Penicillium Diversity from Intertidal Zone in Korea

  • Penicillium species are commonly isolated from various outdoor and indoor environments, including marine environments such as sponges, algae and sand. Penicillium is especially important because numerous bioactive compounds have been isolated. Penicillium was the most common species in intertidal zone in Korea, however the diversity and ecological roles of Penicillium in intertidal zone are not clarified. We explored diversity and ecological roles of marine-derived Penicillium from tidal flat and sea sand in Korea. The diversity of marine-derived Penicillium from Korea was investigated using both culture-dependent and culture-independent approach by ${\beta}$-tubulin sequence. In addition, we evaluated optimal temperature, halo-tolerance, and enzyme activity of Penicillium strains, such as extracellular alginase, endoglucanase, ${\beta}$-glucosidase, and protease. For culture-dependent approach, a total of 182 strains of 62 Penicillium species were isolated, with 53 species being identified. The most common species was Penicillium oxalicum, followed by P. crustosum, P. brasilianum, P. koreense, and P. griseofulvum. Species richness and composition were not significantly different by season, substrates, and seaside. For culture-independent approach using Illumina sequencing, 73 OTUSs were detected. The most frequently observed species was P. antarcticum, followed by P. koreense, P. crustosum, and P. brevicompactum. Diversity of Penicillium was higher during winter season than during summer season and in western sea than in southern sea, respectively. Community structure was significantly different by season and sea side. 52 species were detected by both methods. Unique species were isolated from each of methods - 10 from culture methods and 21 from Illumina sequencing. Furthermore, salinity adaption of the Penicillium varied depending on species. Many Penicillium species showed endoglucanase, ${\beta}$-glucosidase, and protease activity. Some species including P. paneum and P. javanicum degraded the polycyclic aromatic hydrocarbons. Thus, our results demonstrate that intertidal zone in Korea harbors diverse Penicillium community and marine-derived Penicillium play important ecological roles as decomposers of organic material in marine environments.

Re-Identification on Korean Penicillium Sequences in GenBank Collected by Software GenMine

  • Penicillium species have been actively studied in various fields, and many new and unrecorded species continue to be reported in Korea. Moreover, unidentified and misidentified Korean Penicillium species still exist in GenBank. Therefore, it is necessary to revise the Korean Penicillium inventory based on accurate identification. We collected Korean Penicillium nucleotide sequence records from GenBank using the newly developed software, GenMine, and re-identified Korean Penicillium based on the maximum likelihood trees. A total of 1681 Korean Penicillium GenBank nucleotide sequence records were collected from GenBank. In these records, 1208 strains with four major genes (Internal Transcribed Spacer rDNA region, b-tubulin, Calmodulin and RNA polymerase II) were selected for Penicillium reidentification. Among 1208 strains, 927 were identified, 82 were identified as other genera, the rest remained undetermined due to low phylogenetic resolution. Identified strains consisted of 206 Penicillium species, including 156 recorded species and 50 new species candidates. However, 37 species recorded in the national list of species in Korea were not found in GenBank. Further studies on the presence or absence of these species are required through literature investigation, additional sampling, and sequencing. Our study can be the basis for updating the Korean Penicillium inventory.

일균일명 체계에 의한 국내 보고 Aspergillus, Penicillium, Talaromyces 속의 종 목록 정리 (Species List of Aspergillus, Penicillium and Talaromyces in Korea, Based on 'One Fungus One Name' System)

  • 인간의 삶에 큰 영향을 끼치는 Aspergillus, Penicillium 및 그들의 완전세대 속에 속하는 곰팡이의 국내 보고 종 목록을 작성하였다. 기존의 국가 생물종 목록집에는 Aspergillus 14종, Eurotium 4종, Neosartorya 8종, Penicillium 47종, Talaromyces 5종이 보고되었다. 본 논문에서는 국제조류 균류 식물명명규약(ICN)에 따른 일균일명 체계에 따라 Eurotium과 Neosartorya은 Aspergillus 속으로 합치고 Penicillium과 Talaromyces도 새로운 속 개념에 따라 정리하였다. 또한 기존의 국가 생물종 목록에 빠져 있던 77종을 추가하여 Aspergillus, Penicillium 및 그들의 완전세대에 속하는 곰팡이의 국내 보고 종 목록을 Aspergillus 55종, Penicillium 82종, Talaromyces 18종으로 정리하였다.

Detection of Extracellular Enzyme Activity in Penicillium using Chromogenic Media

  • A total of 106 Penicillium species were tested to examine their ability of degrading cellobiose, pectin and xylan. The activity of ${\beta}$-glucosidase was generally strong in all the Penicillium species tested. P. citrinum, P. charlesii, P. manginii and P. aurantiacum showed the higher ability of producing ${\beta}$-glucosidase than other tested species. Pectinase activity was detected in 24 Penicillium species. P. paracanescens, P. sizovae, P. sartoryi, P. chrysogenum, and P. claviforme showed strong pectinase activity. In xylanase assay, 84 Penicillium species showed activity. Strong xylanase activity was detected from P. megasporum, P. sartoryi, P. chrysogenum, P. glandicola, P. discolor, and P. coprophilum. Overall, most of the Penicillium species tested showed strong ${\beta}$-glucosidase activity. The degree of pectinase and xylanase activity varied depending on Penicillium species.

Disinfection of Penicillium-infected Wheat Seed by Gaseous Chlorine Dioxide

  • Seeds of wheat (Triticum aestivum L. cv. Olgeurumil) were infected with Penicillium sp. at mean infection rate of 83%. Penicillium sp. was detected in endosperm with bran but not in embryo. Gaseous chlorine dioxide ($ClO_2$) effectively inhibited growth of Penicillium sp. at concentration of 5 to $20{\mu}g/ml$. As treatment duration was extended from 1 to 3 h, growth of Penicillium sp. was completely suppressed even at $10{\mu}g/ml$. There was no significant reduction in the incidence of Penicillium sp. at 30% relative humidity (RH). However, the incidence of Penicillium sp. was 27.7% at 50% RH, further those were 3.5% and 0.2% at 70% and 80% RH, respectively. Seed germination was not affected by $ClO_2$ treatment at all the RH conditions. Water-soaked seeds (30% seed moisture content) showed a drastic reduction in the incidence of Penicillium sp. when treated at more than $10{\mu}g/ml$ of $ClO_2$. The incidences of Penicillium sp. were 3.3, 1.8 and 1.2% at 10, 15 and $20{\mu}g/ml$, respectively. The incidence of Penicillium sp. in dry seeds with 9.7% seed moisture content did not reduce when treated with 5 and $10{\mu}g/ml$ at 50% RH although it tended to decrease as $ClO_2$ concentration increased to $20{\mu}g/ml$. Seed germination was not affected by $ClO_2$ treatment at the tested concentrations. These results indicated that gaseous $ClO_2$ was effective disinfectant to wheat seeds infected with Penicillium sp. and that the effectiveness of $ClO_2$ strongly increased when moisture content around or inside of the seed was increased.

인삼부패곰팡이의 형태 및 생리학적 특성에 관한 연구 (Studies on the morphological and physiological characteristics of isolated strains from rotting ginseng)

  • 인삼 및 인료제품을 변질시키는 곰팡이를 방제하기 위한 기초조사로서 그 변질곰광이의 생리를 구명하기 위하여 인삼 제품에서 3종의 미생물을 분리동정한 후 이들의 생육조건을 조사한 결과는 다음과 같다. 1. 인삼 및 인삼제품에서 가장 생육하는 곰팡이는 Asoergukkys sp., Penicillium sp. -A, 및 Penicillium sp. -B로 분리동정 되었다. 2. 분리균주의 생육최적온도는 Aspergillus sp.가 $40^{\circ}C$, Penicillium sp. -A와 Penicillium sp. -B가 $30^{\circ}C$이며, 생육최적 pH는 Aspergillus-sp.가 pH 5이고 Penicillium sp. -A와 Penicillium sp. -B는 pH 3 이다. 3. Aspergillus sp.는 saponin 0,7% 첨가구에서 Penicillium sp. -A 및 Penicillium sp. -B는 saponin 0.5% 첨가구에서 가장 높은 균체량을 보였으며 saponin 1% 첨가구에서 3균주 모두 생육이 저해되었다.

수확 후 저장 배에서 분리한 Penicillium속 균의 유전적 다양성 (Genetic Diversity of Penicillium isolates Isolated from Pears with Postharvest Decay in Storage)

  • 평택일원의 6개의 배 저장고로부터 300개의 부패된 배를 수집 하여 병반부위로부터 곰팡이를이분리 하여 조사한 결과 Penicillium 균이 이병배중에서 84%가 분리되었다. 84 Penicillium 균주를 대상으로 URP2R primer로 균주 간의 DNA 다양성을 조사한결과 18 type의 Penicillium 균주를 검출 할 수 있었으며 URP1F, URP2F, URP2R, URP4R의 PCR 다형성밴드를 이용한 유전적 유연관계분석으로 4 group으로 나눌 수 있었다. 18 Penicillium균주의 형태적특징을 조사한결과 3가지의 특징적인 배양적 특징을 갖는 것으로 확인 되었으며 포자형성과 포자형태에서도 3가지 type이 관찰 되었는데 SP-15균주 type, KP-1-1 균주 type, SP-17 type으로 PCR 다형성결과와 유사한 경향을 나타내었다. SP-15형은 형태적, rDNA의 ITS 염기서열 분석에서 P. expansum로 동정 되었으며 분리된 Penicillium 균주 중 70%를 차지하는 우점종인 것으로 나타났으므로 향 후 URP-PCR profile은 종간, 종내 계통 분류에 유용하게 이용 할 수 있다.

Development of Medium for Griseofulvin Production: Part II. Optimization of Medium Constituents Using Central Composite Design

  • Central composite experimental design was employed to determine the optimal concentration of medium constituents for griseofulvin production by Penicillium griseofulvum MTCC 1898 and Penicillium griseofulvum MTCC 2004. The optimal concentration of sucrose, $K_2HPO_4,\;NaNO_3,\;and\;FeSO_4{\cdot}7H_2O$ were found to be 48.08 g/1, 1.228 g/1 , 2.7 g/1, and 0.011 g/1, respectively, for Penicillium griseofulvum MTCC 1898, and for Penicillium griseofulvum MTCC 2004, 23.52 g/1, 43.67 g/1, and 0.0434 g/1 of glucose, lactose, and $MnSO_4{\cdot}H_2O$, respectively. The yield of griseofulvin under optimal composition of medium constituents increased by 1.26 and 1.38 times than prior to optimization, for Penicillium griseofulvum MTCC 1898 and Penicillium griseofulvutn MTCC 2004, respectively.

Three New Records of Penicillium Species Isolated from Insect Specimens in Korea

  • Three Penicillium species have been isolated from insect specimens in Korea; Penicillium sp., P. steckii, and P. polonicum. Penicillium sp. (KNU12-3-2) was isolated from Lixus imperessiventris, while P. polonicum (KNU12-1-8) and Penicillium steckii (KNU12-2-9) were isolated from Muljarus japonicas and Meloe proscarabaeus, respectively. The identification was based on the morphological characteristics of the fungi and in internal transcribed spacer analysis. This is the first report on the isolation of these three species of Penicillium from insects in Korea.
