Development and sustained release effect of implantable formula of somatotropin (Somatotropin 이식 제형 개발과 지속적 방출 효과)

  • The present study was carried out to develop a sustained release implantable formula of bovine somatotropin (SRIF-BST) and to examine its sustained release effect. The SRIF-BST was produced by coating a solid pellet, which was comprised of BST and an excipient, made of a biodegradable polymer and poloxamer, which are capable of regulating the rate of BST release. The coated membrane of SRIFBST was observed with a field emission scanning electron microscope. The thickness of the coated membrane was approximately $1{\mu}m$, and the pore sizes of the coated membrane surface were below $10{\mu}m$. In dissolution test, the release duration of the SRIF-BST maintained for 10 days, whereas the release duration of the control BST formula maintained for 3 days. In weight gain assay and tibia test of hypophysectomized rats, the release duration of the SRIF-BST treated group was 12 days and the net weight gain was 53.16 g, also the tibia length and strength of the SRIF-BST treated group was increased 10.5% and 23.1% compared with those of the control group, respectively.

A Generalized Model for the Prediction of Thermally-Induced CANDU Fuel Element Bowing (CANDU 핵연료봉의 열적 휨 모형 및 예측)

  • The CANDU element bowing is attributed to actions of both the thermally induced bending moments and the bending moment due to hydraulic drag and mechanical loads, where the bowing is defined as the lateral deflection of an element from the axial centerline. This paper consider only the thermally-induced bending moments which are generated both within the sheath and the fuel and sheath by an asymmetric temperature distribution with respect to the axis of an element The generalized and explicit analytical formula for the thermally-induced bending is presented in con-sideration of 1) bending of an empty tube treated by neglecting the fuel/sheath mechanical interaction and 2) fuel/sheath interaction due to the pellet and sheath temperature variations, where in each case the temperature asymmetries in sheath are modelled to be caused by the combined effects of (i) non-uniform coolant temperature due to imperfect coolant mixing, (ii) variable sheath/coolant heat transfer coefficient, (iii) asymmetric heat generation due to neutron flux gradients across an element and so as to inclusively cover the uniform temperature distributions within the fuel and sheath with respect to the axial centerline. As the results of the sensitivity calculations of the element bowing with the variations of the parameters in the formula, it is found that the element bowing is greatly affected relatively with the variations or changes of element length, sheath inside diameter, average coolant temperature and its variation factor, pellet/sheath mechanical interaction factor, neutron flux depression factor, pellet thermal expansion coefficient, pellet/sheath heat transfer coefficient in comparison with those of other parameters such as sheath thickness, film heat transfer coefficient, sheath thermal expansion coefficient and sheath and pellet thermal conductivities.

Microstructure, Electrical Property and Nonstoichiometry of Light Enhanced Plating(LEP) Ferrite Film

  • A magnetic film was deposited on a slide glass substrate from aqueous solutions of $FeCl_2$ and $NaNO_2$ at 363 K. XRD analysis showed that the film was polycrystalline magnetite $(Fe_{3(1-{\sigma})}O_4)$ without impurity phase. The lattice constant was 0.8390 nm. Mossbauer spectrum of the film could be deconvoluted by the following parameters: isomer shifts for tetrahedral $(T_d)$ and octahedral $(O_h)$ sites are 0.28 and 0.68 mm/s, respectively, and corresponding magnetic hyperfine fields are 490 and 458 kOe, respectively. The estimated chemical formula of the film by the peak intensity of Mossbauer spectrum was $Fe_{2.95}O_4$. Low temperature transition of the magnetite (Verwey transition) was not detected in resistivity measurement of the film. Properties of the film were discussed with those of pressed pellet and single crystal of synthetic magnetites. On the surface of the film, magnetite particles of about 0.2 μm in diameter were identified by noncontact atomic force microscopy (NAFM) and magnetic force microscopy (MFM).

  • This study analyzed the fatty acid and amino acid compositions of dorsal and fin muscles collected from olive flounder cultured using a formula feed(extruded pellets, EP) and compared them to those of fish cultured with a raw fish moist pellet(MP) feed as a control. The olive flounder in this study were cultured for 10 months with either the formulated extruded pellets(FEP), commercial extruded pellets(CEP), or the MP feed, and their average weight was 1.15 kg. The proximate compositions of the dorsal muscle were not different among the groups, whereas in the fin muscles of the fish fed with the MP diet, moisture content was higher and crude lipid content was lower. The major fatty acids in the dorsal and fin muscles of the fish fed with the MP, CEP, and FEP diets were palmitic acid and oleic acid. Finally, no significant differences were observed among the diet groups in the results for amino acid content or the sensory and textural properties of the muscle.

Formulation of Omeprazole Preparations using Omeprazole-Ethylendiamine Complex (오메프라졸-에칠렌디아민 복합체를 이용한 제제설계)

  • The study was carried out to develop useful formulation for omeprazole(OMP) through OMP-ethylendiamine complex(OMPED), and the pharmaceutical properties of formula were tested to find out the difference in vivo behaviors of formulations between the free and complexed OMP. Oral and suppository dosage forms were also formulated and the dissolution profiles and pharmacokinetic parameters were measured to observe the difference in bioavailability between the free and complex form, and the correlation between dissolution rate and bioavailability was evaluated. The results are summarized as follows; In the case of formulation for oral administration, the release of OMP from enteric OMPED pellets was found satisfactory to the requirement standard and no decomposition of OMP in the pellets was found in acidic solution. Therefore the enteric OMPED pellets are anticipated to be a stable formulation. The release of OMP from OMPED tablet with chitosan as excipient and coated with cellulose acetate phthalate was found to be significantly retarded. The results of bioavailability test for OMP and OMPED tablets with lactose-excipient showed that the AUC value of OMP tablet was $116.89\;{\mu}g\;{\cdot}\;min/ml$, that of OMPED tablet was $161.10\;{\mu}g\;{\cdot}\;min/ml$, respectively. The reason why was thought that OMP decomposes more readily in body than OMPED, and the AUC of the tablet with chitosan-excipient and coated with cellulose acetate phthalate was most enhanced. In the case of bioavailability for suppositories with OMP, $OMP-{\beta}\;-cyclodextrin$ complex and OMPED, the AUC of OMPED suppository was most increased. From the above results, it is thought that the more stable and bioavailable oral or rectal dosage forms could be developed by using the OMPED as a potential OMP complex.

Antiangiogenic Effect of $AS_2O_3$ on the New Vessels Induced by bFGF in the Rat Cornea (랫드 각막에서 bFGF(basic Fibroblast Growth Factor)로 유발시킨 신생혈관에 대한 $AS_2O_3$의 혈관신생 억제 효과)

  • This study was performed to evaluate the effects of $AS_2O_3$ upon antiangiogenesis in rat cornea, to examine it\`s possible application as an anticancer drug and to provide basic data for further studies of antiangiogenetic mechanism of $AS_2O_3$ . Angiogenesis was induced by cornea micropocket assay, as previously described. Sixteen of forty-eight eyes of Sprague-Dawley rats were randomly assigned to one of three groups, namely, only a bFGF group(control group), and a group treated by $AS_2O_3$ ($AS_2O_3$ group). After pellet implantation, we measured the number of new vessels, vessel length and clock hour of neovascularization, and area of neovascularization was determined using a mathematical formula. New vessels growing began at day 3, number of vessels in $AS_2O_3$ group were significantly more less than those in control group (p<0.05). The length of vessels of $AS_2O_3$ group was significantly shorter than that of control group after day 3(p<0.05). The clock hours of all group were slowly increased at all days but $AS_2O_3$ group was inhibited more than control group. Neovascularization areas of $AS_2O_3$ group were more significantly inhibited than those of control group (p<0.05). This study showed that $AS_2O_3$ had powerful antiangiogenetic effects and it would be useful in the choice of anticancer drug.

Effect of Different Feed Additives on Growth Performance and Blood Profiles of Korean Hanwoo Calves

  • This experiment was conducted on 60 Hanwoo calves comprising five feed additive groups, with 12 calves in each group, to determine the effects of additives at pre- and post-weaning on growth performance and blood profile. The groups were control, antibiotic (Neomycin 110 ppm), illite (2%), fermented green tea probiotics (FGTP, 0.5%), and mixed additives (FGTP 0.25%, illite 1% and licorice 0.1%). The calves were offered experimental pellet feeds ad libitum and after one month were supplied with imported timothy hay. They moved freely within the group and suckled their mother' milk during the pre-weaning stage (birth to 3 months) and were separated from their dam during the post-weaning stage (4-5 months). During the pre-weaning stage, the highest average daily gain (ADG) was recorded in the antibiotic- and mixed additive-fed groups followed by FGTP, control and illite groups. In the post-weaning stage, significantly higher total weight gain and ADG were recorded in both the FGTP and mixed additive groups compared to the other groups (p<0.05). Feed efficiency of mixed additive- and illite-fed calves were almost similar with antibiotic-fed calves compared to the other two groups, but the ADG was lowest in illite-fed calves during the pre-weaning stage. In contrast, post-weaning calves fed FGTP and mixed additives showed better feed efficiency. The values of hematological indices, differential leukocyte count, blood proteins and immunoglobulin among the additive-fed calves were not significantly different (p>0.05), although hemoglobin and hematocrit values were lower in FGTP compared to control, but similar in mixed additive and antibiotic groups. These results indicate no detrimental effects of feed additives on the blood profile of calves at both pre- and post-weaning age. Serum albumin in post-weaning calves of all feed additive groups were similar but significantly lower (p<0.05) than in the control group. Post-weaning, IgM was significantly lower (p<0.05) in illite-fed calves compared to other treatment groups, but there was no difference at pre-weaning. Considering all factors, the mixed feed additives and FGTP can be the replacement feed formula for antibiotic for Hanwoo beef calf production, especially when used post- weaning.

The Studies on synthesis of $SnO_2$ doped $In_2O_3$ (ITO: Indium Tin Oxide) powder by spray pyrolysis (분무열분해법(Spray Pyrolysis)에 의한 주석산화물이 도핑된 $In_2O_3$(ITO: Indium Tin Oxide)의 분말 제조에 대한 연구)

  • The micron-sized ITO(indium tin oxide) particles were prepared by spray pyrolysis from aqueous precursor solutions for indium, and tin and organic additives solution. Organic additives solution with citric acid(CA) and ethylene glycol(EG) were added to aqueous precursor solution for Indium and Tin. The obtained ITO particles prepared by spray pyrolysis from the aqueous solution without organic additives solution had spherical and filled morphologies whereas the obtained ITO particles with organic additives solution had more hollow and porous morphologies with increasing mole of organic additives. The micron-sized ITO particle with organic additives was changed fully to nano-sized ITO particle whereas the micron-sized ITO particle without organic additives was not changed fully to nano-sized ITO particle after post-treatment at $700^{\circ}C$ for 2 hours and wet-ball milling for 24 hours. The size of primary ITO particle by Debye-Scherrer formula and surface resistance of ITO pellet were measured.
