  • The studies on the carotenoids in the viscera the brine shrimp(Penaeus Orientalis Kishinouye) have been carried out. The brine shrimps were caught around the western coastal area of Korea from Aug. to Oct. The individual carotenoids in the acetone extracts were separated and purified by the silica gel TLC. Two isolated carotenoids were investigated and identified by epoxide test, partition test, alkaline hydrolysis, reduction with sodium borohydride and visible absorption spectrophqtometry. As the results of the series of experiments, esterified Astaxanthin was discovered as one of main pigments of Penaeus Orientalis Kishinouye.

  • A protease without tryptic and chymotryptic activities was purified from the hepatopancreas of shrimp, Penaeus orientalis, using Q-Sepharose ionic exchange, benzamidine Sepharose-6B affinity, Mono-Q, and gel chromatography. Molecular weight (M.W.) of the protease was estimated to be 27kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDSPAGE). The amino acid composition of the protease was different from that of protease from P. japonicus or trypsin from P. orientalis. The protease was completely inhibited by benzamidine, $N\alpha-p-tosyl-L-lysine$ chloromethyl ketone (TLCK), and phenylmethylsulfonyl fluoride (PMSF) and was not affected by leupeptin, pepstatin, N-tosyl-L-phenylalanine chloromethyl ketone (TPCK), iodoacetate, and ethylenediamine tetra acetate (EDTA). The enzyme did not have any activity against Na-benzoyl-DL-arginine p-nitroanilide (BAPNA) or N-benzoyl-L-tyrosine ethyl ester (BTEE) which are specific substrates of trypsin and chymotrypsin, respectively. However, the protease showed hydrolytic activity for a carboxyl terminal of Tyr, Trp, Phe, Glu, and Cys.

  • Trypsin-like enzyme was purified from shrimp hepatopancreas through Q-Sepharose ionic exchange, benzamidine Sepharose-6B affinity, and Superdex 75 gel chromatography. Purity of trypsin-like enzyme was increased 69-fold with $44\%$ yield. The enzyme consisted of a single polypeptide chain with a molecular weight (M.W.) of 32 kDa judged by sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE). The enzyme was completely inactivated by serine enzyme inhibitors such as soybean trypsin inhibitor (SBTI), tosyl-Llysine chloromethyl ketone (TLCK), and leupeptin. However, the enzyme was not affected by tosyl-L-phenylalanine chloromethyl ketone (TPCK) which is a chymotrypsin specific inhibitor. The enzyme had no activity against benzoyl-tyrosine ethyl ester (BTEE) which is a chymotrypsin specific substrate. The enzyme showed high activity on the carboxyl terminal of Phe, Tyr. Glu, Arg, and Asp. However. no activity was detected against the carboxyl terminal of Pro, Trp, Cys, Gly, Val, and Ala.

  • Since 1993, sudden and massive death has occurred among penaeid shrimp, Penaeus orientalis cultured in Korea. We investigated the pathological characterizations on the spontaneously and experimentally infected penaeid shrimp. The major gross findings of the spontaneous cases were 2-6mm sized-white spots on the inside of the carapace and reddish discolorization. Histopathologically, massive necrosis of lymphoid organ, degeneration and necrosis of epithelia of epidermis and foregut were observed in both spontaneous and experimental cases. Amphophilic to basophilic intranuclear inclusion bodies were readly observed epithelia of epidermis, foregut and gills. Electron microscopy revealed enveloped, non-occuluded ellipsoid to rod shaped virus particles, within the nucleus, in the lymphoid organs and interstitial cells of hepatopancreas of both spontaneously and experimentally infected shrimps. The size of the virion was $375{\times}167nm$, and the nucleocapsid was $290{\times}75nm$. The causative agent causing massive death in penaeid shrimps in Korea resembles baculovirus associated with white spot syndrome (WSBV) occurred in Taiwan in virus morphology and gross and histological changes of the shrimps.

  • The isolation, purification and characterization of a carotenoprotein from the carapace of Pnaeus orientalis were investigated. The carotenoprotein was purple with broad λmax between 480, 409, 318 and 280 nm. Apparent structures were estimated by using X-ray diffractometry and scanning electron microscope, respectively. The molecular weight of the carotenoprotein complex had been determined by GPC and PAGE. The heavier complex, designated the $\alpha$-form (M.W = 170 KDa), was dissociated to a major subunit, $\beta$-form (M.W = 42 KDa). SDS-PAGE of $\alpha$-form showed apparently oligomeric pattern, and also $\beta$-form gave two polypeptides corresponding to 22 KDa and 19 KDa, respectively. The amino acid of the two proteins $({\alpha}-and\;{\beta})$-form, lipid and free fatty acid compositions were described. The prosthetic groups of the carotenoprotein were confirmed by TLC, IR, $^1H$-NMR, MS and various organic reactions as astaxanthin, astaxanthin monoester and astaxnathin diester.

  • The difference in growth rate of p. orientalis cultured on muddy bottom region and sandy bottom region in a pond filled with sea water were studied. For the investigation of the growth rate, the body length and the body weight were determined. Five individuals from each experimental region were sampled in random. The samples were collected at ten-day intervals. The survival rate, the qredator of p. orientalis, and the water conditions during the experimental period were also determined. The results in this study are summerized as follows: 1. The growth rate of p. orientalis which was cultured in the muddy bottom region was greater than that in the sandy bottom region. This is attributable in either case regardless of whether they were cultured with or without food, 2. The relationship between the culturing period (X) and the body length (Y) is : Muddy bottom, Food Y=0.58497X+25.05210 Sandy bottom, Food Y=0.51030X+26.57900 Muddy bottom, No food Y=0.22352X+32.79360 Sandy bottom, No food Y=0.l1418X+31.20740 3. The relationship between the culturing period (X) and the body weight (Y) is : Muddy bottom, Food Y=0.09062X-2.11140 Sandy bottom, Food Y=0.06992X-1.19640 Muddy bottom, No food Y=0.01615X+0.51150 Sandy bottom, No food Y=0.00495X+0.56150 4. When the predator is not present, $84\%$ of P. orientalis may survive. 5. Acanthogobius flavimanus is one of the predator of P. orientalis. 6. It is considered that the variation of water conditions of the pond, in which water had been exchanged 12 times per month, did not affect to the survival rate of P. orientalis.

  • A series of experiments since 1963 ha sbeen made on the artificial propagation of Penaeus oriental is for the purpose of improving breeding of the prawn. The results are as follows: 1. Spawning takes place in the evening, and almost completes before 10 o'clock p. m.. Number of spawns deposited by a prawn is 30,000 to 150,000. 2. Deposited spawns become nauplius stage after approximately 18 hours (about 13 hours in hatching tanks) at water temperature of $23\~25^{\circ}C$, and grow to first nauplius stage in subsequent 28 hours. 3. It takes about five days from first nauplius stage to zoea stage, and additional four or five days from first zoea stage to first mysis stage. 4. It takes approximately 2 to 3 weeks from spawning to post larva. 5. Hatching rate ranged from $1.4\%$ to $14.9\%$, with the average of $6\%$ in 1966 and $10\%$ to $35\%$ averaging $20\%$ in 1967.
