In this study, the α-amylase inhibitory activity, α-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the α-amylase inhibitory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The α-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50~1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5~8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mushrooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as α-amylase, α-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.
The aim of the present study was to assess the effects of onion (Allium cepa) skin extract (OSE) on pancreatic lipase (PL), the key enzyme of the digestion and absorption of dietary fat in the small intestine, and to evaluate its potential for the inhibition of body-weight gain. OSE inhibited PL with an $IC_{50}$ of 53.70 mg/mL, which means as potent as 0.07635% of the activity of orlistat. At 3 and 4 hr after administration of OSE, the plasma triacylglycerol concentration was significantly lower in the OSE-treated rats than control. Body-weight gain and parametrial adipose tissue weights were significantly lower in mice fed the high-fat diet (fat comprises 31% of total calories) with 5%(w/w) OSE than in control. The results suggest that OSE may be an effective nutraceutical for the inhibition of body-weight gain.
Caffeine and epigallocatechin gallate (EGCG) are major constituents of green tea, the leaves of Camellia sinensis (Theaceae). Although EGCG is well known for diverse beneficial effect, caffeine is sometimes harmful with adverse effects. Therefore, the extraction efficiency was investigated using different extraction method such as extraction solvent, extraction time, extraction method, and repeated extraction. The content of EGCG and caffeine in green tea extract was quantitated by HPLC analysis. The extraction condition exerted difference on the extraction yield. The content of EGCG was also affected by different extraction condition. Especially, the extraction solvent greatly affected the content of EGCG in the extract. However, the content of caffeine was less affected compared to that of EGCG. The inhibitory effect of green tea extract on pancreatic lipase was almost similar regardless of extraction condition. Taken together, optimization of extraction condition will provide best efficacy for further development of green tea as anti-obesity therapeutics.

While searching for pancreatic lipase inhibitors, the active compound was found in a methanol extract of Apis mellifera venome. The active compound was isolated by Diaion HP-20 column chromatography, thin layer chromatography and HPLC. The active compound is stable to the extreme pH and heat. There is no loss of activity both in acidic and alkaline solution in the pH range of 2 to 11 by heating for 15 minutes at $90^{\circ}C$. The rf value of the compound was 0.51 at TLC with butanol : methanol: water (4:1:2) solvent system. The molecular weight of the compound was determined to be 293 by EI-MS.
In the course of screening pancreatic lipase inhibitory activity, total methanolic extract and EtOAc-soluble fraction of Duchesnea chrysantha showed significant inhibitory activity. Further fractionation and isolation of the EtOAc-soluble fraction resulted in five compounds, which were identified as trans-tiliroside (1), isovitexin (2), kaempferol-8-O-${\beta}$-glucoside (3), kaempferol-3-O-${\beta}$-glucoside (4) and quercetin-3-O-${\beta}$-glucoside (5). All the five flavonoids derivatives were first reported from this plant but showed weak inhibitory effects on pancreatic lipase activity.
Cheese milk를 기질로 하여 pronase 등 protease와 palanatase ML 등 lipase를 이용하여 EMC를 제조하였다. Cheese milk 가수분해에 관여하는 각 protease의 반응 조건을 살펴본 결과, pronase-3$0^{\circ}C$, pH 7.0, PANCREATIO-4$0^{\circ}C$, pH 8.0, pacific protease-3$0^{\circ}C$, pH 7.0과 Asp. Sp. 기원 protease-5$0^{\circ}C$, pH 8.0에서 최대의 활성을 보였으며, lipase의 반응 조건은 pancreatic lipase-5$0^{\circ}C$, pH 8.0, palatase ML-5$0^{\circ}C$, pH 7.0과 Candida cylindracea 기원 lipase-4$0^{\circ}C$, pH 6.0 이었다. 최적 반응 조건에서 가수분해를 행한 결과 반응 시간이 증가할수록 유리아미노산의 양과 유리지방산의 양이 증가하는 경향을 보였다. Pacific protease와 pronase에 의해 가수분해시 0.67mg/ml과 0.74 mg/ml의 유리 아미노산을 생성하였으며, 유리 아미노산 중 glutamic acid와 leucine이 가장 많은 함량을 보였다. Candida cylidracea 기원 lipase를 이용하여 가수분해시 가장 많은 유리 지방산 생성량을 보였다. 각 EMC의 지방산 조성을 살펴본 결과 butyric acid, palmitic acid, stearic acid와 oleic acid의 함량이 보였다. Pancreatin에 의해 가수분해 후 각 lipase를 사용하여 제조한 EMC의 관능검사 결과, pancreatic lipase를 이용한 EMC가 가장 쓴맛이 강하였고, palatase에 의한 EMC는 가장 바람직한 cheese 향기를 가지고 있다.
비만은 에너지의 섭취와 소비의 불균형으로 지방조직이 비정상적으로 분화하면서 생기는 대사질환으로 알려져있다. 본 연구에서는 본 연구는 적양파 추출물 처리에 따른 Pancreatic lipase 억제, 지방세포분화 억제 활성을 확인하고자 하였다. 적양파추출물 처리에 따른 활성은 지방세포 분화 및 관련 유전자에 대한 평가는 3T3-L1 지방전구세포를 이용하고 Real-Time PCR을 통하여 확인하였다. 실험 결과, Pancreatic lipase 활성 억제 실험에서 적양파 추출물은 농도 의존적으로 lipase 활성을 억제하였다. 지방세포분화 실험을 수행한 결과, insulin, dexamethasone, 3-isobutyl-1-methylxanthine(MDI)등으로 분화 유도된 3T3-L1 세포에서 적양파 추출물은 지방전구세포의 분화를 억제하였으며 동시에 지방구 형성을 억제하는 것으로 나타났다. 또한, 지방전구세포의 분화 과정과 관련된 C/EBP-α, C/EBP-β, PPAR-γ의 발현을 억제하였다. 본 실험에서 적양파 추출물은 지방분해효소를 억제하며, 지방전구세포 분화와 관련된 유전자 발현을 억제함으로써 지방세포 분화 및 지방구형성을 억제할 수 있는 항비만 소재로의 개발 가능성이 높다고 판단된다.

The object of the present study was to compare abnormal serum canine pancreas-specific lipase results and pancreatic ultrasonographic findings in dogs with pancreatitis. Pancreatitis is a common disease in dogs that is difficult to diagnose. The noninvasive diagnostic procedures, including a serum canine pancreatic-specific lipase (cPL) test and ultrasonographic changes in the pancreas, can be considered for the diagnosis of canine pancreatitis in clinical practice. A retrospective study was performed to assess pancreatitis in dogs. Forty client-owned dogs were suspected to have pancreatitis, which was confirmed by abnormal serum SNAP cPL results. Abdominal ultrasound examinations were also performed. The present study investigated the distribution of clinical signs associated with pancreatitis, and the dogs were divided into two groups: group 1 (clinical signs compatible with pancreatitis; n = 30) and group 2 (no clinical signs; n = 10). Based on this study, an abnormal result on the SNAP cPL assay can still present as a normal pancreas through an ultrasonographic examination, and a normal health status can identify the status of pancreatic ultrasonographic abnormal lesions. Therefore, for dogs with suspected pancreatitis, it is important to repeat an ultrasonographic evaluation. There is no significant difference between clinical symptoms and ultrasonographic changes in the pancreas.

The effect of aqueous methanol on the catalytic properties of porcine pancreatic lipase has been investigated. The k$_{CAT}$, values for the hydrolysis of N$^{alpha}$-benzyloxycarbonyl-Llysine p-nitrophenyl ester at 0$^{circ}$C increased in a linear manner with increasing methanol concentration. However, the K$_{M}$ values were not influenced at methanol concentrations lower than $30\%$ and then began to increase at higher concentrations in an exponential fashion. Based on product analysis, the increase in k$_{CAT}$, with increasing methanol concentration can be accounted for by nucleophilic competition of methanol for the acyl enzyme intermediate, indicating that the rate-limiting step of the porcine pancreatic lipase-catalyzed reaction is deacylation under current experimental conditions. The exponential increase in K$_{M}$ at methanol concentrations higher than $30\%$ is attributed to the hydrophobic partitioning effect on substrate binding. There was no loss of lipase activity over a 4 h period in $60\%$ methanol concentration at pH$^{circ}$ 5.5 and 0$^{circ}$C. By monitoring the intrinsic fluorescence and absorbance, no evidence for structural changes by methanol was observed.
In acute pancreatitis group, all the dogs are showed increase of amylase and lipase after the 1st day of surgery, and amylase and lipase activity were significantly more increased than those of control group. The methemalbumin was increased significantly after the 2nd day of surgery in the acute pancreatitis group. In pancreatitis group, ultrasonographic findings included thickened duodenal wall and poorly circumscribed hyperechoic lesion of pancreatic mass after the 2nd day of surgery. And the lesion was exacerbated until the 4th day and reduced after the 6th day of surgery. To identify the lesion of pancreas, it is considered that transverse view is more useful Than sagittal view. Gross findings showed increase of pancreatic parenchymal consistency, surface nodule, and extensive pancreatic necrosis. Necrosis of peripancreatic fat tissue was also a prominent feature. The microscopic appearance of the pancreas was characterized by pancreatic acinar cell necrosis, hemorrhage, infiltration of the inflammatory cell and fat necrosis and saponification were also observed.
