Tracking of Stem Cells from Human Exfoliated Deciduous Teeth Labeled with Molday ION Rhodamine-B during Periodontal Bone Regeneration in Rats

  • Background and Objectives: Chronic periodontitis can lead to alveolar bone resorption and eventually tooth loss. Stem cells from exfoliated deciduous teeth (SHED) are appropriate bone regeneration seed cells. To track the survival, migration, and differentiation of the transplanted SHED, we used super paramagnetic iron oxide particles (SPIO) Molday ION Rhodamine-B (MIRB) to label and monitor the transplanted cells while repairing periodontal bone defects. Methods and Results: We determined an appropriate dose of MIRB for labeling SHED by examining the growth and osteogenic differentiation of labeled SHED. Finally, SHED was labeled with 25 ㎍ Fe/ml MIRB before being transplanted into rats. Magnetic resonance imaging was used to track SHED survival and migration in vivo due to a low-intensity signal artifact caused by MIRB. HE and immunohistochemical analyses revealed that both MIRB-labeled and unlabeled SHED could promote periodontal bone regeneration. The colocalization of hNUC and MIRB demonstrated that SHED transplanted into rats could survive in vivo. Furthermore, some MIRB-positive cells expressed the osteoblast and osteocyte markers OCN and DMP1, respectively. Enzyme-linked immunosorbent assay revealed that SHED could secrete protein factors, such as IGF-1, OCN, ALP, IL-4, VEGF, and bFGF, which promote bone regeneration. Immunofluorescence staining revealed that the transplanted SHED was surrounded by a large number of host-derived Runx2- and Col II-positive cells that played important roles in the bone healing process. Conclusions: SHED could promote periodontal bone regeneration in rats, and the survival of SHED could be tracked in vivo by labeling them with MIRB. SHED are likely to promote bone healing through both direct differentiation and paracrine mechanisms.

Gaussian Filtering Effects on Brain Tissue-masked Susceptibility Weighted Images to Optimize Voxel-based Analysis (화소 분석의 최적화를 위해 자화감수성 영상에 나타난 뇌조직의 가우시안 필터 효과 연구)

  • Purpose : The objective of this study was to investigate effects of different smoothing kernel sizes on brain tissue-masked susceptibility-weighted images (SWI) obtained from normal elderly subjects using voxel-based analyses. Materials and Methods: Twenty healthy human volunteers (mean $age{\pm}SD$ = $67.8{\pm}6.09$ years, 14 females and 6 males) were studied after informed consent. A fully first-order flow-compensated three-dimensional (3D) gradient-echo sequence ran to obtain axial magnitude and phase images to generate SWI data. In addition, sagittal 3D T1-weighted images were acquired with the magnetization-prepared rapid acquisition of gradient-echo sequence for brain tissue segmentation and imaging registration. Both paramagnetically (PSWI) and diamagnetically (NSWI) phase-masked SWI data were obtained with masking out non-brain tissues. Finally, both tissue-masked PSWI and NSWI data were smoothed using different smoothing kernel sizes that were isotropic 0, 2, 4, and 8 mm Gaussian kernels. The voxel-based comparisons were performed using a paired t-test between PSWI and NSWI for each smoothing kernel size. Results: The significance of comparisons increased with increasing smoothing kernel sizes. Signals from NSWI were greater than those from PSWI. The smoothing kernel size of four was optimal to use voxel-based comparisons. The bilaterally different areas were found on multiple brain regions. Conclusion: The paramagnetic (positive) phase mask led to reduce signals from high susceptibility areas. To minimize partial volume effects and contributions of large vessels, the voxel-based analysis on SWI with masked non-brain components should be utilized.

Characterization of Iron Oxides in Soils of Cheju Island by Mössbauer Spectroscopy and Chemical Techniques (Mössbauer 분광법(分光法)과 선택적(選擇的) 추출방법(抽出方法)에 의한 제주도(濟州道) 토양(土壤)에서의 산화철(酸化鐵)의 특성(特性) 구명(究明))

  • Iron oxide compounds in 8 selected Cheju Island soil samples have been analized by X-ray fluorescence spectrometer(XRF), X-ray diffractometry(XRD), selected chemical techniques, and $M{\ddot{o}}ssbauer$ spectroscopy. The result of this analysis by XRF shows that the rate of quantity of $Fe_2O_3$ in 8 soil samples was from 8.03wt.%(Daejeong paddy soil) to 18.21wt.%(Songag soils). Songag, Heugag and Gueom soils were detected to have lower peaks of intensity of hematite by XRD. In addition, these soils were not detected to have hematite and goethite peaks. Ferrihydrite, which is a short-range-order mineral commonly present in volcanic ash soil, was not detected by XRD due to low concentration and/or poor cristallinity. Ferrihydrite contents estimated from Feo values were 8.8~35.2g/kg for volcanic ash soils and 0.85g/kg for the Daejeong soil. Most of the soil samples represented by the paramagnetic $Fe^{3+}$ doublet obtained from $M{\ddot{o}}ssbauer$ spectra at room temperature and 18K were considered to arise from the presence of ferrihydrite, superparamagnetic goethite, and silicate minerals. Also the paramagnetic $Fe^{2+}$ doublets are attributable to primary minerals such as olivine, illite, chlorite, augite, biotite, and hornblende. Goethite and hematite were identified as the dominant crystalline iron oxides in these soils from $M{\ddot{o}}ssbauer$ spectra obtained at room temperature and 18K. All the soil samples exhibited strong superparamagnetic relaxation. Collapse of the $M{\ddot{o}}ssbauer$ magnetic hyperfine splitting at room temperature was due to the small size(${\sim}180{\AA}$) of the oxide particles and/or Al-subsituted goethite.

Crystall ographic and Magnetic Properties of Ultrafine $CoFe_{1.9}Bi_{0.1}O_4$ Grown by Using a Sol-Gel Method (Sol-gel법에 의한 초미세 분말 $CoFe_{1.9}Bi_{0.1}O_4$의 결정학적 및 자기적 성질 연구)

  • Ultrafine $CoFe_{1.9}Bi_{0.1}O_4$ particles were fabricated by using a sol-gel method and their magnetic and structural properties were investigated with an x-ray diffractometer (XRD), a vibrating sample magnetometer (VSM), and a M$\"{o}$ssbauer spectrometer. The result of x-ray diffraction and M$\"{o}$ssbauer spectroscopy showed that the powders fired at and above 523 K had only cubic spinel structures. M$\"{o}$ssbauer spectra measurements showed that the powders annealed at 523,723 and 823 K possessed ferrimagnetic nature and paramagnetic nature due to superparamagnetism, simultaneously at room temperature and the powders annealed at and above 923 K behaved ferrimagnetically. In the case of the powder annealed at 923 K, the lattice constant was $a_0=8.398$\pm$0.005{\AA}$ and the hyperfine fields were $H_{hf}(A)=479kOe,\; H_{hf}(B)=502kOe$. The isomer shifts indicate that the iron ions are ferric at tetrahedral[A] and octahedral sites [B], respectively. The magnetization as a function of annealing temperature increased as increasing annealing temperature. The largest coercivity values were $H_C=1368\;Oe$ AT 923 K annealing temperature. In the case of the powder annealed at 1123 K, the magnetization value was $M_S=75\;emu/g$ and this value was similler to that of $CoFe_2O_4$.Fe_2O_4$.

Crystallographic and Magnetic Properties of Co, Zn, Ni-Zn Substituted Nano-size Manganese Ferrites Synthesized by Sol-gel Method

  • Cobalt-, zinc-, and nickel-zinc-substituted nano-size manganese ferrite powders, $MnFe_2O_4$, $Mn_{0.8}Co_{0.2}Fe_2O_4$, $Mn_{0.8}Zn_{0.2}Fe_2O_4$ and $Mn_{0.8}Ni_{0.1}Zn_{0.1}Fe_2O_4$, were fabricated using a sol-gel method, and their crystallographic and magnetic properties were subsequently studied. The $MnFe_2O_4$ ferrite powder annealed at temperatures above 523 K exhibited a spinel structure, and the particle size increased as the annealing temperature increased. All ferrites annealed at 773 K showed a single spinel structure, and the lattice constants and particle size decreased with the substitution of Co, Zn, and Ni-Zn. The $M{\ddot{o}}ssbauer$ spectrum of the $MnFe_2O_4$ ferrite powder annealed at 523 K only showed a doublet due to its superparamagnetic phase, and the $M{\ddot{o}}ssbauer$ spectra of the $MnFe_2O_4$, $Mn_{0.8}Co_{0.2}Fe_2O_4$, and $Mn_{0.8}Zn_{0.2}Fe_2O_4$ ferrite powders annealed at 773 K could be fitted as the superposition of two Zeeman sextets due to the tetrahedral and octahedral sites of the $Fe^{3+}$ ions. However, the $M{\ddot{o}}ssbauer$ spectrum of the $Mn_{0.8}Ni_{0.1}Zn_{0.1}Fe_2O_4$ ferrite powder annealed at 773 K consisted of two Zeeman sextets and one quadrupole doublet due to its ferrimagnetic and paramagnetic behavior. The area ratio of the $M{\ddot{o}}ssbauer$ spectra could be used to determine the cation distribution equation, and we also explained the variation in the $M{\ddot{o}}ssbauer$ parameters by using this cation distribution equation, the superexchange interaction and the particle size. Relative to pure $MnFe_2O_4$, the saturation magnetizations and coercivities were larger in $Mn_{0.8}Co_{0.2}Fe_2O_4$ and smaller in $Mn_{0.8}Zn_{0.2}Fe_2O_4$, and $Mn_{0.8}Ni_{0.1}Zn_{0.1}Fe_2O_4$. These variations could be explained using the site distribution equations, particle sizes and magnetic moments of the substituted ions.

  • Lithium niobate single crystals with various [Li]/[Nb] ratios were grown by the Czochralski method from melts having compositions varing between 48.6 ~ 58.0 mol % $Li_2O$. A vapor transport equilibration technique has been used to improve the homogeneity and adjust the [Li]/[Nb] ratio in small $LiNbO_3$ single crystals grown by the Czochralski method. When equilibrated with a Li-rich powder (65 mol%$Li_2O$), containing a mixture of $LiNbO_3$ and $Li_3NbO_4$, crystals of nearly stoichiometric composition can be obtained. This was established by studying the composition dependence of the following properties; lineshape, intensity and linewidth for the electron paramagnetic resonance (EPR) of $Fe^{3+}$ energy of the fundamental absorption edge and $OH^-$ absorption spectra.

The Effects of Rotational Correlation Time of Paramagnetic Contrast Agents on Relaxation Enhancement: Partial Binding to Macromolecules (거대분자에 부분적으로 결합한 상자성 자기공명 조영제의 회전속도가 이완증강에 미치는 영향)

  • Purpose : To evaluate the effect of rotational correlation time (${\tau}_R$) and the possible related changes of other parameters, ${\tau}_M,{\;}{\tau}_S,{\;}and{\;}(\tau}_V$ of gadolinium (Gd) chelate on T1 relaxation enhancement in two pool model. Materials and Methods : The NMRD (Nuclear Magnetic Relaxation Dispersion) profiles were simulated from 0.02 MHz to 800 MHz proton Larmor frequency for different values of rotational correlation times based on Solomon-Bloembergen equation for inner-sphere relaxation enhancement. To include both unbound pool (pool A) and bound pool (pool B), the relaxivity was divided by contribution from unbound pool and bound pool. The rotational correlation time for pool A was fixed at the value of 0.1 ns, which is a typical value for low molecular weight complexes such as Gd-DTPA in solution and ${\tau}_R$ for pool B was changed from 0.1 ns to 20 ns to allow the slower rotation by binding to macromolecule. The fractional factor of was also adjusted from 0 to 1.0 to simulate different binding ratios to macromolecule. Since the binding of Gd-chelate to macromolecule cab alter the electronic environment of Gd ion and also the degree of bulk water access to hydration site of Gd-chelate, the effects of these parameters were also included. Results : The result shows that low field profiles, ranged from 0.02 to 40 MHz, and dominated by contribution from bound pool, which is bound to macromolecule regardless of binding ratios. In addition, as more Gd-chelate bound to macromolecule, sharp increase of relaxivity at higher field occurs. The NMRD profiles for different values of ${\tau}_S$ show the enormous increase of low field profile whereas relaxivity at high field is not affected by ${\tau}_S$. On the other hand, the change in ${\tau}$V does not affect low field profile but strongly in fluences on both inflection fie이 and the maximum relaxivity value. The results shows a fluences on both inflection field and the maximum relaxivity value. The results shows a parabolic dependence of relaxivity on ${\tau}_M$. Conclusion : Binding of Gd-chelate to a macromolecule causes slower rotational tumbling of Gd-chelate and would result in relaxation enhancement, especially in clinical imaging field. However, binding to macromolecule can change water enchange rate (${\tau}_M$) and electronic relaxation ($T_le$) vis structural deformation of electron environment and the access of bulk water to hydration site of metal-chelate. The clinical utilities of Gd-chelate bound to macromolecule are the less dose requirement, the tissue specificity, and the better perfusion and intravascular agents.

Petrochemical and Physical Characteristics of the Cretaceous Pink Granites in the Jinan Area (진안일대에 분포하는 백악기 홍색 화강암류의 암석화학 및 물성특성)

  • The Cretaceous pink granites of the finan area, southwestern Ogcheon belt, are adjacently developed in the eastern part (Keg) and western part (Kwg) as stocks, respectively. Keg of rounded shape occur as mainly medium-coarse grained rocks, whereas Kwg of ellipsoidal shape occurs as medium-coarse grained ones with partly porphyritic and fine-grained textures. Miarolitic cavities of them are often seen and can be observed more frequently in Kwg than Keg. Rose and counter fracture diagrams of the two granites show that Keg and Kwg have more potentiality of non-dimension and dimension to non-dimension stones, respectively. Physical properties such as porosity and absorption ratio have 0.25% and 0.65%, and 0.43% and 1.11%, respectively, which could suggest that emissions of gas phase at later magma stages are abundant in Kwg than those of Keg. From the major and trace elements petrochemisoy, they belong to acidic, peraluminous and calc-alkaline rocks, showing that Kwg are later product than Keg of the same granitic parent magma. REE concentrations normalized to chondrite value have trends of gradual and parallel enriched LREE and depleted HREE. Eu negative anomalies of Kwg are far more severe than those of Keg, which suggest that plagioclase fractionation in Kwg was much stronger than that of Keg. In the magnetic susceptibility vs. petrochemical and modal parameters, they all belong to magnetite-series and I-types, and can be classified as weakly-moderately ferromagnetic rocks. And the above relations could suggest that their susceptibility values are more mainly depended on ferromagnetic opaques than ferromagnetic and paramagnetic assemblages (Bt + Ch + Ser + Op).

Concentrating Effect of Heavy Metals from Heavy Metal Contaminated Soil by Magnetic Separation (중금속오염 토양의 자기분리에 의한 오염농축효과)

  • A magnetic separation study was conducted for a soil sampled from a landfill site where steel slag had been dumped for a long time. Heavy metal concentrating effect was evaluated by analyzing heavy metal content of magnetically separated soil and passed through soil. The effect was compared between soil after soil-washing process and original landfill soil and the effect was also tested between wet condition-magnetic separation and dry condition-magnetic separation. Separated ratio was relatively higher in non-soil washed sample. The water content has no significant effect on the separation rate. The concentrating effect of Fe, Pb, Cu, and Cd were 3.2, 2.1, 12.1, 2.5, 1.5 and 17.4, 7.0, 15.7, 9.6, 7.0 respectively for non-soil washed sample and soil washed sample. We can expect a bigger volume reduction effect from soil-washed samples. The volume reduction effect was obtained from the separation in dry condition. However, when the separation ratio is too high the volume reduction effect decreases. The magnetic separation leads to a volume reduction and concentration of heavy metals into a portion of soil in case of paramagnetic particles contained soil.

Phase Analysis and Magnetic Properties of $Fe_5Si_xB_{5-x}$ (x = 0, 1, 2, 3) Powders Prepared by Mechanical Alloying (기계적합금법으로 제조된 $Fe_5Si_xB_{5-x}$ (x = 0, 1, 2, 3) 분말의 상분석 및 자기적 특성)

  • $Fe_5Si_Xb_{5-x}$ (x=0, 1, 2, 3) powders were prepared by mechanical alloying, and their phases and magnetic properties were investigated by using XRD, TEM, Mossbauer spectroscopy and VSM. Starting elements are incorporated into $\alpha$-Fe in the early stage of mechanical alloying, and the stable phases are formed as the milling proceeds. After the annealing at 80$0^{\circ}C$ for 2 hours, it is found that the FeB and $Fe_2B$ phases coexist for the $Fe_5B_5$(x=0) composition. By substituting Si for B, the formation of $Fe_2B$ phase is restricted and the $Fe_5SiB_2$, $Fe_2Si_{0.4}B_{0.6}$ and paramagnetic phase begin to appear. The FeB phase has wide range of hyperfine magnetic field because it is not fully crystallized on the annealing at 800 $^{\circ}C$. On the contrary, others have good crystalline phases and show well-defined hyperfine magnetic field. Magnetic saturation is highest for the $Fe_5B_5$ composition where the amount of the $Fe_2B$ phase in large.
