The study investigated the effect of carrier composition (ionic strength and pH) on the retention of various proteins in flow field-flow fractionation (Flow FFF) as well as the conformational change of Bovine Serum Albumin (BSA) with urea concentration, storage time and temperature. The study found that the retention of protein in Flow FFF increased with the ionic strength of the carrier liquid. Most proteins were well solubilized at pH = 7-8. The hydrodynamic diameters obtained from Flow FFF retention data agree well with theoretical values. The retention increased and the peak shape became distorted at extreme pH conditions of the carrier solution. The selected carrier composition for comparison between the literature value of proteins was 0.05 M tris buffer solution with a pH of 8. Storing BSA at 4 ±2℃ over a period of three months resulted in slow dimerization. Also, in case of the storage of BSA at 37 ±5℃ for one week, the retention of both BSA monomer and dimer increased with the urea concentration. Finally, the structural composition of specific enzymes: malonyl-CoA decarboxylase (MCDC) and malonyl-CoA synthesis (MCS) was determined by using Flow FFF at specific carrier solutions. The molecular weight of the natural MCDC was determined to be 208 kDa, which means it is a homotetramer, while that of the MCS was determined to be 47 kDa, which means it is a monomer.
During a loss-of-coolant accident (LOCA) in the pressurized water reactor (PWR), there is a possibility that high temperature and internal pressure of the fuel rods lead to ballooning of the cladding, which causes a partial blockage of flow area in a subchannel. Such flow blockage would influence the core coolant flow, thus affecting the core heat transfer during a reflooding phase and subsequent severe accident. However, most of the system analysis codes simulate the accident process based on the assumed channel blockage ratio, resulting in the fact that the simulation results are not consistent with the actual situation. This paper integrates the developed core Fuel Rod Thermal-Mechanical Behavior analysis (FRTMB) module into the self-developed severe accident analysis code ISAA. At the same time, the existing flow blockage model is improved to make it possible to simulate the change of flow distribution due to fuel rod deformation. Finally, the ISAA-FRTMB is used to simulate the QUENCH-LOCA-0 experiment to verify the correctness and effectiveness of the improved flow blockage model, and then the effect of clad ballooning on core heat transfer and subsequent parts of core degradation is analyzed.
The determination of feasible design flood is the most important to control flood damage in river management. Model parameters should be calibrated using observed discharge but due to deficiency of observed data the parameters have been adopted by engineer's empirical sense. Storage coefficient in the Clark unit hydrograph method mainly affects magnitude of peak flood. This study is to estimate the storage coefficients based on the observed rainfall-runoff events at the four stage stations in the Hantan river basin. Model calibration is the process of adjusting model parameter values until model results match historical data. An objective function which is the percent difference between the observed and computed peak flows is available for measuring the goodness-of-fit between computed and observed hydrographs. By sensitivity analysis for the storage coefficient, it has been shown that the storage coefficients affect the peak flows. The Clark parameters adopted in the River Rectification Basic Plan have been estimated through an iterative process designed to produce a hydrograph with the peak flow.
This paper considers internal pressure fluctuations for a range of building volumes and dominant wall opening areas. The study recognizes that the air flow in and out of the dominant opening in the envelope generates Helmholtz resonance, which can amplify the internal pressure fluctuations compared to the external pressure, at the opening. Numerical methods were used to estimate fluctuating standard deviation and peak (i.e. design) internal pressures from full-scale measured external pressures. The ratios of standard deviation and peak internal pressures to the external pressures at a dominant windward wall opening of area, AW are presented in terms of the non-dimensional opening size to volume parameter, $S^*=(a_s/\bar{U}_h)^2(A_W^{3/2}/V_{Ie})$ where $a_s$ is the speed of sound, $\bar{U}_h$ is the mean wind speed at the top of the building and $V_{Ie}$ is the effective internal volume. The standard deviation of internal pressure exceeds the external pressures at the opening, for $S^*$ greater than about 0.75, showing increasing amplification with increasing $S^*$. The peak internal pressure can be expected to exceed the peak external pressure at the opening by 10% to 50%, for $S^*$ greater than about 5. A dominant leeward wall opening also produces similar fluctuating internal pressure characteristics.
Rheological Properties of polypropylenes having different molecular structures (linear polypropylene (PPL) and branched one (PPB)) were studied. Both the extensional flow and oscillatory shear flow properties were checked. Especially, the melt strength of polypropylenes having various shear history were investigated by using in-house-made Rheometer (called SMER). Compared to linear polypropylene, the branched polypropylene shows enhanced melt strength during extensional flow due to the retarded relaxation of molecules. When the slope of melt tension was plotted against take up speed of melt strand, the characteristic peak was observed in case of branched polypropylene, while the linear polypropylene shows only monotonously decreasing pattern. This entanglement was partially disrupted by physical forces such as shear during melt extrusion. However, the melt strength of PPB after multiple extrusion is still higher than PPL, implying the loss of elasticity during multiple extrusion is not so comprehensive. On dynamic experiments, PPB shows typical shear thinning behavior and the tangent delta of PPB is lower than PPL, reflecting high elasticity of PPB.
A numerical analysis based on the three-dimensional Reynolds-averaged Navier-Stokes equation has been conducted to investigate the flow within a NASA rotor 67 transonic fan. General coordinate transformations are used to represent the complex blade geometry and an H-type grid is used. The governing equations are solved using implicit LU-SGS scheme for the time-marching integration and a standard ${\kappa}-{\varepsilon}$ model is used with wall functions for the turbulence modeling. The computations are compared with the experimental data and a detailed study of the flow structures near peak efficiency and near stall is presented. The calculated overall aerodynamic efficiency and three-dimensional shock system agree well with the laser anemometer data.
In this study, pyrocatechol violet (Pcv) is proposed for the first time as an efficient electrocatalyst for oxidation of sulfide and flow injection analysis (FIA) of sulfide. A graphite pencil electrode (GPE) was modified with Pcv via immersion of the GPE into 0.01 M Pcv solution for 15 min. Cyclic voltammograms (CVs) demonstrated that Pcv/GPE exhibits a good electrocatalytic performance due to shift in the potential from +400 at bare GPE to +70 mV at Pcv/GPE and obtaining an enhancement in the peak current compared with the bare GPE. A linear range between 0.25 and 250 μM sulfide with a detection limit of 0.07 μM was obtained from the recorded current-time curves in Flow Injection Analysis (FIA) of sulfide. Sulfide in water samples was also successfully determined using the proposed FI amperometric methods.
This work developed improved slip factor model and correction method to predict flow through impeller in forward-curved centrifugal fan. Both steady and unsteady three-dimensional CFD analyses were performed to validate the slip factor model and the correction method. The results show that the improved slip factor model presented in this paper could provide more accurate predictions for forward-curved centrifugal impeller than the other slip factor models since the present model takes into account the effect of blade curvature. The correction method is provided to predict mass-averaged absolute circumferential velocity at the exit of impeller by taking account of blockage effects induced by the large-scale backflow near the front plate and flow separation within blade passage. The comparison with CFD results also shows that the improved slip factor model coupled with the present correction method provides accurate predictions for mass-averaged absolute circumferential velocity at the exit of impeller near and above the flow rate of peak total pressure coefficient.
The flow characteristics on the centerline in case of free jet, sudden expansion jet and impinging jet have been investigated. Centerline flow behaviors and similaritis with mean velocities, turbulent intensities, shear stresses, isotropic structures and turbulent kinetic energies on the streamwise direction were looked into and compared with three jets, The results show that mean velocities have represented potential core and decayed with similar gradients. The turbulent intensities and shear stresses were presented peak values in the self-preserving region, and then they were in decay. Aeolotropy in the initial region were possible returned to isotropy patterns with asymptotic approach in the downstream region. It has been found that the turbulent kinetic energies for the three cases of jet existed in the similarity and they coincided with Gaussian profile.
We have investigated the blood sucking phenomena of a female mosquito. The main objective of this study is to understand the mosquito's blood sucking mechanism and eventually to develop a bio-mimic technology that can be used to resolve the problem encountered in the transport of infinitesimal biological fluids in various bio-chips and microchips. At first, the consecutive velocity fields of blood-sucking flow in a proboscis were measured using a micro-particle image velocimetry (PIV) system employed with a high-speed camera. The velocity signals of the blood-sucking flow in the proboscis represent a periodic pulsatile flow pattern and spectral analysis on the velocity waveform shows a clear peak at 6.1 Hz.
