Modulation of Quorum Sensing in Acyl-homoserine Lactone-Producing or -Degrading Tobacco Plants Leads to Alteration of Induced Systemic Resistance Elicited by the Rhizobacterium Serratia marcescens 90-166

  • Numerous root-associated bacteria (rhizobacteria) are known to elicit induced systemic resistance (ISR) in plants. Bacterial cell-density-dependent quorum sensing (QS) is thought to be important for ISR. Here, we investigated the role of QS in the ISR elicited by the rhizobacterium, Serratia marcescens strain 90-166, in tobacco. Since S. marcescens 90-166 produces at least three QS signals, QS-mediated ISR in strain 90-166 has been difficult to understand. Therefore, we investigated the ISR capacity of two transgenic tobacco (Nicotiana tabacum) plants that contained either bacterial acylhomoserine lactone-producing (AHL) or -degrading (AiiA) genes in conjunction with S. marcescens 90-166 to induce resistance against bacterial and viral pathogens. Root application of S. marcescens 90-166 increased ISR to the bacterial pathogens, Pectobacterium carotovorum subsp. carotovorum and Pseudomonas syringae pv. tabaci, in AHL plants and decreased ISR in AiiA plants. In contrast, ISR to Cucumber mosaic virus was reduced in AHL plants treated with S. marcescens 90-166 but enhanced in AiiA plants. Taken together, these data indicate that QS-dependent ISR is elicited by S. marcescens 90-166 in a pathogen-dependent manner. This study provides insight into QS-dependent ISR in tobacco elicited by S. marcescens 90-166.

Effect of Bacillus aryabhattai H26-2 and B. siamensis H30-3 on Growth Promotion and Alleviation of Heat and Drought Stresses in Chinese Cabbage

  • Plants are exposed to biotic stresses caused by pathogen attack and complex abiotic stresses including heat and drought by dynamic climate changes. To alleviate these stresses, we investigated two bacterial stains, H26-2 and H30-3 in two cultivars ('Ryeokkwang' and 'Buram-3-ho') of Chinese cabbage in plastic pots in a greenhouse. We evaluated effects of bacterial strains on plant growth-promotion and mitigation of heat and drought stresses; the role of exopolysaccharides as one of bacterial determinants on alleviating stresses; biocontrol activity against soft rot caused by Pectobacterium carotovorum subsp. carotovorum PCC21. Strains H26-2 and H30-3 significantly increased fresh weights compared to a $MgSO_4$ solution; reduced leaf wilting and promoted recovery after re-watering under heat and drought stresses. Chinese cabbages treated with H26-2 and H30-3 increased leaf abscisic acid (ABA) content and reduced stomatal opening after stresses treatments, in addition, these strains stably colonized and maintained their populations in rhizosphere during heat and drought stresses. As well as tested bacterial cells, exopolysaccharides (EPS) of H30-3 could be one of bacterial determinants for alleviation of tested stresses in Chinese cabbages, however, the effects were different to cultivars of Chinese cabbages. In addition to bacterial activity to abiotic stresses, H30-3 could suppress incidence (%) of soft rot in 'Buram-3-ho'. The tested strains were identified as Bacillus aryabhattai H26-2 and B. siamensis H30-3 based on 16S rRNA gene sequence analysis. Taken together, H26-2 and H30-3 could be candidates for both plant growth promotion and mitigation of heat and drought stresses in Chinese cabbage.

무 품종의 세균성 무름병 저항성 생물검정법 평가 (Evaluation of Bioassay Methods to Assess Bacterial Soft Rot Resistance in Radish Cultivars)

  • 세균성 무름병 균[Pectobacterium carotovorum subsp. carotovorum (Pcc)]에 의해서 일어나는 무름병은 아시아 국가에서 재배되는 무에 있어서 심각한 질병 중의 하나로 알려져 있다. 이번 연구의 목적은 상업적으로 시판되는 무 품종의 세균성 무름병의 저항성에 대한 효율적인 생물검정법을 확립하고자 하였다. 첫째, 무 품종에 대해 세균성 무름병의 효율적인 생물검정 방법을 조사하였다. 6개의 무 품종을 다양한 조건[두 가지 온도(25℃와 30℃), 3가지 접종방법(관주, 분무, 침지), 두 발생단계(2와 4잎 단계)]으로 조사하였다. 연구 결과는 무름병균 1×106 cfu/ml 농도를 분무한 4잎 단계와 30℃에서 배양한 생물검정 방법이 무 품종에 대해 가장 효율적인 방법임을 알 수 있었다. 둘째, 41개의 무 품종에 5가지 세균(KACC 10225, KACC 10343, KACC 10421, KACC 10458, KACC 13953)을 접종하여 저항성을 조사하였다. KACC 10421가 세균성 무름병의 감수성 및 저항성 질병 정도를 가장 잘 나타냈다. 41개의 무 품종 중 13개는 무름병 균에 대해 중도 저항성을 나타냈고 28개는 감수성을 나타냈다. 이 연구에서 중도저항성 무 품종은 세균성 무름병 저항성 육종을 위한 저항성 자원으로 활용 가능하고 육종가, 농민, 연구자, 최종 소비자에 의해서 다양한 목적을 위해 사용 가능할 것으로 사료된다.

칼라의 무름병 저항성 품종 선발을 위한 검정 방법 개발 (A New Screening Method for the Selection of Calla Lily Zantedeschia aethiopica Cultivars Resistant to Calla Lily Soft Rot)

  • 본 연구는 P. carotovorum subsp. carotovorum에 의해 발생하는 칼라의 무름병 저항성 품종 선발을 위한 효율적인 검정 방법을 구명하기 위해 수행되었다. Z. aethiopica 4품종에 대하여 $1{\times}10^7cfu/mL$, $1{\times}10^8cfu/mL$, $1{\times}10^9cfu/mL$ 농도의 EccNHRI-21 균주 현탁액을 잎과 엽병에 상처접종하고, 접종 후 4, 18, 26시간 후에 무름병 발생 정도를 조사하였다. 그 결과, $1{\times}10^9cfu/mL$ 농도의 균주 현탁액을 잎에 접종한 후 18시간 이후에 저항성 정도를 평가하는 것이 칼라 무름병 저항성 검정 및 품종 선발을 위해 가장 적합한 것으로 나타났다. 상처접종법을 이용하여 Z. aethiopica 재배품종 10종과 야생종 4종 등 총 14종에 대하여 무름병 저항성 정도를 평가한 결과, 재배품종의 경우 'Crowbrough'와 'White Cutie'가 가장 강한 저항성을 보였으며, 'Mont Blanc'과 'Silky White'도 저항성 품종으로 선발되었다. 또한 'Wedding March'와 'Kiwi blush'는 중도저항성 품종으로 평가되었고, 'Childsiana' 등 4품종은 감수성 품종으로 평가되었다. 야생종의 경우 'Z. aethiopica wildtype 11'이 저항성을 보이는 등 4종 모두 중도저항성 이상의 무름병 저항성을 가지는 것으로 평가되었다. 지금까지 경종적 방제 및 생물학적 방제, 화학적 방제 등 여러 가지 방제 방법이 시도되었으나, 무름병 발생 피해를 최소화하기 위해서는 내병성 품종을 재배하는 것이 가장 효율적이다. 따라서 상처접종법을 이용한 무름병 저항성 검정 기술이 칼라의 무름병 저항성 품종 개발에 유용하게 쓰일 것으로 기대된다.

Enhanced bacterial resistance in transgenic tobacco expressing a BrRZFP1 encoding a C3HC4-type RING zinc finger protein from Brassica rapa

  • C3HC4-type RING zinc finger proteins essential in the regulation of plant processes, including responses to abiotic stresses. We previously isolated and examined the C3HC4-type RING zinc finger protein (BrRZFP1) from Brassica rapa under abiotic stresses. To elucidate the role of the BrRZFP1 transcription factor in gene regulation, we transformed tobacco plants with the BrRZFP1 gene. Plants were regenerated from 82 independently transformed callus lines of tobacco and analysed for transgene expression. Transgene integration and expression was confirmed by Southern and RT-PCR analyses, respectively. T2 plants displayed more tolerance to the bacterial pathogens Pectobacterium carotovorum and Ralstonia solanacearum, and the tolerance levels were correlated with BrRZFP1 expression levels. These results suggest that the transcription factor BrRZFP1 is an important determinant of stress response in plants and its overexpression in plants could increase biotic stress resistance.

Proteomic Analysis of a Global Regulator GacS Sensor Kinase in the Rhizobacterium, Pseudomonas chlororaphis O6

  • The GacS/GacA system in the root colonizer Pseudomonas chlororaphis O6 is a key regulator of many traits relevant to the biocontrol function of this bacterium. Proteomic analysis revealed 12 proteins were down-regulated in a gacS mutant of P. chlororaphis O6. These GacS-regulated proteins functioned in combating oxidative stress, cell signaling, biosynthesis of secondary metabolism, and secretion. The extent of regulation was shown by real-time RT-PCR to vary between the genes. Mutants of P. chlororaphis O6 were generated in two GacS-regulated genes, trpE, encoding a protein involved in tryptophan synthesis, and prnA, required for conversion of tryptophan to the antimicrobial compound, pyrrolitrin. Failure of the trpE mutant to induce systemic resistance in tobacco against a foliar pathogen causing soft rot, Pectobacterium carotovorum SCCI, correlated with reduced colonization of root surfaces implying an inadequate supply of tryptophan to support growth. Although colonization was not affected by mutation in the prnA gene, induction of systemic resistance was reduced, suggesting that pyrrolnitrin was an activator of plant resistance as well as an antifungal agent. Study of mutants in the other GacS-regulated proteins will indicate further the features required for biocontrol-activity in this rhizobacterium.

Potato Soft Rot Caused by Psychrotolerant Pseudomonas sp. from Subarctic Tundra Soil

  • Agricultural activities and the number of farms in the subarctic regions have been increasing annually after the coronavirus disease 2019 pandemic to achieve food self-sufficiency. Potatoes are vulnerable to soft rot bacteria at all stages of production, storage, and transportation. A novel bacterium, Pseudomonas sp. N3-W, isolated from Alaska tundra soil, grows at 5-25℃ and produces extracellular protease(s). N3-W caused necrotic spots (hypersensitivity) in hot pepper leaves and soft rot disease (pathogenicity) in potato tubers. The psychrotolerant N3-W caused significant soft rot symptoms on potatoes at a broad temperature range (5℃, 15℃, and 25℃). In contrast, mesophilic Pectobacterium carotovorum KACC 16999 induced severe rotting symptoms in potatoes at their optimal growth temperature of 15℃ and 25℃. However, it barely produced symptoms at 5℃, which is the appropriate storage and transportation temperature for potatoes. The results of pathogenicity testing imply that psychrotolerant soft rot pathogens from polar regions may cause severe soft rot not only during the crop growing season but also during storage and transportation. Our study indicates the possibility of new plant pathogen emergence and transmission due to the expansion of crop cultivation areas caused by permafrost thawing in response to recent polar warming.

탄닌산을 함유한 친환경농자재(공시번호 2-4-064)의 식물병원세균 기내 억제효과 (In Vitro Screening of Tannic Acid-based Eco-friendly Farming Material (notice no. 2-4-064) against Plant Pathogenic Bacteria)

  • 지금까지 주요 식물병원세균에 대한 방제대책은 화학적 방제방법에 의존하여왔다. 그러나 항생제와 같은 화학적 방제방법은 저항성 균주의 출현으로 인해 효율성이 감소함에 따라 대체방법의 개발이 요구되어지고 있는 실정이다. 따라서 본 연구에서는 주요 7종(그램음성 - 무름병, 감귤 궤양병, 풋마름병, 가지검은마름병 및 과실썩음병; 그램 양성 - 토마토 궤양병 및 더뎅이병)의 식물병원세균에 대한 탄닌산 함유 친환경농자재(공시번호 2-4-064)의 억제효과를 고체배지 확산법과 액체배지 배양법으로 검정하였다. 고체배지 확산법에서는 가지검은마름병원세균을 제외한 6종의 병원세균에 대한 억제효과를 나타내었으며, 액체배지 검정에서는 액체배지법으로 실험가능한 6종의 병원세균의 생장을 억제하였다. 전자현미경을 통한 표면변화를 관찰한 결과, 모든 병원세균의 표면구조의 변화를 발생하여 생장저해 또는 세포사멸을 유도하는 현상이 관찰되었다.

절화수량이 우수한 무름병 저항성 조생 백색칼라 'White Cutie' 육성 (Breeding of a Multi-flowering and Early-flowering White Calla Lily Cultivar 'White Cutie' Resistant to Soft Rot Disease)

  • 국립원예특작과학원에서는 무름병에는 감수성이지만 다화성인 'Childsiana' 품종과 무름병에 강하지만 절화수량이 적은 'Wedding March' 품종을 각각 모본과 부본으로 인공 교배하여 얻은 실생계통으로부터 2005년부터 2011년까지 구근양성 및 생육개화특성검정, 무름병저항성 검정, 소비자 기호도 평가를 수행하여 2011년 무름병 저항성 백색칼라 신품종 'White Cutie'를 육성하였다. 백색(RHS W155C) 화색의 'White Cutie'는 화포의 길이와 폭이 각각 8.6cm, 8.7cm인 중형 품종이다. 개화소요일수가 85.6일로 짧은 조기개화성 품종으로, 연간 채화수량도 주당 6.2개 정도로 많은 다화성 품종이다. 무름병 저항성 품종으로 구근증식량(13.4개/주) 매우 우수하고 소비자기호도가 높다.

Biological Control of Soil-borne Diseases with Antagonistic Bacteria

  • Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.
