Alcohol fermentation conditions were investigated using 'Cheondobansi' astringent persimmon (Diospyros kaki T.) for the study of persimmon wine and distilled liquor, The optimal yeast strain for 'Cheongdobansi' astringent persimmon alcohol fermentation was Saccharomyces kluyveri DJ97, which showed 10.8% of alcohol concentration, 96.25% of alcohol yield, and 935 ppm of methanol. The initial conditions of $22^{\circ}Brix$ and 120%(v/w) water addition resulted in the highest alcohol concentration of 10.7%. The alcohol concentration was higher in pectinase non-treated samples than in pectinase-teated samples. Lower concentrations of acetaldehyde and n-propanol were measured for the pectinase-treated sample than for the non-treatment samples. However, the methanol concentration of the pectinase-teated sample was higher than that of the pectinase non-treatment sample.
The study was aimed to saccharify callulosic waste by pectinase produced from strain KL34 isolated from soil. The enzyme activity in the culture using 1%(w/v) fruit waste as carbon source reached to 3.8 U/ml. In the enzymatic hydrolysis of cellulosic waste, we obtained 9.5g/L reducing sugar in the condition of supernatant containing 5 U/ml enzyme and 10%(w/v) apple rind as substrate. Additionally, in enzymatic hydrolysis of food waste using pectinase from KL34, reducing sugar of 12.7g/L was obtained, indicating enhancement of 1.6 fold compared with that of only cellulase employment.
In this study, we tried to establish the best method for fresh blueberry beverage production using enzyme treatment as well as low temperature extraction. During extraction of physiologically functional materials, we used low temperature to prevent nutritional loss by heat. In addition, we investigated optimal blueberry extraction conditions using various enzyme treatments (cellulase, pectinase, cellulase:pectinase (1:1) mixture) to increase extraction efficiency and reduce turbidity. A variety and ratio of enzymes, extraction temperature, extraction time, and shaking speed were considered for the best extraction efficiency rate. We observed high extraction efficiency rates of 85.72-86.55% and 87.06-87.93%, respectively, upon cellulase or pectinase treatment. In addition, a mixture of cellulase:pectinase (1:1) showed an extraction efficiency rate of 86.84-88.14%. The best extraction efficiency rate was observed when crude blueberry was treated at $45^{\circ}C$ (87.91%), for 3 h (87.88%), in a 90 rpm shaker (89.19%). Sugar content and acidity of blueberry extract were not affected by the various treatments. However, total phenolic compounds were detected upon pectinase treatment (18.62 mg/g). Only fructose and glucose as free sugars were found in all samples regardless of treatments and extraction conditions.

This study examined the extraction yields, total phenolic compounds content and the antioxidant activities on V79-4 cells of white ginseng extracts prepared by enzyme treatment. Yields of crude extract were 29.5-76%, and total phenolic compounds content showed 0.45-2.2% according to enzyme treatments. Pectinase treatment group showed the highest values of extraction yields and total phenolic compounds content. Pectinase and a-amylase treatment groups protected V79-4 cell viability(above 50%) against $H_2O_2$-induced oxidative damage. In the result of antioxidant enzyme activity evaluation in cells, enzyme treatments did not show the significant difference of SOD activity (p>0.05). However, pectinase treatment group exhibited increased CAT and GPx activities (p>0.05). Also, pectinase and protease treatment group inhibited MDA formation (>50%) in the lipid peroxidation protection experiment.
The thermostable pectinase gene TmPec isolated from Thermotoga maritima was introduced into the NdeI site of pRSET-B vector and expressed in its intact form in Escherichia coli BL21. The overexpressed intact form of pectinase (TmPecN protein) was partially purified by heat-denaturation procedure. TmPecN showed the highest activity between 85 and $95^{\circ}C$, and at approximately pH 6.5. Enzyme activity was stably maintained at temperatures below $85^{\circ}C$. In the presence of $Ca^{2+}$, pectinase activity of TmPecN increased to 128.4% of normal level. In contrast, $Ba^{2+}$, $Zn^{2+}$, and $Mn^{2+}$ strongly inhibited TmPecN activity. We conclude that the biochemical properties of the intact form of TmPecN are comparable to those of the recombinant protein TmPec reported previously.

Use of a pectinase during preparing handmade papers from bast fiber of paper mulberry(Broussonetia kazinoki Sieb.) was investigated in order to decrease cooking chemicals and environmental pollution. For this purpose, four kinds of commercial pectinases, Rapidase LIQ(RLP), Rapidase Press(RP), Rapidase C80L Max(RCM) and Pectinase SS Kyowa(PSK) were used. And the durability of handmade papers before and after pectinase treatment was determined. RP and PSK had higher pectinase activity ad lower cellulase activity. The bast fiber was not defibered when pectinase was used. In order to increase the efficiency of enzymes, the bast fiber were treated ammonium oxalate(AO) or $K_2CO_3$under mild conditions. The AO pretreatment with those produced by $K_2CO_3$. The RP treated pulps after mild $K_2CO_3$cooking of the bast fiber were defibrated more easily than untreated pulp. The handmade paper prepared with the RP treated pulps after mild $K_2CO_3$cooking has good strength properties such as breaking length and folding endurance. Also, it has higher durability on heat aging, though its brightness was slightly lower than that of untreated paper.
Suspension cells of Angelica gigas Nakai was cultivated to produce extracellular polysaccharide(ECP) as immunostimulating agents. In this study, effects of pectinolytic enzymes such as pectinase. Pectinex on cell growth and ECPs production were investigated in suspension cultures. Optimum concentration of pectinase for ECP production ,vas found to be 0.1 U/L. at which ECP production was increased 1.39 fold (0.97 g/L) compared to control culture.

Treatment with the enzyme pectinase has been reported to lower the cationic demand of thermomechanical pulp(TMP) bleached with alkaline peroxide in the laboratory. We have extended this discovery to bleached TMP produced industrially, and shown that commercial enzyme preparations can treat pulp within 15 minutes at the at the temperature and pH values prevalent in paper mills. About half of the cationic demand in the bleached pulp can be destroyed by pectinase. Dynamic drainage jar experiments show that the enzyme treatment improves the effectiveness of several cationic polymers to increase retention in the absence of retention aids or with non-ionic polymers, and does not damage the strength properties of the pulp. Pectinase could be easily incorporated into paper machine stock preparation systems to lower the charges of cationic retention aids needed in furnishes containing peroxide-bleached mechanical pulp.
A total of 106 Penicillium species were tested to examine their ability of degrading cellobiose, pectin and xylan. The activity of ${\beta}$-glucosidase was generally strong in all the Penicillium species tested. P. citrinum, P. charlesii, P. manginii and P. aurantiacum showed the higher ability of producing ${\beta}$-glucosidase than other tested species. Pectinase activity was detected in 24 Penicillium species. P. paracanescens, P. sizovae, P. sartoryi, P. chrysogenum, and P. claviforme showed strong pectinase activity. In xylanase assay, 84 Penicillium species showed activity. Strong xylanase activity was detected from P. megasporum, P. sartoryi, P. chrysogenum, P. glandicola, P. discolor, and P. coprophilum. Overall, most of the Penicillium species tested showed strong ${\beta}$-glucosidase activity. The degree of pectinase and xylanase activity varied depending on Penicillium species.

This study examined the effect of surfactant pretreatment on the pectinase-treated cotton/chitosan blends by weight loss and properties such as water absorbency, dyeability, tensile strength, pilling property, and surface morphology. The weight loss of cotton/chitosan blends was 1.5% by the surfactant pretreatment/pectinase treatment. The water absorbency and dyeability of samples showed a significant improvement by the surfactant pretreatment/pectinase treatment. The tensile strength and pilling property of treated fabrics showed no change. The water absorbency and dyeability of pectinase treated samples improved with the pretreatment of the surfactant without damaging the fibers.
