist-stimulated pepsinogen secretion in chief cells has been considered to be mediated in adenosine 3',5'-cyclic monophosphate pathway and/or guanosine 3', 5'-cyclic monophosphate (cGMP) pathway. Taken together, the above results suggest that partial decrease of exocrine secretion following treatment of NOS inhibitor may result from the inactivation of NOS and subsequent guano- late cyclase, and NO/cGMP pathway may play a pivotal role in exocrine secretion.

CHANGES IN CONTENTS AND LOCALIZATIONS OF CARBONIC ANHYDRASE II, PROCHYMOSIN AND PEPSINOGEN IN ABOMASAL MUCOSAE DURING LONG TERM MILK FEEDING GOATS

  • The present paper describes temporal changes of immunohistochemical localization and quantities of carbonic anhydrase isozyme II (CA-II) prochymosin (PC) and pepsinogen (PN) in goat's abomasal mucosae during long term milk feeding. The CA-II was not detected by day 14 after birth and then became positive on day 34 in the parietal cells, suggesting that the excretion of the hydrochloric acid (HCl) begins between days 14 and 34 under a feeding condition without solid materials. The quantity of the PC in the gastric chief cells detected by the ELISA showed rapid increase from the day of birth, making a peak on day 8 and then gradually decreased with age. The decrease in quantity of PC became started during the time period when HCl excretion had not started yet. The quantities of PN in the gastric chief cells were almost stable during the whole period examined. Expressions of these gastric enzymes did not seem to be regulated by the change of feeding condition.
