Manufacture of Hydrogen and C2+ Chemicals from Methane using Microwave Plasma and Catalyst (마이크로웨이브 플라즈마와 촉매를 이용한 메탄으로부터 수소 밀 C2+ 화학원료 제조에 환한 연구)

  • The microwave plasma and catalytic reaction have been employed to investigate the activation of methane to hydrogen and higher hydrocarbons at low gas temperature. The catalytic activity of Fe, Ni, Pt Pd metal catalysts were also studied in this reaction system. With increasing plasma power at a $CH_{4}$ flow rate of 20 ml/min, C2+ products increased from 29 to $42\%$, whereas hydrogen from 60 to $65\%$. When catalysts were loaded below the plasma region, the selectivitity of ethylene md acetylene increased but the yield of C2+ products remained constant. The usage of ECR electric fie3d and Pd-Ni bimetal catalyst produced a minimum C2+ yield of $64\%$.

A Study on Contamination of Hydrogen Permeable Pd- based Membranes (Pd 계열 수소 분리막의 오염에 관한 연구)

  • $H_2$ permeation flux though a $100{\mu}m-thick$ Pd-Ru (6wt%) membrane was measured at various temperatures and pressures. The permeation flux followed the Sievert's law and thus the rate-limiting step of the hydrogen permeation was the bulk atomic diffusion step. The activation energy of the permeation flux was obtained at 17.9 kJ/mol and this value is consistent with those published previously. While no degradation of the permeation flux wasfound in the membrane exposed to the $O_2$ and $CO_2$ environments for 100 hours, the membrane exposed to $N_2$ environment for 100 hours showed the degradation in the $H_2$ permeation flux. The $H_2$ permeation was decreased as the exposure temperature to $N_2$, environment was increased. The $H_2$ permeation flux was fully recovered after the membrane was kept in the $H_2$ environment for certain time. The permeation flux degradation might be caused by the formation of metal nitride on the membrane surface.

Functional role of Forskolin and PD166285 in the development of denuded mouse oocytes

  • Objective: cAMP and mature promoting factor (MPF) play critical roles during the maturation of mammalian oocytes. The aim of this study was to produce the offspring from denuded oocytes (DOs) in mice by regulating cAMP and MPF. Methods: In this study, we used DOs at the germinal vesicle (GV) stage in mice and regulated levels of cAMP and MPF in DOs by adding Forskolin and PD166285 during in vitro maturation without follicle stimulating hormone and luteinizing hormone, respectively. Results: Combined use of $50{\mu}M$ Forskolin for 3 h and $2.5{\mu}M$ PD166285 for additional 21 h enhanced the developmental competence of DOs, maturation rate of DOs was $76.71%{\pm}4.11%$, blastocyst rate was $18.33%{\pm}4.44%$ after parthenogenetic activation (PA). The DOs could successfully be fertilized with sperm in vitro, cleavage rate was $17.02%{\pm}5.82%$ and blastocyst rate was $5.65%{\pm}3.10%$. Besides, 2-cell in vitro fertilization embryos from DOs produced 4 normal live offspring (4/34). Conclusion: The results confirmed that the combination of Forskolin and PD166285 can induce DOs to complete meiosis process and produce normal offspring.

Ceramic barrier coated Pd hydrogen membrane on a porous nickel support (수소 분리용 팔라듐계 분리막의 세라믹 코팅 영향)

  • A highly performed Pd-based hydrogen membrane has prepared successfully on a modified porous nickel support. The porous nickel support modified by impregnation method of $Al(NO_3)_3{\cdot}9H_2O$ (Aldrich Co.) over the nickel powder showed a strong resistance to hydrogen embrittlement and thermal stability. Plasma surface modification treatment was introduced as a pre-treatment process instead of conventional HCl wet activation. Ceramic barrier was coated on the external surface of the prepared nickel supports to prevent intermetallic diffusion and to enhance the affinity between the support and membrane. Palladium and copper were deposited at thicknesses of $4\mu}m$ and $0.5{\mu}m$, respectively, on a barrier-coated support by DC sputtering process. The permeation measurement was performed in pure hydrogen at $400^{\circ}C$. The single gas permeation of our membrane was two times higher than that of the previous membrane which do not have ceramic barrier.

[ ${\alpha}$ ]Synuclein Induces Unfolded Protein Response Via Distinct Signaling Pathway Independent of ER-membrane Kinases

  • Parkinson's disease (PD) is a neurodegenerative disease caused by selective degeneration of dopaminergic neurons in the substantia nigra. Mutations in ${\alpha}$-synuclein have been causally linked to the pathogenesis of hereditary PD. In addition, it is a major component of Lewy body found in the brains of sporadic cases as well. In the present study, we examined whether overexpression of wild type or PD-related mutant ${\alpha}$-synuclein induces unfolded protein response (UPR) and triggers the known signaling pathway of the resulting endoplasmic reticulum (ER) stress in SH-SY5Y cells. Overexpression of wild type, A30P, and A53T ${\alpha}$-synuclein all induced XBP-1 mRNA splicing, one of the late stage UPR events. However, activation of ER membrane kinases and upregulation of ER or cytoplsmic chaperones were not detected when ${\alpha}$-synuclein was overexpressed. However, basal level of cytoplsmic calcium was elevated in ${\alpha}$-synuclein-expressing cells. Our observation suggests that overexpression of ${\alpha}$-synuclein induces UPR independent of the known ER membrane kinase-mediated signaling pathway and induces ER stress by disturbing calcium homeostasis.

A Study of Carbon Monoxide Oxidation on Pt & Pt-Pd Catalysts (귀금속촉매 (Pt, Pd)를 이용한 일산화탄소 산화반응에 관한 연구)

  • This study is concerned with the oxidation of carbon monoxide on platinum and platinum-palladium catalysts. Catalysts were made by the impregnation method and flow reactor was used in the catalytic reaction. As for the mixed gases, carbon monoxide concentration varied from 1 to 4% and that of oxygen from 1 to 4%. $N_2$ was used as carrier gas and GHSV varied from 24, 000 $h^{-1} to 60, h^{-1}$. The temperature range was from 200 to $600^\circ$C. It was also taken into consideration that the heat and mass transfer resistance of our catalysts was negligible in the study. Experimental results showed that platinum-palladium catalyst was about 1.5-3.9% superior to platinum catalyst in conversion yield. When we used platinum-palladium catalyst, we observed that carbon monoxide oxidation was found to be 1 st order with respect to carbon monoxide concentration. Activation energy of the catalyst was 23.5 kcal/mol.

Sphigosine-1-Phosphate-Induced ERK Activation Protects Human Melanocytes from UVB-Induced Apoptosis

  • Ultraviolet B (UVB) is known to induce apoptosis in human melanocytes. Here we show the cytoprotective effect of sphingosine-1-phosphate (S1P) against UVB-induced apoptosis. We also show that UVB-induced apoptosis of melanocytes is mediated by caspase-3 activation and poly(ADP-ribose) polymerase (PARP) cleavage, and that S1P prevents apoptosis by inhibiting this apoptotic pathway. We further investigated three major mitogen-activated protein (MAP) kinases after UVB irradiation. UVB gradually activated c-Jun N-terminal kinase (JNK) and p38 MAP kinase, while extracellular signal-regulated protein kinase (ERK) was inactivated transiently. Blocking of the p38 MAP kinase pathway using SB203580 promoted cell survival and inhibited the activation of caspase-3 and PARP cleavage. These results suggest that p38 MAP kinase activation may play an important role in the UVB-induced apoptosis of human melanocytes. To explain this cytoprotective effect, we next examined whether S1P could inhibit UVB-induced JNK and p38 MAP kinase activation. However, S1P was not found to have any influence on UVB-induced JNK or p38 MAP kinase activation. In contrast, S1P clearly stimulated the phosphorylation of ERK, and the specific inhibition of the ERK pathway using PD98059 abolished the cytoprotective effect of S1P. Based on these results, we conclude that the activation of p38 MAP kinase plays an important role in UVB-induced apoptosis, and that S1P may show its cytoprotective effect through ERK activation in human melanocytes.

NSA9, a human prothrombin kringle-2-derived peptide, acts as an inhibitor of kringle-2-induced activation in EOC2 microglia

  • In neurodegenerative diseases, such as Alzheimer' and Parkinson', microglial cell activation is thought to contribute to CNS injury by producing neurotoxic compounds. Prothrombin and kringle-2 increase levels of NO and the mRNA expression of iNOS, IL-1$\beta$, and TNF-$\alpha$ in microglial cells. In contrast, the human prothrombin kringle-2 derived peptide NSA9 inhibits NO release and the production of pro-inflammatory cytokines such as IL-1$\beta$, TNF-$\alpha$, and IL-6 in LPS-activated EOC2 microglia. In this study, we investigated the anti-inflammatory effects of NSA9 in human prothrombin- and kringle-2-stimulated EOC2 microglia. Treatment with 20-100 ${\mu}M$ of NSA9 attenuated both prothrombin- and kringle-2-induced microglial activation. NO production induced by MAPKs and NF-$\kappa$B was similarly reduced by inhibitors of ERK (PD98059), p38 (SB203580), NF-$\kappa$B (N-acetylcysteine), and NSA9. These results suggest that NSA9 acts independently as an inhibitor of microglial activation and that its effects in EOC2 microglia are not influenced by the presence of kringle-2.
