Rapid Isolation Method for Preparation of Immuno-Stimulating Rhamnogalacturonans in Citrus Peels (귤피 유래 면역활성 람노갈락투로난류의 신속 분리방법)

  • We developed a rapid isolation method for fractionation of polysaccharides with different characteristics, and optimized it for the polysaccharide mixture from Korean citrus peels. A crude polysaccharide mixture, citrus-peel-enzyme (CPE) fraction was isolated from the citrus peels digested with pectinase and ethanol precipitation. CPE was further fractionated with serially diluted ethanol solution (ethanol:deionized water=8:1, 4:1, 3:1, 2:1, 1.5:1, 1:1, and 0.5:1) to produce seven fractions labeled from CPE8 to CPE0.5. Fraction from CPE8 to CPE1 were mostly composed of 11 different sugars, including rhamnogalacturonan (RG) I and II, and the sugars contained arabino-${\beta}$-3,6-galactan moiety. However, CPE0.5 did not contain RG-II and arabino-${\beta}$-3,6-galactans. Treatment of macrophages with fractions CPE8-CPE1 led to a dose-dependent increase in interleukin-6 production (IL-6), while treatment with CPE1 and CPE0.5 fractions resulted in decreased levels of IL-6. These results indicate that this isolation method may be useful for the rapid fractionation of bioactive RGs from polysaccharide mixtures.

Growth Performance and Nutrient Composition in the White-spotted Flower Chafer, Protaetia brevitarsis (Coleoptera: Scarabaeidae) Fed Agricultural By-product, Soybean Curd Cake (비지박 첨가 먹이원 급여에 따른 흰점박이꽃무지 유충의 생육과 영양성분 변화)

  • Insects are gaining recognition as an alternative source of protein. As a result, more and more domestic farms have begun mass rearing of edible insects. In order to produce high quality insects, studies on the development of safe and nutritious feed sources are needed. Given the cost of rearing insects, agricultural and industrial by-products are good sources for feed. The efficient utilization of these by-products can help in reducing the cost of production and preventing environmental pollution. In the current study, Citrus unshiu peel (CP), soybean curd cake (SCC), soybean oil meal (SOM), and brewers dried grain (BDG) were investigated for their effects on larval growth and development of Protaetia brevitarsis. Interestingly, the feed with 10% SCC increased larval weight by 3.5 times. For the larval period, the group of 10% SCC was significantly shorter than the control. Furthermore, minerals such as Zn, Ca, K, Mg, Na, and P were recorded to be high in 10% SCC. A total of 17 amino acids were present in 10% SCC, of which tyrosine and arginine were predominant. The heavy metal contents were very small amounts or not detected in any of the investigated groups. These findings provided a scientific basis for the utilization of soybean curd cake as a nutritional feed source to promote larval growth and produce quality insects.

Extending shelf-life of Oriental Melon(Cucumis melo L.) by Modified Atmosphere Packaging (MA 포장기법에 의한 참외의 신선도 유지)

  • Modified atmosphere packaging was applied to oriental melon(Cucumis melo L. var. makuwa Mak.) to extend its freshness during distribution. Gas composition and ethylene content in the film bags changed rapidly at the early stage of storage. Within 10 days, weight loss of the unpackaged increased upto 7.68% while those of the packaged remained less than 1.0% except LDPE film modified by addition of 5%(w/w) zeolite with $20\;{\mu}m$ thickness(20CK, 2.38%). Firmness was effectively maintained in the LDPE film modified by addition of 5%(w/w) zeolite with $40\;{\mu}m$ thickness(40CK) and the LDPE film with $40\;{\mu}m$ thickness containing ethylene absorber sachet(40LP). Peel color of the fruit changed rapidly in control and a little in 40CK. Oriental melon packages using 40CK provided better visual and sensory quality retention compared with others. Results suggested that packaging treatment such as 40CK could be used for extending freshness of oriental melon during transport period at ambient temperature.

Quality and Nutrition Labeling Study of Domestic Fruit (Plum) (국내산 청과물 (자두)의 품질 영양표시에 관한 연구)

  • In order to develop a model on the quality and nutrition label of Korean fruit plum, we first examined the quality and nutritional characteristics of Korean Plum, ChooHee. The contents of moisture, crude protein, crude fat, crude fiber, crude ash and N-free extinct were 92% 0.74% 0.64% 1.65%, 0.32% 4.29% respectively. Thus the total calorie of the sample was 32 kcal/100 g. The content of sugar, acidity and Vitamin C were $33{\pm}0.85 Bx^{\circ}$, $1.08{\pm}0.12%$ and $7.037{\pm}0.317mg%$ respectively. The principle minerals were S, K, Ca, Na, and Mg. Major free sugars were fructose, glucose and sucrose and major organic acid were succinic acid, malic acid and citric acid. The difference of contents of total phenols between peel and flesh pare were considerable. The total phenolic contents of whole fruit were $75.55{\pm}0.73mg%$. On the above results, the quality and nutrition label of Korean plum were developed in the first time. We expect that this study on labeling could play an important role to the quality control and marketing of Korean agricultural products.

Quality Changes of Over-wintering Satsuma Mandarin during Storage by Chitosan and Calcium Treatment and Storage Warehouse (키토산 및 칼슘 처리와 저장고 형태에 따른 월동 온주밀감의 저장 중 품질변화)

  • Quality changes of over-wintering satsuma mandarin (Citrus unshiu Marc. var. miyagawa) during storage by chitosan and calcium treatment and type of storage warehouse were investigated. Citrus were treated with 2000-folds diluted iminoctadime-triacetate solution and 1.5% chitosan with 0.5% CaCl$_2$ solutions and were at 30$\^{C}$ far 24 h before storage. The citrus of about 12 kg/26 L plastic container were stored at room temperature, and at 4$\^{C}$ with 87% relative humidity. Chitosan and CaCl$_2$ solution treated citrus fruits were showed lower in decay ratio than the ones without treatment. Also, these chitosan and calcium treated citrus fruits skewed less in weight loss, that seems it also has restraining effect of fruits' transpiration. Decay ratio of citrus with precise temperature and humidity control were lower than the others during storage. Weight loss, moisture content of peel and flesh were decreased slowly during storage. 0.84 ∼0.90% of acid content were decreased on 120 days' storage. Reducing sugar of citrus was decreased rapidly after 90 days, and vitamin C content were also decreased rapidly after 60 days during storage.

Physiological activities of natural color powders and their mixtures (천연소재로부터 분리한 색소분말과 혼합물의 생리활성)

  • Seven kinds (acacia, cochineal, catechu, grape peel, persimmon, gallnut and clove) of color powders obtained from natural resources and their mixtures were evaluated for their biological activities, such as antioxidant (ABTS and DPPH radicals scavenging), cholesterol absorption, and COX-2 inhibitory activities. Catechu, gallnut and clove were selected for the further studies due to its the best activities. The cholesterol absorption, COX-2 inhibitory, ${\alpha}$-amylase inhibitory and ${\alpha}$-glucosidase inhibitory activities were measured using the mixtures of catechu, gallnut and clove. The ABTS radical scavenging activity of the seven types of natural color powders were higher than the DPPH radical scavenging activity. The cholesterol absorption activity was significantly higher in cloves. The COX-2 inhibitory activity was significantly lower in acacia. The ${\alpha}$-amylase inhibitory activity was higher in catechu and gallnut, indicating that there were no significant difference between two mixtures. The ${\alpha}$-glucosidase inhibitory activity was the highest in catechu, which was higher than that of the catechu-containing mixtures. From all these results, a synergistic effect could be obtained when utilizing a mixture of powders rather than using only individual type. Since the activity of each powder was different, further studies will be required for clarifying the interactions between mixtures.

Conservation for Wooden Objects Excavated From Imdang, KyungSan - Wooden frame of Armor and Lacquer Wares - (경산 임당유적 목제품 보존 - 갑옷틀 및 칠기 -)

  • The wooden armor frame and various lacquerware artifacts excavated from the low swamp remains in Imdang, Gyeongsan have very weak material and lacquer coating; in case they are exposed in the air, they are likely to be shrunken and deformed immediately. The wooden armor frame has large size and it might be dissolved during freeze-drying. The lacquer coating of lacquerware blocks the penetration of chemicals and it is very likely to swell or peel off during the treatment. Therefore, to prevent the dissolution during vacuum freeze-drying, the wooden armor frame was replaced with t-butanol and impregnated in t-butanol solution with PEG#3, 350 43%, and lacquerware was impregnated in PEG#3, 350 40% solution at room temperature and freeze-dried. According to the analysis of the lacquer fragment from the lacquerware, the lid and the mounted cup was pasted with the mixture of lacquer and black pigment (soot) on the base layer after the first coating and pasted two or three times more with natural lacquer; the red lacquered wooden cup was pasted with the mixture of lacquer and black pigment on the base layer and pasted once on the middle layer. The top lacquer (red) was pasted with the mixture of iron oxide(Fe2O3).

Magnetoresistance Properties in Synthetic CoFe/Ru/CoFe/FeMn Spin Valves with Different Pinned Layer Thicknesses (합성형 반강자성체인 CoFe/Ru/CoFe/FeMn에서 고정층의 두께 차이에 따른 스핀 밸브 구호의 자기저항 특성)

  • Top synthetic spin valves wi th structure Ta/NiFe/CoFe/Cu/CoFe(Pl)/Ru/CoFe(P2)/FeMn/Ta on Si (100) substrate with SiO$_2$ of 1500 were prepared by dc magnetron sputtering system. We have changed only the thickness of the free layer and the thickness difference (Pl-P2) in the two ferromagnetic layers separated by Ru, and investigated the effect of magnetic film thickness on the GMR properties and the interlayer coupling field in a spin valve with a synthetic antiferromagnet. As thickness difference of pinned layer was decreased from +25 to -25 , MR ratio was decreased gradually. However, there was a dip zone indicating a big change of MR ratio around Pl = P2, which can be due to the large canting of pinned layers. The modified Neel model was suggested for the top synthetic spin valve to explain the interlayer coupling field according to the thickness change of ferromagnetic layers. The interlayer coupling field was decreased due to the magnetostatic coupling (orange peel coupling) as suggested by model. However, the interlayer coupling field was not explained at the dip zone by the modified Neel model. The deviation of modified Neel model at the dip zone could be due to the largely canting of the pinned layers as well, which depends on different thickness in synthetic antiferromagnetic structure.

According to the decrease of free layer thickness, interlayer coupling field was increased due to the magnetostatic coupling(orange peel coupling). In case of t$\_$P1/>t$\^$P2/, interlayer coupling field agreed well with the modified Neel model suggested in conventional spin valve structures by Kools et al. However, in case of t$\_$P1/>t$\^$P2/, it was found that the interlayer coupling field was not explained by the Modified Neel Model and was confirmed the necessity of further remodeling. The dependence of Cu thickness on the interlayer coupling field was investigated and 10 Oe of interlayer coupling field was obtained when the Cu thickness is 32 $\AA$.

Effects of Storage Temperature and Blanching Treatment on the Storage Stability of Citron (Citrus junos) (저장온도 및 Blanching 처리가 유자의 저장성에 미치는 영향)

  • Experiments were conducted to investigate the effects of storage temperature and blanching treatment on the storage stability in order to extend the shelf life of Citrus junos. Chilling injury of Citrus junos was not observed at the temperature range of $0^{\circ}C{\sim}5^{\circ}C$. Respiration rate of Citrus junos classified into climacteric characteristics as storage time goes by increase of respiration rate. The $Q_{10}$ value of Citrus junos during storage appeared to have a narrow range of $2.3{\sim}2.4$ The decaying rate increased drastically at $20^{\circ}C$ ranging from 71.4% to 86.5%. It was not much influenced by the pretreatments but by the storage temperature. There was no difference in weight loss between the control and blanching treatment. The weight loss of Citrus junos at $20^{\circ}C$ appeared to be high during the 12 weeks period, and consequently the overall quality of Citrus junos was rapidly decreased. The ratio of brix to acid showed a slow increase for Citrus junos stored at $3^{\circ}C.\;At\;20^{\circ}C$, however, it increased rapidly and then decreased. This phenomenon was mainly due to the relatively high increase of brix, compared to the increase in acid content. Development of browning color in Citrus junos was mainly associated with Citrus junos peel, rather than with Citrus junos pulp.
