Screening of the Antifungal Activity from Natural Products against Candida albicans and Penicillium avellaneum (Candida와 Penicillium 속 진균에 대한 천연물의 항진균 효과 검색)

  • For the research of antifungal active constituents from natural products, 226 plants were extracted with ether and methanol, separately, and screened antifungal activity against Candida albicans and Penicillium avellaneum cells. The results demonstrated that 30 samlpes showed antifungal activity in ether or methanol extracts and 17 samples in ether extracts and 20 samples in methanol extracts against C. albicans. Against P. avellaneum, 19 samlpes showed antifungal activity in ether or methanol extracts and 17 samples in ether extracts and 11 samples in methanol extracts, respectively. The antifungal activity of natural products against C. albicans were showed more sensitive than P. avellaneum, and the polarity of the solvent was not specific in antifungal activity.

  • Dextranase는 dextran의 $\alpha-1,6-glycoside$ 결합을 끊는 효소로써 치석을 형성하는데 요인이 되는 dextran을 분해 제거하는 역할을 한다. 본 실험에서는 이러한 dextranase 생산 곰팡이류 중에서 Penicillium 속에 속하는 균주를 선정하여 이 효소 생산의 최적조건을 실험 검토하였다.

Diversity Analysis of Endophytic Fungi Isolated from the Roots of Coastal Plants in Taean Peninsula (태안반도에 자생하는 해안식물 뿌리에서 분리한 내생진균의 다양성 분석)

  • Forty-two endophytic fungi were isolated from the roots of coastal plants, such as Aster sphathulifolius Maxim., Aster tripolium L., Phragmites australis, and Puccinellia nipponica Ohwi, naturally growing in Taean peninsula. The identity of the endophytic fungal strains was analyzed using the sequence of internal transcribed spacer regions. All fungi belonged to the phylum Ascomycota, and they were classified into eight orders (Botryosphaeriales, Capnodiales, Diaporthales, Dothideales, Eurotiales, Helotiales, Hypocreales, and Pleosporales) and thirteen genera (Alternaria, Aureobasidium, Cadophora, Cladosporium, Davidiella, Diaporthe, Fusarium, Gibberella, Macrophomina, Metarhizium, Neosartorya, Penicillium, and Phoma). Among the analyzed fungi, the fungi belonging to the genus Penicillium in Eurotiales were the most widely distributed. The host plant Aster tripolium L. was found to contain the most diverse endophytic fungal species among the coastal plants.

  • Halophyte samples, such as Suaeda japonica, Phragmites australis, Limonium tetragonum, Suaeda maritima were collected from Go-chang salt marsh. Thirty-nine endophytic fungal strains were isolated from the roots of halophytes naturally growing in salt marsh. All endophytic fungal strains isolated were analyzed by internal transcribed spacer (ITS) containing ITS1, 5.8 s and ITS2 region. Endophytic fungal strains belong to eight orders, i.e., Eurotiales (36%), Pleosporales (26%), Hypocreales (18%), Incertae sedis (8%), Glomerellales (5%), Sordariales (2%), Xylariales (2%), and Capnodiales (3%). On genus level, they were composed of Alternaria, Aspergillus, Chaetomium, Cladosporium, Colletotrichum, Coniothyrium, Dothideomycete, Fusarium, Gibberella, Macrophoma, Penicillium, Pestalotiopsis, Phaeosphaeria, Phoma, Pleosporales, Pseudozyma, Talaromyces, and Termitomyces. Of them, Penicillium (26%), Fusarium (13%) of Eurotiales and Hypocreales were predominant.

  • A mycological survey was carried out with the soil samples collected in Korea from Sptember, 1978 to December, 1979. Special attention was paid fungus genus Penicillum. One hundred twenty three isolates, as a result, were obtained from the Korean soils. Among these, sixteen species were iedentified and described in this paper. Almost all of the fungi reported here are new to Korea. One of them is an asomycete cleisothecia of $120~50\mu$ in diameter. Ascospores of this fungus were found lenticular with an equatorial furrow as indicated in the previous descriptions.

  • Five coastal plant species, Artemisia fukudo, Aster sphathulifolius, Plantago camtschatica, Sedum oryzifolium, and Setaria viridis, were collected from the coastal region of Ulleung Island (Ulleung-Do, South Korea). Thirty-six endophytic fungi were isolated from the roots of these plants, and all were identified by using PCR with the following specifications: internal transcribed spacer 1 (ITS1), 5.8S rRNA, and ITS2 regions. Phylogenetic analysis indicated that all fungal strains belong to the phylum Ascomycota and comprise four orders (Capnodiales, Eurotiales, Hypocreales, and Pleosporales). Among all the identified species, the Eurotiales species were more abundant than species in the other orders. Nine different genera (Alternaria, Aspergillus, Cladosporium, Exserohilum, Fusarium, Neosartorya, Penicillium, Phoma, and Pyrenochaeta) in the four orders were confirmed. Penicillium and Aspergillus species were the most dominant species among the endophytic fungi isolated from the coastal plants. Shannon's diversity index (H') ranged from 0.684 to 1.609, and the endophytic fungi in S. oryzifolium was more diverse compared to the endophytic fungi in the other plants.

Isolation and Cultivation of Microorganism Producing Lipoxygenase Inhibitor (Lipoxygenase Inhibitor를 생산하는 미생물의 분리 및 배양)

  • The microorganisms producing a lipoxygenase inhibitor were screened from a wide variety of sources. The isolated strain was assigned to genus Penicillium by its cultural and morphological characteristics. The proper medium for the production of lipoxygenase inhibitor was composed of glucose 3.0%, ammonium sulfate 0.4%, and potassium phosphate (dibasic) 0.1%. The cultivation for lipoxygenase inhibitor production was carried out in 500m1 Erlenmyer flasks containing 100m1 of the medium at $30^{\circ}C$ by cultivating reciprocally. The highest lipoxygenase inhibitor production was observed after 8 days of cultivation. The inhibitor was the low molecular weight substance and inhibited specifically soybean origin lipoxygenase.

  • The soil microfungal flora of the paddy fields in Korea was investigated at four different seasons. The fungi were isolated by the dilution plate method from soil samples of two selected sites around Seoul. A total of 85 isolates was obtained as pure cultures and 30 species 13 genera were identified and 11 isolates were unidentified. Among these, 6 species of Deuteromycetous fungi, Penicillium spp., were found to be dominant in paddy field soils. Penicillium funiculosum, P. piceum, P. roqueforti and P. verruculosum were described as new to Korea. P. piceum has remarkable characteristics of the typical columnar head similar to a compact spruce-like evergreen tree. P. requeforti has penicilli variable in pattern with compactly branched metulae and appressed or Paecilomyces variotii were also described. In addition, Zygorhynchus moelleri, a remarkably dominant Zygomycete in Korean paddy soils, produces subglobose sporangia with oval columella and dark colored zygospores of about $35\mu\textrm{m}$ in diameter.

  • Halophytes of seven species, Carex scabrifolia Steud., Limonium tetragonum Bullock, Salicornia europaea L., Suaeda glauca Bunge, Suaeda japonica Makino, Suaeda maritima Dumort., and Triglochin maritimum L. were collected from the Taean Peninsula. Thirty-seven endophytic fungi were isolated from the root of halophytes, and analyzed using the DNA sequences of internal transcribed spacers (ITS). The diversity of all endophytic fungi was analyzed using diversity indices. The endophytic fungi associated with the halophytes belonged to seven orders; Eurotiales (78%), Capnodiales (5%), Hypocreales (5%), Agaricales (3%), Corticiales (3%), Glomerellales (3%), and Pleosporales (3%). At the phylum level, the endophytic fungi were composed of Ascomycota and Basidiomycota. At the genus level, Alternaria, Aspergillus, Cladosporium, Paecilomyces, Penicillium, Phanerochaete, Schizophyllum, Talaromyces, and Verticillium were confirmed. Among them, Penicillium was the most abundant in the roots of the halophytes. This study analyzed the distribution and diversity of endophytic fungi on halophytes in the Taean Peninsula.

  • The evolutionary relationships of the genus Trichoderma and related taxa were assessed using partial sequencing of 18S ribosomal RNA. Phylogenetic tree divided into three major groups; 1. Saccharomyces cerevisiae-Geotrichum klebahnii-Alternaria mali group; 2. Neurospora crassa-Aspergillus-Penicillium-Chrysosporium pannorum-Scopulariopsis sp. group; 3. Trichoderma group. The genus Trichoderma seemed to be phylogenetically separated from Saccharomyces cerevisiae, Aspergillus and Penicillium groups, and have passed through it's own evolutionary pathway.
