Immunoregulative Action of Bangpoongtongsungsan (방풍통성산(方風通聖散) 전탕액(煎湯液)의 면역(免疫) 조절작용(調節作用))

  • The purpose of this research was to investigate effects of Bangpoongtongsungsan water extract(BTSE) on the immune reaction, anti-allergy action and anti-inflammatory action in BALB/c mice. The administration of BTSE (500mg/kg) enhanced the cell viability of thymocytes and the population of helper T cells in splenic T-lymphocytes. BTSE suppressed the production of nitric oxide, but enhanced the phagocytic activity in peritoneal macrophages. BTSE enhanced hemagglutination titer in mice. BTSE inhibited passive cutaneous anaphylaxis induced by egg albumin in rat, the lethal anaphylaxis induced by platelet activating factor and compound 48/80 in mice, and then inhibited the degranulation of peritoneal mast cells induced by compound 48/80. BTSE did not inhibit Arthus reaction, but inhibited the delayed type hypersensitivity induced by SRBC and contact dermatitis induced by DNFB. BTSE inhibited the acute hind paw edema induced by histamine after 30 minutes, the permeability of evans blue into peritoneal cavity induced by acetic acid and the writhing syndrome induced by acetic acid. These results suggest that BTSE has an immunopotentiative action, anti-allergy action and anti-inflammatory action via the inhibition of histamine release.

  • Three newly synthesized alkanol esters of d-2-(6-methoxy-2-naphthyl) propionic acid, NAPROXEN were examined for physicochemical properties and biopharmaceutical characteristics. These esters were very stable in solid state, but more than 90% of these esters were hydrolysed to the parent, naproxen in rabbit's liver hornogenates. They showed higher dissolution rate in the artificial gastric and intestinal juice, and significantly greater partition coefficient in n-octanol, when compared with naproxen. The absorption rate constants of these esters were increased, while the elimination rate constants were decreased, comparing with naproxen. The ulcerogenic doses on gastric and intestinal mucosa were increased remarkably, and the antiinflammatory dose against carrageenininduced edema on rat hind paw was decreased markedly in these esters, and thus the safety indexes of these esters were higher than that of naproxen.

  • Verbascoside has inhibition activity on carrageenan induced edema in rat paw, enhancement of bile juice secretion, and hepatoprotective activity. The hepatoprotective functions were improved on serumtransaminase activities (s-GPT and s-GOT), liver weight, and even sleeping time in mice treated with verbascoside. In addtion, the mice toxicated with carbon tetrachloride are signiflcantly recovered by the administration of verbascoside. This is the first report shown their biological activity of verbascoside.

  • Three components, fraxetin-8-glucoside, esculetin-6-glucoside and mannitol, were isolated from the stem bark of Fraxinus chiisanensis var. stenophylla, F. japonica var. intermedia and F. densata. The MeOH extract of the cortex of F. densata had the antiinflammatory activity on the carrageenin-induced paw edema in rat. The MeOH extracts of the cortex of all three Fraxinus spp. have the potent analgesic activity on the HOAc-induced writhing syndrome in mouse and the hepatoprotective activity on the $CCl_4-induced$ fatty liver in rat: protection of ballooning formation and inhibition of sGPT and sGOT increased by $CCl_4$.

  • Many kinds of phenylpropanoids(PP), C6-C3 compounds, are widely distributed in many plants. In this experiments, effect of PP on sheep red bood cells (sRBC)-induced Arthus reaction (AR) were studied in ICR male mice. SRBC were challenged by i.p. injection two weeks after sensitization of Lp. injection of sRBC. Five days after the challenge of antigen, paw edema induced 3 hours after the last challenge by AR.   • As an attempt to search for bioactive natural products exerting antiinflammatory activity, we have evaluated the anti-inflammatory and antinociceptive activities of the methanol extract from the Fomes fomentarius (MEFF). MEFF (50, 100 mg/kg/day, p.o.) significantly reduced an acute paw edema induced by carrageenan in rats. When analgesic activity was measured by acetic acid-induced writhing test and hot plate test, MEFF showed a dose-dependent inhibition in animal models. In addition, MEFF potently inhibited the LPS-induced production of NO, $PGE_2$ and TNF-$\alpha$production of macrophages. (omitted)

  • Dictyophora indusiata is an edible mushroom belongs to Family Phallaceae of Phallales, Basidiomycota. The purpose of this study was to investigate the antioxidant and anti-inflammatory activities of methanol and hot water extracts prepared from fruiting bodies of Dictyophora indusiata. Besides measuring of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity for those extracts. To measure the anti-inflammatory activities for the extracts, nitric oxide(NO) production from lipopolysaccharide(LPS) treated RAW 264.7 macrophage cells and carrageenan-induced acute hind paw edema of rats were investigated. The results showed that the extracts have excellent DPPH scavenging and chelating activity on the ferrous ions compared with positive control. The nitric oxide(NO) production in LPS-stimulated RAW 264.7 macrophage cells were decreased as we increased the concentration of the mushroom extracts. Significant reduction of paw edema of rats were observed at 2~6 h after treatment of methanol and hot-water extracts with 50 mg/kg concentration to the rats which are induced acute hind paw edema by carrageenan administration. Therefore, the experimental results suggested that methanol and hot-water extracts of Dictyophora indusiata fruiting bodies might be used for natural sources of antioxidant and anti-inflammatory agents.
